Characteristics of Organic Acid Secretion Associated with the Interaction between Burkholderia multivorans WS-FJ9 and Poplar Root System

Guan-Xi Li,Xiao-Qin Wu,Jian-Ren Ye,He-Chuan Yang

The objective of this study was to investigate whether plant-bacteria interaction affects the secretion of organic acids by both organisms and to assess whether the production of IAA by the bacterium increases the secretion of organic acids by root exudates, and if the stress produced by low available phosphorus (P) affects the production of organic acids by bacteria, by roots, or by root exudates in presence of bacterial cultures. With this purpose, we used as a biological model poplar plants and one strain of Burkholderia multivorans able to solubilize P. High performance liquid chromatography was utilized to measure organic acids. The tests, the inductive effects of exogenous indole-3-acetic acid (IAA) on secretion of organic acids, the 2 × 4 × 2 factorial design experiment, and the ability of organic acids to solubilize tricalcium phosphate were performed to investigate the interactive effects. The results showed that, after B. multivorans WS-FJ9 interacted with the poplar root system, the key phosphate-solubilizing driving force was gluconic acid (GA) which was produced in three ways: (1) secreted by the root system in the presence of IAA produced by B. multivorans WS-FJ9; (2) secreted by B. multivorans WS-FJ9; and (3) secreted by the poplar root system in the presence of phosphorus stress. When phosphorus stress was absent, the GA was produced as outlined in (1) and (2) above. These results demonstrated that inoculating B. multivorans WS-FJ9 into the poplar root system could increase the amount of GA secretion and implied that the interaction between B. multivorans WS-FJ9 and the poplar root system could contribute to the increase of P available fraction for poplar plants.

The RhLOL1–RhILR3 module mediates cytokinin-induced petal abscission in rose

Chuyan Jiang,Yue Liang,Shuning Deng,Yang Liu,Haohao Zhao,Susu Li,Cai-Zhong Jiang,Junping Gao,Chao Ma

Summary In many plant species, petal abscission can be considered the final step of petal senescence. Cytokinins (CKs) are powerful suppressors of petal senescence; however, their role in petal abscission is ambiguous. Here, we observed that, in rose ( Rosa hybrida ), biologically active CK is accumulated during petal abscission and acts as an accelerator of the abscission process. Using a combination of reverse genetics, and molecular and biochemical techniques, we explored the roles of a LESION SIMULATING DISEASE1 (LSD1) family member Rh LOL1 interacting with a bHLH transcription factor Rh ILR3 in CK-induced petal abscission. Silencing Rh LOL1 delays rose petal abscission, while the overexpression of its ortholog Sl LOL1 in tomato ( Solanum lycopersicum ) promotes pedicel abscission, indicating the conserved function of LOL1 in activating plant floral organ abscission. In addition, we identify a bHLH transcription factor, Rh ILR3, that interacts with Rh LOL1. We show that Rh ILR3 binds to the promoters of the auxin signaling repressor auxin/indole-3-acetic acid ( Aux/IAA ) genes to inhibit their expression; however, the interaction of Rh LOL1 with Rh ILR3 activates the expression of the Aux/IAA genes including Rh IAA4-1 . Silencing Rh IAA4-1 delays rose petal abscission. Our results thus reveal a Rh LOL1– Rh ILR3 regulatory module involved in CK-induced petal abscission via the regulation of the expression of the Aux/IAA genes.

Intestinal cGAS–STING–IFN signalling promotes obesity by downregulating microbiota-derived IAA in male mice

Deng Jiangming,Meng Wen,Yang Yuanqin,Xiao Ting,Jin Zhangliu,Wang Jing,Li Qing-xin,Xie Lingxiang,Hu Jingyi,Song Rong,Hu Yibo,Chu Yi,Li Qing,Jin Ping,Wang Zheng,Wu Irene XY,Liu Feng,Xiao Yang

