Synergistic effect of N-Ho on photocatalytic CO2 reduction for N/Ho co-doped TiO2 nanorods

Xiya Wang,Zhaoguo Zhang,Zhengfeng Huang,Peimei Dong,Xiaoxiao Nie,Zhi Jin,Xiwen Zhang

The photoreduction of CO 2 by using Ho and N co-doped TiO 2 was explored. The doping elements can further improve the separation efficiency of photogenerated electron-hole pairs via refining grains, causing lattice distortion and adsorbing more hydroxyl radicals(·OH). We found that there exists product selectivity such that the photoreduction products of Ho/N-TiO 2 mainly consist of CO and methanol, while the counterparts of pristine TiO 2 are CO and methane. The possible mechanism of CO 2 reduction is depicted involving the generation and combination of specific radicals. Thus, the amount of ·OH adsorbed on the surface of the catalyst makes a significant difference in the selectivity in the reduction products. Ho/N-TiO 2 catalyst with the synergic effect of Ho and N showed the best photoreductive performance for CO 2 with H 2 O to CO and CH 4 , and its formation rate of CH 4 was 29 μmol/gcatal./hour.

Ho doping-induced ferroelectric polarization enhances UV photodetector performance of YbMnO3

Shan Zhang,Xin Song,Xing Gao,Chunxiao Lu,Pei Han,Yunying Liu,Xiaowei Li,Junyan Zhang,Yong Li

The ferroelectric photovoltaic (PE-PV) effect has been extensively explored and plays a crucial role in constructing self-powered photodetectors. Ferroelectric materials have been attracting great interest in self-powered ultraviolet (UV) photodetectors due to their polarization-induced photovoltaic effect. In this paper, the ultraviolet self-powered photodetection properties of polycrystalline rare-earth Yb 1-x Ho x MnO 3 (0 ≤ x ≤ 0.08, Δx = 0.02) ferroelectric materials have been investigated. The substitution of Ho atoms for Mn atoms has a great influence on the ferroelectric polarization, and Ho doping increases the photocurrent density. The photodetectors show excellent reproducibility responsivity (R) and detectivity (D*) under illumination at 365 nm. In addition, at the light density of 1 mW/cm 2 , the responsivity and detectivity of the photodetectors are as high as 0.09 W/A and 3.359 × 10 11 Jones, respectively. These outstanding photodetection performances are due to the relatively high short-circuit current density ( J sc ). High J sc implies that more photogenerated carriers can be produced under illumination, which makes the device more sensitive to optical signals. This study presents a feasible method for improving the light-detection performance of the ultraviolet self-powered detector.

Inhibition of MALAT1 facilitates ROS accumulation via the Keap1/HO-1 pathway to enhance photodynamic therapy in secondary hyperparathyroidism

Ying Wen,Yitong Li,Danhua Zhang,Ziru Liu,Hong Liu,Xiejia Li,Wei Wu,Liyun Zeng,Qiongyan Zou,Wenjun Yi

The prevalence of secondary hyperparathyroidism (SHPT) in advanced chronic kidney disease (CKD) exceeds 80%. Our previous study indicated that photodynamic therapy (PDT) has potential for treating SHPT. Long noncoding RNA (lncRNA) is involved in various oxidative stress and apoptotic processes, but the molecular mechanism remains unreported. In this study, we found that PDT induced apoptosis in SHPT through reactive oxygen species (ROS) accumulation. The expression of metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) and heme oxygenase 1 (HO-1) within SHPT was upregulated after PDT. Inhibition of MALAT1 increased PDT-induced ROS, which promoted the apoptosis. Pearson correlation analysis confirmed that there was a positive correlation between MALAT1 and HO-1, and MALAT1 inhibition down-regulated HO-1, whereas concomitant overexpression of HO-1 was able to eliminate the PDT-induced ROS and inhibit apoptosis. The direct binding of MALAT1 to Kelch-like ECH-associated protein 1 (Keap1) protein was confirmed by high-throughput sequencing, RNA pulldown, silver staining and western blotting assays. Si-Keap1 was able to rescue the down-regulation of HO-1 caused by MALAT1 inhibition, restoring the elimination of ROS by HO-1 and attenuating the effect of PDT. In addition, PDT effectively reduced parathyroid hormone (PTH) secretion in SHPT rats, and this effect was further enhanced in combination with MALAT1 inhibitors. Overall, MALAT1 activates downstream HO-1 expression by binding to Keap1, thereby reducing ROS and inhibiting apoptosis, which in turn mediates PDT resistance in SHPT. Inhibition of MALAT1 significantly enhanced the efficacy of PDT, suggesting a potential therapeutic target for improving PDT for SHPT outcomes.

