Preliminary enrichment and separation of chlorogenic acid from Helianthus tuberosus L. leaves extract by macroporous resins

Peng-Cheng Sun,Ying Liu,Yue-Tao Yi,Hong-Juan Li,Ping Fan,Chuan-Hai Xia

In the present study, a simple and efficient method for the preparative separation of 3-CQA from the extract of Helianthus tuberosus leaves with macroporous resins was studied. ADS-21 showed much higher adsorption capacity and better adsorption/desorption properties for 3-CQA among the tested resins. The adsorption of 3-CQA on ADS-21 resin at 25   °C was fitted best to the Langmuir isotherm model and pseudo-second-order kinetic model. Dynamic adsorption/desorption experiments were carried out in a glass column packed with ADS-21 to optimise the separation process of 3-CQA from H. tuberosus leaves extract. After one treatment with ADS-21, the content of 3-CQA in the product was increased 5.42-fold, from 12.0% to 65.2%, with a recovery yield of 89.4%. The results demonstrated that the method was suitable for large-scale separation and manufacture of 3-CQA from H. tuberosus leaves.

Phenolics and antifungal activities analysis in industrial crop Jerusalem artichoke (Helianthus tuberosus L.) leaves

Fujia Chen,Xiaohua Long,Mengni Yu,Zhaopu Liu,Ling Liu,Hongbo Shao

Spoilage of industrial fruits and vegetables in storage and transportation due to fungal infection results in significant losses, and new natural antifungal treatments would have a large economic value. The extracts of antifungal compounds and phenolic acids from Jerusalem artichoke ( Helianthus tuberosus L.) leaves were investigated for potential use in enhancing preservation of fruits and vegetables in storage. Either crude leaf extract or n -butanol fraction was active against Botrytis cinerea, Colletotrichum gloeosporioides, Phytophthora capsici Leonian and Rhizoctonia cerealis , with the values of IC 50 (half of the maximum inhibitory concentration) ranging from 2.166 to 2.534   g   L −1 for the crude leaf extract and 0.232–1.911   g   L −1 for n -butanol fraction. The severity of grey mould caused by B. cinerea was significantly reduced by n -butanol fraction applied at 1 and 2   g   L −1 (the control efficiency of 71.3% and 77.8%, respectively, compared with commercial preparation Carbendazim . Following in vitro activity-guided fractionation by bioautography, six phenolic acids were separated from n -butanol fraction. Among them, caffeic acid, 3,4-dicaffeoylquinic acid and 1,5-dicaffeoylquinic acid played a dominant role and were active in bioassays against Gibberella zeae , with respective minimum inhibitory concentrations (MIC) being 108, 60 and 4.2   μg   mL −1 . These results imply that Jerusalem artichoke leaves might be a potential source of natural fungicides.

Evaluation of Antifungal Phenolics from Helianthus tuberosus L. Leaves against Phytophthora capsici Leonian by Chemometric Analysis

Fu-Jia Chen,Xiao-Hua Long,En-Zhong Li

Phytophthora capsiciLeonian causes destructive economical losses in pepper production, and a promising source of natural fungicides-Helianthus tuberosusleaves was reported. The antifungal activities of different extracts and compounds fromH. tuberosusleaves against the phytopathogen,P. capsiciLeonian, were examined by chemometric analysis, including HPLC-MS/MS and multivariate data analyses. Principal component analysis and orthogonal partial least squares-discriminate analysis were applied to examine the four groups ofH. tuberosusleaves samples, including crude extracts obtained by different methods, including refluxing, macerating, and refluxing under vacuum; four fractions, namely, petroleum ether (PE), chloroform (Chl), ethyl acetate (EA), and n-butanol (NB) fractions; the samples of threeH. tuberosuscultivars; and the samples at three growth stages of cultivar Nan Yu. The phenolics contents were categorized based on 3,5-Dicaffeoylquinic acid (3,5-DiCQA), 1,5-Dicaffeoylquinic acid (1,5-DiCQA), 3-O-Caffeoylquinic acid (3-CQA), and 4,5-Dicaffeoylquinic acid (4,5-DiCQA), which were predominant in all the samples. Antifungal activity assay revealed that Chl and NB fractions were more active againstP. capsiciLeonian with lower IC50(half of maximal inhibitory concentration) values, whereas partial least squares-discriminate analysis suggested caffeoylquinic acid isomer(4-CQA), methyl-quercetin glycoside(MQG), and caffeic acid(CA) might be the main active components inH. tuberosusleaves againstP. capsiciLeonian. Furthermore, microscopic evaluation demonstrated structural deformities inP. capsiciLeonian treated with Chl and NB fractions, indicating the antifungal effects ofH. tuberosusleaves. These results imply thatH. tuberosusleaves with a high concentration of phenolics might be a promising source of natural fungicides. Keywords:Helianthus tuberosus;antifungal activity;phenolic acids;chemometric analysis;PLS-DA

