In China, Hyphantria cunea (Drury) is an invasive phytophagous pest; it attacks nearly all species of defoliated trees. To develop integrated pest management programs (IPM) for H. cunea, we need to ensure the availability of insects by mass-rearing them on artificial diets under laboratory conditions. This study compared the growth characteristics, nutritional indices, growth indices, and digestive enzyme activity of insects reared on Pterocarya stenoptera C.DC (Fagales: Juglandaceae), the Chinese wingnut, and an artificial diet. We also investigated the correlation between diet components and growth indices using principal components analysis and Pearson correlation analysis. We found that mass-rearing of H. cunea on an artificial diet was feasible. It led to a shorter developmental period, with heavier larvae and pupae than natural diets. The principal components analysis indicated that the growth indices and α-Amylase were significantly positively associated with PC1, which explained 82.45% of the total data variability. Pearson correlation analysis showed a significant correlation between digestion, absorption parameters, and growth. Developing a mass-rearing program to produce H. cunea on an artificial diet will be valuable for improving IPM strategies. Understanding the mechanism of the responses of phytophagous insect populations to anthropogenic diet regulation can provide new ideas and methods for pest control.

The toxicity of heavy metals on various trophic levels along the food chain has been extensively investigated, but no studies have focused on parasitic natural enemy insects. Herein, we constructed a food chain consisting of soil– Fraxinus mandshurica seedlings– Hyphantria cunea pupae– Chouioia cunea to analyze the effects of Cd exposure through food chain on the fitness of parasitic natural enemy insects and its corresponding mechanism. The results showed that the transfer of Cd between F. mandshurica leaves and H. cunea pupae and between H. cunea pupae and C. cunea was a bio-minimization effect. After parasitizing Cd-accumulated pupa, the number of offspring larvae, and the number, individual size (body weight, body length, abdomen length) and life span of offspring adults decreased significantly, while the duration of embryo development extended significantly. The contents of malondialdehyde and H 2 O 2 in Cd-exposed offspring wasps increased significantly, accompanied by a significantly decrease in antioxidant capacity. The cellular immunity parameters significantly decreased in Cd-accumulated pupae, including the number of hemocytes, melanization activity and the expression level of cellular immunity genes (e.g. Hemolin-1 and PPO1 ). The humoral immunity disorder was found in the Cd-accumulated pupae, as evidenced by that the expression level of immune recognition gene ( PGRP-SA ), signal transduction genes ( IMD , Dorsal , and Tube ), as well as all antimicrobial peptide genes (e.g. Lysozym and Attacin ) decreased significantly. Cd exposure decreased the content of glucose, trehalose, amino acid, and free fatty acid in H. cunea pupae. The expression of Hk2 in glycolysis pathway and the expression of Idh2 , Idh3 , Cs , and OGDH in TCA cycle pathway were significantly down-regulated in Cd-accumulated pupae. Taken together, exposure to Cd through the food chain causes oxidative damage on the offspring wasps and disrupts energy metabolism of the host insect, ultimately reducing the parasitic fitness of C. cunea to H. cunea pupae.

Strong multi-host adaptability significantly contributes to the rapid dissemination of Hyphantria cunea. The present study explores the involvement of cytochrome P450 monooxygenase (P450) in the multi-host adaptation of H. cunea and aims to develop RNA pesticides targeting essential P450 genes to disrupt this adaptability. The results showed that inhibiting P450 activity notably reduced larval weight and food-intake across seven plants groups. The P450 gene HcCYP6AE178 was highly upregulated in H. cunea larvae from medium- and low-preference host plant groups. Silencing HcCYP6AE178 significantly decreased H. cunea larval body weight, increased larval mortality, inhibited energy metabolism genes expression and interfered with growth regulatory genes expression. Overexpression of HcCYP6AE178 enhanced the tolerance of Drosophila and Sf9 cells to the plant defensive substances cytisine and coumarin. The RNA pesticide CS-ds HcCYP6AE178 constructed using chitosan (CS) exhibited remarkable stability. Treatment with CS-ds HcCYP6AE178 effectively reduced H. cunea larval body weight, heightened larval mortality, and disrupted growth regulatory genes expression in low-preference host plant groups. Combined treatment of CS-ds HcCYP6AE178 and coumarin significantly elevated H. cunea larval mortality compared to coumarin alone, accompanied by the inhibition of growth regulatory genes expression and an abnormal increase in energy metabolism genes expression. Taken together, HcCYP6AE178 is essential for the adaptation of H. cunea to multiple host plants, and RNA pesticides targeting HcCYP6AE178 can effectively impair the performance of H. cunea in different host plants.