Activation of Nrf2/HO-1 signaling pathway exacerbates cholestatic liver injury

Wang Yi,Fu Xiaolong,Zeng Li,Hu Yan,Gao Rongyang,Xian Siting,Liao Songjie,Huang Jianxiang,Yang Yonggang,Liu Jilong,Jin Hai,Klaunig James,Lu Yuanfu,Zhou Shaoyu

Nuclear factor erythroid 2-related factor-2 (Nrf2) antioxidant signaling is involved in liver protection, but this generalization overlooks conflicting studies indicating that Nrf2 effects are not necessarily hepatoprotective. The role of Nrf2/heme oxygenase-1 (HO-1) in cholestatic liver injury (CLI) remains poorly defined. Here, we report that Nrf2/HO-1 activation exacerbates liver injury rather than exerting a protective effect in CLI. Inhibiting HO-1 or ameliorating bilirubin transport alleviates liver injury in CLI models. Nrf2 knockout confers hepatoprotection in CLI mice, whereas in non-CLI mice, Nrf2 knockout aggravates liver damage. In the CLI setting, oxidative stress activates Nrf2/HO-1, leads to bilirubin accumulation, and impairs mitochondrial function. High levels of bilirubin reciprocally upregulate the activation of Nrf2 and HO-1, while antioxidant and mitochondrial-targeted SOD2 overexpression attenuate bilirubin toxicity. The expression of Nrf2 and HO-1 is elevated in serum of patients with CLI. These results reveal an unrecognized function of Nrf2 signaling in exacerbating liver injury in cholestatic disease.

Growth and magneto-optical properties of Ho-doped Tb3Sc2Al3O12 single crystal for faraday isolators

Yuankai Hao,Xiuwei Fu,Xianhui Xin,Xianxian Yang,Zhen Zhang,Zhitai Jia,Xutang Tao

In this work, a novel Ho-doped Tb 3 Sc 2 Al 3 O 12 magneto-optical single crystal has been successfully grown by the Czochralski method for the first time. Phase composition of the crystal was tested by XRD measurements. The distribution coefficient of Ho 3+ in the crystal was obtained. The crystal exhibits excellent optical transmittance without inclusions. It shows a uniform elemental distribution and excellent magneto-optical properties. The Verdet constant is comparable to Tb 3 Sc 2 Al 3 O 12 crystal and higher than that of traditional Tb 3 Ga 5 O 12 crystal. Therefore, the Ho-doped Tb 3 Sc 2 Al 3 O 12 single crystal is a promising material for visible near-infrared Faraday applications.

Chemical bond features and microwave dielectric properties of olivine-type LiMSiO4 (M = Ho, In) ceramics

Congxue Su,Haiyan Che,Wenlong Tao,Yang Yang,Ruijuan Wang,Zhaohui Wang,Xingliu Qin,Haibin Li,Jun Fan,Liang Fang

Orthorhombic olivine-structured LiMSiO 4 (M = Ho, In) ceramics were synthesised using the solid-state reaction. Promising microwave dielectric properties of ε r  = 10.04, τ f  = 23.3 ppm/°C and a high-quality factor ( Q  ×  f ) of 11,415 GHz for LiHoSiO 4 were obtained at 1050 °C, and ε r  = 7.78, τ f  = −46.8 ppm/°C and Q  ×  f  = 23,390 GHz for LiInSiO 4 were achieved at 1140 °C. According to the bond valence result, Ho 3+ presented a slightly compressed state but Li + and Si 4+ exhibited a strong rattling state in LiHoSiO 4 , which led to higher ε r than the ε theo . The integrated effect of rattling cations (Li + and In 3+ ) and a compressed cation (Si 4+ ) in LiInSiO 4 resulted in a small positive deviation Δ ε r . The larger packing fraction of LiInSiO 4 contributed to a higher Q  ×  f than LiHoSiO 4 . The Si–O bond in both ceramics played a leading role in influencing the Q × f according to the Phillips−Van Vechten−Levine chemical bond theory.

Curcumin induces ferroptosis in hepatocellular carcinoma by regulating PERK/Nrf2/HO-1 signaling pathway

