The effect of non-thermal (HPP and semi-HPP-CO 2 ) and thermal (flash pasteurization, FP) treatments on phytonutrients of flowable smoothie prepared from quadrable vegetable blends (FQVS) was investigated using multidimensional methods. First, FQVS gained an acceptability sensorial index (85.7%) compared with other formulas. FQVS/semi-HPP-CO 2 showed a greater microbial stability during storage (0–30 d) compared to HPP and FP. Fructose and glucose highly declined than sucrose in all smoothies, where semi-HPP-CO 2 steadily declined this reduction during storage. LC/MS-MS analysis showed that semi-HPP-CO 2 preserved most of FQVS's phytonutrients and their antioxidant effects measured by ORAC and oxidative enzymes inhibition assays. Semi-HPP-CO 2 acquired the lowest apparent viscosity among different FQVS smoothies, showing its post-processing flowability behavior. Most importantly, semi-HPP-CO 2 predicted a reduced power consumption for HPP and reduced the gas emission. In conclusion, blending different vegetables assisted with semi-HPP-CO 2 could be a novel approach to produce storage-stable smoothies with adequate amounts of phytonutrients and sensorial scores.

“High-pressure processing (HPP) plus” combined technologies are applied to overcome the limitation of single HPP and to produce juices with more stable quality during storage. This research explored the potential of HPP in combination with centrifugation to produce cloud stable orange juice during refrigerated storage. The results indicated that HPP combined processing technology significantly improved the cloud stability of orange juice, which was related to removed large particles, reduced viscosity, decreased protein contents, and inactivated pectin methylesterase activity induced by centrifugation ( P < 0.05). Besides, chelator solubilized pectin (CSP) decreased but water solubilized pectin (WSP) maintained in the juice after centrifugation. During storage, the conversion of pectin fraction from WSP to CSP, resulting in sedimentation appeared in centrifugation treated orange juice when stored for 28 days. In general, pectin characteristics changes and pectin fractions conversion were the main driving forces affecting cloud stability of orange juice pasteurized by HPP in combination with centrifugation and during chilled storage.

The cyclonic eddy uplifts nutrient-rich seawater into the euphotic zone, typically directly enhancing phytoplankton abundance and primary production. However, its impact on heterotrophic prokaryotic production (HPP) remains unclear due to the complex interplay of multiple indirect factors governing this process. Here, we conducted a comprehensive investigation of the distribution of picophytoplankton and heterotrophic prokaryotes, prokaryotic community structure, and HPP within a cyclonic eddy in the western North Pacific subtropical gyre. The results indicated that despite the higher abundance of picophytoplankton accompanied by nutrient upwelling at the eddy core compared to the edge, higher levels of HPP were observed at the eddy edge between 100 and 200 m, consistent with the distribution of the low nucleic acid content (LNA) prokaryotes. The significant positive correlation between HPP and the proportion of LNA group in total heterotrophic prokaryotes suggested a primary contribution from the LNA group over the high nucleic acid content (HNA) group. SAR11, a typical member of the LNA group, may primarily contribute to the elevated HPP observed at the eddy edge. The changes in temperature, nutrients, and light intensity induced by the cyclonic eddy may significantly influence the distribution and activity of HNA and LNA groups, potentially exerting a greater impact on HPP compared to phytoplankton-related factors. These findings contribute to understanding the underlying mechanisms of HPP responses to cyclonic eddies in the oligotrophic open ocean. Key Points Higher heterotrophic prokaryotic production (HPP) occurred at the eddy edge, while picophytoplankton predominantly occurred in the core The elevated HPP at the eddy edge was attributed to low nucleic acid prokaryotes, such as SAR11 The altered temperature, nutrients, and light intensity may impact the distribution and HPP in the eddy Plain Language Summary Mesoscale eddies are common features of global ocean circulation. Cyclonic eddies uplift isopycnals at their centers, bringing cold, nutrient-rich waters from the lower layers to the surface within the eddy, thereby promoting higher phytoplanktonic abundance and primary production typically observed at the eddy core. However, the response of heterotrophic prokaryotic distribution and heterotrophic prokaryotic production (HPP) to the upwelling in the cyclonic eddy remains unclear, as limited studies have not yet reached a consensus. We tracked a cyclonic eddy in the western North Pacific subtropical gyre to compare prokaryotic abundance, community composition, and HPP between eddy core and edge. The results indicate that, while picophytoplankton abundance was higher at the eddy core compared to the edge, HPP was predominantly elevated at the edge in response to the cyclonic eddy. The low nucleic acid content (LNA) prokaryotes dominated by SAR11 may significantly contribute to the increased HPP, as opposed to the traditionally assumed high nucleic acid content (HNA) group. The main factors influencing the distribution of LNA and HNA groups, as well as HPP, include temperature, nutrients, and light intensity altered by the eddy. This study advances the understanding of heterotrophic prokaryotic responses to cyclonic eddy dynamics.

The discovery and application of analyte-triggered fluorophore generation or fluorogenic reaction are significant and beneficial to the development of novel fluorescence (FL) analysis method. In this study, for the first time, we have reported a fluorogenic reaction to prepare fluorescent silicon-containing polymer dots (Si-PDs) by simply mixing N -[3-(trimethoxysilyl)propyl]ethylenediamine (DAMO) and hydroquinone (HQ) in aqueous solution at ambient temperature. Inspired by the alkaline phosphatase (ALP)-catalyzed hydrolysis of the substrate sodium 4-hydroxyphenyl phosphate (4-HPP) into HQ and the resultant HQ-controlled intense green Si-PDs generation, we have established a straightforward ALP activity assay by innovatively employing commercially available 4-HPP as the substrate. More significantly, the specific preparation method, clear formation mechanism and excellent performance enable the Si-PDs as well as its generation process to develop facile and attractive FL immunoassay. With the help of the universal ALP-based enzyme-linked immunosorbent assay (ELISA) platform and corresponding antibody, a convenient and conceptual ALP-based fluorescent ELISA has been constructed and applied in sensing cardiac troponin I (cTnI), a well-known biomarker of acute myocardial infarction. Our research via in situ formation of fluorescent nanomaterials has great potential application in ALP activity assay, inhibitor screening, and disease diagnosis.
