This study focused on enhancing the stability and bioaccessibility of pterostilbene (PTS) by encapsulating it with hydroxypropyl-β-cyclodextrin (HP-β-CD) using high-pressure homogenization (HPH) and pH-shift methods. The encapsulation process was performed at pressures of 50, 100, and 150 MPa along with three different treatments, achieving a maximum encapsulation efficiency of 94.34% and a loading capacity of 14.82% at 150 MPa. The SEM, XRD, and FTIR analyses confirmed encapsulation by revealing significant morphological changes, crystal structure modifications, and host-guest interactions. PTS-HP-β-CD complexes retained 82.22% of PTS at 4°C (28 days) and 76% under UVB (120 min), compared to 56.03% and 21% for free PTS, respectively. Additionally, these complexes exhibited enhanced DPPH scavenging rates (44.95% at 0.02 mg/mL), nearly twice that of free PTS. In the release studies, the complexes’ release behavior aligned with the Higuchi model. Encapsulated PTS revealed lower digestion rates in in-vitro gastric (32.24% vs. 49.88%) and intestinal phases (63.98% vs. 91.01%). Additionally, there was no observable toxicity for PTS-HP-β-CD to human epidermal keratinocytes, supporting its potential use in cosmetics, food, and pharmaceuticals. These results demonstrated that HPH combined with pH-shift methods effectively improved the stability and bioaccessibility of PTS, offering a promising strategy for its formulation.

Effects of hydroxypropyl-β-cyclodextrin grafted polyethylenimine (HP-β-CD-PEI) including HP-β-CD-PEI 600 , HP-β-CD-PEI 1800 , HP-β-CD-PEI 10000 on the pulmonary absorption of insulin, calcitonin, 5(6)-carboxyfluorescein (CF) and fluorescein isothiocyanate dextrans (FDs) with various molecular weights (FD4, FD10 and FD70) were examined by an pulmonary absorption study in rats. Pulmonary absorptions of these poorly absorbable drugs were significantly enhanced by HP-β-CD-PEI 1800 and HP-β-CD-PEI 10000 , and HP-β-CD-PEI 1800 with the concentration of 5% (w/v) provided maximal absorption enhancing effect on pulmonary absorption of these model drugs. The toxicity study demonstrated that HP-β-CD-PEI did not induce any toxic action to rat pulmonary membranes. In addition, zeta potential of insulin solution changed to positive by addition of various HP-β-CD-PEI, meanwhile, the degree of positive charge was linearly correlated with absorption enhancing effect of HP-β-CD-PEI, suggesting that positive charge of HP-β-CD-PEI might be related to their absorption enhancing mechanisms for enhancing pulmonary absorption of insulin in rats. In conclusion, HP-β-CD-PEI is a potential and safe absorption enhancer for improving absorption of hydrophilic macromolecules especially peptide and protein drugs by pulmonary delivery.

In this study, a tumor-targeting and pH-sensitive inclusion complex based on the host–guest recognition between the chitosan and folic acid grafted HP-β-CD (FA–CS–CD) and stearic acid modified 2-benzimidazolemethanol (BM-SA) was designed and fabricated for the controlled delivery of paclitaxel (PTX). Through the combination of computational simulations and experiments, the interaction between FA–CS–CD, BM-SA, and PTX was investigated, and the optimized preparation method was obtained. For the optimized PTX-loaded FA–CS–CD/BM-SA inclusion complex, the particle size and zeta potential were 146 nm and +15.4 mV, respectively. In vitro drug release study revealed the pH-triggered drug release behavior of the inclusion complex. Both in vitro and in vivo evaluations demonstrated that the PTX-loaded FA–CS–CD/BM-SA inclusion complex exhibited enhanced antitumor efficiency and minimized systemic toxicity. This system might be a promising carrier for PTX.