Yuan Cheng,Liu Ruiwen,Xu Xinming,Sun Jun,Zhu Yi,Lu Rongzhu,Liu Yueyu,Chen Jian

Background Because of the high malignancy and subtle early symptoms of hepatocellular carcinoma (HCC), most patients lose the opportunity for surgery and opt for drug therapy. However, the current drugs for HCC treatment remain suboptimal, underscoring the urgent need to develop a novel anti-HCC agent. Curcumin is a natural chemical compound that has anti-cancer effects on various tumor cells, attracting sustained attention from researchers and clinicians. The present study aims to further elucidate curcumin’s anti-HCC mechanisms, thereby offering a promising therapeutic candidate for HCC patients. Methods The viability of HCC cells was evaluated by Cell Counting Kit-8 (CCK-8) assay. The levels of glutathione (GSH), malondialdehyde (MDA), and total iron in the cells were detected by biochemical kits. The levels of reactive oxygen species (ROS) and Fe 2+ in the cells were detected by fluorescent probes. The protein expressions of protein kinase RNA-like endoplasmic reticulum kinase (PERK), nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and glutathione peroxidase 4 (GPX4) in the cells were detected by western blotting. Results Curcumin exerted a notably suppressive impact on HCC cells, which could be reversed by ferrostatin-1 (Fer-1) and desferrioxamine (DFO). By detecting cellular metabolic products, we observed an increase in total iron, Fe 2+ , MDA, ROS, and a reduction in GSH levels in HCC cells after treatment with curcumin, and these effects could be attenuated by Fer-1. Western blot analysis showed that curcumin significantly downregulated GPX4 levels in HCC cells while upregulating PERK, Nrf2 and HO-1 expression. Additionally, using PERK inhibitor, HO-1 inhibitor, and endoplasmic reticulum (ER) stress inhibitor could partially reverse the inhibition of HCC cells viability by curcumin and alleviate the reduction of GPX4 expression caused by curcumin. Conclusions Curcumin may induce ferroptosis in HCC cells through the PERK/Nrf2/HO-1 signaling pathway, thereby exerting its anti-cancer effects, suggesting that curcumin could potentially be used as a drug for treating HCC.

Puerarin prevents cataract development and progression in diabetic rats through Nrf2/HO‑1 signaling

Duzhen Zhang,Man Li

Puerarin is the major bioactive ingredient isolated from the dry root of Pueraria lobata, a plant used in traditional Chinese medicine. Puerarin has been used to treat diabetes and cataracts in China; however, its underlying mechanism of action remains unclear. The aim of the present study was to investigate the effectiveness and mechanism of puerarin in preventing cataracts in diabetic rats. Diabetes was induced by streptozocin (STZ) administration and rats were intraperitoneally injected with puerarin (25, 50 and 100 mg/kg). Blood glucose levels and cataract development were examined in the different experimental groups. In addition, the expression levels of markers associated with oxidative stress, including nuclear factor erythroid 2 like 2 (Nrf2) and heme oxygenase‑1 (HO‑1), were analyzed. The present results suggested that treatment with puerarin at 25, 50 and 100 mg/kg significantly reduced blood glucose levels and the incidence of cataract in STZ‑induced diabetic rats. Additionally, puerarin treatment reduced oxidative stress, restoring the levels of malondialdehyde and glutathione, and the activity of glutathione peroxidase. Furthermore, puerarin administration decreased the expression levels of retinal vascular endothelial growth factor and interleukin‑1β and increased the mRNA expression levels of Nrf2 and HO‑1, thus inhibiting oxidative stress. The present findings suggested that puerarin had hypoglycemic effects and that it prevented cataract development and progression in diabetic rats by reducing oxidative stress through the Nrf2/HO‑1 signaling pathway.

Self-Protonated Ho-Doped Zn(O,S) as a Green Chemical-Conversion Catalyst to Hydrogenate Nitro to Amino Compounds

Hairus Abdullah,Noto Susanto Gultom,Hardy Shuwanto,Worku Lakew Kebede,Dong-Hau Kuo

Zn(O,S) has been successfully doped with different amounts of Ho and characterized with X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), diffuse reflectance spectroscopy (DRS), X-ray photoelectron spectroscopy (XPS), electrochemical impedance spectroscopy (EIS), and transient photocurrent (TPC). The as-prepared Ho-doped Zn(O,S) catalysts with different Ho amounts are evaluated for hydrogen evolution reaction. The catalyst with the best performance in evolving hydrogen is further utilized to hydrogenate 4-nitrophenol (4-NP) to 4-aminophenol (4-AP). It is found that doping with Ho obviously enhanced charge transfer and photoresponse properties of the catalyst. Therefore, the modified Zn(O,S) can optimally evolve hydrogen by 18 624 μmol/g, which is 20% higher than that of pristine Zn(O,S). Subsequently, the in situ generated hydrogen ions on catalyst surfaces also play an important role as a hydrogen source to hydrogenate 4-NP to 4-AP without any reducing agents such as NaBH4, which is commonly used as a hydrogen source. As Ho is doped in the lattices of Zn(O,S), it acts not only to separate photocarriers and to enhance the charge transfer but also to shorten the diffusion time of nitrophenolate ions to catalyst surfaces for further photocatalytic hydrogenation reaction (PHR) process. A plausible PHR mechanism has been provided to elucidate the great performance of Ho-modified Zn(O,S) for photocatalytic hydrogenation.

