Selective sorption of hippuric acid and 4-methylhippuric acid by polyethylene glycol modified ZIF based molecular imprinted polymer

Junlei Zheng,Haifeng Gao,Chengcheng Jin,Miaomiao Tian

A novel solid phase extraction (SPE) adsorbent, polyethylene glycol (PEG) immobilized zirconium ion-modified zeolite-imidazole framework (ZIF) molecular-imprinted polymer (ZIF@PEG@Zr@MIPs), was developed for detecting hippuric acid (HA) and 4-methylhippuric acid (4-MHA) in human urine using HPLC-UV. Zirconium ions (Zr 4+ ) anchored on the porous ZIF act as binding sites, facilitating coordination and electrostatic interaction with HA and 4-MHA carboxyl groups. Dopamine (DA) imprinting introduces additional sites, enabling π-π conjugation and hydrogen bonding with HA and 4-MHA, enhancing specific adsorption. The adsorbent, using HA as a template, demonstrated adsorption capacities of 37.98 mg g −1 for HA and 32.99 mg g −1 for 4-MHA. Imprinted factors ( IF ) were 2.68 for HA and 2.17 for 4-MHA, with a limit of detection (LOD) of 0.015 mg L −1 for HA and 0.011 mg L −1 for 4-MHA. The SPE-HPLC-UV method achieved 84.8–96.2% recovery and 0.3–2.2% relative standard deviation (RSD), illustrating its potential in health monitoring and environmental safety.

Multiple modulator-induced syntheses, crystal structures, and luminescent properties on hippuric acid and bovine serum albumin of Ln(III) complexes with 2,2′-oxybis(benzoic acid)

De-Chun Zhang,Dou Ma,Xia Li

Six new coordination complexes, Ln2(2,2′-oba)2(phen)2(ox)(H2O)2 (Ln = Eu 1, Tb 2), Ln4(2,2′-oba)6(phen)2 (Ln = Eu 3, Tb 4), Eu4(2,2′-oba)6(phen)2(H2O) (5), and K[Eu(2,2′-oba)2(phen)2] (6) [2,2′-H2oba = 2,2′-oxybis(benzoic acid), phen = 1,10-phenanthroline, H2ox = oxalic acid] were synthesized by hydrothermal reactions with the same compound molar ratios but different modulatory reagents (MRs). Complexes 1–5 have different 1-D chain structures and 6 shows a mononuclear structure. These complexes form diverse 3-D supramolecular networks through hydrogen bonds. The interaction between these complexes and hippuric acid (HA) or bovine serum albumin (BSA) was investigated by fluorescence spectral analysis. Interestingly, the hippuric acid could quench the luminescence of these complexes while the fluorescence of BSA could be quenched by these complexes. Results suggested that the complexes may be potential luminescent testing reagents for HA or BSA by significant fluorescence quenching of Ln3+ or BSA, respectively, through a static and dynamic quenching process.

Hippuric Acid Promotes Renal Fibrosis by Disrupting Redox Homeostasis via Facilitation of NRF2–KEAP1–CUL3 Interactions in Chronic Kidney Disease

Bowen Sun,Xifan Wang,Xiaoxue Liu,Longjiao Wang,Fazheng Ren,Xiaoyu Wang,Xiaojing Leng

Chronic kidney disease (CKD) is characterized by the accumulation of protein-bound uremic toxins (PBUTs), which play a pathophysiological role in renal fibrosis (a common pathological process resulting in CKD progression). Accumulation of the PBUT hippuric acid (HA) is positively correlated with disease progression in CKD patients, suggesting that HA may promote renal fibrosis. Oxidative stress is the most important factor affecting PBUTs nephrotoxicity. Herein, we assessed the ability of HA to promote kidney fibrosis by disrupting redox homeostasis. In HK-2 cells, HA increased fibrosis-related gene expression, extracellular matrix imbalance, and oxidative stress. Additionally, reactive oxygen species (ROS)-mediated TGFβ/SMAD signaling contributed to HA-induced fibrotic responses. HA disrupted antioxidant networks by decreasing the levels of nuclear factor erythroid 2-related factor 2 (NRF2), leading to ROS accumulation and fibrotic responses, as evidenced by NRF2 activation and knockdown. Moreover, NRF2 levels were reduced by NRF2 ubiquitination, which was regulated via increased interactions of Kelch-like ECH-associated protein 1 with Cullin 3 and NRF2. Finally, renal fibrosis and redox imbalance promoted by HA were confirmed in rats. Importantly, sulforaphane (NRF2 activator) reversed HA-promoted renal fibrosis. Thus, HA promotes renal fibrosis in CKD by disrupting NRF2-driven antioxidant system, indicating that NRF2 is a potential therapeutic target for CKD.Keywords:hippuric acid;protein-bound uremic toxin;renal fibrosis;oxidative stress;antioxidation;NRF2;NRF2 ubiquitination