As a nutritious cold chain food , cheese is susceptible to Listeria monocytogenes contamination. In order to prevent and control Listeria monocytogenes in cheese safely and efficiently, maintain the sensory quality during sale, this paper developed a novel nanofiber membrane with good mechanical strength, hydrophobicity , thermal stability , release performance and bacteriostatic performance as food active packaging. In this study, zein was introduced and electrospun to form a reliable carrier to embed and deliver citral in the cavity of HP-β-CD (Hydroxypropyl-β-cyclodextrin). We found that the zein-based nanofiber membranes showed preferable tensile properties and hydrophobicity with 4% (w:v) addition of citral/HP-β-CD inclusion complex. Meanwhile, the thermal stability and release property of citral were significantly improved by embedding in HP-β-CD and loading into zein-base nanofiber membranes. Moreover, after packaged by Zein nanofiber membrane encapsulated with citral/HP-β-CD inclusion complex under the storage conditions of 4 °C and 12 °C, the growth of Listeria monocytogenes in four kinds of cheeses was inhibited to a certain degree. The texture and sensory quality of the four cheeses at the two temperatures and the color quality under the low temperature condition of 4 °C did not change significantly. In summary, Zein nanofiber membranes encapsulated with citral/HP-β-CD inclusion complex has potential application prospects in the field of food active packaging.

Context: Oral bioavailability of some hydrophilic therapeutic macromolecules was very poor, thus leading to their limited application in clinic. Objective: To investigate the efficacy, safety and mechanism of HP-β-CD-PEI polymers on the intestinal absorption of some poorly absorbable drugs in rats. Methods: Effects of HP-β-CD-PEI polymers on the intestinal absorptions of drugs were investigated by an in situ closed loop method in rats. The safety of HP-β-CD-PEI polymer was evaluated by measurement of lactate dehydrogenase (LDH) activity and amount of protein released from rat intestinal perfusate. The absorption enhancing mechanisms were explored by the measurement of zeta potential, transepithelial electrical resistance (TEER) and in vitro transport of FD4 (a paracellular marker) across rat intestinal membranes, respectively. Results: HP-β-CD-PEI polymers, especially HP-β-CD-PEI1800, demonstrated excellent absorption enhancing effects on drug absorption in a concentration-dependent manner and the enhancing effect was more efficient in the small intestine than that in the large intestine. Five percent (w/v) HP-β-CD-PEI1800 obviously decreased the TEER, accompanied with increase in the intestinal transport of FD4, indicating that absorption enhancing actions of HP-β-CD-PEI polymers were possibly performed by loosening tight junctions of intestinal epithelium cells, thereby increasing drug permeation via a paracellular pathway. A good liner relationship between absorption enhancing effects of HP-β-CD-PEI polymers and their zeta potentials suggested the contribution of positive charge on the surface of these polymers to their absorption enhancing effects. Conclusion: HP-β-CD-PEI polymers might be potential and safe absorption enhancers for improving oral delivery of poorly absorbable macromolecules including peptides and proteins.

Daidzein is an isoflavone of naturally abundance existing in plants and foods which has attracted much attention for its significant benefits on human health. However, its application was severely limited by its poor solubilities, instability and low bioavailability. To overcome these drawbacks, inclusion complexes of daidzein with two cyclodextrin (CD) derivatives, i.e., 2-hydropropyl-β-cyclodextrin (HP-β-CD) and sulfobutyl ether-β-cyclodextrin (SBE-β-CD) were prepared and characterized both in solution and solid state by 1D and 2D NMR , XRD, SEM and elemental analyses. Fluorescence spectroscopy and the Job plot were used to demonstrate a mainly 1:1 inclusion mode between daidzein and CDs. Their thermal stabilities were evaluated with TG and DSC experiments. Moreover, water solubility of daidzein was significantly improved by inclusion complexation with CDs. These results might suggest valuable approaches to developments of new pharmaceutical formulations of daidzein.