Multifunctional astragalus polysaccharide composited microneedle promotes oral ulcers healing by inhibiting ferroptosis via Nrf2/HO-1/SLC7A11 axis

Qiang Zhang,Tuo Wang,Junyu Ren,Yanru Guo,Ziyi Yin,Chenrui Zhang,Honglin Fan,Yinting Ren,Junlong Da,Narisu Hu

Oral ulcers, a prevalent mucosal disorder with high recurrence rates, remain challenging to treat due to the oral cavity’s dynamic environment and the multifactorial nature of ulcer progression. Here, we report a multifunctional microneedle (MN) patch loaded with astragalus polysaccharide (APS-MN) that integrates rapid mucosal penetration, reactive oxygen species (ROS) scavenging, and ferroptosis inhibition to accelerate ulcer healing. Unlike conventional MN systems reliant on synthetic drugs or prolonged dissolution, APS-MN leverages the natural antioxidant and immunomodulatory properties of APS-a traditional Chinese medicine-derived polysaccharide-enabling complete drug release within 2 minutes and targeted delivery to ulcer bases. In vitro and in vivo studies demonstrate that APS-MN promotes M2 macrophage polarization, suppresses pro-inflammatory cytokines (TNF-α, IL-1β), and inhibits ferroptosis via the Nrf2/HO-1/SLC7A11 axis, achieving 88.2% wound closure in rats within 6 days. Notably, this work combined herbal polysaccharides with MN technology for oral ulcer therapy, offering a biocompatible, non-invasive alternative to corticosteroids and antibiotics. The rapid dissolution, dual antioxidant/anti-ferroptotic action, and scalability of APS-MN position it as a transformative platform for managing recurrent oral ulcers and other redox-imbalanced mucosal diseases.

Tetrachlorobenzoquinone induces acute liver injury, up-regulates HO-1 and NQO1 expression in mice model: The protective role of chlorogenic acid

Demei Xu,Lihua Hu,Xiaomin Xia,Jianbo Song,Lingrui Li,Erqun Song,Yang Song

Tetrachlorobenzoquinone (TCBQ) is an active metabolite of pentachlorophenol (PCP). Although PCP has been investigated extensively, there are only a few reports describing the toxicity effect of TCBQ, and no report regarding TCBQ-induced liver injury in vivo . In the current study, we aimed to examine the acute hepatic toxicity of TCBQ in the mice model. Chlorogenic acid (CGA) exhibits promising antioxidant activity in the past studies, thus, the second aim of this study was to evaluate the protective effect of CGA on TCBQ-induced liver injury. Our results indicated TCBQ-intoxication caused marked liver cell necrosis and inflammation but not apoptosis, and this damage was alleviated by CGA treatment. Meantime, TCBQ-intoxication enhanced serum ALT, AST activities, TBIL content, hepatic oxidative stress and lipid peroxidation, decreased GSH content and inhibited the activities of antioxidant enzymes. Western blot and immunohistochemical analysis showed that TCBQ marked up-regulated HO-1 and NQO1 expression. On the other hand, pretreatment of CGA reduced TCBQ-induced liver damage remarkably. Taking together, these results revealed that TCBQ has strong hepatic toxic effect, and at least a part of this effect is initiated by free radical and relieved with CGA administration.

Protective effect of rutin on ferroptosis-induced oxidative stress in aging laying hens through Nrf2/HO-1 signaling

Yangyang Wu,Shuo Zhou,An Zhao,Yuling Mi,Caiqiao Zhang

Oxidative stress is a major cause of ovarian aging and follicular atresia. There is growing evidence that showed potential roles of rutin in antidiabetic, anti-inflammatory, antitumor, antibacterial and antioxidant, although it is yet unclear what the underlying mechanism is. Here, we looked into the potential effects of rutin on oxidative stress in the prehierarchical small white follicles (SWFs) from 580-day-old (D580) laying chickens. According to the findings, aging D580 layer ferroptosis was much higher than it was for laying hens during the peak period (280-day-old, D280). In both naturally aged and d -gal-induced chicken SWFs, rutin treatment concurrently boosted cell proliferation and prevented apoptosis. In addition, rutin inhibited the increased ferroptosis in aging hens. Meanwhile, rutin markedly suppressed the elevated ferroptosis and descending antioxidant capacity of D280-culture-SWFs from chicken elicited by d -gal. Rutin's activation of the Nrf2/HO-1 pathway hastened the SWFs' verbal battle with oxidative damage and reduced ferroptosis. Furthermore, activation of the ferroptosis signal increased the oxidative damage in SWFs. In conclusion, rutin alleviated oxidative stress that was induced by ferroptosis in aging chicken SWFs through Nrf2/HO-1 pathway. These findings point to a novel mechanism by which rutin protects SWFs from oxidative stress by suppressing ferroptosis, which is presumably a fresh approach to slowing ovarian aging in laying hens.