Preparation of chitosan-modified magnetic Schiff base network composite nanospheres for effective enrichment and detection of hippuric acid and 4-methyl hippuric acid

Jingjing Han,Huiqi Wang,Zhanfeng Li,Zonghua Wang

Chitosan-modified magnetic Schiff base network composite nanospheres (Fe 3 O 4 @SNW@Chitosan) were prepared for the enrichment and detection of hippuric acid (HA) and 4-methyl hippuric acid (4-MHA) via magnetic solid phase extraction (MSPE) connected with HPLC. The SNW was one of the covalent organic framework, which constructed through covalent bonds, shown comprising solvent stability, low density and accessible pores. The obtained Fe 3 O 4 @SNW@Chitosan has many merits as a magnetic sorbent, including a hydrophilic surface, uniform pore size, unique ordered channel structure, and superparamagnetism. The favourable linearity of this MSPE-HPLC method was in the range of 1-1000 μg L −1 , and LODs of HA and 4-MHA were 0.3 μg L −1 and 0.2 μg L −1 , respectively. The recoveries in urine samples were range from 95.3 to 109.0 % with the RSD less than 9.6 %. When employed for the enrichment of HA and 4-MHA, Fe 3 O 4 @SNW@Chitosan exhibited great potential as a candidate for preconcentration.

Alistipes indistinctus-derived hippuric acid promotes intestinal urate excretion to alleviate hyperuricemia

Ying-Xi Xu,Lu-Di Liu,Jiang-Yuan Zhu,Shan-Shan Zhu,Bing-Qi Ye,Jia-Lu Yang,Jing-Yi Huang,Zhi-Hao Huang,Yi You,Wen-Kang Li,Jia-Lin He,Min Xia,Yan Liu

Hyperuricemia induces inflammatory arthritis and accelerates the progression of renal and cardiovascular diseases. Gut microbiota has been linked to the development of hyperuricemia through unclear mechanisms. Here, we show that the abundance and centrality of Alistipes indistinctus are depleted in subjects with hyperuricemia. Integrative metagenomic and metabolomic analysis identified hippuric acid as the key microbial effector that mediates the uric-acid-lowering effect of A. indistinctus. Mechanistically, A. indistinctus-derived hippuric acid enhances the binding of peroxisome-proliferator-activated receptor γ (PPARγ) to the promoter of ATP-binding cassette subfamily G member 2 (ABCG2), which in turn boosts intestinal urate excretion. To facilitate this enhanced excretion, hippuric acid also promotes ABCG2 localization to the brush border membranes in a PDZ-domain-containing 1 (PDZK1)-dependent manner. These findings indicate that A. indistinctus and hippuric acid promote intestinal urate excretion and offer insights into microbiota-host crosstalk in the maintenance of uric acid homeostasis.

Analyzing urinary hippuric acid as a metabolic health biomarker through a supramolecular architecture

Wenhui Li,Hua Dong,Kejing Niu,Huan-Yu Wang,Wenqian Cheng,Hualong Song,An-Kang Ying,Xiaobing Zhai,Kefeng Li,Huijuan Yu,Dong-Sheng Guo,Yuefei Wang

The prevalence of metabolic disorders has been found to increase concomitantly with alternations in habitual diet and lifestyle, indicating the importance of metabolic health monitoring for early warning of high-risk status and suggest effective intervention strategies. Hippuric acid (HA), as one of the most abundant metabolites from the gut microbiota, holds potential as a regulator of metabolic health. Accordingly, it is imperative to establish an efficient, sensitive, and affordable method for large-scale population monitoring, revealing the association between HA level and metabolic disorders. Upon systematic screening of macrocycle•dye reporter pair, a supramolecular architecture (guanidinomethyl-modified calix [5] arene, GMC5A) was employed to sense urinary HA by employing fluorescein (Fl), whose complexation behavior was demonstrated by theoretical calculations, accomplishing quantification of HA in urine from 249 volunteers in the range of 0.10 mM and 10.93 mM. Excitedly, by restricted cubic spline, urinary HA concentration was found to have a significantly negative correlation with the risk of metabolic disorders when it exceeded 0.76 mM, suggesting the importance of dietary habits, especially the consumption of fruits, coffee, and tea, which was unveiled from a simple questionnaire survey. In this study, we accomplished a high throughput and sensitive detection of urinary HA based on supramolecular sensing with the GMC5A•Fl reporter pair, which sheds light on the rapid quantification of urinary HA as an indicator of metabolic health status and early intervention by balancing the daily diet.

