Plant volatiles mediate the interactions within plant-insect communities and influence the distribution and behavior of insects. Natural enemies often utilize the volatiles that released from plants to locate the profitable patches for foraging. The lady beetle Harmonia axyridis (Pallas) is a dominant generalist predator of various pests in agriculture fields, yet little is known about the chemical cues which the predator employs to locate its prey. In this study, we tested the attractiveness of volatiles emanating from healthy and aphid-infested insectary plant Cnidium monnieri (L.) Cuss. to H. axyridis adults in a Y-tube olfactometer. Next, using coupled gas chromatography – electroantennogram detection (GC-EAD) and gas chromatography – mass spectrometry (GC–MS) identified the volatile components which elicited electrophysiological activity in H. axyridis adults. Lastly, Y-tube olfactometer tests in laboratory and field traps were used to verify the effectiveness of the volatile components. In olfactometer trials, H. axyridis adults preferred healthy and aphid-infested plants versus control. In GC–MS and GC-EAD trials, two antenna-active components (1,2-diethylbenzene and p-diethylbenzene) were detected among the volatile blends emanating from healthy and aphid-infested plants; and in ensuing behavioral assays, H. axyridis adults were attracted to two synthetic volatile compounds. Under field conditions, the number of H. axyridis adults as recorded in traps for two synthetic volatile components at three different concentrations (100 mg/ml, 10 mg/ml, and 1 mg/ml) were significantly higher than those for mineral oil (control). These findings could provide an effective means for the cultivation of insectary plants to develop natural enemy attractants and support the biological control in the field.

BACKGROUND Harmonia axyridis Pallas (Coleoptera: Coccinellidae) is an important natural enemy of aphids. Plant species and plant health conditions can affect the behavior of H. axyridis . To determine plant effects on this lady beetle, we examined beetle responses to four cover crops: coriander ( Coriadrum sativum L., Apiales: Apiaceae), marigold ( Tagetes erecta L., Asterales: Asteraceae), sweet alyssum ( Lobularia maritima L., Brassicales: Brassicaceae), and alfalfa ( Medicago sativa L . , Fabales: Fabaceae). Our goal was to better understand this predator's ovipositional behavior in response to different plants and its olfactory response to the aphid-induced volatiles from these plants. RESULTS We found that this lady beetle did not have any significant oviposition preference among the four plant species, but H. axyridis preferred to lay eggs on the lower surface of leaves, regardless of the plant species. H. axyridis females had a significant preference for aphid-infested marigolds, but were not attracted by any of the other three cover plants or marigolds without aphid damage. Compared to the uninfested marigold plants, the emission of 12 compounds significantly increased on the aphid-infested marigolds, and two of them were attractive to H. axyridis under suitable concentrations. CONCLUSION H. axyridis did not show any significant oviposition preference among the four cover crops. Aphid-infested marigolds can attract H. axyridis . Indole and terpinen-4-ol mediated lady beetle attraction. These synomones have potential for manipulating populations of H. axyridis as a component of conservation biological control. © 2022 Society of Chemical Industry.

Simple SummaryTheHarmonia axyridisis an important natural enemy insect that has received widespread attention in research and the application of biological pest control. Currently, suitable artificial feed for the mass-rearing of theH. axyridishas not been found, which affects the application of this natural enemy in biological control. This study found that the egg production of artificial feed without added egg yolk is similar to that of artificial feed without added egg yolk supplemented with juvenile hormone III. Compared to the control artificial feed, the egg production significantly increased, which is of great significance for further exploring the effects of non-insect source artificial feed on the growth and development of theH. axyridis.Artificial diets applied in the mass-rearing propagation ofH. axyridiscan improve reproductive ability by optimizing the feeding formula. This study used transcriptome data to investigate the effects of various artificial diets on the growth and development ofH. axyridis. Results indicate that spawning increased with the low-fat and JH III-supplemented artificial diet (Diet 3). Furthermore, the highest glycogen content found in Diet 3 was significantly different from the other two groups. Triglyceride content decreased as adult feeding time increased in the three artificial diet groups, with the fastest decrease observed in the low-fat diet (Diet 2). Protein content increased gradually in the high-fat diet (Diet 1) group compared to the other treatment groups. The adults reared on low-fat artificial diets, when compared to those on artificial diets supplemented with juvenile hormones at the transcriptome level, were found to have upregulated genes enriched in ubiquitin-mediated proteolysis, ribosome biogenesis, and the hedgehog signaling pathway. In contrast, the genes upregulated in the latter group were enriched in oxidative phosphorylation, amino acid biosynthesis, and the metabolism of other amino acids. The results suggest that nutritional status significantly affects the growth and development ofH. axyridisand has practical implications for the artificial feeding of natural pest enemies.

