Synthesis of yolk–shell NaA@HMSNs nanoparticles for bifunctional Cd2+ adsorption and curcumin delivery

Libin Wang,Min Li,Kaixuan Liu,Chunyuan Liu,Bo Ren

An RF spacer creates a void between NaA and a mesoporous silica shell. • Shell thickness tunes curcumin loading in yolk–shell NaA@HMSNs. • NaA@HMSN-2 loads 124.0 mg/g curcumin and adsorbs Cd 2+ in vitro. • Curcumin release and Cd 2+ adsorption occur in the same buffer system. • HepG2 viability remains above 90% up to 64 μg/mL after 48 h. Cadmium contamination is an environmental and health concern. Here, a yolk–shell composite was constructed using sodium A-type zeolite (NaA) as the microporous yolk and a hollow mesoporous silica nanoparticle (HMSN) layer as the separated shell. Removal of a resorcinol–formaldehyde spacer generated an internal void, and three shell thicknesses were prepared. The NaA component provided Cd 2+ adsorption sites, whereas the amino-functionalized HMSN compartment was used for curcumin loading. N-NaA@HMSN-2, with an approximately 100 nm shell, loaded 124.0 mg/g curcumin. In buffer containing 100 mg/L Cd 2+ , Cur/N-NaA@HMSN-2 released 34.8% and 39.5% curcumin and adsorbed 73.8% and 67.8% Cd 2+ at pH 7.4 and 1.2, respectively. In HepG2 assays, all three NaA@HMSN carriers maintained more than 90% relative cell viability at concentrations up to 64 μg/mL after 48 h. These findings demonstrate coupled Cd 2+ adsorption and curcumin release under in vitro conditions and support further evaluation of this bifunctional material platform. Tunable pH-responsive HMSNs-g-PDEAEMA as integrated corrosion inhibition and emulsification multi-functional nanocontainer

Huiling Su,Zhikun Wang,Jianqiang Zhang,Nannan Cui,Fengting Li,Miantuo Li,Wentao Qu,Xupeng Li,Jiaqi Feng,Songqing Hu

Responsive nanocontainers have dual functions in targeted delivery of corrosion inhibitors and emulsion development of shale oil in oil and gas fields, exhibiting potential for simultaneously achieving metal protection and efficient oil and gas development from a material perspective. Here, we propose the preparation of a pH-responsive nanocontainer, HMSNs- g -PDEAEMA (poly [2-(N, N-diethyl amino)-ethyl-methacrylate] (PDEAEMA) grafted onto hollow mesoporous spherical silica (HMSNs)), to integrate the delivery of 2-mercaptobenzothiazole (MBT)) for targeted corrosion inhibition and the emulsification of oil as Pickering emulsifiers. Under acidic conditions (reduced pH value caused by localized corrosion or high concentration acidic gases), PDEAEMA chains are protonated and extended by electrostatic repulsion, exposing pores on HMSNs surface and allowing the controlled release of loaded MBT molecules. Once transforming into a neutral or alkaline environment, the responsive release of the MBT process is inhibited. After the fluid passes through the wellbore and enters the shale layer, the HMSNs- g -PDEAEMA nanocontainers act as Pickering emulsifiers to achieve emulsification. The emulsified oil can be extracted onto the ground more efficiently, and a following pH-responsive demulsification process can be achieved. Overall, through a pH-responsive nanocontainer material, the dual function of corrosion inhibition and emulsification in oil and gas development is possible to be simultaneously achieved.

Preparation of Hollow Mesoporous Silica-Based Carriers for Loading and Release of Fenofibrate

Lv J.,Yang S.,Zhang W.,Wei Y.,Qu Y.,Lv C.

