Flash Extraction, Characterization, and Immunoenhancement Activity of Polysaccharide from Hippophae rhamnoides Linn

Yunwen Zhu,Meifu Wu,Xue Li,Yahong Wang,Mei Li,Hongli Zhou

Hippophae rhamnoides L. polysaccharide was optimized with flash extraction by response surface design. The optimum process conditions were: rotation rate 5000 r/min, extraction time 15 s, extraction temperature 90 °C and liquid-to-material ratio 38 mL/g, the extraction yield was 15.28±0.02 %. HRP-1 and HRP-2 obtained by 40 % and 60 % graded alcohol precipitation were characterized. The results indicated that HRP-1 and HRP-2 both composed of mannose, ribose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, xylose, arabinose with different molar ratio and the molecular weights were 380.59 kDa and 288.24 kDa, respectively. In addition, the in vitro antioxidant and immunoenhancement activities of HRP-1 and HRP-2 were analyzed, and the two fractions showed good free radical scavenging activity against ⋅OH, ABTS⋅ + , DPPH⋅, and extremely strong immunomodulatory activity against RAW264.7 cells. Indicating that flash extraction is suitable for extraction of HRP, the structural study of HRP provides a scientific theoretical basis for the development of Hippophae rhamnoides .

Ultrasound-assisted natural deep eutectic solvents extraction of flavonoids from Hippophae rhamnoides: Optimization and kinetics studies

Chenlu Han,Ziwei Yang,Xiaoqiang Chen,Lin Yan,Ying Zhang

Flavonoids are important bioactive components in Hippophae rhamnoides . In this study, flavonoids were extracted from H. rhamnoides leaves with an ultrasonic-assisted natural deep eutectic solvent. The optimal conditions included a molar ratio of 1:3, a liquid–solid ratio of 40 mL/g, and an extraction time of 30 min, and the extraction yield was 18.70 % ± 0.25 %. A model of the kinetics was developed to describe the extraction of flavonoids using Fick's law as the basis, where the rate constant and diffusion coefficient increased with the temperature, highlighting the pronounced influence of the temperature on the extraction efficiency. Following simulated digestion, the bioaccessibility index showed that 78.21 % of the flavonoids could be absorbed by tissues. These results will be beneficial for achieving the efficient extraction of flavonoids and the full exploitation and utilization of sea buckthorn resources.

Fabrication of Nanosuspensions to Improve the Oral Bioavailability of Total Flavones from Hippophae rhamnoides L. and their Comparison with an Inclusion Complex

Tian Rui,Wang Hui,Xiao Yi,Hu Pengyue,Du Ruofei,Shi Xiufeng,Wang Zhengtao,Xie Yan

The aim of this work was to increase the solubility and oral bioavailability of isorhamnetin, kaempferol, and quercetin in the total flavones of Hippophae rhamnoides L. (TFH) by preparing their nanosuspensions (NSs) and an inclusion complex. Based on the particle size and zeta potential, P407, Soluplus, SDS, PEG-6000, and HP-β-CD were selected as stabilizers. TFH NSs and a TFH/HP-β-CD inclusion complex were prepared, and their morphology, crystallinity, molecular interactions, and cytotoxicity were investigated. Furthermore, the saturation solubility, dissolution, and pharmacokinetics of the three flavonoids in the TFH, TFH NSs, and TFH/HP-β-CD inclusion complex were compared. The five obtained TFH NSs were physically stable, and their particle sizes were all below 200 nm. The solubility and dissolution of the three active components were obviously enhanced by the formation of the TFH NSs and TFH/HP-β-CD inclusion complex. Correspondingly, the oral bioavailability of isorhamnetin, kaempferol, and quercetin increased up to 4.11-, 3.85-, and 6.73-fold, respectively, in the TFH NSs and 2.89-, 3.71-, and 9.51-fold, respectively, in the TFH/HP-β-CD inclusion complexes compared to those in the raw TFH. In brief, both NSs and inclusion complexes can improve the oral bioavailability of the three flavonoids in TFH. Taking the drug loading and the stable ratio of the multiple components into consideration, the NSs is a more promising strategy than the inclusion complex for increasing the oral bioavailability of multiple water-insoluble components in herbal extracts. Protective Effects of Hippophae rhamnoides L. Phenylpropanoids on Doxorubicin-Induced Cardiotoxicity in Zebrafish

Gang Li,Ming Chu,Yingying Tong,Yuexin Liang,Shenghui Wang,Chengjun Ma,Zhenhua Wang,Wenna Zhou