Objectives: To study the encapsulation of resveratrol by three cyclodextrin derivatives, investigate the impact of encapsulation technology on its anti-fatigue effect, and explore the anti-fatigue mechanism of the encapsulated resveratrol via transcriptomics analysis of exosomal miRNAs. Methods (1) UV spectroscopy and nuclear magnetic resonance (NMR) were used to investigate the encapsulation and encapsulation modes of resveratrol by β -CD, HP- β -CD and CM- β -CD; and the phase solubility method was used to calculate the solubilisation multiplicity of resveratrol by the three cyclodextrin derivatives. (2) Male C57BL/6 J mice were randomly divided into: fatigue model group (FM), FM+resveratrol group (SF+Res), FM+ β -CD, FM+HP- β -CD, FM+CM- β -CD, FM+ β -CD+Res, FM+HP- β -CD+Res, FM+CM- β -CD +Res, and after 30 days of rearing, the mice were subjected to exhaustion swimming experiment to detect fatigue-related indexes and observe the effects of the three cyclodextrin derivatives on the anti-fatigue effects of resveratrol. (3) Male C57BL/6 J mice were randomly divided into Control, FM, and FM+HP- β -CD+Res. After 30 days of rearing, except for the Control group, the remaining mice were subjected to exhaustion swimming experiments, and exosomes were extracted from cardiac tissue, skeletal muscle tissue, and liver tissue of the three groups of mice, and the transcriptomics technology was applied to sequence the exosomal miRNAs and to explore the mechanism of the effect of the HP- β -CD+Res envelope on fatigue. Results (1) The inclusion ratios of β -CD, HP- β -CD, and CM- β -CD encapsulated resveratrol were 1:1, 1:2, and 1:1, and the solubilisation multiplicities of resveratrol were 23, 39, and 21 fold, respectively; β -CD and CM- β -CD encapsulated Res, which was the benzene ring with two hydroxyl‑terminal ends of Res entered the cavity from the small-ported end of the cyclodextrins; HP- β -CD encapsulated Res, which was the Res containing one hydroxyl‑terminated benzene ring enters the cavity from the wide-mouth end of HP-β-CD. (2) Compared with the FM+Res group, all three inclusions were able to prolong the exhaustion swimming time of mice, with HP- β -CD+Res being the most effective. (3) After HP- β -CD+Res acted on the fatigued mouse model, cardiac tissue showed 1726 differentially expressed exosomal miRNAs, of which 840 were up-regulated and 886 were down-regulated, reversing the fatigue-induced changes in 64.96 % of the exosomal miRNAs (1170 shared differential exosomal miRNAs); Skeletal muscle tissue showed 1948 differentially expressed exosomal miRNAs, of which 718 were up-regulated and 1230 were down-regulated, reversing 80.57 % of the fatigue-induced changes in exosomal miRNAs (1576 shared differential exosomal miRNAs); Liver tissue showed 1442 differentially expressed exosomal miRNAs, of which 451 were up-regulated and 991 were down-regulated, reversing 84.31 % of fatigue-induced changes in exosomal miRNAs (1214 shared differential exosomal miRNAs). Target gene enrichment analysis of differentially expressed exosomal miRNAs in cardiac tissue, skeletal muscle tissue, and liver tissue had similar results, and the biological processes involved in HP- β -CD+Res were related to biological_process, signal transduction, G protein-coupled receptor signaling pathway, etc., and cellular components such as membrane, cytoplasm, integral component of membrane, etc., and molecular functions are closely related to protein binding, metal ion binding, molecular_function, etc.; mainly enriched in Axon guidance, MicroRNAs in cancer, Apoptosis and other signalling pathways. The aim of this study was to develop and evaluate hollow sodium chloride microspheres with enhanced salinity perception as a strategy to reduce overall salt intake in foods. This experimental systematically evaluated the water content, hygroscopicity, particle size distribution, conductivity, scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and electron tongue of hollow salt microspheres prepared from hydroxypropyl-β-cyclodextrin (HP-β-CD), soluble soybean polysaccharide (SSPS), maltodextrin (MD), gum arabic (GA), and β-glucan (β-glucan). The results showed that there were significant differences in the characteristics of hollow sodium chloride microspheres prepared by different carriers. The salt microspheres prepared with HP-β-CD as the carrier exhibited low water content (1.89 %), anti-hygroscopicity (1.67 g/100 g), and fast solubility. The conductivity test results showed that the release rate of sodium ions in the salt microspheres prepared with HP-β-CD was nearly six times higher than that of ordinary sodium chloride, which could rapidly evoke the perception of saltiness. EDS analysis further confirmed that the salt microspheres prepared with HP-β-CD carrier had a higher sodium chloride content on the surface (26.25 %), thus contributing to enhancing salty taste perception. In addition, the salt microspheres prepared with soybean-soluble polysaccharide as the carrier showed a perfectly spherical hollow structure, and the caking property was lower than that of the other salt microspheres. The results of the electronic tongue analysis showed that the salt microsphere solution prepared using soybean-soluble polysaccharide had a stronger salty taste perception ability. In summary, the hollow salt microspheres prepared using HP-β-CD and SSPS showed outstanding advantages in salt taste perception properties. In the later stage, the compound ratio between the carriers will be studied, which provides a new technical reference for the salt reduction strategy of the food industry.

