The ultrasonic-assisted extraction of pigments from the Hylocereus undatus flowers was modeled using response surface methodology. A three-level, three-factor Box–Behnken design was employed to optimize three extraction variables, including liquid–solid ratio (X1), extraction time (X2), and ethanol concentration (X3), for the achievement of high absorbance. The statistical analysis shows that the model is significant to optimize the extraction variables (p < 0.01), and the independent variable (X3) and the quadratic term (X32) have a significant effect on the absorbance (p < 0.01). The optimized conditions are X1 of 42 : 1 mL g−1, X2 of 33.2 min, and X3 of 77.5%. Under these conditions, the experimental absorbance is 2.240 ± 0.095% (n = 3), which is in good agreement with the predicted value of 2.243. The evaluation of antioxidant activity by DPPH and hydroxyl free radicals, reducing power, and metal chelating ability indicate that the pigments from the H. undatus flowers possess significant antioxidant activity. HPLC analysis reveals that gallic acid, rutin, quercetin, kaempferol and isorhamnetin are the major components in the pigments.

Pitaya ( Hylocereus undatus ) fruit is susceptible to pathogens that induce fruit decay after harvest. Cuticular waxes covering the outermost plant surfaces are the first site of plant-environment interactions; however, their changes in response to fruit decay are under-researched. Herein, the surface microstructures and chemical composition of wax on pitayas, as well as their changes in response to fruit decay, were comparatively studied. The varieties ‘Baishuijing’ (BSJ) and ‘Hongshuijing’ (HSJ) were examined after harvest. The fruit appearance, morphology, and wax composition differed between cultivars. The surface of both pitaya cultivars was smooth, although traces of granules and a thicker lipid layer were observed using histochemical staining in HSJ than in BSJ, as well as a higher cuticular wax content. Fruit decay rate increased rapidly in both cultivars under room storage conditions, and greater sensitivity to pathogens was found in BSJ than HSJ. As well as the occurrence of fruit decay, the total wax coverage, very-long-chain primary alcohols, aldehydes, fatty acids, and triterpenoids decreased in BSJ, while remaining relatively stable in HSJ. Correspondingly, the relative gene expression levels of HuKCS11 (−1, 2, and 3) , HuKCS17 , and HuKCS19 (for the elongation of very-long-chain fatty acids), HuCER1 (for aldehydes or n -alkanes), HuFAR (−1 and 2) (for primary alcohols), HuBAS1 (−1, 3, and 4) (for β-amyrin), and HuLUS1 (for lupeol) were considerably modified following fruit senescence during storage. The difference in surface topography and wax accumulation between cultivars might have affected pathogen susceptibility after harvest. These results may help when choosing varieties of pitaya based on surface characteristics and cuticular chemicals. They may also help to optimize strategies for postharvest preservation and storage.

Dragon fruit is a tropical fruit with good taste. It can bring health benefits to human body. As one of the major bioactive components in this fruit, the polysaccharides might contribute to the health benefits. However, the precise structure information remains unknown. A leading polysaccharide of dragon fruit pulp, DFPP, was purified and identified by NMR and GC–MS. →4-β- d -Glc p A-1→, →6-β- d -Gal p -1→ and →4-α- l -Rha p -1→ constituted the backbone and α- l -Ara f -1 → 5-α- l -Ara f -1→ formed the branch chain. The precise structure was putatively identified as below. The molecular weight was 2.2 × 10 3 kDa. The structure information of polysaccharides will be helpful to understand this fruit. Insulin resistance and hepatic steatosis are the most common complications of obesity. Pitaya is an important source of phytochemicals such as polyphenols, flavonoid and vitamin C which are related to its antioxidant activity. The present study was conducted to evaluate the influence of white pitaya juice (WPJ) on obesity-related metabolic disorders (e.g. insulin resistance and hepatic steatosis) in high-fat diet-fed mice. Forty-eight male C57BL/6J mice were assigned into four groups and fed low-fat diet with free access to water or WPJ, or fed high-fat diet with free access to water or WPJ for 14 weeks. Our results showed that administration of WPJ improved high-fat diet-induced insulin resistance, hepatic steatosis and adipose hypertrophy, but it exerted no influence on body weight gain in mice. Hepatic gene expression analysis indicated that WPJ supplement not only changed the expression profile of genes involved in lipid and cholesterol metabolism ( Srebp1 , HMGCoR , Cpt1b , HL , Insig1 and Insig2 ) but also significantly increased the expression levels of FGF21 -related genes ( Klb , FGFR2 , Egr1 and cFos ). In conclusion, WPJ protected from diet-induced hepatic steatosis and insulin resistance, which was associated with the improved FGF21 resistance and lipid metabolism.

Natural bioactive compounds in food have been shown to be beneficial in preventing the development of obesity, diabetes, and other metabolic diseases. Increasing evidence indicates that betacyanins possess free-radical-scavenging and antioxidant activities, suggesting their beneficial effects on metabolic disorders. The main objective of this study was to isolate and identify the betaycanins from Hylocereus undatus (white-fleshed pitaya) peel and evaluate their ability to ameliorate obesity, insulin resistance, and hepatic steatosis in high-fat-diet (HFD)-induced obese mice. The purified pitaya peel betacyanins (PPBNs) were identified by liquid chromatography/tandem mass spectrometry (LC/MS/MS), and the male C57BL/6 mice were fed a low-fat diet, HFD, or HFD supplemented with PPBNs for 14 weeks. Our results showed that the white-fleshed pitaya peel contains 14 kinds of betacyanins and dietary PPBNs reduced HFD-induced body weight gain and ameliorated adipose tissue hypertrophy, hepatosteatosis, glucose intolerance, and insulin resistance. Moreover, the hepatic gene expression analysis indicated that PPBN supplementation increased the expression levels of lipid-metabolism-related genes (AdipoR2, Cpt1a, Cpt1b, Acox1, PPARγ, Insig1, and Insig2) and FGF21-related genes (β-Klotho and FGFR1/2) but decreased the expression level of Fads2, Fas, and FGF21, suggesting that the protective effect of PPBNs might be associated with the induced fatty acid oxidation, decreased fatty acid biosynthesis, and alleviated FGF21 resistance.

Polysaccharides isolated from the peel ofHylocereus undatusexhibit promising anti-inflammatory activity; however, the underlying mechanisms—particularly their modulatory effects on cutaneous microbiota composition and host immune responses—remain incompletely characterized. This study investigates the therapeutic potential of polysaccharides isolated from the peel ofHylocereus undatusin the management of inflammatory cutaneous wounds. The polysaccharide extracted from the peel ofHylocereus undatusvia ultrasound-assisted extraction is an acidic heteropolysaccharide, with galacturonic acid and rhamnose as its dominant monosaccharide components. It exhibits low crystallinity, a porous structure, and good thermal stability. In a mouse wound model, treatment with the polysaccharide extracted from the peel ofHylocereus undatussignificantly accelerated wound closure as early as day 3 (**p< 0.01). By day 9, the wound closure rate approached that of the positive control group and remained significantly higher than that of the untreated group (**p< 0.01), exceeding 90%. Treatment with the polysaccharide advanced the inflammatory peak, as evidenced by elevated anti-inflammatory cytokines (IL-10 and TGF-β) and suppression of the pro-inflammatory cytokine IL-6. Immunofluorescence staining confirmed that polysaccharide promoted cell proliferation and neovascularization at the wound site. In conclusion, polysaccharides isolated from the peel ofHylocereus undatusaccelerate skin wound healing by modulating the skin microbiota, enhancing the anti-inflammatory response, and promoting tissue regeneration, highlighting its potential as a natural wound dressing.