Hippophae rhamnoidesL. is a deciduous shrub that contains many unique bioactive substances. This sea buckthorn possesses anticancer, antioxidant, anti-inflammatory, and cardiovascular protective properties. Herein, the effects of phenylpropyl compounds extracted fromH. rhamnoidesL. on doxorubicin (Dox)-induced cardiotoxicity were evaluated in zebrafish. Cardiac injury in zebrafish was induced using 35 μM Dox for 96 h, and 30 μM phenylpropanoid compounds were used as the protective treatment. The cardioprotective effects and mechanisms of the four phenylpropanoids were investigated using microscopy, behavioral analysis, acridine orange staining, western blotting, flow cytometry, and real-time quantitative polymerase chain reaction. The extracted phenylpropanoids could significantly relieve Dox-induced cardiac injury in zebrafish and inhibit cardiomyocyte apoptosis. The mechanisms of action were mainly related to the stability of mitochondrial biogenesis and function maintained by phenylpropanoids in zebrafish. To our knowledge, this is the first report on the protective effect of sea buckthorn against myocardial injury in zebrafish. Our findings provide support for the further research and development of sea buckthorn and its components. Keywords:sea buckthorn;zebrafish;doxorubicin;cardiotoxicity;mitochondria

Chen Shen,Teng Wang,Feng Guo,Kunlai Sun,Bin Wang,Jie Wang,Zefeng Zhang,Xing Zhang,Yuqin Zhao,Yin Chen

The sea buckthorn ( Hippophae rhamnoides L.) berries are rich in various bioactive components and widely used as fruit and traditional medicine. In this study, a novel heteropolysaccharide fraction (SP0.1-1) was isolated from Sea buckthorn berries. SP0.1-1 is composed of mannose , glucose, galactose , and arabinose in the molar ratio of 1:2.3:1.9:11.2 with a core structure containing 1,4-linked-α- d -Glc p , 1,4,6-linked-α- d -Glc p and 1,4-linked-α- d -Man p residues as the backbone. And the side-chains comprised of 1,3,5-linked-α- l -Ara f , 1,5-linked-α- l -Ara f , terminal α-Ara f and 1,4-linked-β- d -Gal p . Furthermore, a diet supplemented with SP0.1-1 extended the mean lifespan, enhanced antioxidant enzyme (superoxide dismutase, SOD; glutathione peroxidase , GSH-Px; and catalase , CAT) activities, and decreased the malondialdehyde (MDA) level and hydrogen peroxide (H 2 O 2 )-induced mortality rate in fruit flies ( Drosophila melanogaster ). To summarize, the study's findings will provide evidence for the development of sea buckthorn polysaccharide products.

Succinylation-driven structural modification of Hippophae rhamnoides L. protein enhances interfacial properties and emulsion performance

Cheng Xuehuan,Ge Shuya,Liu Wanlu,Zhang Junhang,Wang Bing,Blanchard Chris,Strappe Padraig,Zhou Zhongkai,Zhao Guohua

Sea buckthorn protein (SBP) is an underutilized plant protein with limited aqueous dispersibility and interfacial functionality. In this study, SBP was modified by succinylation, and the resulting changes in structural, physicochemical, and functional properties were systematically characterized. Succinylation markedly improved the dispersibility of SBP, as evidenced by an increase in solubility from 25.25% to 68.99%, a shift in zeta potential from − 33.82 to − 51.62 mV, and reduced particle aggregation. Structural analysis indicated that succinylation induced moderate conformational rearrangement, including decreased α-helix content, increased random coil content, and changes in the tertiary microenvironment. These changes enhanced the emulsifying performance of SBP, with the emulsifying activity index increasing from 25.02 to 35.71 m²/g. β-carotene-loaded emulsions stabilized by succinylated SBP showed smaller and more uniform droplets, with droplet size decreasing from 2085.47 to 700.53 nm, together with improved encapsulation efficiency and storage stability. During in vitro digestion, free fatty acid release increased from 20.92% to 26.02%, while β-carotene bioaccessibility increased from 10.17% to 27.67%. These results suggest that succinylation improves the functional performance of SBP through a charge-conformation-interface mechanism, thereby enhancing its potential as a plant-based emulsifier for β-carotene-loaded emulsion systems. Phenolic compounds, antioxidant activity and sensory evaluation of sea buckthorn (Hippophae rhamnoides L.) leaf tea

