For decades, infectious bronchitis (IB), caused by the infectious bronchitis virus (IBV), has posed a significant threat to poultry health and is mainly controlled by vaccines. However, the currently available commercial vaccines do not provide adequate protection against new IBV strains that emerge due to ongoing evolution. Therefore, new antiviral strategies need to be explored. Cepharanthine (CEP), tetrandrine (TET), and berbamine hydrochloride (BBM) are natural plant-derived bis-benzylisoquinoline alkaloids (BBAs) with proven antiviral activities against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), porcine epidemic diarrhea virus (PEDV), and other viruses. Despite this, their potential antiviral activities against IBV remain unknown. As a proof-of-concept study, we aimed to investigate the inhibitory effects of CEP, TET, and BBM on the laboratory-adapted IBV Beaudette strain in Vero cells and of CEP in chicken embryos. Our research demonstrated that CEP, TET, and BBM effectively suppressed IBV infection in Vero cells, with CEP showing a particularly high selective index of 309.6. These compounds dose-dependently decreased IBV RNA levels and N protein expression and lowered intracellular and extracellular viral titers. Notably, CEP also exhibited antiviral activity against IBV infection in chicken embryos, resulting in reduced mortality and fewer lesions. While these findings highlight the potential of CEP, TET, and BBM as candidates for further development, further studies are required to evaluate their efficacy against field-prevalent IBV strains (e.g., Massachusetts, QX-like) and clarify the specific antiviral mechanisms.

Infectious bronchitis (IB) is an acute respiratory disease caused by the infectious bronchitis virus (IBV), spreads rapidly and manifests with diverse clinical signs, posing a major significant to the poultry industry. Metal ions such as copper are known to possess notable antiviral properties. Therefore, we evaluated the potential efficacy of copper-based pharmaceuticals against IBV. Results demonstrated that the copper-based drugs significantly protected DF-1 cells from IBV-M41 infection, markedly increasing cell activity and reducing IBV N-gene level. Furthermore, these drugs enhanced antiviral immunity and exhibited anti-apoptotic activity in vitro . In addition, these drugs increased the survival rates of infected chicken embryos and chicks while reducing developmental disruptions, thereby demonstrating robust antiviral effects. Pathological examinations revealed that lesions in the lungs, trachea, kidneys, spleen and other organs of treated groups improved to varying degrees, with a down-regulation of IBV N-gene level in chicks. Our study showed that the copper-based drugs exhibited anti-IBV-M41 both in vitro and in vivo , which provides a theoretical and experimental basis for exploring the anti-coronaviral effects of metal-based drug candidates.

Infectious bronchitis (IB) poses a significant global economic threat to the poultry industry. Phillygenin (PHI), a lignan compound possessing antiviral activity, has its clinical application limited by poor aqueous solubility and low oral bioavailability. To overcome these limitations, a self-nanoemulsifying drug delivery system loaded with PHI (PHI SNEDDS) was prepared. The resulting formulation exhibited uniform spherical nanoparticles with a size of 35.06 ± 0.64 nm, a zeta potential of -2.22 ± 0.27 mV, and a polydispersity index (PDI) of 0.180 ± 0.006, indicating high physical stability. In vitro release studies showed that PHI SNEDDS significantly enhanced both the rate and extent of drug release. A pharmacokinetic study in broilers revealed that the relative oral bioavailability of PHI SNEDDS reached 388.34% compared to the PHI suspension, accompanied by significantly increased maximum plasma concentration (C max ) and area under the curve (AUC 0-t ) ( P < 0.001), which confirms a substantial enhancement in absorption. In an IBV-infected broiler model, the SNEDDS formulation potentiated the antiviral efficacy of PHI. Compared to the IBV group and the PHI suspension group, PHI SNEDDS markedly enhanced antiviral efficacy, alleviating weight loss, clinical signs, and pathological damage in the trachea, lungs, and kidneys. It also significantly reduced viral load, virus shedding, and IBV-N protein expression ( P < 0.05), while increasing anti-IBV antibody levels ( P < 0.05), collectively indicating effective suppression of viral replication and enhancement of the host immune response. In conclusion, PHI SNEDDS effectively improved the solubility, bioavailability, and in vivo anti-IBV activity of PHI, demonstrating considerable potential as a therapeutic strategy for IB and offering a robust solution for administering hydrophobic active compounds.

ABSTRACT Vitamin A is a fat-soluble vitamin that is a crucial mediator of the immune system. In this study, we evaluated the effect of oral vitamin A supplementation on host immune responses to infectious bronchitis virus ( IBV ) infection in chickens. Forty 1-day-old specific pathogen-free ( SPF ) chickens were fed a basal diet and randomly divided into 2 groups ( n = 20 birds per group). Chickens in the experimental group were treated orally with vitamin A (dissolved in 0.1 mL soybean oil, at a dose of 8,000 IU per kg diet) daily. Birds in the control group were orally administered 0.1 mL soybean oil without vitamin A until 21 d of age. On d 21 after birth, all chickens were infected with 0.1 mL of 10 6.5 50% median embryo infectious dose of a pathogenic IBV strain (CK/CH/LDL/091022) by intraocular and intranasal routes. The results demonstrated that oral vitamin A supplementation did not affect the clinical course of disease and growth performance of SPF chickens. However, vitamin A supplementation increased the IBV-specific IgG serum levels and decreased the viral load in some tissues of IBV-infected chickens. In addition, the results demonstrated that vitamin A supplementation decreased the expression levels of most immune-related molecules in some tissues of IBV-infected chickens. Vitamin A supplementation decreased the mRNA expression levels of some avian β-defensins ( AvBD2, 3, 6, 7, 11 , and 13 ) and increased the expression levels of AvBD9 and AvBD12 in some tissues of IBV-infected chickens. Similarly, vitamin A supplementation decreased the mRNA expression levels of some cytokines (interferon-γ, interleukin-1β [ IL-1β ], and IL-6 ) and increased the mRNA expression levels of IL-2 in some tissues of IBV-infected chickens. Furthermore, vitamin A supplementation decreased the mRNA expression levels of myeloid differentiation primary response protein 88, nuclear factor-κB p65, toll-like receptor 3, toll-like receptor 7 , and CD4. In summary, the present study suggests that vitamin A supplementation enhances the immune function of SPF chickens against IBV infection by inhibiting viral replication, increasing the IBV-specific antibody titer, and suppressing the excessive inflammatory responses to IBV infection.