The fall webworm Hyphantria cunea (Lepidoptera: Erebidae) is an invasive pest that severely damages
forest resources and disrupts ecological stability. Chlorantraniliprole
(CAP) is widely applied to control this pest, yet conventional formulations
exhibit poor foliar retention, droplet spreading, and rainfastness.
To address these limitations, we prepared tannic acid (TA)-modified
mesoporous silica nanoparticles (MSN) loaded with CAP (TA@CAP@MSN)
to enhance insecticidal efficacy and foliar adhesion. TA@CAP@MSN were
uniform mesoporous nanospheres with 162.95 m 2 /g specific
surface area, 0.241 cm 3 /g pore volume, 27.83% drug loading,
and sustained-release performance. After 72 h of exposure, the LC 50 of TA@CAP@MSN against third-instar H. cunea was 2.130 mg/L, corresponding to a 3.66-fold increase in toxicity
relative to technical CAP. When sprayed on Populus × euramericana leaves, this
nanoformulation achieved 62.33% larval mortality at 120 h. Notably,
TA@CAP@MSN exhibited superior retention, wettability, and rainfastness
on three representative host plants with distinct leaf surface properties.
After simulated rainfall, mortalities under LC 50 /LC 80 were significantly higher than those of pure CAP and CAP@MSN.
Sublethal studies revealed that TA@CAP@MSN initially modulated key
detoxification and antioxidant enzymes, followed by suppression of
superoxide dismutase (SOD) and carboxylesterase (CarE) activities,
consistent with altered antioxidant and detoxification responses.
This tannic-acid-functionalized mesoporous silica nanocarrier provides
an interfacial engineering strategy to construct high-efficiency nanopesticides
against H. cunea .

Chlorbenzuron is a kind of benzoylphenylureas (BPUs), which plays a broad role in insect growth regulators (IGRs), with an inhibitory effect on chitin biosynthesis. However, BPUs how to regulate glycolysis and insect growth remains largely unclear. Here, we investigated the effects of chlorbenzuron on growth, nutritional indices, glycolysis, and carbohydrate homeostasis in Hyphantria cunea , a destructive and highly polyphagous forest pest, to elucidate the action mechanism of chlorbenzuron from the perspective of energy metabolism. The results showed that chlorbenzuron dramatically restrained the growth and nutritional indices of H. cunea larvae and resulted in lethality. Meanwhile, we confirmed that chlorbenzuron significantly decreased carbohydrate levels, adenosine triphosphate (ATP), and pyruvic acid (PA) in H. cunea larvae. Further studies indicated that chlorbenzuron caused a significant enhancement in the enzyme activities and mRNA expressions of hexokinase (HK), phosphofructokinase (PFK), and pyruvate kinase (PK), resulting in increased glycolytic flux. Expressions of genes involved in the AMP-activated protein kinase (AMPK) signaling pathway were also upregulated. Moreover, chlorbenzuron had remarkable impacts on H. cunea larvae from the perspective of metabolite enrichment, including the tricarboxylic acid (TCA) cycle and glycolysis, indicating an energy metabolism disorder in larvae. The findings provide a novel insight into the molecular mechanism by which chlorbenzuron abnormally promotes glycolysis and eventually interferes with insect growth and nutritional indices.

Heavy metals are significant environmental pollutants that contribute to widespread contamination. This study demonstrated that Cd transfer through the food chain imparts cross-tolerance to λ-cyhalothrin in Hyphantria cunea larvae. Cd-induced activation of the P450 detoxification system, particularly HcCYP6AE179 and HcCYP9A286, was identified as a critical mechanism for the development of pesticide cross-tolerance in Cd-exposed H. cunea larvae. The dual-luciferase reporter assays, RNAi, and agonist treatment experiment confirmed that HcAhr (Aryl hydrocarbon receptor) upregulated the expression of HcCYP6AE179 and HcCYP9A286. Additionally, H2O2 generation under Cd stress served as a key upstream signal for activating the HcAhr-HcCYP6AE179/HcCYP9A286 detoxification pathway. The nucleic-acid-based pesticides targeting HcAhr, HcCYP6AE179, and HcCYP9A286 exhibited superior stability, biosafety, and gene silencing efficacy. Treatment with these nucleic acid pesticides significantly reduced the tolerance of H. cunea larvae to λ-cyhalothrin in the CK and Cd groups. This work introduces novel approaches for mitigating pesticide cross-tolerance induced by heavy metals.