Qian He,Kailin Yang,Xinyan Wu,Chunhong Zhang,Chunnian He,Peigen Xiao

Sea buckthorn leaf tea, an emerging potential functional beverage product, has not yet had appropriate product standards and corresponding quality evaluation methods, and its poor taste directly affects the acceptance of the population, thus limiting its market consumption potential. In this study, two major packaging forms of sea buckthorn leaf tea available in the Chinese market were selected. The contents of total phenolics, total flavonoids, and 10 phenolic compounds, as well as the in vitro antioxidant capacity and sensory characteristics of sea buckthorn leaf tea were analyzed. Results showed that the quality of sea buckthorn leaf tea in the Chinese market varied widely. The total phenolic content, total flavonoid content, antioxidant activity, and consumer acceptance of bagged sea buckthorn leaf tea were higher than those of bulk sea buckthorn leaf tea. Multifactorial statistical analysis showed that the taste astringency of sea buckthorn leaf tea was closely related to ellagic acid and isorhamnetin-3-O-neohesperidin. Furthermore, isorhamnetin-3-O-neohesperidin had a greater effect on the antioxidant activity of sea buckthorn leaf tea. Therefore, ellagic acid and isorhamnetin-3-O-neohesperidin can be used as potential quality markers for sea buckthorn leaf tea. This work provides a reference for taste improvement and quality control of sea buckthorn leaf tea.

Converting Hippophae rhamnoides fruit extraction residue into porous carbons with ultra-high surface area for superior-performance supercapacitors

Zhentao Bian,Yaxuan Wang,Bingqian Zhu,Ru Ye,Fan Yuan,Ying Zhang,Taoqin Liu,Pinghua Zhang,Chong Chen,Guang Zhu,Keying Zhang

Scalable production of carbon materials from sustainable and renewable biomass as electrode materials in supercapacitors (SCs) has garnered considerable attention. However, the practical applications of the carbon-based SCs are still been hindered by the low energy density . Herein, hierarchical porous carbons (HPCs) with very high specific surface area and controlled porosity were synthesized by a facile method, which employed Hippophae rhamnoides fruit (HRF) extraction residue as raw materials, and KOH as the activation agent. The prepared carbon (HRF-NPC-2) exhibited a well-developed hierarchical pore structure , which gave an ultra-large specific surface area (SSA) of 2362.8 m 2  g −1 with a pore volume of 1.27 cm 3  g −1 . Electrochemical studies showed that the HRF-NPC-2 based electrode achieved an ultra-high specific capacity of 385.0 F g −1 at 1 A g −1 , and the symmetric supercapacitor in an ionic liquid electrolyte (1 M EMIMBF 4 ) achieved superior energy densities of 36.28 Wh kg −1 at 687.5 W kg −1 , and 43.82 Wh kg −1 at 1715 W kg −1 . Most importantly, the HRF-NPC-2//HRF-NPC-2 symmetric SCs can simultaneously light 88 LED lights for over 10 min, indicating great potential for practical application. The proposed simple fabrication strategy could be extended to convert other biomass waste into useful energy storage materials for supercapacitors .

Hippophae Rhamnoides-derived Phytomedicine Nano-System Modulates Bax/Fas Pathways to Reduce Proliferation in Triple-Negative Breast Cancer

Jabeen Farheen,M. Zubair Iqbal,Asim Mushtaq,Yike Hou,Xiangdong Kong

Triple-negative breast cancer (TNBC) is the most common primary tumor of the breast with limited effectual drug availability. Therefore, the aim of the study is to develop an innovative phyto-nanomedicine (PNM) to cure TNBC with the least genotoxicity. Hereinafter, the sea buckthorn’ extracted polyphenols (SBP), combine with metformin (MET), are synthesized as a novel PNM to evaluate its anti-tumor properties, effectiveness, and mechanism of action in TNBC in vitro and in vivo models. The SBP exhibits 16 new kinds of polyphenols that are been reported earlier which regulated cell development, proliferation, and programmed cell death (PCD) effectively. SBP-MET PNM inhibits MDA-MB-231 (47%), MDA-MB-436 (46%), and 4T1 (46%) cell proliferation but does not affect L929 normal murine cell development and successfully induce PCD (73.19%) in MDA-MB-231 cells. Mechanistically, in vivo SBP-MET proteome expression profiling reveals upregulation of proapoptotic Bax protein and activation of Fas signaling pathways convince downstream Daxx and FADD proteins, which further triggers Caspase-3 that prompts apoptosis in human TNBC cells by cleaving PARP-1 protein. Current findings establish innovative highly biocompatible phyto-nanomedicine that has significant potential to inhibit TNBC cell growth and induce regulated cell death (RCD) in vivo model, thereby opening a new arena for TNBC therapy.