The gut constantly interacts with both pathogens and dietary signals, but how it balances immune and metabolic responses remains unclear. Here we show that intestinal cGAS, a key DNA sensor, acts as a regulator linking gut immunity to whole-body metabolism. We show that cGAS signalling is activated in the intestines of humans and male mice with obesity, leading to increased type I interferon production and heightened immune activity in intestinal cells. Strikingly, deleting cGAS specifically in intestinal epithelial cells enhances energy expenditure, protects against diet-induced obesity and improves metabolic health. These effects depend on the gut microbiota, particularly Lactobacillus murinus and its metabolite indole-3-acetic acid (IAA), which promotes adipose thermogenesis. Our findings position intestinal cGAS as a key driver of obesity through gut-to-fat signalling and suggest that targeting the intestinal cGAS–microbiota IAA axis could offer promising strategies to combat obesity and related metabolic diseases.

Characterization and synthesis of indole-3-acetic acid in plant growth promoting Enterobacter sp.

Bi-Xian Zhang,Pei-Shan Li,Ying-Ying Wang,Jia-Jun Wang,Xiu-Lin Liu,Xue-Yang Wang,Xiao-Mei Hu

Indole-3-acetic acid (IAA) plays an important role in the growth and development of plants. In this study, a series of predominant strains were isolated and identified as Enterobacter sp. with remarkable IAA-producing capabilities. The IAA-producing strains are mainly tryptophan-dependent and have significantly high yields of IAA (3477 μg mL−1 and 3378 μg mL−1). The ipdC gene encoding indole-3-pyruvate decarboxylase was identified by genomic analysis and RT-qPCR analysis, indicating the involvement of the indole-3-pyruvic acid (IPyA) pathway of IAA biosynthesis. The IPyA pathway was also confirmed by the intermediate assay. The IAA product of microbial metabolites was isolated, purified and characterized. These microbes exhibiting IAA production significantly promoted the growth of maize, increasing root length, plant height, fresh weight and dry weight. Thus, Enterobacter sp. with high IAA production has great prospects in agricultural and industrial applications.

RgTAA1-RgYUCCA6-IAA axis mediates hydrogen gas-induced adventitious root formation in Rehmannia glutinosa root cuttings

Linlin Xiao,Lijuan Yang,Meng Liu,Chengsi Lv,Xingyu Li,Hongying Duan,Jiuchang Su

H 2 could induce AR formation in R. glutinosa root cuttings. • H 2 elevates auxin levels via upregulating RgTAA1 and RgYUCCA6 in R. glutinosa . • H 2 -induced R. glutinosa AR formation depends on auxin synthesis and polar transport. • H 2 modulates multiple rooting processes relying on auxin signalling. Adventitious root (AR) formation is critical for the vegetative propagation and yield of Rehmannia glutinosa , a valuable medicinal plant. However, the molecular mechanisms governing R. glutinosa AR formation have not been documented previously. Here, we provide the first evidence that RgTAA1-RgYUCCA6-IAA axis mediated the hydrogen gas (H 2 )-induced AR formation in R. glutinosa root explants. Phenotypic and transcriptomic analysis demonstrated that H 2 triggered R. glutinosa AR emergence, and auxin biosynthesis genes ( RgYUCCA6 ), transport gene ( RgPIN-LIKES ), and early auxin response genes ( RgSAUR71 ) might be the target gene for H 2 . Next, we found that R. glutinosa AR exhibited an increase in endogenous H 2 and auxin levels during AR formation, and exogenous H 2 increased auxin levels. However, the application of 1-naphthylphthalamic acid and L -kynurenine ( L -kyn) blocked the auxin accumulation and AR formation. Further, H 2 upregulated the expression of RgTAA1 and RgYUCCA6 , leading to increased accumulation of indole-3-pyruvic acid (IPA) and IAA, but these were suppressed by L -kyn application. Meanwhile, H 2 substantially promotes AR formation in R. glutinosa by triggering AR primordium development, remodeling cell wall components, regulating rooting-related enzyme activities, maintaining cell viability, and all these processes are strictly dependent on auxin biosynthesis and polar transport. This is the pioneering study documenting the H 2 -induced R. glutinosa AR formation, shedding new light on the improvement of its vegetative propagation efficiency, and deepening our understanding of gasotransmitter-phytohormone crosstalk in plants. Indole-3-Acetic Acid Biosynthesis Pathways in the Plant-Beneficial Bacterium Arthrobacter pascens ZZ21