An HP-β-CD-functionalized hydrogel SERS strategy was developed for white tea storage-year analysis. • HP-β-CD-assisted capture and Ag-hotspot SERS sensing of tea polyphenols were mechanistically discussed. • Agarose improved nanoparticle dispersion and signal uniformity. • Sample-level machine learning enabled internal storage-year discrimination of the tested white tea samples. • External validation using multi-origin samples is still required to confirm practical robustness. Accurate discrimination of white tea storage years is important for quality evaluation, authenticity control, and market regulation. In this study, an HP-β-CD-functionalized hydrogel surface-enhanced Raman scattering (SERS) substrate was developed for rapid storage-year discrimination of white tea. Hydroxypropyl-β-cyclodextrin-functionalized silver nanoparticles (HP-β-CD/Ag NPs) were incorporated into an agarose hydrogel to construct an HP-β-CD/Ag NPs/AG substrate. HP-β-CD was designed to promote the local capture of aromatic and polyphenolic tea components near SERS-active Ag hotspots, while the agarose hydrogel improved nanoparticle dispersion and signal uniformity. The optimized substrate retained distinguishable SERS responses for epigallocatechin gallate and quercetin at 10 −7 mol L −1 and showed good signal uniformity, with a relative standard deviation of 6.96%. White tea samples from seven storage years between 2011 and 2023 were analyzed, including 10 independent samples per year and 10 spectra per sample. Principal component analysis showed that the first three principal components explained 98.94% of the spectral variance. Under 50 repeated tea-sample-level splits, machine-learning models achieved stable internal storage-year discrimination, with sample-level accuracies ranging from 97.62% to 100.00%. A sample-level permutation test further indicated that the classification performance was unlikely to result from random label association. These results suggest that the proposed hydrogel SERS strategy can provide a rapid and fingerprint-based approach for white tea storage-year discrimination, although further external validation using more diverse samples is still required. Neohesperidin (NH), a bioactive flavanone glycoside, exhibits multifaceted pharmacological properties including antioxidant and anti-inflammatory activities. However, its clinical application is severely constrained by inherent physicochemical limitations such as poor aqueous solubility and instability under physiological conditions. To address these challenges, this study developed a dual-carrier nano-liposomal system through the synergistic integration of phospholipid complexation and hydroxypropyl-β-cyclodextrin (HP-β-CD) inclusion technologies. Two formulations—NH-PC (phospholipid complex) and NH-PC-CD (phospholipid/HP-β-CD hybrid)—were fabricated via ultrasonication-assisted ethanol precipitation. Comprehensive characterization using FTIR and PXRD confirmed the amorphous dispersion of NH within lipid bilayers, with complete elimination of crystalline diffraction peaks, indicative of molecular-level interactions between NH’s hydroxyl groups and phospholipid polar moieties. The engineered nanosystems demonstrated remarkable solubility enhancement, achieving 321.77 μg/mL (NH-PC) and 318.75 μg/mL (NH-PC-CD), representing 2.01- and 1.99-fold increases over free NH. Encapsulation efficiencies exceeded 95% for both formulations, with sustained release profiles revealing 60.81% (NH-PC) and 80.78% (NH-PC-CD) cumulative release over 72 h, governed predominantly by non-Fickian diffusion kinetics. In vitro gastrointestinal simulations highlighted superior bioaccessibility for NH-PC-CD (66.35%) compared to NH-PC (58.52%) and free NH (20.85%), attributed to enhanced stability against enzymatic degradation. Storage stability assessments further validated the robustness of HP-β-CD-modified liposomes, with NH-PC-CD maintaining consistent particle size (<3% variation) and encapsulation efficiency (>92%) over 30 days. Antioxidant evaluations demonstrated concentration-dependent DPPH radical scavenging, wherein nanoencapsulation significantly amplified NH’s activity compared to its free form. This study establishes a paradigm for dual-functional nanocarriers, offering a scalable strategy to optimize the delivery of hydrophobic nutraceuticals while addressing critical challenges in bioavailability and physiological stability.

