With the development of social society, sleep deprivation has become a serious and common issue. Previous studies documented that there is a correlation between sleep deprivation and oxidative stress . However, the information of sleep deprivation related ROS has rarely been obtained. Also, it has been demonstrated that sleep deprivation can induce endoplasmic reticulum (ER) stress. As such, for a better understanding of sleep deprivation as well as its related diseases, it is important to develop probes with ER-targeting ability for detecting ROS generated in this process. Herein, a novel two-photon fluorescent molecular probe , JX-1 , was designed for sensing HClO in live cells and zebrafish. The investigation data showed that in addition to real-time response (about 150 s), the probe also exhibited high sensitivity and selectivity . Moreover, the probe JX-1 demonstrated two-photon fluorescence, low cytotoxicity and ER targeting ability. These prominent properties enabled the utilization of the probe for monitoring exogenous and endogenous HClO in both live cells and zebrafish. Using this useful tool, it was found that sleep deprivation can induce the generation of HClO in zebrafish.

Hypochlorous acid (HClO) as well as its ionic form (ClO − ), representative of reactive oxygen species (ROS), are essential players in all sorts of biological processes. The abnormal level of each can lead to the onset of various diseases. Besides, Sodium hypochlorite , a commonly-used bleaching agent in our daily lives, could also result in breathing and skin problems when overexposed. Therefore, developing a molecular chemosensor for sensing HClO is of biological and environmental importance. Though many such chemosensors have been reported, new HClO chemosensors with different sensing performances may still come in handy in certain situations. In this work, we have developed a new coumarin-based chemosensor, CM-hbt , for realizing both ratiometric and colorimetric imaging detection of HClO in live cells. Notably, we further explored its application in sensing HClO in plant mung beans as well as fabricated an easy-to-use paper strip apparatus for facilitating its quick detection, which is seldomly seen in other HClO chemosensors. All the analysis results confirmed the high sensitivity and selectivity of this novel chemosensor. DFT calculations were used to decipher the underlying sensing mechanism of CM-hbt . Overall, this work presents a novel chemosensor, CM-hbt , as a colorimetric and ratiometric chemosensor for realizing imaging detection of HClO in a variety of different model systems, which highlights its broad spectrum of application potentials.

Hypochlorous acid (HClO) along with its ionic form, hypochlorite anion (ClO − ) are critical reactive oxygen species (ROS), which play vital roles in biological systems. Dysregulated production of HClO/ClO − can result in tissue damage and cause a variety of diseases. Besides, Sodium hypochlorite has been widely used as a bleaching agent for water disinfection , surface cleanin g in daily life. Excessive exposure to sodium hypochlorite will lead to symptoms of severe breathing and skin problems. Therefore, developing a state-of-the-art (simple, highly sensitive, highly selective and super fast-response) sensor for tracking HClO is of biological, toxicological, and environmental importance. Though many HClO probes have been reported so far, this big aim still presents a challenge. Researchers around the world are continuing to develop new HClO probes that could improve their sensitivity, selectivity , the limit of detection, response time, easiness to use, etc. Herein, with coumarin as the fluorophore molecule, we rationally developed a novel chemosensor ( CMTH ) for detecting HClO with both ratiometric and colorimetric responses resulted from the oxidation reaction of C N bond. Further analysis results indicated that CMTH can realize highly sensitive with low limit of detection (256 nM, among the best of its kind) and highly selective (over a bunch of interfering analytes) imaging detection of HClO in multiple organisms with low cytotoxicity, and good cell and tissue permeability as well. In particular, compared to other fluorescent HClO probes reported so far, CMTH excels in the response time to HClO (< 40 s), being the top-notch of its kind. Besides, owing to its excellent water solubility, CMTH can also be applied to track HClO in the environmental system. Taken together, we have presented here a novel chemosensor , CMTH , as a colorimetric and ratiometric chemosensor for highly sensitive and ultrafast imaging detection of HClO in aqueous solutions, eukaryotic cells, prokaryotic bacteria and vertebrate zebrafish.

Reactive oxygen species (ROS) are an important component for maintaining normal physiological activities in organisms, and abnormal changes in their level are often accompanied by many diseases. As the two most representative components of ROS, HClO and H2O2 play vital roles in many physiological and pathological processes and are interdependent and mutually transformable. Although there is a lot of work that has specifically detected HClO or H2O2, there are few reports on the simultaneous differential detection of HClO and H2O2. Here, we report a ratio-based fluorescent probe capable of simultaneously distinguishing HClO and H2O2 based on making the best use of the untapped potential of coumarin derivatives. This probe was triumphantly put into use in the discriminative identification of HClO and H2O2 in aqueous media with high sensitivity and selectivity, and the probe was appropriate in a wide pH range. Furthermore, the imaging experiment for HClO and H2O2 in cells and zebrafish was eventually proven to be feasible. Importantly, this probe was qualified for monitoring the variation of HClO and H2O2 levels in organisms with alcoholic liver injury.