Mengsha Li,Rui Guo,Fei Yu,Xu Chen,Haiyan Zhao,Huixin Li,Jun Wu

Arthrobacter pascensZZ21 is a plant-beneficial, fluoranthene-degrading bacterial strain found in the rhizosphere. The production of the phytohormone indole-3-aectic acid (IAA) by ZZ21 is thought to contribute to its ability to promote plant growth and remediate fluoranthene-contaminated soil. Using genome-wide analysis combined with metabolomic and high-performance liquid chromatography-mass spectrometry (HPLC-MS) analyses, we characterized the potential IAA biosynthesis pathways inA. pascensZZ21. IAA production increased 4.5-fold in the presence of 200 mg·L−1tryptophan in the culture medium. The transcript levels ofprrandaldH, genes which were predicted to encode aldehyde dehydrogenases, were significantly upregulated in response to exogenous tryptophan. Additionally, metabolomic analysis identified the intermediates indole-3-acetamide (IAM), indole-3-pyruvic acid (IPyA), and the enzymatic reduction product of the latter, indole-3-lactic acid (ILA), among the metabolites of ZZ21, and subsequently also IAM, ILA, and indole-3-ethanol (TOL), which is the enzymatic reduction product of indole-3-acetaldehyde, by HPLC-MS. These results suggest that the tryptophan-dependent IAM and IPyA pathways function in ZZ21.Keywords:Arthrobacter;IAA biosynthesis pathway;genome-wide analysis;metabolomics;HPLC-MS

Rhizosphere microbes enhance plant resistance to cadmium through a root ROS-microbial IAA-root DNA methylation interkingdom signaling pathway

Xihui Xu,Yue Dou,Shangjun Zhao,Chunfang Zhao,Yahua Chen,Mingyi Jiang,Zhenguo Shen,Chen Chen

Plants deploy a “cry-for-help” strategy to recruit beneficial microbes, thereby enhancing stress resistance and expanding their ecological niches in harsh environments. However, the molecular mechanisms driving this plant-microbe communication remain poorly understood. To uncover the underlying dialogue, we explore interactions between two plant-growth-promoting bacteria (PGPBs) and their host plants, rice and Solanum nigrum , under heavy metal (HM) stress. We identify an interkingdom signaling pathway, characterized by “root reactive oxygen species (ROS)-microbial indole-3-acetic acid (IAA)-root DNA methylation,” which mediates plant-PGPB interactions. Under HM stress, root-derived ROS triggers IAA synthesis in PGPBs, which subsequently suppresses ROS production in the roots, thereby mitigating HM damage in plants. Furthermore, PGPB-derived IAA induces root DNA methylation modifications via ROS-dependent pathways, establishing a cross-kingdom signaling mechanism that links microbial metabolism to plant DNA methylation. This interkingdom signaling pathway is widely observed in the rhizosphere, providing insights into plant-microbe interactions in adverse environments.

CsTCP14-CsIAA4 module-mediated repression of auxin signaling regulates citrus somatic embryogenesis

Peng-Bo Wang,Yao-Yuan Duan,Ru-Meng Quan,Meng-Qi Feng,Jie Ren,Yong-Yu Tang,Mei Qing,Kai-Dong Xie,Wen-Wu Guo,Xiao-Meng Wu

