Simultaneously achieving high sensitivity, long-term stability, and recyclability in surface-enhanced Raman scattering (SERS) substrates remains a major challenge that limits their practical deployment in biochemical sensing. Here, we present a fully solid-state SERS substrate that realizes hierarchical hotspot engineering within an Au@TiO 2 @Ag multilayered nanoarchitecture. An ultrathin TiO 2 spacer layer deposited by atomic layer deposition conformally separates the Ag nanorod array and sharp-tipped Au nanoparticles, generating a cascade of plasmonic hotspots from the atomic scale to the microscale. The multilevel electromagnetic couplings, including inter-rod gap coupling, mirror-coupling, and tip-induced field concentration, act cooperatively to provide strong and uniform Raman enhancement while maintaining surface integrity under complex environments. The Au@TiO 2 @Ag substrate exhibits excellent reproducibility and long-term durability, enabling ultrasensitive antibiotic detection down to 10 −12 M directly in various human body fluids. This solid-state and hierarchically engineered SERS platform provides a general and robust pathway toward practical, high-performance biochemical sensing applications.

Surface-enhanced Raman spectroscopy (SERS) offers exceptional sensitivity but faces a critical trade-off in living systems: rigid substrates lack biological adaptability, while colloidal nanoprobes suffer from poor signal reproducibility. Herein, we present a bioadaptive SERS platform using magnetically guided swarming nanoprobes. These probes integrate a magnetic core, plasmonic gold/silver layers, and a biocompatible silica coating, enabling programmable assembly under magnetic fields into chain-like nanostructures with interparticle gap-dependent hotspots, followed by coordinated reconfiguration into dynamically stable swarms. Multiphysics simulations reveal that cyclic assembly-disassembly generates transient electromagnetic hotspots while inducing convective flows to actively recruit analytes. This dual mechanism achieves reproducible enhancement factors exceeding 2.9×10 7 , an order of magnitude higher than colloidal systems. In vivo, swarming nanoprobes deployed in rabbit models demonstrate over 10.3-fold Raman signal amplification during intravascular detection. By leveraging active matter physics to synergize nanoscale sensing, this work establishes a new paradigm for in vivo molecular diagnostics.

Microplastics (MPs) are critical vectors for the dissemination of antibiotic resistance genes (ARGs); however, the prevalence and ecological risks of high-risk ARGs in mangrove ecosystems—globally vital yet understudied coastal habitats—remain poorly understood. To address this gap, this study investigated polyethylene, polystyrene, and polyvinyl chloride incubated in mangrove sediments for one month, focusing on high-risk ARGs, virulence gene (VGs), and pathogenic antibiotic-resistant bacteria within the mangrove plastisphere. High-throughput PCR and metagenomic analyses revealed that high-risk ARGs, VGs, and mobile genetic elements (MGEs) were significantly enriched on MPs compared to surrounding sediments. Pathogenic bacteria and MGEs were also more abundant in the plastisphere, highlighting its role as a hotspot for ARG dispersal. Metagenome-assembled genome analysis identified Pseudomonas and Bacillus as key hosts for ARGs, MGEs, and VGs, particularly multidrug resistance genes, integrase genes, and adherence factors. Notably, polystyrene harbored the highest abundance of pathogenic bacteria carrying ARGs, MGEs, and VGs, and mangrove root exudates were found to amplify horizontal gene transfer on MPs, uncovering a previously overlooked mechanism driving antibiotic resistance in coastal ecosystems. These findings not only elucidate how MPs accelerate the spread of ARGs, but also underscore the urgent need for targeted mitigation strategies to address the adverse impacts microplastic pollution on human, animal, and environmental health.

