Propolis, a well-known folk medicine in the world, has drawn growing attention due to the various bioactivities. An on-line method of coupling HPLC with 2,2′-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) cationic radical scavenging detection (HPLC–ABTS) was developed to screen the single polyphenol possessing ABTS cationic radical scavenging activity in ethanol extracts of Chinese propolis (EECP). Ten common polyphenols were selected from the active fingerprints of ten EECP samples obtained from different provinces of China by the on-line HPLC–ABTS method and quantified by Trolox equivalent. Their contributions to the antioxidant property of EECP were compared. Furthermore, Pearson’s correlation analysis was applied to determine whether there was a correlation between the antioxidant activity of individual polyphenol and total antioxidant activity of EECP. The results indicated that the composition of polyphenols in ten EECP samples obtained from China was similar. But the different polyphenols contributed in different way. Caffeic acid, kaempferol, and galangin simultaneously were the most prominent contributors to the antioxidant properties of ten EECP. The contributions of pinobanksin 3-acetate, caffeic acid phenethyl ester (CAPE), and caffeic acid cinnamyl ester varied largely with the different geographical regions. They, except galangin, significantly correlated with the antioxidant property of propolis. However, pinobanksin, pinocembrin, chrysin, p -coumaric acid, and ferulic acid rarely contributed to the antioxidant property of Chinese propolis. Therefore, the on-line HPLC–ABTS method was convenient, effective, and comprehensive to evaluate the contributions of polyphenols to the antioxidant property for Chinese propolis.

In the present study, an online liquid extraction coupled with high-performance liquid chromatography-2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (HPLC-ABTS) system for rapid screening of antioxidants in tea samples was proposed. As an example, the tea samples were firstly extracted by online HPLC extractor with mobile phase at 70°C, then the hyphenated HPLC-ABTS was used for the chromatographic separation on a Poroshell EC C18 column by 0.3% aqueous formic acid and acetonitrile with a gradient elution at 1.5 mL·min−1, and the UV and antioxidant chromatograms with detection wavelengths at 270 nm and 750 nm were recorded, respectively. The established system integrated the processes of online HPLC sample extraction, HPLC separation and online antioxidants detection, the total analysis time of which was <20 min. The developed method was successfully applied to samples of green tea, oolong tea and black tea. As a result, 11 antioxidants were found in tea samples, including gallocatechin, epigallocatechin, catechin, chlorogenic acid, epicatechin, epigallocatechingallate, epicatechingallate, rutin, 1,4,6-trigalloylglucose, quercetin-3-glycoside and kaempferol-3-glucoside. The combined online liquid microextraction and online HPLC-ABTS method is a rapid and green approach for the quality evaluation of tea.

The root of Polygonum bistorta (PB) is a traditional Chinese medicinal plant material widely used in China. It has been commonly used for the treatment of hemostasis, detumescence, diarrhea, snake bite, and acute gastroenteritis. However, the research on the antioxidant properties and bioactive compounds from PB is inadequate. In the current research, an online microextraction (OLME) coupled with a high-performance liquid chromatography coupled with the 2,2-nitrogen-di (3-ethyl-benzothiazole-6-sulfonic acid) diammonium salt antioxidant assay (HPLC-ABTS) system for rapid analysis of antioxidants from PB was proposed. The PB sample (0.17 mg) was online extracted by mobile phase (acetonitrile and 0.2% acetic acid); a Poroshell 120 SB-Aq column was used for separation; then, an online ABTS assay system was used for screening the antioxidants. Finally, ten components were found in PB, and among them, eight components possessed antioxidant activities. Furthermore, five components (gallic acid, neochlorogenic acid, caffeic acid, chlorogenic acid, and an unknown compound) were proved as major antioxidants when compared with rutin as an antioxidant marker. The results showed that the developed OLME-HPLC-ABTS system was a simple, rapid, green, and efficient instrument for the screening of antioxidants from PB, which provides a powerful tool for the discovery of natural antioxidants in Chinese medicines.

In this study, an online antioxidant assay based on HPLC-ABTS was mainly developed for screening the antioxidants of flavonoids from Astragali Radix (AR), and comparing the antioxidant capacity between traditional decoction pieces (TDP) of AR and cell wall-broken decoction pieces (CDP) of AR. The experimental results showed that the overall antioxidant capacity of CDP of AR was about twice as much as that of TDP of AR, which was specifically expressed as the antioxidant capacity of the screened antioxidants extracted from CDP was equivalent to 1.9–5.1 times that of those extracted from TDP, and three antioxidants were successfully screened, which were calycosin-7-O-β-D-glucoside, calycosin, and formononetin. The method established in this study is characterized by high efficiency and accuracy, which can simultaneously accomplish the screening of antioxidant components and the comparison of antioxidant capacity between samples, and provides a new method for the quality evaluation of AR from the perspective of antioxidant activity.

In the present study, a simple and efficient approach based on the online extraction-high performance liquid chromatography coupled with ABTS antioxidant assay (OLE-HPLC-ABTS) was established to quickly and directly analyze the antioxidants in S. baumii. Through this system, the HPLC mobile phase via a guard column packed with a S. baumii sample was used for online extraction (OLE). The separation was performed on an Agilent Poroshell EC-C18 column with a gradient elution using 0.1% formic acid (A) and 0.1% formic acid–acetonitrile (B) as mobile phase systems and detected at a wavelength of 254 nm. Then, the separated compounds were reacted with the antioxidant solution (ABTS), and the response was recorded at a wavelength of 400 nm. The developed analytical method was successfully applied to S. baumii samples, and eight antioxidants were identified. The established system integrated the online extraction, separation and online antioxidant detection, which is rapid, efficient, and suitable for the rapid screening of antioxidant compounds from solid sample mixtures.

This study developed an online HPLC-ABTS method to screen antioxidant phenolic acids in Pandani tectorii radix et rhizoma (PT) from 20 geographical origins. Six common constituents were screened and assigned by standards: neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, and isochlorogenic acid C. Using these six components as variables, four regression models were constructed to predict ABTS radical scavenging rate: support vector regression (SVR), decision tree regression, K-nearest neighbors (KNN) regression, and extreme gradient boosting (XGBoost) regression. From 80 samples, 64 were used for model development, and 16 were reserved for hold-out validation. Model selection employed 5-fold, nested, and leave-one-out cross-validations, complemented by permutation tests and learning curve analysis. The SVR model performed best, with nested CV R 2 = 0.9298, RMSE = 4.1207, MAE = 2.2313, and an optimism gap of 0.0137. After final tuning (C = 100, gamma = 0.1, and epsilon = 0.5), it achieved R 2 = 0.9000, RMSE = 4.0602, and MAE = 2.2986 on the hold-out set. This efficient and accurate method enables simultaneous screening of antioxidant constituents and prediction of radical scavenging activity, providing a new technical tool and scientific basis for PT quality evaluation.
