Proteomic analysis suggests that monoterpenes in lemongrass disrupt Ca2+ homeostasis in Haemaphysalis longicornis leading to mitochondrial depolarization and cytotoxicity

Desmond O. Agwunobi,Mengxue Li,Ningmei Wang,Guomin Chang,Xiaojing Zhang,Xiaomin Xue,Zhijun Yu,Hui Wang,Jingze Liu

Complex mixtures of bioactive ingredients in plant essential oils present complex chemistries which involve different modes of action. An increasing body of scientific reports has recently focused on the acaricidal activities of plant essential oils attributed to their monoterpene components, but information about their underlying molecular mechanism of action is scarce. Here, after the chemical analysis of lemongrass oil, a proteomic analysis of the ovary, salivary gland, and midgut of Haemaphysalis longicornis exposed to Cymbopogon citratus (lemongrass) essential oil was performed via data-independent acquisition mass spectrometry (DIA-MS) technology to further elucidate the molecular mechanisms involved. Pathway analysis reveals the activation of metabolic pathways mediated by oxidoreductases and transferases. Furthermore, the upregulation of various calcium-associated proteins and the upregulation of cytochrome c1, cytochrome c oxidase polypeptide IV, and programmed cell death protein 6-like isoform X1 suggest a cytotoxic mode of action via the formation of reactive oxygen species (ROS), mitochondrial Ca 2+ overload, mitochondrial uncoupling, and depolarization, and ATP depletion leading to either apoptotic or necrotic death. Morphological alterations observed after the RNAi of a major detoxification enzyme (glutathione S-transferase) merit further investigation. Hence, the cytotoxic mode of action exhibited by C. citratus oil could be vital for the development of eco-friendly acaricide.

Acaricidal activity of Cinnamomum cassia (Chinese cinnamon) against the tick Haemaphysalis longicornis is linked to its content of (E)-cinnamaldehyde

Nwanade Chuks F.,Wang Min,Wang Tianhong,Zhang Xiaoyu,Wang Can,Yu Zhijun,Liu Jingze

Background The tick Haemaphysalis longicornis (Neumann) is a well-known vector of numerous pathogens of veterinary and medical importance. Various control strategies, including the use of synthetic pesticides, have been developed to control this tick species. However, demand for effective and safe alternative pesticides is increasing due to the adverse effects associated with the intensive and injudicious use of synthetic pesticides, which include undesirable effects on non-target species and environmental pollution. Hence, the acaricidal activity of the extract and the essential oil of Cinnamomum cassia (Chinese cinnamon) and their major components, and the underlying mechanisms of this activity, were evaluated against unfed larvae and nymphs of H . longicornis . Methods The components of the extract and essential oil of C. cassia were determined by gas chromatography-mass spectrometry, and their larvicidal and nymphicidal activity were evaluated using the larval and nymphal packet test. The underlying detoxification mechanism was elucidated by targeting in vivo esterase and monooxygenase activity, and the toxicological effect was assessed on non-target Tenebrio molitor and Harmonia axyridis by topical application in open Petri dishes. Results ( E )-cinnamaldehyde was the predominant component of the extract (50.79%) and essential oil (89.95%). The 50% lethal concentration (LC 50 ) for larvae and nymphs treated with the extract was 11.56 and 49.18 mg/mL, respectively. The essential oil, ( E )-cinnamaldehyde and fenvalerate exhibited acaricidal activity, with LC 50 values of 3.81, 3.15, and 0.14 mg/mL, respectively, against the larvae, and 21.31, 16.93, and 1.89 mg/mL, respectively, against the nymphs. ( E )-cinnamaldehyde significantly increased esterase and monooxygenase activity in both larvae and nymphs. Unlike fenvalerate, C . cassia essential oil and ( E )-cinnamaldehyde did not cause mortality of T . molitor or H . axyridis adults. Conclusions This study demonstrates that C . cassia essential oil and ( E )-cinnamaldehyde have the potential to be developed into botanical-based larvicidal and nymphicidal agents for tick control. Symbiont-regulated serotonin biosynthesis modulates tick feeding activity

Zhengwei Zhong,Ting Zhong,Yeqing Peng,Xiaofeng Zhou,Zhiqian Wang,Huiru Tang,Jingwen Wang

Summary Ticks are obligate hematophagous arthropods. Blood feeding ensures that ticks obtain nutrients essential for their survival, development, and reproduction while providing routes for pathogen transmission. However, the effectors that determine tick feeding activities remain poorly understood. Here, we demonstrate that reduced abundance of the symbiont Coxiella (CHI) in Haemaphysalis longicornis decreases blood intake. Providing tetracycline-treated ticks with the CHI-derived tryptophan precursor chorismate, tryptophan, or 5-hydroxytryptamine (5-HT; serotonin) restores the feeding defect. Mechanistically, CHI-derived chorismate increases tick 5-HT biosynthesis by stimulating the expression of aromatic amino acid decarboxylase (AAAD), which catalyzes the decarboxylation of 5-hydroxytryptophan (5-HTP) to 5-HT. The increased level of 5-HT in the synganglion and midgut promotes tick feeding. Inhibition of CHI chorismate biosynthesis by treating the colonized tick with the herbicide glyphosate suppresses blood-feeding behavior. Taken together, our results demonstrate an important function of the endosymbiont Coxiella in the regulation of tick 5-HT biosynthesis and feeding.