Surface-enhanced Raman spectroscopy (SERS) offers unique molecular fingerprinting capability and enhanced sensitivity. However, the application of SERS in therapeutic drug monitoring (TDM) is limited due to the occurrence of unstable plasmonic hotspots, poor reproducibility, and the lack of molecular specificity. In this study, we present “PlasmoBridge”, a dual-function aptamer–nanoparticle assembly that bridges adjacent silver nanoparticles with thiolated methotrexate (MTX) aptamers. This design generates uniform nanogaps (∼6.5 nm) that enable stable and reproducible hotspot formation while selectively enriching MTX molecules in the electromagnetic field. Consequently, PlasmoBridge achieves ultrasensitive MTX detection (10 −8 M) with relative standard deviations below 10 % and the requisite linearity across clinically relevant concentration ranges. Integrating liquid–liquid and solid-phase extraction delivers recoveries in serum that exceed 90 %. Convolutional neural network–assisted spectral analysis serves to further enhance quantitative accuracy (R 2 > 0.99). In a preclinical osteosarcoma model, PlasmoBridge-guided TDM preserved antitumor efficacy while reducing systemic toxicity via optimized dosing. When compared with enzyme-multiplied immunoassay technique (EMIT) and liquid chromatography-tandem mass spectrometry (LC–MS/MS) procedures, PlasmoBridge uniquely balances sensitivity, speed, and clinical practicality, enabling near-real-time and point-of-care monitoring. Moreover, this generalizable biosensing framework, combining nanoscale engineering, molecular recognition, and AI-driven quantification, offers a promising route to precision pharmacotherapy of a range of small-molecule drugs.

Pancreatic lipase (PL) is a validated peripheral target for limiting dietary fat absorption, yet structurally diverse natural inhibitors remain scarce. We assembled a PL inhibitor dataset from public resources and trained a random-forest classifier using PubChem fingerprints (test AUC = 0.9134) to prioritize a natural product library. After drug-likeness and toxicity filtering, docking, and experimental validation, three hits were identified: Licochalcone C (IC 50 = 62.05 ± 1.77 μM), Neoglycyrol (IC 50 = 95.21 ± 0.57 μM), and Notopterol (IC 50 = 104.27 ± 3.19 μM). Interaction fingerprint and molecular dynamics analyses showed that binding was dominated by hydrophobic interactions, with Val260/Ala261 acting as key residues across the three PL-ligand complexes. Dissociation free-energy profiles from steered molecular dynamics and umbrella sampling were consistent with the potency ranking. Collectively, this data-driven pipeline identified new natural PL inhibitors and provided residue-level insights for further optimization.

Surface enhanced Raman scattering (SERS) enabled trace molecules detection has important application prospects. By structuring/modifying the surface of SERS substrate, molecules in highly-diluted solution can be concentrated into localized active area for highly sensitive detection. However, subject to the difficulty of fabrication process, it remains challenging to balance hot-spots construction and concentration capacity to molecules simultaneously. Therefore, preparing SERS substrate with dense ordered hot-spots and efficient concentration capacity is of great significance for highly sensitive detection. Herein, we propose the Ag and fluoroalkyl modified hierarchical armour substrate (Ag/F-HA), which has a double-layer stacking design to combine analyte concentration with hotspot construction. The micro armour structure fabricated by femtosecond-laser processing to serve as a superhydrophobic and low-adhesive surface to concentrate molecules, while anodic aluminum oxide (AAO) template creates nanopillars array serving as dense and ordered hot spots. Under the synergy action of hot-spots and molecule concentration, Ag/F-HA achieves the detection limit down to 10−7 M of Doxorubicin (DOX) molecules with a relative standard deviation (RSD) of 7.69%. Additionally, Ag/F-HA exhibits the excellent robustness to resist external disturbance such as liquid splash or abrasion. Based on our strategy, the SERS substrates with directional analyte concentration are further explored by patterning microcone array with a defect. This work opens a way to the realistic implementation of SERS in diverse scenarios.

Rapid urbanization and economic development have led to the development of heavy industry and structural re-equalization in mainland China. This has resulted in scattered and disorderly layouts becoming prominent in the region. Furthermore, economic development has exacerbated pressures on regional resources and the environment and has threatened sustainable and coordinated development in the region. The NASA Land Science Investigator Processing System (Land-SIPS) Visible Infrared Imaging Radiometer (VIIRS) 375-m active fire product (VNP14IMG) was selected from the Fire Information for Resource Management System (FIRMS) to study the spatiotemporal patterns of heavy industry development. Furthermore, we employed an improved adaptive K-means algorithm to realize the spatial segmentation of long-order VNP14IMG and constructed heat source objects. Lastly, we used a threshold recognition model to identify heavy industry objects from normal heat source objects. Results suggest that the method is an accurate and effective way to monitor heat sources generated from heavy industry. Moreover, some conclusions about heavy industrial heat source distribution in mainland China at different scales were obtained. Those can be beneficial for policy-makers and heavy industry regulation.