As a key reactive oxygen species (ROS), hypochlorous acid (HClO) plays an important role in many physiological and pathological processes. The mitochondria-targeting probes for the highly sensitive detection of HClO are desirable. In present work, we designed and synthesized an original mitochondria-localizing and turn-on fluorescent probe for detecting HClO. 4-Aminonaphthalimide was employed as the fluorescent section, the (2-aminoethyl)-thiourea unit was utilized as a typical sensing unit, and the quaternized pyridinium moiety was used as a mitochondria-targeted localization group. When HClO was absent, the probe showed weak fluorescence. In the existence of HClO, the probe revealed a blue fluorescence. Moreover, the turn-on fluorescent probe was able to function in a broad pH scope. There was an excellent linearity between the fluorescence emission intensity at 488 nm and the concentrations of HClO in the range of 5.0 × 10–7 to 2.5 × 10–6 mol·L–1. Additionally, the probe had almost no cell toxicity and possessed an excellent mitochondria-localizing capability. Furthermore, the probe was able to image HClO in mitochondria of living PC-12 cells. The above remarkable properties illustrated that the probe was able to determine HClO in mitochondria of living cells.

A novel dual-responsive fluorescence probe, 2-[(1E)-2-[(6-dimethylthiocarbamoylnaphthalen)-2-yl]-ethenyl]-3-ethylbenzothiazolium iodide (TNB) has been developed for the discriminative sensing of viscosity and hypochlorous acid (HClO). Under high-viscous conditions, TNB's green fluorescence at 506 nm was switched on due to the inhibition of intramolecular bond rotation. Whereas in PBS aqueous buffer, the release of thiocarbamate moiety from TNB triggered by HClO forms a phenolic compound (NB-OH), which was chlorinated by excessive HClO to the final product (NB-Cl-OH) with near-infrared emission at 676 nm. Consequently, TNB enables the dual-colour visualization of viscosity or HClO fluctuations, showcasing good selectivity and photo-stability, high sensitivity, and quick response to HClO (∼ 4 s). Moreover, the fluorescence of NB-Cl-OH is highly responsive to viscosity changes, achieving dynamic monitoring of HClO and viscosity simultaneously. Possessing mitochondria-targeting capability, TNB realizes the dual-channel imaging of viscosity/HClO alterations in the process of oxidative stress and inflammation in live cells and zebrafish.

The conversion of excess ammonia N into harmless N2 is a primary challenge for wastewater treatment. We present here a method to generate ClO• directionally for quick and efficient decomposition of NH4+ N to N2. ClO• was produced and enhanced by a bifacial anode, a front WO3 photoanode and a rear Sb–SnO2 anode, in which HO• generated on WO3 reacts with HClO generated on Sb–SnO2 to form ClO•. Results show that the ammonia decomposition rate of Sb–SnO2/WO3 is 4.4 times than that of WO3 and 3.3 times than that of Sb–SnO2, with achievement of the removal of NH4+ N on Sb–SnO2/WO3 and WO3 being 99.2 and 58.3% in 90 min, respectively. This enhancement is attributed to the high rate constant of ClO• with NH4+ N, which is 2.8 and 34.8 times than those of Cl• and HO•, respectively. The steady-state concentration of ClO• (2.5 × 10–13 M) is 102 times those of HO• and Cl•, and this is further confirmed by kinetic simulations. In combination with the Pd–Cu/NF cathode to form a denitrification exhaustion system, Sb–SnO2/WO3 shows excellent total nitrogen removal (98.4%), which is more effective than WO3 (47.1%) in 90 min. This study provides new insight on the directed ClO• generation and its application on ammonia wastewater treatment.

Previous studies have developed numerous peroxymonosulfate (PMS) activation technologies and demonstrated remarkable decontamination efficiency for emerging contaminants represented by antibiotics in aqueous environments. However, most research has focused on investigating the decontamination effect and mechanisms using simulated solutions prepared with pure water, lacking in-depth exploration of actual wastewater treatment efficacy and mechanisms. In this study, practical aquaculture wastewater from a shrimp culture farm was selected, innovatively leveraging its high salinity characteristics to reveal the endogenous Cl − /PMS self-activation pathway for hypochlorous acid (HClO) generation. Results show that the inherent chloride and cobalt ions in the wastewater react with PMS to form active species, achieving sulfamethoxazole (SMX) degradation efficiency that surpasses systems with externally added catalysts. Through radical quenching experiments, specific probes, and electron paramagnetic resonance technology, the active species in the endogenous substances/PMS system were identified as HClO and Co(IV) = O, with contribution ratios determined to be about 80 % and 20 % respectively. Combined with density functional theory calculations and intermediate analysis, the Cl − mediated PMS activation pathway and SMX degradation intermediate formation routes were elucidated. Finally, the system also enabled synchronous removal of antibiotic-resistant bacteria (ARBs). This study provides new insights into utilizing endogenous pollutants and mechanistic interpretation during PMS-based treatment of practical wastewater, and develops a new idea of “waste treats waste” strategies.