Bairesi Pills ameliorates vitiligo-like depigmentation by reducing hippuric acid accumulation and regulating MAPK14/β-catenin mediated melanogenesis

Zhuorui Li,Ningbo Zhang,Limei Dai,Xiangzhen Meng,Jiaqi Huang,Qingwen Hei,Zhinan Mei,Hui Xiong

Bairesi Pills promote repigmentation in hydroquinone-induced vitiligo mice. • Bairesi Pills reduce hippuric acid levels and restore gut-skin axis homeostasis. • Bairesi Pills and their active constituents including piperine and quercitrin, enhance melanogenesis through modulation of the MAPK14/β-catenin pathway. Background Bairesi Pills is a traditional Uyghur medicinal formula prescribed for vitiligo. However, its bioactive constituents and specific pharmacological mechanisms remain to be fully elucidated. Purpose This study aims to elucidate the underlying mechanisms and identify the active constituents of Bairesi Pills in the treatment of vitiligo. Methods A hydroquinone-induced vitiligo mouse model was established to evaluate the effects of Bairesi in vivo. Serum untargeted metabolomics and gut microbiome analyses were integrated to identify key differential metabolites and explore the association between gut dysbiosis and skin depigmentation. In vitro, melanocyte oxidative injury models were established using H₂O₂ or hippuric acid. Transcriptome sequencing was performed to screen differentially expressed genes and related signaling pathways. MAPK14 overexpression and pharmacological inhibition were further used to verify the effects and mechanisms of Bairesi and its active constituents in alleviating hippuric acid-induced oxidative stress. Results In vivo, Bairesi ameliorated oxidative stress and promoted pigment deposition in vitiligo-like mice, which was associated with enhanced intestinal mucus barrier integrity, reshaped gut microbiota, and reduced hippuric acid accumulation. In vitro, H₂O₂ or hippuric acid induced oxidative injury and MAPK activation in melanocytes, whereas Bairesi, piperine, and quercitrin suppressed MAPK14 activation, alleviated oxidative stress, and restored melanin synthesis, but these protective effects were attenuated by MAPK14 overexpression. Conclusion Bairesi Pills ameliorates vitiligo-like depigmentation by reshaping the gut microbiota–metabolite profile, reducing hippuric acid accumulation, and reversing HA-induced oxidative stress through inhibition of MAPK14 phosphorylation. Heterometallic ZnHoMOF as a Dual-Responsive Luminescence Sensor for Efficient Detection of Hippuric Acid Biomarker and Nitrofuran Antibiotics

Jingrui Yin,Wenqian Li,Wencui Li,Liying Liu,Dongsheng Zhao,Xin Liu,Tuoping Hu,Liming Fan

Developing efficient and sensitive MOF-based luminescence sensors for bioactive molecule detection is of great significance and remains a challenge. Benefiting from favorable chemical and thermal stability, as well as excellent luminescence performance, a porous Zn(II)Ho(III) heterometallic–organic framework (ZnHoMOF) was selected here as a bifunctional luminescence sensor for the early diagnosis of a toluene exposure biomarker of hippuric acid (HA) through “turn-on” luminescence enhancing response and the daily monitoring of NFT/NFZ antibiotics through “turn-off” quenching effects in aqueous media with high sensitivity, acceptable selectivity, good anti-interference, exceptional recyclability performance, and low detection limits (LODs) of 0.7 ppm for HA, 0.04 ppm for NFT, and 0.05 ppm for NFZ. Moreover, the developed sensor was employed to quantify HA in diluted urine samples and NFT/NFZ in natural river water with satisfactory results. In addition, the sensing mechanisms of ZnHoMOF as a dual-response chemosensor in efficient detection of HA and NFT/NFZ antibiotics were conducted from the view of photo-induced electron transfer (PET), as well as inner filter effects (IFEs), with the help of time-dependent density functional theory (TD-DFT) and spectral overlap experiments. Keywords:antibiotics detection;bifunctional luminescence sensor;biomarker detection;heterometallic metal–organic framework;sensing mechanism

Quaternary Ammonium Modified Mg–Al Layered Double Hydroxide Based Composite Aerogel Adsorbents for Removal of Hippuric Acid During Hemodialysis

