In this work, the effect of drought on K+uptake in root and its translocation from root to shoot was investigated using six barley genotypes contrasting in drought tolerance. Results showed that drought conditions caused significant changes in K+uptake and translocation in a time- and genotype-specific manner, which consequently resulted in a significant difference in tissue K+contents and drought tolerance levels between the contrasting barley genotypes. The role of K+transporters and channels and plasma membrane (PM) H+-ATPase in barley’s adaptive response to drought stress was further investigated at the transcript level. The expression of genes conferring K+uptake (HvHAK1,HvHAK5,HvKUP1,HvKUP2andHvAKT1) and xylem loading (HvSKOR) in roots were all affected by drought stress in a time- and genotype-specific manner, indicating that the regulation of these K+transporters and channels is critical for root K+uptake and root to shoot K+translocation in barley under drought stress. Furthermore, the barley genotypes showed a strong correlation between H+efflux and K+influx under drought stress, which was further confirmed by the significant up-regulation ofHvHA1andHvHA2. These results suggested an important role of plasma membrane H+-ATPase activity and/or expression in regulating the activity of K+transporters and channels under drought stress. Taken together, it may be concluded that the genotypic difference in drought stress tolerance in barley is conferred by the difference in the ability to regulate K+transporters and channels in root epidermis and stele. Keywords:drought tolerance;ion flux;transmembrane K+transport;plasma membrane H+-ATPase;barley

The regulation solutions and mechanisms of reducing pesticide phytotoxicity to nontarget plants are not well-defined and detailed. Here, we have proposed a new detoxification strategy to control the toxic effects of herbicide imazethapyr (IM) induced in wheat seedlings from the perspective of the plasma membrane (PM) H+-ATPase. We found that the changes in PM H+-ATPase activity have a regulatory effect on the phytotoxic effects induced by IM in plants. Treatment with PM H+-ATPase activators restored the reduced auxin content and photosynthetic efficiency caused by IM, thereby promoting plant growth. Application of a PM H+-ATPase inhibitor further reduced phosphorus content and significantly increased 2,4-dihydroxy-7-methoxy-2H,1,4-benzoxazin-3(4H)one (DIMBOA) and jasmonic acid levels. These effects indicate that auxin and DIMBOA may regulate plant growth trends and detoxification effects mediated by PM H+-ATPase. This work opens a new strategy for regulating herbicide toxicity to nontarget plants from the PM H+-ATPase.

Fulvic acid (FA) has been widely used as a plant growth regulator to alleviate salt stress. However, the mechanism by which it enhances salt tolerance in rice remains unclear. This study systematically investigated the chemical structure of FA and its role in enhancing rice salt tolerance through the regulation of plasma membrane (PM) H + -ATPase. The results demonstrated that the increased molecular weight (MW) of FA companied by progressive aromaticity enhancement and functional group depletion. Principal component analysis revealed that low-MW FA fractions (< 3 kDa), enriched in carboxylic-C, phenolic-C, and lignin-like substances, exhibited a strong positive correlation with root growth, membrane potential (MP) hyperpolarization, and Na + efflux under 100 mmol/L NaCl stress. Mechanistically, FA directly activated PM H + -ATPase (activity increased by 30.3%‒86.2%), generating a proton gradient that energized SOS1 to mediate Na + efflux, leading to salt tolerance in rice. Genetic validation using osa1 knockout mutants confirmed the indispensable role of PM H + -ATPase in FA-induced responses, as both MP and Na + efflux remained unaltered upon FA1‒FA5 treatment. This research found that FA fractions with high functional group density and low aromaticity can serve as optimal plant biostimulants for salt stress mitigation, providing a new strategy for sustainable lignite valorization in precision agriculture.

Toxicants are generally harmful to biotechnology in wastewater treatment. However, trace toxicant can induce microbial hormesis, but to date, it is still unknown how this phenomenon affects nutrient removal during municipal wastewater treatment process. Therefore, this study focused on the effects of hormesis induced by cetyltrimethyl ammonium bromide (CTAB), a representative quaternary ammonium cationic surfactant, on nutrient removal by Chlorella vulgaris F1068. Results showed that when the concentration of CTAB was less than 10 ng/L, the cellular components chlorophyll a , proteins, polysaccharides, and total lipids increased by 10.11, 58.17, 38.78, and 11.87 %, respectively, and some enzymes in nutrient metabolism of algal cells, such as glutamine synthetase (GS), acid phosphatase (ACP), H + -ATPase, and esterase, were also enhanced. As a result, the removal efficiencies of ammonia nitrogen (NH 4
+ ) and total phosphorus (TP) increased by 14.66 and 8.51 %, respectively, compared to the control during a 7-day test period. The underlying mechanism was mainly due to an enhanced photosynthetic activity of C. vulgaris F1068 indicated by the increase in chlorophyll fluorescence parameters (the value of Fv / Fm , ΦII, Fv / Fo , and rETR increased by 12.99, 7.56, 25.59, and 8.11 %, respectively) and adenylate energy charge (AEC) (from 0.68 to 0.72). These results suggest that hormesis induced by trace toxicants could enhance the nutrient removal, which would be further considered in the design of municipal wastewater treatment processes.