Heavy metals are prevalent environmental contaminants in pest habitats. This study investigates the tolerance of Hyphantria cunea larvae to β-cypermethrin under cadmium (Cd) stress. Exposure to Cd markedly enhanced larval tolerance to β-cypermethrin. The P450 enzyme was found to be crucial in triggering this cross-tolerance. Notably, Cd exposure significantly upregulated the expression of HcCYP333A29 and HcCYP6AE184 both before and after β-cypermethrin treatment. Silencing these genes reduced the β-cypermethrin tolerance in Cd-exposed larvae. Furthermore, overexpression of HcCYP333A29 and HcCYP6AE184 significantly improved the tolerance of Sf9 cells and Drosophila to β-cypermethrin. Silencing of HcDicer-1, a key gene in the miRNA synthesis pathway, significantly diminished the β-cypermethrin tolerance in Cd-treated larvae. The miR-2739 controlled overexpression of HcCYP333A29, while miR-2854 controlled HcCYP6AE184 overexpression, both of which were essential for β-cypermethrin cross-tolerance in H. cunea larvae under Cd exposure. Collectively, Cd exposure induces cross-tolerance to β-cypermethrin in H. cunea larvae through miRNA-mediated post-transcriptional regulation.

Developing effective insecticidal strategies is an important means of reducing the spread and host plant damage by Hyphantria cunea . In this study, key metabolites with insecticidal activity against H. cunea were screened by targeted metabolomics in Tilia amurensis , a low-preference host plant. Subsequently, the potential of key metabolites that could be used as botanical pesticides was evaluated. The results showed that coumarin was the key insecticidal metabolite of T. amurensis and had a significant insecticidal effect and weight inhibition effect on H. cunea larvae. Coumarin treatment significantly decreased the larval nutrient content and the gene expression of rate-limiting enzymes in the glycolytic pathway and tricarboxylic acid cycle . A significantly enhanced detoxification enzyme activity (CarE and GST), antioxidant oxidase activity (SOD and CAT), non-enzymatic antioxidant levels (GSH), and total antioxidant capacity were observed in coumarin-treated larvae. Coumarin treatment resulted in a significant increase in the expression levels of detoxification enzyme genes ( CarE1 , CarE2 , CarE3 , GST2 , and GST3 ) and antioxidant oxidase genes ( SOD1 , CAT1 , and CAT2 ) in H. cunea larvae. Coumarin treatment significantly increased the levels of MDA and H 2 O 2 in larvae but did not cause pathological changes in the ultrastructure of the larval midgut. Coumarin solution sprayed directly or as a microcapsule suspension formulation with coumarin as the active ingredient had significant insecticidal activity against the H. cunea larvae. Overall, coumarin, a key anti-insect metabolite identified from T. amurensis , can significantly inhibit the growth and survival of H. cunea larvae and has the potential to be developed as a botanical pesticide.

Hyphantria cunea , a globally significant quarantine pest, causes severe ecological and economic impacts in invaded regions. Attractants are environment-friendly and valuable pest management tools for H. cunea control. In this study, we identified that the combination of β-ocimene with sex pheromone components was strongly attractive to H. cunea males. This attractant combination activates the Ras signaling pathway and stimulates Ca 2+ in male adults following exposure. Feeding and oviposition behavior results demonstrated that H. cunea prefers Morus alba over Platanus × acerifolia , Metasequoia glyptostroboides , and Taxodium distichum . In the fresh branches of M. alba with leaves, the relative abundance of β-ocimene (36.33 ± 0.41 %) was significantly higher than the other hosts. Electroantennography recordings, behavioral choice assays, and field trapping trials showed that the combination of β-ocimene with sex pheromone components significantly enhanced male response and attraction. The number of trapped males using β-ocimene/ sex pheromone blend was 3.7-fold higher than that of sex pheromone or β-ocimene alone in the field, indicating a synergistic effect of adding β-ocimene to sex pheromone. The molecular mechanism of β-ocimene/sex pheromone synergism was further analyzed. After exposure to the attractant, the Ras signaling pathway in the heads of males are activated ( HcILP and HcPLCε1 are upregulated, while HcRasGAP and HcPLD are downregulated), which further stimulates the expression of IP3 and Ca 2+ . The activation of Ca 2+ may be the key reason for its higher attraction to males. These findings provide a theoretical basis for the selection and mechanistic understanding of attractants for H. cunea , offering insights for attractant-based pest control strategies.