As an important signal molecule in the living body, hypochlorous acid (HClO) participates in various physiological processes . However, excessive hypochlorous acid will cause some oxidative damage. The organic aggregation-induced emission luminogens (AIEgens) based on tetraphenylethylene (TPE) structures have recently become research hotspots as fluorescent probe materials. In this paper, a kind of water-soluble fluorescent probe TPE-M based on TPE and morpholine-thiourea structures were designed and synthesized for the detection of HClO. Due to the introduction of morpholine structures, the TPE-M showed excellent water solubility. The fluorescence properties of the TPE-M were strongly affected by the polarity of the solvent. Besides, the cation-anion and reactive oxygen species recognition experiments indicated that the fluorescent probe TPE-M displayed a single recognition characteristic for HClO because of the existence of thiourea structures. The limit of detection for HClO was calculated to be 0.25 μM. The particle size distribution measurement under the water phase revealed that the particle size of the TPE-M was mainly centered at 150 nm. The recognition mechanism of HClO by the probe TPE-M was proposed and explained by mass spectra and Gaussian calculation. Furthermore, a cell imaging experiment proved that the TPE-M probe successfully detected HClO in Hela cells.

To date, dynamics monitoring oxidative stress on time resolution in vivo has not been achieved due to the limitation of probes, causing the difficulty to comprehensively understand its biological behaviors and functions. Herein, we propose a novel probe, namely highly bright and color tunable “multicenter-emitting” carbon dots (C-dots), for dynamics monitoring oxidative stress in vivo. Combined with experiments and theoretical calculations, we reveal that each C-dot contains multiple conjugated polyaromatic units on their surfaces, which act as fluorescent emitting centers and response sites of hypochlorous acid (HClO) to achieve an instantaneously color-change readout (107 nm, from yellow-green to blue). Accordingly, the proposed C-dots enable the observation of HClO burst and dynamics evolution at fresh wounds of zebrafish larvae. Such a burst occurs only at 1–5 s after cutting, which is an unprecedented thought regarding oxidative stress dynamics. Furthermore, the present study indicates that HClO molecules are principally generated in situ rather than migration.

Hypochlorous acid (HClO) and sodium hypochlorite (NaClO) are widely present in living organisms and the environment and are closely associated with health or disease in living organisms. Therefore, it is necessary to develop new materials and methods that can detect HClO/ClO − quickly, easily and sensitively. Based on the oxidative property of hypochlorite on the amine bond of diaminomaleonitrile Schiff bases, we have designed and synthesized a ‘Turn-ON’ fluorescent/colourimetric dual-mode probe with high selectivity for HClO/ClO − . In the detection system (DMF: water=1:1, v/v), the addition of HClO/ClO − to the probe was accompanied by a gradual change in the colour of the solution from pink to colourless, with a concomitant 97-fold enhancement of fluorescence intensity at 522 nm (Tun-On), a Stokes shift of 96 nm and a detection limit of 250 nM. A novel HClO/ClO − sensing mechanism in which the imine bond of the probe is oxidized to a carboxyl group by HClO/ClO − was demonstrated by high-resolution mass spectrometry and 1 H NMR spectroscopy. In addition, the probe was applied to the spiked recovery detection of HClO/ClO − in real water samples and the imaging of hypochlorite in cells.

Carbon dots (CDs) as a remarkable fluorescent nanomaterial have the advantages of easy preparation, good photostability and high sensitivity. However, the poor aqueous solution stability of carbon dots largely limited their practical application due to the characteristic of easily forming precipitation for long time storage. Here, a kind of cationic fluorescent carbon dots CDs-P(Ph) 3 was designed by introducing a cationic compound, (4-carboxybutyl) triphenyl phosphonium bromide, to construct an electrostatic shell outside the dots. Such electrostatic shell could highly improve carbon dots stability in an aqueous solution to make CDs-P(Ph) 3 stable for long-term storage with negligible aggregation. Meanwhile, the sensitivity of CDs-P(Ph) 3 for hypochlorous acid (HClO) was also enhanced on the basis of the electron-withdrawing effect of cationic substituents on the surface of carbon dots. The limit of detection of CDs-P(Ph) 3 for HClO was as low as ∼0.32 μM. Additionally, the fluorescence of CDs-P(Ph) 3 could be rapid quenched by HClO with a quenching efficiency of more than 80% within 30 s. The excellent stability of CDs-P(Ph) 3 in an aqueous solution made it suitable for on-site detecting HClO in real samples, such as tap, well and lake water. Such designed fluorescent nanomaterial would provide a practical application pathway for optical sensing detection in environmental samples.
