The oxygen sensing enhancement based on room temperature phosphorescence (RTP) of Gd-HMME adjusted by imidazole was studied. The phosphorescence intensity I P0 and the Stern-Volmer equations under different imidazole concentration were obtained, and the physical mechanism of imidazole regulating the oxygen quenching constant K SV was analyzed. It was found that the K SV value increased by ∼46 folds in the range of 12.4(1)–576.1(5) kPa −1 , and the large-scale variation of K SV is conducive to the realization of high precision oxygen concentration measurement in a wide range. In addition, the standard deviation σ of continuous measurement results was given, and the limit of detection (LOD) was determined to be 6.6 ppm.

Immune checkpoint blockade (ICB) therapies, such as targeting programmed death-1 (PD-1) and programmed cell death protein ligand-1 (PD-L1), are often ineffective in low-immunogenic tumors, limiting their widespread use. In this study, an emulsion hydrogel is developed to encapsulate the sonosensitizer hematoporphyrin monomethyl ether (HMME), aimed at enhancing sonodynamic immunotherapy for low-immunogenic cancers. The water-in-oil emulsion hydrogel, characterized by its shear-thinning rheological properties, demonstrates excellent ultrasound (US)-triggered and lipase-triggered release, followed by sustained release. The emulsion hydrogel loaded with HMME (Gel@HMME), when combined with US, induces an immune response by promoting dendritic cell (DC) maturation and M1-tumor-associated macrophage (M1-TAM) polarization. This, in turn, enhances the infiltration of helper T lymphocyte 1 (Th1) cells and cytotoxic T lymphocytes (CTLs), thereby amplifying the antitumor immune response. Furthermore, Gel@HMME + US enhances the expression of PD-L1 in tumor cells. The combination of ICB (anti-PD-1, αPD-1) with Gel@HMME + US exerts a significant therapeutic effect on pancreatic cancer by upregulating the expression of calreticulin. Thus, this study presents a promising strategy for treating low-immunogenic cancers using an injectable HMME-loaded emulsion hydrogel, which enhances sonodynamic immunotherapy.

The enhancement of the room-temperature phosphorescence (RTP) emissions of phosphorescent metalloporphyrins is critical toward extending their applications in many fields. Imidazole is widely used as a molten solvent in the synthesis of metalloporphyrins and can influence their RTP emissions. Here, the effects of imidazole on the RTP emissions of a series of metalloporphyrins (hematoporphyrin monomethyl ether coordinated to gadolinium (Gd-HMME), lutetium (Lu-HMME), and palladium (Pd-HMME) ions) were studied. It was found that imidazole effectively enhanced the RTP emissions of these metalloporphyrins both on a solid matrix and in solution. For metalloporphyrins on a solid matrix (filter paper), the RTP intensities of samples immersed in an imidazole solution were much higher than those immersed in methanol. Imidazole enhanced the phosphorescence quantum yields ( Φ P ) of Gd-, Lu- and Pd-HMME on filter paper by 23-, 5.0-, and 3.5-fold, respectively. For metalloporphyrins in solution (methanol), the RTP intensities increased gradually with the concentration of imidazole. These results indicate that imidazole has potential as an RTP enhancer by protecting the triplet states of phosphorescent metalloporphyrins.

Gaseous oxygen detection is essential in numerous production and manufacturing sectors. To meet the varying oxygen detection requirements across different fields, techniques that offer a wide oxygen detection range should be developed. In this study, a wood-based oxygen sensing material was designed using balsa wood as the supporting matrix and gadolinium hemoporphyrin monomethyl ether (Gd-HMME) as the oxygen-sensitive indicator. The wood-based Gd-HMME exhibits a cellular porous structure, which not only facilitates the loading of a substantial number of indicator molecules but also enables the rapid interaction between indicators and oxygen molecules.OPis defined as the ratio of the phosphorescence intensity of the oxygen-sensing material in the anaerobic and aerobic environment. A linear relationship betweenOPand oxygen partial pressure ([O2]) was obtained within the whole range of [O2] (0–100 kPa). The wood-based Gd-HMME exhibited excellent resistance to photobleaching, along with a rapid response time (3.9 s) and recovery time (4.4 s). It was demonstrated that the measurement results obtained using wood-based Gd-HMME were not influenced by other gaseous components present in the air. An automatic oxygen detection system was developed using LabVIEW for practical use, and the limit of detection was determined to be 0.01 kPa.

A mechanism for the enhanced room-temperature phosphorescence (RTP) of gadolinium-coordinated hematoporphyrin monomethyl ether (Gd-HMME) in the presence of imidazole and free gadolinium ions (Gd3+) is revealed. Imidazole, the molten solvent used in the synthesis of Gd-HMME, was effective in enhancing the Gd-HMME RTP. In the presence of imidazole, further enhancement of the Gd-HMME RTP was observed upon adding Gd3+. Overall, a 40-fold enhancement of Gd-HMME RTP intensity was achieved by adding both imidazole and Gd3+. In addition, there was an increase in the RTP lifetime. Through spectroscopic analysis, we deduced that a protective medium is formed by the imidazole and the degree of this protection is further increased by Gd3+. The protective medium enhances the Gd-HMME RTP by partially inhibiting energy transfer from the lowest triplet state of Gd-HMME to oxygen. This was demonstrated by the presence of lower levels of singlet oxygen in the Gd-HMME solution after the addition of imidazole. These results indicate that imidazole could have potential application as an RTP enhancer or triplet state protector.