Sintering inhomogeneities caused by hotspots are one of the factors hindering the development of flash sintering technology. Herein, to overcome this problem for a high-entropy pyrochlore-structured (La 0.2 Nd 0.2 Sm 0.2 Eu 0.2 Gd 0.2 ) 2 Zr 2 O 7 ceramic (abbreviated as RE 2 Zr 2 O 7 ), we propose a simple strategy, namely, addition of Al 2 O 3 to the ceramic and achieved a significant effect. During flash sintering, rare earth ions in RE 2 Zr 2 O 7 reacted with Al 2 O 3 to form REAlO 3 and RE 2−x Zr 2 O 7–3x/2 , which substantially improved the sintering homogeneity. With an increase in the Al 2 O 3 content, REAlO 3 transformed into REAl 11 O 18 . The crack deflection and bridging effect caused by the formation of these phases and their intrinsic high hardness and high modulus improved the mechanical properties of the composite ceramic. The use of an internal reaction to improve the electrical conductivity of a ceramic body is an economical and effective strategy to prevent hotspot formation during flash sintering.

To effectively extract target analytes from complex sample surfaces is of great significance for the practical application of surface-enhanced Raman scattering (SERS) spectroscopy. A plasmonic substrate with multiple “hotspots” for highly sensitive detection of pesticide residues were prepared successfully by assembling gold nanoparticles on buckypaper (AuNPs-BP). The substrate exhibited high SERS enhancement and excellent detection sensitivity, with a detection limit (LOD) of 2.03 × 10 -11 M and 6.88 × 10 -12 M for the probe molecule R6G and MB, respectively. Combined with 3D finite-difference time-domain (3D-FDTD) simulation, the excellent SERS performance of the substrate was attributed to the enhancement of the electromagnetic field around the “hotspots”. Additionally, the substrates exhibited excellent flexibility, allowing easy contact with irregular surfaces and facilitating the collection of target molecules on the sample surface. Using a portable Raman spectrometer, the substrate achieved in situ analysis of chlorpyrifos residues on peach, with a LOD as low as 6.8 × 10 -11 M. The method showed high accuracy, with a recovery value ranging from 94.2 % to 115.5 %. The results indicate that the substrate has great potential for rapid and highly sensitive detection of pollutants, especially on non-planar surfaces.