Summary Somatic embryogenesis (SE) is an important in vitro regeneration approach for plants, especially in biotechnological manipulations. However, SE capability is difficult to modulate and deteriorates over time. Glycerol medium is effective in SE induction of citrus, while the mechanisms remain unclear. We found that auxin signaling reduced soon after the citrus embryogenic callus (EC) was transferred to glycerol medium, and the expression of CsTCP14 and AUX/IAA gene CsIAA4 was induced by glycerol. Overexpression of CsIAAm that encodes a stable indole-3-acetic acid (IAA) protein suppressed auxin signaling in EC and enhanced SE. CsTCP14 bound to the promoter of CsIAA4 and activate CsIAA4 expression in EC with strong SE competence. Overexpression of CsTCP14 activated CsIAA4 expression and reduced auxin signaling in citrus EC, and thus enhanced SE. Application of exogenous IAA inhibits SE, while the auxin signaling inhibitor p -chlorophenoxyisobutyric acid (PCIB) enhances SE in citrus. The SE enhancement effect of CsIAA4 and CsTCP14 overexpression on EC was alleviated by exogenous IAA, but reinforced by PCIB. We uncover the regulatory pathway of CsTCP14-CsIAA4 module-mediated repression of auxin signaling in glycerol-induced citrus SE, which deepens our understanding of SE mechanisms in plants and supports modulation of SE in citrus breeding via biotechnological approaches.

Multi-Omics analysis reveals the important role of indole-3-acetic acid homeostasis in male fertility of cotton

Tao Zhang,Li-hong Ma,Qian-hao Zhu,Xin-yu Zhang,Chu-yi Nie,Jing-ya Zhou,Jie Sun,Feng Liu

Auxin (Indole-3-acetic acid, IAA) is essential for anther development and male fertility, but little is known about its homeostasis in developing anthers due to its complex and diverse synthesis pathways. Here, we employed transcriptomic, targeted metabolomic, and amino acid analyses on a wild type (WT) cotton ( Gossypium hirsutum ) line SD98–6 and its nearly-isogenic male sterile (MS) line SD98–6A, to delve into the relationship between IAA and anther fertility. In WT anthers, IAA homeostasis was achieved mainly through self-synthesis, conjugate hydrolysis, and polar transport, and accumulation of IAA exhibited an inverted "U" pattern with the highest content observed at the tetrad stage. In MS anthers, no IAA accumulation was observed after the meiosis stage and the IAA content remained constantly low, mainly due to reduction of the substrates (phenylalanine and tryptophan) for IAA synthesis, and significantly decreased expression of the genes involved in IAA synthesis. Meanwhile, carbohydrate metabolism pathways directly contributing to IAA synthesis and synthesis of amino acids were malfunctional in MS anthers based on the expression profile of the metabolic genes and content measurement. These results provide valuable insights into the IAA homeostasis in cotton anthers and imply that genes related to IAA homeostasis are potential targets for development of IAA-sensitive male sterile cotton lines.

The Characterization of an Efficient Phenylpyruvate Decarboxylase KDC4427, Involved in 2-Phenylethanol and IAA Production from Bacterial Enterobacter sp. CGMCC 5087

Wenzhi Bao,Xing Li,Jinfeng Liu,Rong Zheng,Lijuan Liu,Haibo Zhang

Phenylpyruvate decarboxylase (PPDC) is a crucial enzyme that plays important roles in 2-phenylethanol (2-PE) biosynthesis. In our previous study, we screened a highly efficient PPDC KDC4427 from the novel 2-PE-producing strain Enterobacter sp. CGMCC 5087. Meanwhile, its decarboxylation activity of indolylpyruvate (IPyA) was also higher than other indolylpyruvate decarboxylases (IPDCs) reported so far. In this study, KDC4427 protein was purified and characterized, and its catalytic mechanisms were analyzed by biological methods. The optimum pH and temperature of KDC4427 was pH 6.5 and 35°C, respectively. The enzyme activity was relatively stable between pH 6 and 8 and over the range of temperatures from 25°C to 45°C. KDC4427 showed the highest catalytic efficiency on phenylpyruvic acid (PPA); meanwhile, it also showed high activity for IPyA and 2-ketobutanoic acid, and it was found that KDC4427 belongs to IPDCs by phylogenetic tree analysis. The coverage of the three-dimensional structure of KDC4427 and EcIPDC from Enterobacter cloacae was 96%. Leucine 542, one of the residues in the substrate-binding pocket, is replaced by isoleucine in KDC4427 compared with EcIPDC. Site-directed mutagenesis showed that the transition from leucine to isoleucine was unlikely to make KDC4427 have high catalytic activity for PPA and IPyA; the mutants at glutamate 468 almost completely lost catalytic activities for both PPA and IPyA, indicating that this glutamate was essential for the catalytic activity. Additionally, alanine 387 plays an important role in the substrate selectivity of KDC4427.IMPORTANCE Compared with the chemical synthesis of 2-phenylethanol (2-PE) by condensation of ethylene oxide and benzene, the biological synthesis of 2-PE is a potential method to replace the traditional process. This makes biotransformation gradually become the main way to produce high-quality 2-PE. Phenylpyruvate decarboxylase (PPDC) is the critical enzyme in 2-PE biosynthesis, and it is a momentous point of penetration to increase the production of 2-PE. In this regard, KDC4427 can catalyze phenylpyruvic acid (PPA) to phenylacetaldehyde more efficiently than any other PPDC previously reported. Moreover, it has high activity of indolepyruvate decarboxylases (IPDCs), which will be a great breakthrough in the synthesis of indole-3-acetic acid (IAA). With this study, we offer insights into the KDC4427 catalytic mechanism and significantly expand the toolbox of available α-ketoacid decarboxylases for application in biosynthesis.