Bofan An,Hui Ye,Xuefen Wang

As the global population of chronic kidney disease (CKD) patients continues to expand, high-performance adsorbents for uremic toxins have emerged as a critical solution to alleviate dialysis treatment challenges. In this work, a layered double hydroxide (Mg–Al LDH) was prepared via a facile hydrothermal approach and subsequently functionalized for adsorption of protein-bound toxins by organosilicon quaternary ammonium salt (Q-Mg–Al LDH). The synthesized Q-Mg–Al LDH was then incorporated into sodium alginate (SA) solution to achieve a three-dimensional Q-Mg–Al LDH/SA composite aerogel (SA-Q) by freeze-drying for easy maneuverability as adsorbents. Specifically, the organosilicon quaternary ammonium-grafted LDH (Q-Mg–Al LDH) offered both long alkyl chain and positively charged quaternary ammonium moieties, enabling synergistic adsorption through electrostatic and hydrophobic interactions, thereby exhibiting superior adsorption capacity (222.7 mg/g) for hippuric acid (HA, one typical protein-bound uremic toxin). The optimized SA-Q possessed outstanding adsorption capabilities (141.8 mg/g); more importantly, it could maintain a 95% removal rate even after 8 cycles, and also could selectively remove the diffused HA in the dialysate during hemodialysis, indicating its excellent operability and recyclability. This work demonstrated a promising strategy for constructing advanced adsorbents to eliminate uremic toxins from dialysate to facilitate the development and popularization of miniaturized hemodialysis equipment.

Preparation of activated carbon and β-cyclodextrin modified magnetic nanoparticles for extraction and determination of hippuric acid in urine samples

Xiaoyue Shan,Ji Shao,Ling Zhang,Yan Jin,Siwei Tan,Haipeng Ye,Luoxian Yang

In this study, a novel magnetic adsorbent (AC/β-CD/Fe 3 O 4 ) was prepared for the magnetic solid phase extraction (MSPE) of hippuric acid (HA) from urine. The adsorbent combined the high surface area of activated carbon (AC), the host-guest recognition of β-cyclodextrin (β-CD), and the magnetic responsiveness of Fe 3 O 4 . Under the optimal conditions, the method demonstrated excellent performance for HA detection, with a linearity range of 0.0064-1.0 µg/mL, low limit of detection of 1.9 µg/L, and high precision (RSDs < 5%). Recoveries from spiked urine matrices ranged from 95% to 99%, confirming the method's accuracy and practicality for monitoring HA in real biological matrices.

Dietary supplementation with red cabbage extract rich in anthocyanins increases urinary hippuric acid excretion and consequently decreases nitrous oxide emissions in beef bulls

Jian Gao,Bingbing Cheng,Yufeng Liu,Meng M. Li,Guangyong Zhao

Two consecutive experiments were conducted to investigate the effects of dietary supplementation with red cabbage extract (RCE) rich in anthocyanins on the nitrogen (N) metabolism in beef bulls and the nitrous oxide (N 2 O) emissions from the soil applied with the urine of beef bulls. In Experiment 1, 8 Simmental beef bulls (body weight 387.9 ± 40.7 kg) were used as experimental animals . Two levels of RCE, i.e. 0 and 114 g dry matter per day, were supplemented to a basal ration as experimental treatments. The animals and the treatments were allocated in a 2 × 2 crossover design. In Experiment 2, the static incubation technique was used to determine the N 2 O-N emissions from the soil applied with the urine samples collected from Experiment 1. The results of Experiment 1 indicated that RCE supplementation did not affect the N excretion, N retention, and N retention rate in beef bulls ( P  > 0.10). However, RCE supplementation increased the ratio of hippuric acid-N/urinary N ( P  < 0.05) and tended to increase the ratio of creatinine-N/urinary N ( P  = 0.081) without changing the excretion of other urinary nitrogenous components ( P  > 0.10). Plasma metabolome analysis indicated that RCE supplementation upregulated 12 metabolites while downregulated 18 metabolites ( P  < 0.05). The results of Experiment 2 indicated that RCE supplementation reduced the estimated urine N 2 O-N emissions by 33.1% through decreasing the ratio of N 2 O-N/urine-N ( P  < 0.05). In conclusion, RCE supplementation did not affect the N retention and N excretion of beef bulls, whereas it attenuated the urine N 2 O-N emissions of beef bulls through increasing urinary hippuric acid excretions.

Design of a calix[4]arene-functionalized metal-organic framework probe for highly sensitive and selective monitor of hippuric acid for indexing toluene exposure

Yaran Du,Xiqian Li,Haijiao Zheng,Xueju Lv,Qiong Jia

In the present work, a novel metal-organic framework (MOF) fluorescent probe was prepared by post-synthetic modification of MIL-53-NH 2 (Al) with carboxylatocalix[4]arene (CC[4]A). The introduced CC[4]A could not only enhance the fluorescence performance and the recognition ability of the probe, but also sustain the high stability under UV light and moisture conditions. A method based on the as-synthesized CC[4]A@MIL-53-NH 2 (Al) probe was established for sensing hippuric acid . The detection limit was determined to be as low as 3.7 μg mL −1 . Over the concentration range of 0.005–3 mg mL −1 , the calibration curve was obtained with a satisfactory linearity ( R 2  = 0.993). The method was successfully used for rapid and highly selective direct monitor of hippuric acid in human urine. The sensor had great potential to be used as a simple diagnostic tool for hippuric acid in human urine which is regarded as a biological index of toluene exposure.