Many animals, including insects, exhibit plasticity of body colour in response to environmental changes. Varied expression of carotenoids, major cuticle pigments, significantly contributes to body colour flexibility. However, the molecular mechanisms by which environmental cues regulate carotenoid expression remain largely unknown. In this study, we used the ladybird Harmonia axyridis as a model to investigate the photoperiodic-responsive plasticity of elytra coloration and its endocrine regulation. It was found that H. axyridis females under long-day conditions develop elytra that are much redder than those under short-day conditions, resulting from the differential accumulation of carotenoids. Exogenous hormone application and RNAi-mediated gene knockdown indicate that carotenoid deposition was directed through the juvenile hormone (JH) receptor-mediated canonical pathway. Moreover, we characterized an SR-BI/CD36 ( SCRB ) gene SCRB10 as the carotenoid transporter responding to JH signalling and regulating the elytra coloration plasticity. Taken together, we propose that JH signalling transcriptionally regulates the carotenoid transporter gene for the photoperiodic coloration plasticity of elytra in the beetles, which reveals a novel role of the endocrine system in the regulation of carotenoid-associated animal body coloration under environmental stimuli.

Aphids are among the most destructive phytophagous pests of host plants, because of their rapid reproduction, parthenogenesis, extensive crop damage, and the transmission of many plant viruses. Since lady beetles are important predatory natural enemies of aphids, developing lady beetle attractants to increase their field abundance is vital for aphid control. Floral volatiles and other semiochemicals are reportedly attractive to lady beetles. In this research, a total of 58 floral volatiles were tested by Y-tube olfactometer assays, among which 29 were highly attractive to both Harmonia axyridis (Coleoptera: Coccinellidae) and Coccinella septempunctata (Coleoptera: Coccinellidae). Meanwhile, the results of wind tunnel trials showed that only isoamyl acetate, α -humulene, trans -3-hexen-1-ol, methyl salicylate, and β -pinene lure these two species. Thereafter, 15 semiochemicals from pests, natural enemies, and pest-infested crops were mixed with the selected floral volatiles, to determine optimum formulations for attracting lady beetles through wind tunnels and further field trials. Eventually, formulas (1) α -humulene: β -pinene: methyl salicylate: trans -3-hexen-1-ol = 1:3:3:10; (5) α -humulene: β -pinene: methyl salicylate:1-nonanal = 1:3:3:10; (8) α -humulene: β -pinene: methyl salicylate: indole = 1:3:3:10 ( 1 , 5 , and 8 are labels used for the various formulations in field trials) were successfully verified to be attractive to lady beetles in both pumpkin and wheat fields. These formulations will be of great significance in developing integrated pest management strategies for aphid control.

Background The tick Haemaphysalis longicornis (Neumann) is a well-known vector of numerous pathogens of veterinary and medical importance. Various control strategies, including the use of synthetic pesticides, have been developed to control this tick species. However, demand for effective and safe alternative pesticides is increasing due to the adverse effects associated with the intensive and injudicious use of synthetic pesticides, which include undesirable effects on non-target species and environmental pollution. Hence, the acaricidal activity of the extract and the essential oil of Cinnamomum cassia (Chinese cinnamon) and their major components, and the underlying mechanisms of this activity, were evaluated against unfed larvae and nymphs of H . longicornis . Methods The components of the extract and essential oil of C. cassia were determined by gas chromatography-mass spectrometry, and their larvicidal and nymphicidal activity were evaluated using the larval and nymphal packet test. The underlying detoxification mechanism was elucidated by targeting in vivo esterase and monooxygenase activity, and the toxicological effect was assessed on non-target Tenebrio molitor and Harmonia axyridis by topical application in open Petri dishes. Results ( E )-cinnamaldehyde was the predominant component of the extract (50.79%) and essential oil (89.95%). The 50% lethal concentration (LC 50 ) for larvae and nymphs treated with the extract was 11.56 and 49.18 mg/mL, respectively. The essential oil, ( E )-cinnamaldehyde and fenvalerate exhibited acaricidal activity, with LC 50 values of 3.81, 3.15, and 0.14 mg/mL, respectively, against the larvae, and 21.31, 16.93, and 1.89 mg/mL, respectively, against the nymphs. ( E )-cinnamaldehyde significantly increased esterase and monooxygenase activity in both larvae and nymphs. Unlike fenvalerate, C . cassia essential oil and ( E )-cinnamaldehyde did not cause mortality of T . molitor or H . axyridis adults. Conclusions This study demonstrates that C . cassia essential oil and ( E )-cinnamaldehyde have the potential to be developed into botanical-based larvicidal and nymphicidal agents for tick control.