A peroxisomal cinnamate:CoA ligase-dependent phytohormone metabolic cascade in submerged rice germination

Yukang Wang,Gaochen Jin,Shuyan Song,Yijun Jin,Xiaowen Wang,Shuaiqi Yang,Xingxing Shen,Yinbo Gan,Yuexing Wang,Ran Li,Jian-Xiang Liu,Jianping Hu,Ronghui Pan

The mechanism underlying the ability of rice to germinate underwater is a largely enigmatic but key research question highly relevant to rice cultivation. Moreover, although rice is known to accumulate salicylic acid (SA), SA biosynthesis is poorly defined, and its role in underwater germination is unknown. It is also unclear whether peroxisomes, organelles essential to oilseed germination and rice SA accumulation, play a role in rice germination. Here, we show that submerged imbibition of rice seeds induces SA accumulation to promote germination in submergence. Two submergence-induced peroxisomal Oryza sativa cinnamate:CoA ligases (OsCNLs) are required for this SA accumulation. SA exerts this germination-promoting function by inducing indole-acetic acid (IAA) catabolism through the IAA-amino acid conjugating enzyme GH3. The metabolic cascade we identified may potentially be adopted in agriculture to improve the underwater germination of submergence-intolerant rice varieties. SA pretreatment is also a promising strategy to improve submerged rice germination in the field.

AUXIN RESPONSE FACTOR7 integrates gibberellin and auxin signaling via interactions between DELLA and AUX/IAA proteins to regulate cambial activity in poplar

Hu Jian,Su Huili,Cao Hui,Wei Hongbin,Fu Xiaokang,Jiang Xuemei,Song Qin,He Xinhua,Xu Changzheng,Luo Keming

Cambial development in the stems of perennial woody species is rigorously regulated by phytohormones. Auxin and gibberellin (GA) play crucial roles in stimulating cambial activity in poplar (Populus spp.). In this study, we show that the DELLA protein REPRESSOR of ga1-3 Like 1 (RGL1), AUXIN RESPONSE FACTOR 7 (ARF7), and Aux/INDOLE-3-ACETIC ACID 9 (IAA9) form a ternary complex that mediates crosstalk between the auxin and GA signaling pathways in poplar stems during cambial development. Biochemical analysis revealed that ARF7 physically interacts with RGL1 and IAA9 through distinct domains. The arf7 loss-of-function mutant showed markedly attenuated responses to auxin and GA, whereas transgenic poplar plants overexpressing ARF7 displayed strongly improved cambial activity. ARF7 directly binds to the promoter region of the cambial stem cell regulator WOX4 to modulate its expression, thus integrating auxin and GA signaling to regulate cambial activity. Furthermore, the direct activation of PIN-FORMED 1 expression by ARF7 in the RGL1–ARF7–IAA9 module increased GA-dependent cambial activity via polar auxin transport. Collectively, these findings reveal that the crosstalk between auxin and GA signaling mediated by the RGL1–ARF7–IAA9 module is crucial for the precise regulation of cambial development in poplar.