Abstract Hollow mesoporous silica-based materials are ideal drug carriers applied in controlled drug delivery systems due to their unique hollow core and tunable shell composition. Two types of hollow mesoporous materials, hollow mesoporous silica nanoparticles (HMSNs) and hollow periodic mesoporous organosilicas (HPMOs), were synthesized and applied as drug carriers to regulate insoluble drug release. Fenofibrate (FNB) was used as a model drug to study the effect of framework difference for HMSNs and HPMOs materials on the drug loading and release processes. FNB/HPMOs had a higher drug loading (35.39%) than FNB/HMSNs (30.22%) because of hydrophobic bridging C 2 H 4 groups within the mesoporous skeleton. HMSNs showed fast release behavior, resulting from their hydrophilic surface with a large number of silanol groups favorable for dissolution media diffusion and drug dissolution. HPMOs showed sustained release behavior, attributed to their hydrophobic surface which possessed stronger hydrophobic-hydrophobic van der Waals attraction to drug molecules. The drug release of HMSNs and HPMOs in different dissolution media followed the first order kinetic model with decreased release rate vs . time. Regulation of the organic-inorganic skeleton structure of the mesoporous materials could change their surface properties especially hydrophilicity or hydrophobicity, furtherly controlling the loading capacity and release behavior of the loaded drugs.

Spatiotemporally Programmed VEGF/IL-4 Delivery via HMSNs Enhances Endothelialization and Immune-Mediated Matrix Remodeling in Acellular Vascular Grafts

Zeguo Chen,Zhongshi Wu,Can Huang,Qiying Wu,Chao Xie,Sicheng Chen,Liang Yi,Haoyong Yuan,Sixi Liu,Abdulraheem Mustapha,Siyao Chen,Wansong Chen,Ting Lu,Zhenjie Tang,Yuhong Liu

Delayed endothelialization and inadequate matrix remodeling remain major obstacles in the development of small-diameter vascular grafts (SDVGs; <6 mm). To address these challenges, we developed an immunomodulatory tissue-engineered vascular graft (iTEVG) by integrating hollow mesoporous silica nanoparticles (HMSNs) with tailored mesopore sizes as delivery carriers for immunoregulatory factors. VEGF was selectively immobilized on the intimal surface, while the adventitia incorporated a sequential release system for VEGF and IL-4. VEGF exhibited rapid release (64.04 ± 4.44% in 7 days), promoting monocyte recruitment and adventitial neovascularization, while IL-4 showed sustained release (65.94 ± 2.06% over 28 days), driving long-term M2 macrophage polarization. In rats, iTEVGs achieved 88% endothelial coverage and smooth muscle cell infiltration within 1 month, full-thickness cellularization with a complete trilaminar structure by 3 months, and maintained mechanical integrity without aneurysm formation up to 6 months. This spatially partitioned platform, built on an acellular vascular scaffold, enables precise spatiotemporal regulation of the immune microenvironment and offers a design paradigm that synergistically promotes endothelialization and vascular matrix remodeling. The “intima-targeted regeneration and adventitia-sequential-release” strategy provides a promising template for SDVG design that may be extended to other complex organ constructs.

A versatile gas-generator promoting drug release and oxygen replenishment for amplifying photodynamic-chemotherapy synergetic anti-tumor effects

Xiudan Wang,Yuling Mao,Changshan Sun,Qinfu Zhao,Yikun Gao,Siling Wang

Excellent efficiency of combinational therapy of chemotherapy and photodynamic therapy (PDT) highly depends on the amounts of drug and oxygen in tumor tissue. However, how to cleverly promote drug release accompanied with improving oxygen concentration remains a challenge. Herein, we proposed a gas-generator that realized a high drug loading and integrated facilitation of drug release with oxygen replenishment into a single and simple system, utilizing huge cavities and mesoporous channels of hollow mesoporous silica nanoparticles (HMSNs) for encapsulating oxygen (O 2 ) saturated perfluoropentane (PFP) droplets, indocyanine green (ICG) and doxorubicin (DOX), biocompatible polydopamine (PDA) as the gatekeepers. Under irradiation of 808 nm laser , the thermal effect of PDA caused PFP droplets occur liquid-gas phase transition that triggered the burst release of DOX and O 2 , finally amplifying the synergetic effects of PDT and chemotherapy both in vitro and in vivo. The influence of PFP, GSH and laser on drug release kinetic was explored through mathematical models. Notably, the mechanism of gas-generator on accelerating drug release under irradiation based on doing volume work and enhancing diffusion coefficient was clarified by researching the relation between DOX release, PFP release and temperature change. Additionally, the way of replenishing O 2 did not rely on intracellular components but timely offered abundant “fuels” for producing reactive oxygen species (ROS) when compared with traditional manners. This work provides a new research strategy for boosting drug release and opens an avenue for constructing multifunctional controlled delivery systems.
