GSH-responsive bithiophene Aza-BODIPY@HMON nanoplatform for achieving triple-synergistic photoimmunotherapy

Siao Yang,Xiaoxiao Hu,Zhengze Yong,Qingqing Dou,Cuilu Quan,Hong-Bo Cheng,Mo Zhang,Jing Wang

Photoimmunotherapy represents an innovative approach to enhancing the efficiency of immunotherapy in cancer treatment. This approach involves the fusion of immunotherapy and phototherapy (encompassing techniques like photodynamic therapy (PDT) and photothermal therapy (PTT)). Boron-dipyrromethene (BODIPY) has the potential to trigger immunotherapy owing to its excellent PD and PT efficiency. However, the improvements in water solubility, bioavailability, PD/PT combined efficiency, and tumor tissue targeting of BODIPY require introduction of suitable carriers for potential practical application. Herein, a disulfide bond-based hollow mesoporous organosilica (HMON) with excellent biocompatibility and GSH-responsive degradation properties was used as a carrier to load a bithiophene Aza-BODIPY dye (B5), constructing a sample chemotherapy reagent-free B5@HMON nanoplatform achieving triple-synergistic photoimmunotherapy. HMON, involving disulfide bond , is utilized to improve water solubility, tumor tissue targeting, and PD efficiency by depleting GSH and enhancing host-guest interaction between B5 and HMO. The study reveals that HMON’s large specific surface area and porous properties significantly enhance the light collection and oxygen adsorption capacity. The HMON’s rich mesoporous structure and internal cavity achieved a loading rate of B5 at 11 %. It was found that the triple-synergistic nanoplatform triggered a stronger anti-tumor immune response, including tumor invasion, cytokine production, calreticulin translocation, and dendritic cell maturation, eliciting specific tumor-specific immunological responses in vivo and in vitro . The BALB/c mouse model with 4T1 tumors was used to assess tumor suppression efficiency in vivo , showing that almost all tumors in the B5@HMON group disappeared after 14 days. Such a simple chemotherapy reagent-free B5@HMON nanoplatform achieved triple-synergistic photoimmunotherapy.

Fe0Ni0 bimetal co-immobilized horseradish peroxidase on ZIF-8@HMON for degradation of trichloroacetic acid

Yuxiang Yang,Xiaocui Huang,Mengyang Dong,Ziling Chang,Hongming Yuan,Yan Huang,Xiangnong Liu,Jianbo Zhang,Anbang Dai

As a common pollutant in the environment, trichloroacetic acid (TCA) is difficult to be completely degraded by common advanced oxidation processes (AOPs) due to its high C-Cl bond content. Advanced oxidation–reduction processes (AORPs) may improve the degradation performance of TCA. In this work, we use Fe 0 Ni 0 bimetal as reducing catalyst, hollow mesoporous silicone nanoparticles (HMONs) modified by ZIF-8 as immobilized carrier, horseradish peroxidase (HRP) and Fe 0 Ni 0 were fixed in the carrier to form a AORP system for degradation of TCA. The effects of temperature, pH, H 2 O 2 concentration, doping ratio on the degradation performance of the catalyst and the degradation mechanism of the composite catalyst were investigated. Degradation experiments showed that H· and OH· were present in the dual system, dechlorination and decarboxylation exist at the same time. Under the condition of 55 °C and pH = 4, 30 mg Fe 0 Ni 0 and 20 mg immobilized enzyme shows the best degradation on TCA, and 81.48 % of 20 mg·L −1 TCA could be degraded by 2 h.

Biodegradable, pH-Sensitive Hollow Mesoporous Organosilica Nanoparticle (HMON) with Controlled Release of Pirfenidone and Ultrasound-Target-Microbubble-Destruction (UTMD) for Pancreatic Cancer Treatment

Feng Gao,Jianrong Wu,Shiwei Niu,Ting Sun,Fan Li,Yun Bai,Lifang Jin,Lizhou Lin,Qiusheng Shi,Li-Min Zhu,Lianfang Du

The dense extracellular matrix (ECM) and hypovascular networks were often found in solid pancreatic tumors form an impenetrable barrier, leading to limited uptake of chemotherapeutics and thus undesirable treatment outcomes. Methods : A biodegradable nanoplatform based on hollow mesoporous organosilica nanoparticle (HMON) was designed as an effective delivery system for pirfenidone (PFD) to overcome the challenges in pancreatic tumor treatment. By varying pH producing a mildly acidic environment to emulate tumor cells, results in cleavage of the acetal bond between HMON nanoparticle and gating molecular, gemcitabine (Gem), enabling its controlled release. Results : The in vitro and in vivo immunocytochemistry evaluations demonstrated an excellent ECM regulation efficacy of the nanoplatform and therefore the improved penetration of drug into the cells. The technique employed was especially enhanced when mediated with ultrasound target microbubble destruction (UTMD). Evaluations culminated with pancreatic cancer bearing mice and demonstrated therapeutic efficacy, good biodegradability, and negligible systemic toxicity. Conclusion : the designed Gem gated biodegradable nanosystem is expected to provide an alternative way of improving antitumor efficacy by down-regulation of ECM levels and offers a passive-targeted therapy for pancreatic cancer treatment.