The remarkable host adaptability of Hyphantria cunea contributes to its invasiveness and destructive impact. This study investigates the molecular mechanisms underlying the multi-host adaptability in H. cunea larvae, focusing on UDP-glucuronosyltransferases (UGT). The results revealed a significant increase in UGT enzyme content in H. cunea larvae feeding on low-preference host plants compared to those on high-preference hosts. UGT inhibitor treatment markedly reduced larval body weight and food intake across all host plant groups. Additionally, qPCR analysis indicated distinct expression patterns of UGT family genes in larvae feeding on different host plants. Notably, HcUGT40R20 was highly expressed specifically in larvae consuming intermediate- and low-preference hosts. Functional analysis showed that silencing HcUGT40R20 led to a significant decrease in larval body weight and disrupted the expression of genes involved in energy metabolism and growth regulation across all host plant groups, along with a marked increase in larval mortality in intermediate- and low-preference hosts. Moreover, the bio-toxicity of the host plant defensive compounds, cytisine and coumarin, was significantly reduced in transgenic Drosophila and Sf9 cells overexpressing HcUGT40R20 . Furthermore, an RNAi-based formulation, CS-ds HcUGT40R20 , was successfully developed using chitosan (CS) nanomaterials and dsRNA, demonstrating stability in the gut fluid of H. cunea . CS-ds HcUGT40R20 treatment significantly reduced the body weight of larvae fed on different host plants and markedly increased the mortality rate of larvae under coumarin stress. In conclusion, HcUGT40R20 plays a critical role in the multi-host adaptability of H. cunea , and RNAi-based formulation targeting HcUGT40R20 provide an effective strategy for controlling H. cunea .

Heavy metal pollution is an abiotic factor that can affect the efficiency of pest control. In this study, two microbial pesticides , Bacillus thuringiensis and Mamestra brassicae nuclear polyhedrosis virus (MbNPV), were used to treat Hyphantria cunea larvae with Cd pre-exposure, and the humoral and cellular immunity of H. cunea larvae with Cd exposure were evaluated. The results showed that Cd exposure increased the susceptibility of H. cunea larvae to microbial pesticides B. thuringiensis and MbNPV, and the lethal effect of Cd exposure and microbial pesticides on H. cunea larvae was synergistic. Cd exposure significantly decreased the expression of pathogen recognition genes (GNBP1 and GNBP3), signal transduction genes (Relish, Myd88, Tube, and Imd), and antimicrobial peptide gene (Lebocin) in the humoral immunity of H. cunea larvae compared with the untreated larvae. Parameters of cellular immunity, including the number of hemocytes , phagocytic activity , melanization activity, encapsulation activity, and the expression of three phagocytic regulatory genes (HEM1, GALE1, GALE2), were also found to decrease significantly in Cd-treated larvae. TOPSIS analysis showed that humoral immunity, cellular immunity, and total immunity levels of H. cunea larvae with Cd exposure were weaker than those in untreated larvae. Correlation analysis showed that the mortality of two microbial pesticides investigated in H. cunea larvae was negatively correlated with the humoral and cellular immunity of larvae. Taken togther, Cd exposure results in immunotoxic effects on H. cunea larvae and the use of microbial pesticides are an effective strategy for pest control in heavy metal-polluted areas.

BACKGROUND Cuticular proteins (CPs) play essential roles in forming cuticular structures in insects. However, the specific functions and regulatory mechanisms of CPs remain largely unexplored. In this study, the Larval cuticular protein 17 ( HcLCP-17 ) gene was identified from Hyphantria cunea , a highly destructive and polyphagous forest pest. To investigate the role of HcLCP-17 in cuticular function and transcriptional regulation mediated by 20E-responsive transcription factors (ERTFs), we employed RNA interference (RNAi) and yeast one-hybrid assay techniques. Additionally, we examined the molecular mechanism by which chlorbenzuron, a type of benzoylphenylurea (BPU) that functions as a chitin synthesis inhibitor (CSI), affects the 20E signaling pathway and ultimately regulates HcLCP-17 expression. RESULTS HcLCP-17 encodes a polypeptide consisting of 393 amino acids, which includes a chitin-binding domain. Silencing HcLCP-17 resulted in a disturbance in the structural organization of the larval cuticle and a notable reduction in chitin levels. HcLCP-17 expression was controlled by the interaction between Broad-Complex (Br-C) and beta Fushi Tarazu Factor-1 (βFTZ-F1) with its promoter fragment. Furthermore, the inhibitory effect of chlorbenzuron on HcLCP-17 expression was found to be potentially mediated by Br-C and βFTZ-F1. CONCLUSION The study presents a novel mode of action for the 20E signaling pathway in regulating the expression of CPs and reveals the potential mode-of-action of BPUs in insect cuticles. These findings provide a theoretical basis for future utilization of LCP-17 as a pesticide target making a significant contribution to the development of effective pest management strategies. © 2024 Society of Chemical Industry.
