Protein from Hylocereus polyrhizus protects MRC-5 cells against hydrogen peroxide (H2O2)-induced damage

Ding Haomiao,Zhang Yuzhe,Zha Yue,Zhou Sijia,Cao Chaoqing,Zhu Huajian,Xu Feng,Zhang Xiuqiang,Wang Caisheng

The cytoprotective and potential molecular mechanisms of Hylocereus polyrhizus protein (RFPP) were investigated on the hydrogen peroxide (H 2 O 2 )-triggered damage in normal human embryonic lung (MRC-5) cells. An MTT assay was conducted to assess the MRC-5 cell viability after exposure to H 2 O 2 or RFPP. Cell cycle distribution and apoptosis were explored via flow cytometry. The contents of related proteins were assessed via western blot. MRC-5 cells exhibited markedly decreased cellular viability after treatment with H 2 O 2 ; however, treatment with RFPP suppressed this decrease. Additionally, RFPP interference dampened H 2 O 2 -triggered intracellular apoptosis levels and increased H 2 O 2 -triggered intracellular S phase. In these processes, the contents of phosphorylated ( p )-AKT along with p -mTOR proteins were downregulated in 120 µM H 2 O 2 -treated cells compared with vehicle-treated cells. Nevertheless, in MRC-5 cells inoculated with RFPP, the levels expression of these proteins were reversed. To conclude, RFPP protected MRC-5 cells from H 2 O 2 -triggered damage via activation of the PI3K/AKT/mTOR cascade.

Chemical Composition of the Cuticle Membrane of Pitaya Fruits (Hylocereus Polyrhizus)

Hua Huang,Yueming Jiang

This study comprehensively analysed the chemical composition of the cuticle in pitaya fruits. The total coverage amount of the waxes versus cutin monomers accumulated at a ratio of 0.6, corresponding to masses per unit of 30.3 μg·cm−2and 50.8 μg·cm−2, respectively. The predominant wax mixtures weren-alkanes in homologous series of C20–C35, dominated by C31and C33; as well as triterpenoids with an abundant amount of uvaol, lupenon, β-amyrinon, and β-amyrin. The most prominent cutin compounds were C16- and C18-type monomers, in which 9(10),16-diOH-hexadecanoic acid and 9,10-epoxy-ω-OH-octadecanoic acid predominated, respectively. The average chain length (ACL) of aliphates in pitaya fruit cuticle (30.5) was similar to that estimated in leaf waxes, and higher than that in most of the fruit and petal waxes that have been reported. We propose that the relatively high ACL and wax/cutin ratio might enhance the cuticular barrier properties in pitaya fruit cuticle to withstand drought. Keywords:pitaya fruit;cuticle;wax;cutin;ACL

HubHLH36 promotes oxalate degradation through HuAAE3 to enhance salt tolerance in pitaya (Hylocereus polyrhizus)

Weijuan Xu,Jianyi Li,Shuguang Jian,Mingyong Zhang,Yongxia Jia,Kuaifei Xia

Pitaya ( Hylocereus polyrhizus ) is one of the subtropical and tropical fruits, has gained global cultivation interest due to its nutritional, medicinal, and economic value. To improve the adaptability of pitaya on islands or soil salinization, it is of great significance to explore the salt tolerance mechanisms of pitaya. While oxalate has been conventionally characterized as an antinutrient metabolite, emerging evidence underscores its dual role as a stress signaling molecule through homeostatic regulation. Here, we demonstrate that dynamic control of oxalate catabolism is essential for pitaya's salt adaptation. Transcriptomic profiling revealed coordinated upregulation of the oxalate-degrading gene HuAAE 3 under salt stress, accompanied by enhanced oxalate accumulation. Notably, the transcription factor HubHLH36 emerged as a central regulator responding to both ionic stress and exogenous oxalate. Molecular validation through yeast one-hybrid and dual-luciferase assays unveiled confirmed that HubHLH36 directly activates HuAAE3 transcription by binding to its promoter. Transient overexpression of HuAAE3 or HubHLH36 in pitaya plants significantly reduced oxalate levels, enhanced salt/oxalate tolerance, and alleviated reactive oxygen species (ROS) accumulation. Conversely, virus-induced gene silencing (VIGS) of these genes elevated oxalate content, compromised stress tolerance, and exacerbated ROS production. Heterologous expression in Arabidopsis further validated the conserved function of HubHLH36-HuAAE3 module in conferring salt/oxalate tolerance with ROS mitigation. Our findings establish a novel transcriptional regulatory module where HubHLH36 modulates oxalate homeostasis via HuAAE3 activation, thereby mitigating oxidative stress under salt stress. These mechanistic insights provide genetic targets and theoretical frameworks for developing salt-tolerant pitaya cultivars, offering promising solutions for sustainable agriculture in saline environments.

Hylocereus polyrhizus Pulp Residues Polysaccharide Alleviates High-Fat Diet-Induced Obesity by Modulating Intestinal Mucus Secretion and Glycosylation

Guanghui Li,Kit-Leong Cheong,Yunhua He,Ahluk Liew,Jiaxuan Huang,Chen Huang,Saiyi Zhong,Malairaj Sathuvan

AlthoughHylocereus polyrhizuspulp residues polysaccharides (HPPP) have shown potential in improving metabolic disorders and intestinal barrier function, the mechanism by which they exert their effects through regulating O-glycosylation modifications in the mucus layer remains unclear. Therefore, this study established a HFD-induced obese colitis mouse model (n= 5 per group) and combined nano-capillary liquid chromatography-tandem mass spectrometry (nanoLC-MS/MS) technology to quantitatively analyze the dynamic changes in O-glycosylation. Additionally, through quantitative O-glycosylation proteomics and whole-proteome analysis, we identified 155 specifically altered O-glycosylation sites in colon tissue, with the glycosylation modification level of the MUC2 core protein increased by approximately 2.1-fold. The results indicate that HPPP alleviates colonic mucosal damage by regulating interactions between mucus O-glycosylation. Overall, we demonstrated that HPPP increases HFD-induced O-glycosylation sites, improves intestinal mucosal structure in obese mice, and provides protective effects against obesity-induced intestinal mucosal damage.