To develop a novel food packaging material, hydroxypropyl-β-cyclodextrin (HP-β-CD) with good water solubility was used to encapsulate cinnamaldehyde (CA). The prepared CA/HP-β-CD inclusion complex (CA/HP-β-CD-IC) was further embedded into the nanofibers, which were composed of phospholipid and poly (ethylene oxide). FTIR and molecular structure modeling indicated that CA was encapsulated in HP-β-CD most probably by directly inserting the phenyl of CA into the cavity of HP-β-CD. Subsequently, the preferable mechanical properties with the uniform diameter distribution of the mat of nanofibers were realized. Meanwhile, the thermal stability of CA was further improved after being embedded into HP-β-CD and the mat of nanofibers. For the application on fresh-cut cucumber, the mat of nanofibers encapsulated with CA/HP-β-CD-IC exhibited antibacterial activity against Listeria monocytogenes at 4 °C and 25 °C for 4 days, without significant effect on the sensory indicators of fresh-cut cucumber. Thus, the mat of nanofibers encapsulated with CA/HP-β-CD-IC could be a promising and practical food packaging material.

Mangoes are highly susceptible to post-harvest spoilage, leading to significant losses. This study aimed to optimize the encapsulation of lemongrass essential oil (LEO) using ultrasonic-assisted hydroxypropyl-β-cyclodextrin (HP-β-CD) and response surface methodology (RSM). The ultrasonic-assisted inclusion complex (UIC) was characterized through various techniques including particle size analysis, SEM, UV–vis, FTIR, XRD, and TGA. The antibacterial activity of the UIC was evaluated, and its effectiveness in prolonging mango shelf life was assessed by monitoring parameters such as weight loss, firmness, total soluble solids (TSS), titratable acidity (TA), malondialdehyde (MDA), vitamin C (VC) content, and external appearance. The results indicated that ultrasonic-assisted encapsulation significantly improved the encapsulation efficiency (EE), with optimal conditions of a core-wall ratio of 9:1, ultrasonic power of 150 W, and temperature of 36 °C. The UIC exhibited sustained release of LEO and enhanced antimicrobial activity, resulting in significant reductions in weight loss, firmness degradation, and metabolic changes during mango storage. Treated mangoes also showed improved appearance, with delayed onset of black spots and browning. These findings suggest that ultrasonic-assisted HP-β-CD encapsulation of LEO is an effective, eco-friendly strategy for postharvest preservation of mangoes, offering substantial potential for extending shelf life and maintaining fruit quality.