Analysis of Phenolic Acids of Jerusalem Artichoke (Helianthus tuberosus L.) Responding to Salt-Stress by Liquid Chromatography/Tandem Mass Spectrometry

Fujia Chen,Xiaohua Long,Zhaopu Liu,Hongbo Shao,Ling Liu

Plant phenolics can have applications in pharmaceutical and other industries. To identify and quantify the phenolic compounds in Helianthus tuberosus leaves, qualitative analysis was performed by a reversed phase high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS) and quantitative analysis by HPLC. Ten chlorogenic acids (CGAs) were identified (3-o-caffeoylquinic acid, two isomers of caffeoylquinic acid, caffeic acid, p-coumaroyl-quinic acid, feruloylquinic acid, 3,4-dicaffeoyquinic acid, 3,5-dicaffeoylquinic acid, 1,5-dicaffeoylquinic acid, and 4,5-dicaffeoylquinic acid) by comparing their retention times, UV-Vis absorption spectra, and MS/MS spectra with standards. In addition, four other phenolic compounds, including caffeoyl glucopyranose, isorhamnetin glucoside, kaempferol glucuronide, and kaempferol-3-o-glucoside, were tentatively identified in Helianthus tuberosus leaves for the first time. The 3-o-caffeoylquinic acid (7.752 mg/g DW), 4,5-dicaffeoylquinic acid (5.633 mg/g DW), and 3,5-dicaffeoylquinic acid (4.900 mg/g DW) were the major phenolic compounds in leaves of Helianthus tuberosus cultivar NanYu in maturity. The variations in phenolic concentrations and proportions in Helianthus tuberosus leaves were influenced by genotype and plant growth stage. Cultivar NanYu had the highest concentration of phenolic compounds, in particular 3-o-caffeoylquinic acid and 4,5-dicaffeoylquinic acid compared with the other genotypes (wild accession and QingYu). Considering various growth stages, the concentration of total phenolics in cultivar NanYu was higher at flowering stage (5.270 mg/g DW) than at budding and tuber swelling stages. Cultivar NanYu of Helianthus tuberosus is a potential source of natural phenolics that may play an important role in the development of pharmaceuticals.

The Role of Inulin in Maintaining Antioxidant Capacity and Enzymatic Activities of Jerusalem Artichoke (Helianthus tuberosus L.) Cultivars During Cold Storage

Yuwen Mu,Bohua Zhang,Shiqi Lv,Fencan Li,Changming Zhao

Jerusalem artichoke ( Helianthus tuberosus L.) is valued for its high inulin content and adaptability to marginal lands. This study investigated the changes in inulin content, antioxidant capacity, polyphenol concentrations, and enzymatic activities of eight cultivars during 60 days of cold storage. Inulin levels ranged from 582.43 g/kg (LZJ006) to 809.70 g/kg (LZJ055), with LZJ047 maintaining the highest content throughout storage. The antioxidant potential, as measured by ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, declined across all cultivars, correlating with the reduction in inulin content. The polyphenol content varied significantly, with LZJ119 having 2.17 times more than LZJ010. POD activity increased, while catalase (CAT) and superoxide dismutase (SOD) activities fluctuated during the storage period. Hierarchical Cluster Analysis identified three distinct antioxidant clusters, revealing significant correlations between inulin content and key antioxidant parameters (CAT, FRAP, DPPH). These findings highlight the pivotal role of inulin in preserving the antioxidant system and bioactive properties of Jerusalem artichoke tubers during extended cold storage, providing valuable insights for post-harvest management and cultivar selection.