Individual and synergistic toxicity of cinnamon essential oil constituents against Haemaphysalis longicornis (Acari: Ixodidae) and their potential effects on non-target organisms

Chuks Fidelis Nwanade,Min Wang,Hongxia Li,Abolfazl Masoudi,Zhijun Yu,Jingze Liu

Plant essential oils (EOs) are usually complex mixtures of volatile compounds, and these compounds can vary in composition as a result of biotic and abiotic factors. Thus, understanding the contribution of the individual compounds particularly their interactions with respect to acaricidal activity is important for the use of EOs as an alternative to synthetic acaricides . Herein, the acaricidal activity of constituents from EOs of cinnamon ( Cinnamomum zeylanicum and Cinnamomum cassia ) against Haemaphysalis longicornis was investigated using the nymphal and adult immersion tests. Among the test compounds, benzyl benzoate showed the highest toxicity against the nymphs (LC 50 = 24.41 µL/mL) and adults (LC 50 = 3.99 µL/mL). All combinations at their sublethal concentration (LC 30 ) exhibited synergy. The binary mixture of benzyl benzoate +  cinnamaldehyde presented the highest synergy ratio (3.42) while tertiary mixtures of cinnamaldehyde +  eugenol  +  linalool showed the least synergy ratio (1.20). Enzyme assays revealed that benzyl benzoate significantly inhibited AChE activity ( p  < 0.05). The ecotoxicological assessments of benzyl benzoate and cinnamaldehyde indicate likely minimal effects on non-target terrestrial invertebrate, Tenebrio molitor . On the other hand, they were toxic to non-target aquatic species , Daphnia magna . Based on our results, the acaricidal activity of cinnamon EO against H . longicornis may be linked to the content of benzyl benzoate whose presence may be synergistic to the activity of other compounds.

Biochemical and molecular mechanisms involved in the response of Haemaphysalis longicornis (Acari: Ixodidae) to Cinnamomum cassia essential oil and its major constituent

Nwanade Chuks Fidelis,Wang Min,Yu Zhijun,Liu Jingze

To develop an appropriate program for essential oils (EOs) used in pest control, it is important to acquire information about the effect of the EO when used at a low concentration. Previously, we demonstrated the enzymatic activity of Haemaphysalis longicornis (the main livestock pest in Australia and New Zealand) exposed to Cinnamomum cassia EO and ( E )-cinnamaldehyde (its major compound) when applied at 50% lethal concentration (LC 50 ). However, H . longicornis respond to toxic substances via different mechanisms and at different times of exposure. Herein, the enzymes involved in the defense response were studied by monitoring glutathione-S-transferase, acetylcholinesterase, and Na + /K + -adenosine triphosphatases (ATPase) activities in H . longicornis larvae exposed to C . cassia EO and ( E )-cinnamaldehyde across different times. Further, the molecular changes related to the defense mechanisms were studied by monitoring adenosine triphosphate (ATP)-binding cassette (ABCB10) gene expression. In addition, we investigated the acaricidal activity of the test substances against the engorged larvae and nymphs of H . longicornis . Our results showed that the activities of these enzymes were significantly affected by the EO and/or ( E )-cinnamaldehyde depending on the exposure time. We observed up-regulation of the ABCB10 gene in the larvae. Also, our results revealed that C . cassia EO (LC 50  = 1.46 and 16.69 mg/mL) and ( E )-cinnamaldehyde (LC 50  = 3.86 and 17.96 mg/mL) were effective against the engorged larvae and nymphs, respectively. Our results provided a deeper understanding of the defense mechanisms of H . longicornis, which may provide an important clue for the development of EO-based acaricides that are cheaper and environmentally friendly.

The acaricidal activity and enzymatic targets of the essential oils of Cinnamomum cassia and Cinnamomum camphora and their major components against Haemaphysalis longicornis (Acari: Ixodidae)

Lingqian Bai,Zhihua Gao,Xiaofeng Xu,Wenxia Lv,Yikui Wang,Kexin Dong,Zhijun Yu,Xiaolong Yang

Haemaphysalis longicornis is a notorious vector tick species widely distributed in China, with its current control relying heavily on synthetic chemicals. The disadvantages of chemical acaricides including single mechanism of action, environmental pollution, and easy-to-induce acaricidal resistance have long been criticized. As plant extracts and essential oils (EOs) have been proven to show good insecticidal activity and environmental safety, hence, in this study, the chemical constituents of EOs from Cinnamomum cassia and Cinnamomum camphora were analyzed and their potential for application in the control of H. longicornis was explored. Gas chromatography–mass spectrometry analysis showed that the Trans -cinnamaldehyde and Diethyl phthalate were the main components of the EOs of C. cassia and C. camphora , respectively. Larval packet test and nymph immersion test showed that the EOs of C. cassia and C. camphora had significant acaricidal activity against unfed larvae and nymphs of H. longicornis , and the EO of C. cassia showed more toxic against unfed adults than C. camphora EO at the same concentration. Enzyme assays revealed that the C. cassia EO significantly inhibited the activity of acetylcholinesterase (AchE), and C. camphora EO and Trans -cinnamaldehyde had no significant effect on AchE. Cinnamomum cassia EO and C. camphora EO significantly inhibited the activity of carboxylesterase (CarE). Trans -cinnamaldehyde significantly inhibited the activity of catalase (CAT), and C. cassia EO and C. camphora EO had no obvious effect. The above results suggested that the EOs of C. cassia and C. camphora and their major components target different enzymes, which may potentially mitigate the acaricidal resistance development in tick, indicating the prospect that these EOs may become an effective substitute for chemical synthetic acaricides in tick control.
