This study aimed to evaluate the safety profile and beneficial effects of Heyndrickxia coagulans strain BC99 (BC99) for potential use in functional foods and pharmaceuticals. We began with whole genome sequencing of BC99, followed by a comprehensive safety assessment comprising genome analysis, hemolysis, cytotoxicity, antibiotic susceptibility tests, and cell adhesion and tolerance studies, along with acute and subacute oral toxicity studies in animal models. BC99 was isolated from a well-characterized collection originating from the feces of a healthy infant. Our results indicated no hemolytic activity on Columbia blood agar plates and broad antibiotic sensitivity, including to gentamicin, ampicillin, chloramphenicol, ciprofloxacin, and others. Cytotoxicity testing confirmed no adverse effects on HT-29 cells and significant adhesive properties to intestinal epithelial cells. Tolerance tests demonstrated over 90% viability of BC99 under simulated gastrointestinal conditions. In vivo studies in mice and rats confirmed the absence of adverse effects following oral administration. Collectively, these findings support BC99’s robust tolerance to gastrointestinal environments, strong adhesion capabilities, and a broad spectrum of antibiotic resistance, underlining its potential as a safe and effective agent for gut microbiota modulation and host health enhancement.

With the increasing global demand for protein sources to feed the ever-growing population, peanut meal—a by-product of peanut oil extraction—holds promise as a valuable feed ingredient for animal protein production. However, its utilization is constrained by anti-nutritional factors and limited digestibility. This study employed solid-state fermentation using Heyndrickxia coagulans (formerly Bacillus coagulans ) to enhance the nutritional quality of peanut meal. Among three evaluated strains, Heyndrickxia coagulans S17 was selected for its superior fermentation performance. Optimal fermentation conditions were established: 1 g/L of CaCl 2 , 45 °C, material-to-liquid ratio of 1:2.1, 14% inoculation, and 36-h fermentation. Under these conditions, crude protein, small peptides, and acid-soluble protein contents increased from 53.75%, 3.35%, and 1.80% to 60.11%, 8.93%, and 5.37%, respectively. In vitro protein digestibility improved from 54.18% to 64.91%. Additionally, aflatoxin B 1 levels decreased from 43.87 μg/kg to 9.23 μg/kg, while trypsin inhibitors and phytic acid were reduced from 2.97 mg/g to 0.56 mg/g and 14.36 g/kg to 2.57 g/kg, respectively. These findings suggest that solid-state fermentation with Heyndrickxia coagulans S17 effectively enhances the nutritional quality of peanut meal, offering a viable approach for producing high-quality protein feed and contributing to sustainable livestock nutrition.

Hyperuricemia (HUA) and gout are primarily caused by excessive uric acid production resulting from the catabolism of purine nucleosides, notably inosine and guanosine. In this study, a candidate probiotic strain capable of biodegrading nucleosides (both inosine and guanosine) was successfully isolated fromJiaosuand identified asHeyndrickxia coagulansNJ01 by average nucleotide identity analysis. In vitro assays showed 100% biodegradation of 1.0 g/L of both inosine and guanosine by NJ01 cells and its cell-free extracts within 24 h. HPLC and genomic analyses confirmed that inosine and guanosine were biodegraded by purine-nucleoside phosphorylase into hypoxanthine and guanine, which are terminal metabolites not further degradable by the strain, and the key gene was confirmed by PCR. Furthermore, evaluation of the probiotic properties and safety of NJ01 at both genotypic and phenotypic levels confirmed that this strain is a promising probiotic candidate for further in vivo validation. This study provides a crucial foundation for the potential application ofH. coagulansNJ01, a strain capable of biodegrading uric acid precursors, in the development of functional foods for HUA management.