Interfacial assembly enables bilayer plasmonic SERS films; • Dynamic hotspot regulation governs multilayer SERS performance; • Ultrasensitive quantitative detection demonstrated in serum. Interfacial assembly of plasmonic nanostructures provides a versatile strategy for engineering Surface-enhanced Raman Spectroscopy (SERS) substrates, yet the fundamental mechanism governing multilayer SERS enhancement remains poorly understood. Here, we report interfacially assembled bilayer plasmonic films with precisely controlled Au nanoparticle layer numbers and uncover a previously unrecognized nonmonotonic enhancement behavior, in which the bilayer configuration unexpectedly outperforms both monolayer and multilayer counterparts through a dynamic hotspot regulation mechanism. The bilayer architecture exhibits optimal SERS performance, achieving ultralow limits of detection of 1.05 × 10 −14 M for crystal violet (CV) and 1.0 × 10 −11 M for paclitaxel (PTX) with excellent reproducibility (RSD < 5%). Time-resolved SERS measurements and Raman mapping reveal that solvent-driven analyte enrichment and interlayer nanogap evolution generate temporally stabilized plasmonic hotspots, producing a robust signal plateau window and establishing a direct structure–dynamics–performance relationship fundamentally distinct from conventional static hotspot amplification models. Benefiting from this mechanism, the bilayer films enable quantitative PTX detection in human serum with a limit of detection of 6.9 × 10 −10 M and show strong agreement with LC–MS analysis in 50 clinical samples (R 2 = 98.78%). Furthermore, in an orthotopic breast cancer mouse model, SERS-guided dose adjustment maintains antitumor efficacy while reducing systemic toxicity. This work not only provides insights into a previously unexplored mechanism of hotspot regulation in multilayer plasmonic assemblies but also provides a mechanistically guided interfacial engineering strategy for designing robust SERS substrates for ultrasensitive quantitative detection in complex biological environments. Scheme 1. Dynamic hotspot regulation arising from interlayer nanogap evolution and analyte enrichment establishes a structure–dynamics–performance correlation in bilayer plasmonic films. This mechanism produces a stable signal plateau window and enables ultrasensitive and quantitative paclitaxel monitoring for precision chemotherapy. Engineering plastics are essential to modern industry, yet their recycling remains challenging because conventional processes are energy-intensive and often degrade material value. Polycarbonate (PC), a representative engineering plastic, exemplifies this problem due to its widespread use and growing waste accumulation. Here, we propose a hotspot-catalytic site synergy strategy for the photothermal closed-loop upcycling of waste PC into high-performance flame-retardant composites. In the designed CNT-PEI hybrid, carbon nanotubes generate localized photothermal hotspots under light irradiation, while polyethyleneimine provides amine-rich catalytic sites for carbonate-bond cleavage. This synergy markedly accelerates depolymerization under mild conditions, affording bisphenol A (BPA) in >90% yield within 40 min under a light intensity of 400 mW/cm 2 , with depolymerization efficiency improved by 138.0% and 54.8% relative to thermal depolymerization and CNT-based photothermal depolymerization, respectively. Furthermore, the recovered BPA was repolymerized into PC, followed by blending with recovered CNT-PEI to fabricate regenerated PC/CNT-PEI composites. As expected, PC/CNT-PEI exhibit a flame-retardant rating of UL-94 V-0, 18.9% higher tensile strength, and 71.0% higher thermal conductivity than pristine PC. Moreover, the PC/CNT-PEI composites can be depolymerized again to BPA in 97.5% yield, demonstrating a closed-loop upcycling process. This work provides an energy-efficient strategy for upcycling waste PC into high-performance flame-retardant materials.

Tailwater treatment from high-standard plastic-cleaning wastewater and red mud (RM) resource recycling pose major challenges. In this study, a heterogeneous microwave-responsive catalyst, RM/biochar (RM/BC), was successfully prepared to synergistically integrate magnetic and dielectric losses for activation of sodium percarbonate (SPC) to degrade the plastic additive diethyl phthalate (DEP). Characterization analyses revealed that under oxygen-limited conditions at 600 °C, poplar leaf powder was fully carbonized, while Fe 2 O 3 in RM was reduced to FeO and further transformed into Fe 3 O 4 , forming a composite dominated by amorphous carbon and Fe 3 O 4 . In RM/BC-600, Fe 3 O 4 and amorphous carbon collaboratively enabled dual hot-spot effects under microwave irradiation via magnetic and dielectric losses, accompanied by enriched oxygen vacancies. Hydroxyl radicals (•OH) were identified as the dominant reactive oxygen species (ROS) in SPC activation. Through single-factor and response surface optimization, under conditions of 40 μM DEP concentration, 4.03 mM SPC dosage, 6.24 g/L RM/BC-600 dosage, 87.2 °C reaction temperature, and 100 W microwave power, the optimal DEP removal efficiency reached 96.1 %. In-situ FTIR/Raman spectroscopy and EPR analysis demonstrated that RM/BC primarily facilitated SPC decomposition to hydrogen peroxide, with oxygen vacancies catalyzing hydrogen peroxide to generate •OH as the main ROS, supplemented by carbonate radicals (•CO 3 − ), superoxide radicals (•O 2 − ), and singlet oxygen ( 1 O 2 ), while dual hot-spot effects accelerated this process. HPLC-QTOF-MS identified 12 intermediate products, and plausible degradation pathways were proposed. ECOSAR software predicted reduced acute toxicity of degradation products, and seed germination assays confirmed minimal ecotoxicity of RM/BC to soil-plant systems. This study achieves RM resource recovery and hazard mitigation via a green strategy, enhancing plastic wastewater treatment through DEP detoxification and stability.