Sonodynamic therapy (SDT), which involves the activation of sonosensitizers to generate cytotoxic reactive oxygen species under ultrasound irradiation, is a promising noninvasive modality for cancer treatment. However, the clinical translational application of SDT is impeded by the lack of efficient sonosensitizers, the inefficient accumulation of sonosensitizers at tumor sites, and the complicated immunosuppressive tumor microenvironment. Herein, we developed a facilely synthesized multifunctional porous organic polymer nanosonosensitizer (mHM@HMME) for enhanced SDT. Specifically, mHM@HMME nanosonosensitizers were prepared by incorporating chemotherapeutic mitoxantrone into the one-step synthesis process of disulfide bond containing porous organic polymers, followed by loading with organic sonosensitizer (HMME) and camouflaging with a cancer cell membrane. Due to the cancer cell membrane camouflage, this multifunctional mHM@HMME nanosonosensitizer showed prolonged blood circulation and tumor targeting aggregation. Under ultrasound irradiation, the mHM@HMME nanosonosensitizer exhibited a satisfactory SDT performance both in vitro and in vivo. Moreover, the potent SDT combined with glutathione-responsive drug release in tumor cells induced robust immunogenic cell death to enhance the antitumor effect of SDT in turn. Overall, this facilely synthesized multifunctional mHM@HMME nanosonosensitizer shows great potential application in enhanced SDT.

en This link goes to a English section zh This link goes to a Chinese section Diabetic wound infections are a common complication of diabetes, which severely impact patients' quality of life. Effective treatment of diabetic wound infections remains one major challenge in the clinic, partially due to the formation of bacterial biofilm and antibiotic resistance. It is imperative to develop non-antibiotic-dependent strategies to efficiently eradicate biofilm infections in diabetic wounds. In this work, an innovative nanospray (CDs-HM) was successfully prepared by linking one acoustic sensitizer (hematoporphyrin monomethyl ether, HMME) to CDs fabricated with hemin, nickel(II) chloride, and polymer ethylene imine. CDs-HM exhibited catalase-enhanced sonodynamic properties and photothermal-enhanced chemodynamic properties. In vitro experiments and transcriptomic analysis demonstrated that CDs-HM successfully killed the bacteria and destroyed bacterial biofilm by disrupting bacterial cell membrane integrity, inducing oxidative stress, and inhibiting ATP production through synergistic photothermal therapy/chemodynamic therapy/sonodynamic therapy (PTT/CDT/SDT) effects. In the diabetic wound infection mouse model, CDs-HM remarkably eradicated MRSA biofilm, reduced inflammation levels, and promoted angiogenesis/collagen deposition so as to accelerate wound healing. It is noteworthy that CDs-HM displayed superior bacteria killing and wound healing effects compared with conventional vancomycin and nanosilver dressing interventions. The practicality and efficiency of CDs-HM endowed it with broad clinical translation prospects. It paved the way for developing novel strategies for combating diabetic wound infections. 摘要 en This link goes to a English section zh This link goes to a Chinese section 糖尿病伤口感染是糖尿病的常见并发症,严重影响患者的生活质量。

Synergistic photodynamic and photothermal therapy (PDT-PTT) has emerged as an appealing effective antitumor approach. However, clinical utilization of PDT-PTT is plagued by aggregation-caused photobleaching, sequential double irradiations, unsatisfying balance between single oxygen ( 1 O 2 ) quantum yield and photothermal conversion efficiency. Here, an anchored tumor-homing cell-penetrating peptide (PEGA- p VEC) and PANI-ES/HMME loaded FRET nanobullet (AHP-P) are reported. Within nanobullet, HMME (donor) and PANI-ES (acceptor) spontaneously form a förster resonance energy transfer (FRET) pair. Upon 660 nm laser irradiation, HMME convert near-infrared fluorescence (NIRF) to PANI, thus produce FRET-amplified photoacoustic imaging guided PTT. In addition, AHP-P with pH-sensitivity can gradually release HMME within acidic tumor environment, boosts the 1 O 2 regeneration alongside with highly efficient photothermal conversion for photoinduced cancer PTT-PDT. Furthermore, the AHP-P nanobullet can home in on the tumor site and penetrate into cytoplasm through PEGA- p VEC, inducing remarkable tumor regression with an ≈80% tumor volume reduction and decreased skin phototoxicity in vivo during FRET-amplified PTT-PDT.

Helicobacter pylori ( H. pylori ) has infected more than half of the world's population, and is the major cause of gastric cancer. The efficacy of standard antibiotic-based triple therapy is declining due to drug resistance development. Herein, a pH-responsive reactive oxygen species (ROS) nanogenerator (Fe-HMME@DHA@MPN) composed of acid-responsive metal polyphenol network (MPN) shell and mesoporous metal-organic nanostructure core [Fe-HMME (hematoporphyrin monomethyl ether, sonosensitizer)] loaded with dihydroartemisinin (DHA) is reported. These nanoparticles generate more ROS singlet oxygen than sonosensitizer HMME under ultrasonication, and this sonodynamic process is fueled by oxygen generated through Fenton/Fenton-like reactions of the degraded product in gastric acid Fe (II) and hydrogen peroxide (H 2 O 2 ) in the infection microenvironment. The encapsulated DHA, as a hydroperoxide source, is found to enhance the peroxidase-like activity of the Fe-HMME@DHA@MPN to generate ROS hydroxyl radical, beneficial for the microenvironment without sufficient H 2 O 2 . In vitro experiments demonstrate that the ROS nanogenerators are capable of killing multidrug-resistant H. pylori and removing biofilm, and ROS nanogenerators show high therapeutic efficacy in a H. pylori infection mouse model. Unlike the triple therapy, the nanogenerators display negligible side effects toward the normal gut microbiota. Taken together, these self-enhanced ROS nanogenerators have a great potential for treatment of H. pylori infection.