Electroantennogram and Olfactory behavioral responses of Trabala vishnou gigantina (Lepidoptera: Lasiocampidae) to herbivore induced Hippophae rhamnoides leaf volatiles

Yonghua Liu,Kexu An,Shuo Tang,Jiangshuai Feng,Xiongfei Yan

The moth Trabala vishnou gigantina Yang, 1978 (Lepidoptera: Lasiocampidae), a leaf-eating pest, had a severe outbreak in the sea buckthorn, Hippophae rhamnoides L. (Elaeagnaceae) plantation in North China. This study aims to investigate the influence of volatiles emitted by T. vishnou gigantina -infested H. rhamnoides on the behavioral responses of T. vishnou gigantina , laying a basis for the development of plant-based elicitors. The chemical basis of herbivore insect host dynamics between these species were investigated, providing information for herbivore insect control methods. After identifying the compounds produced by the differences between healthy and infested plants, six critical volatile compounds were identified to explore their attractions to T. vishnou gigantina imagoes by using electroantennogram (EAG) and olfactory behavioral response experiments. The results showed that the EAG responses of T. vishnou gigantina imagoes did not only vary in these six different herbivore-infested volatiles, but also between different concentrations of the same volatile. The EAG responses to the herbivore-infested volatiles peaked at a stimulus concentration of 100 µg/µL, with Hexyl 2-methyl butyrate having the highest EAG response. The test results of olfactory behavioral responses had significant differences in the olfactory behavioral responses of female and male imagoes to the six herbivore-infested volatiles. Hexyl 2-methyl butyrate had the strongest attraction to female and male imagoes, followed by 2-ethylhexanol and longifolene. 2-ethylhexanol had a significant attraction to female imagoes, while there was no significant attraction to males.

Phytic acid enhances the oral absorption of isorhamnetin, quercetin, and kaempferol in total flavones of Hippophae rhamnoides L.

Yan Xie,Huilin Luo,Jingze Duan,Chao Hong,Ping Ma,Guowen Li,Tong Zhang,Tao Wu,Guang Ji

Aim Total flavones of Hippophae rhamnoides L. (TFH) have a clinical use in the treatment of cardiac disease. The pharmacological effects of TFH are attributed to its major flavonoid components, isorhamnetin , kaempferol , and quercetin . However, poor oral bioavailability of these flavonoids limits the clinical applications of TFH. This study explores phytic acid (IP 6 ) enhancement of the oral absorption in rats of isorhamnetin, kaempferol, and quercetin in TFH. Methods In vitro Caco-2 cell experiments and in vivo pharmacokinetic studies were performed to investigate the effects of IP 6 . The aqueous solubility and lipophilicity of isorhamnetin, quercetin, and kaempferol were determined with and without IP 6 , and mucosal epithelial damage resulting from IP 6 addition was evaluated by MTT assays and morphology observations. Results The P app of isorhamnetin, kaempferol, and quercetin was improved 2.03-, 1.69-, and 2.11-fold in the presence of 333   μg/mL of IP 6 , respectively. Water solubility was increased 22.75-, 15.15-, and 12.86-fold for isorhamnetin, kaempferol, and quercetin, respectively, in the presence of 20   mg/mL IP 6 . The lipophilicity of the three flavonoids was slightly decreased, but their hydrophilicity was increased after the addition of IP 6 in the water phase as the logP values of isorhamnetin, kaempferol, and quercetin decreased from 2.38   ±   0.12 to 1.64   ±   0.02, from 2.57   ±   0.20 to 2.01   ±   0.04, and from 2.39   ±   0.12 to 1.15   ±   0.01, respectively. The absorption enhancement ratios were 3.21 for isorhamnetin, 2.98 for kaempferol, and 1.64 for quercetin with co-administration of IP 6 (200   mg/kg) in rats. In addition, IP 6 (200   mg/kg, oral) caused neither significant irritation to the rat intestines nor cytotoxicity (400   μg/mL) in Caco-2 cells. Conclusions The oral bioavailability of isorhamnetin, kaempferol, and quercetin in TFH was enhanced by the co-administration of IP 6 . The main mechanisms are related to their enhanced aqueous solubility and permeability in the presence of IP 6 . In summary, IP 6 is a potential absorption enhancer for pharmaceutical formulations that is both effective and safe.
