Mitochondrial fusion reduces T cell susceptibility to HIV infection through citrate modulation

Song Zichen,Wang Jiangrong,Zheng Zhihang,He Zhixiang,Xun Jingna,Gu Ling,Shen Yinzhong,Chen Jun

Inhibiting the metabolic activity of CD4+ T cells can effectively reduce HIV infection. Mitochondria, as critical organelles in eukaryotic metabolism, play a significant role in the progression of many diseases. The change of mitochondrial dynamics is an important process of mitochondrial regulation of cell metabolic activity. However, it remains uncertain whether regulating mitochondrial dynamics is a viable approach to reducing HIV infection. In this study, we demonstrated that promoting mitochondrial fusion in Jurkat cells through treatment with the mitochondrial fusion promoter M1 and the dynamin-related protein 1 (Drp1) inhibitor Mdivi1 conferred resistance to single-round VSVG-HIVNL4-3-GFP viral infection. Targeted metabolomics analysis revealed and subsequently confirmed the potential involvement of citrate in reducing HIV infection, which has been subsequently verified. And we found that plasma citrate level was negatively associated with HIV disease progression. Multi-omics results showed that citric acid leads to a decrease in the level of nucleotide metabolism in Jurkat cells. In conclusion, increased citrate levels resulting from mitochondrial fusion significantly impair the ability of HIV to infect cells, which may due to regulate nucleotide metabolism

Alterations of Natural Killer Cell and T-Lymphocyte Counts in Adults Infected with Human Immunodeficiency Virus through Blood and Plasma Sold in the Past in China and in Whom Infection Has Progressed Slowly over a Long Period

Yongjun Jiang,Hong Shang,Zining Zhang,Yingying Diao,Di Dai,Wenqing Geng,Min Zhang,Xiaoxu Han,Yanan Wang,Jing Liu

Natural killer (NK) cells, natural killer T (NKT) cells, and T lymphocytes were analyzed by using a flow cytometer in 225 human immunodeficiency virus (HIV)-positive individuals infected through the past sale of blood and plasma without receiving antiretroviral therapy in the People’s Republic of China. According to CD4 T-cell counts these HIV-infected adults were stratified into three groups: long-term slow progressors, HIV-infected subjects, and AIDS patients. NK cell counts in long-term slow progressors were higher compared to HIV infection and AIDS patients (P < 0.05) and lower compared to normal controls (P < 0.05), whereas NKT cell counts in slow progressors and the HIV infection group were not different from those of normal controls. NK cell counts in HIV-seropositive subjects were positively correlated with CD4 T-cell counts (P < 0.05), and NKT cell counts were positively correlated with CD4 T-cell and CD8 T-cell counts (P < 0.05). The CD8 T-cell counts were higher in slow progressors compared to those with HIV infection, AIDS patients, and normal controls. These results indicated that HIV infection causes alterations of NK cells and T cells in slow progressors, HIV-infected subjects, and AIDS patient groups, but no difference was found in NKT cell counts and percentages in slow progressors and the HIV-infected group compared to normal controls.

An in-situ gold growth self-catalytic signal amplification system enhanced hollow gold immunochromatography for ultrasensitive detection of p24 antigen in HIV infection

Chuanjiang Ran,Qingjie Zhang,Zhenglin Long,Yuhao Li,Xinyue He,Shengzhou Li,Chen Chen,Cong Cheng,Ying Xu,Wenli Zhang,Wei Guo,Yan Shen

In recent years, the global public health landscape has faced unprecedented challenges, notably with the COVID-19 outbreak and the wide spread of HIV. The rapid spread and serious harm of the virus has highlighted the critical need for fast, accurate, and accessible diagnostic tools. These requirements make lateral flow immunochromatographic assay (LFIA) become the most prominent point-of-care testing (POCT) technology at present. In order to find the balance between sensitivity, ease of operation, detection speed and cost of LFIA, hollow gold nanoparticles (HGNPs) with stronger optical signal, stability and simple synthesis process were used as immune labeling materials to construct LFIA platform (HGNPs-LFIA). HGNPs exhibited elevated catalytic activity in relation to the reaction between chloroauric acid (HAuCl 4 ) and hydroxylamine hydrochloride (NH 2 OH-HCl). This heightened activity consequently resulted in the deposition of new gold nanoparticles on the surface of the original HGNPs. At lower analyte levels, a deepening of the colors of the test line and the control line can be observed with naked eyes. Utilizing this special catalytic activity, an in-situ gold growth method was further used to amplify the detection signal of HGNPs-LFIA by self-catalytic, signal amplified HGNPs-LFIA (sa-HGNPs-LFIA) platform for ultrasensitive and rapid detection of p24 antigen in serum was constructed. The platform struck a good balance between high sensitivity and low false positives, and was capable of detecting p24 antigen as low as 1 pg/mL in serum within 30–40 min, which is 10 times more sensitive than the original HGNPs-LFIA and 20 times more sensitive than the commercially available Alere test strips, and further shortened the window of detection for acute HIV infection.

Identification and validation of CCL5 as a key gene in HIV infection and pulmonary arterial hypertension

Yang Mengyue,Bi Wen,Zhang Zhijie

Background: The relationship between human immunodeficiency virus (HIV) infection and pulmonary arterial hypertension (PAH) has garnered significant scrutiny. Individuals with HIV infection have a higher risk of developing PAH. However, the specific mechanism of HIV-associated PAH remains unclear. Our study aims at investigating the shared biomarkers in HIV infection and PAH and predicting the potential therapeutic target for HIV-associated PAH.Methods: Data for HIV infection and PAH were downloaded from Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) analysis was performed to detect shared genes in HIV infection and PAH. Enrichment analysis was conducted to identify the function of common DEGs. Protein-protein interaction (PPI) analysis was used to detect key genes. These crucial genes were subsequently verified by RT-qPCR. Finally, candidate drugs were identified by using the Drug Signatures Database (DSigDB).Results: Nineteen common DEGs were identified in HIV infection and PAH. Enrichment analysis exhibited that the functions of these genes were mainly enriched in inflammatory responses, mainly including cellular immunity and interaction between viral proteins and cytokines. By constructing PPI networks, we identified the key gene CC-type chemokine ligand 5 (CCL5), and we verified that CCL5 was highly expressed in hypoxia induced human pulmonary artery endothelial cells (hPAECs) and human pulmonary artery smooth muscle cells (hPASMCs). In addition, we predicted 10 potential drugs targeting CCL5 by Autodock Vina. Conclusion: This study revealed that CCL5 might be a common biomarker of HIV infection and PAH and provided a new therapeutic target for HIV-associated PAH. However, further clinical validation is still indispensable.
