Real time monitoring peripheral nerve function with ICG and BDA-ICG by NIR-II fluorescence imaging

Xiaoqi Yang,Yuanyi Wang,Chunrong Qu,Boyu Tan,Minjin Wang,Senrui Li,Jinsheng Huang,Jiangnan Li,Mengyuan Fang,Zhen Cheng,Nan Zhou

Neuroanatomical tract tracers are important for studying axoplasmic transport and the complex interconnections of the nervous system. Though traditional fluorescent tracers are widely used, they have several prominent drawbacks when imaging, including low resolutions and low tissue penetrations and inability to be supervised dynamically within a long peripheral nerve during the long term. Here, we explored the potential of ICG as a neural tracer for axoplasmic transport and for the first time demonstrated that ICG could be used to detect transport function within peripheral nerve by near-infrared region II (NIR-II) imaging. On basis of this finding, a novel bi-directional neural tracer biotinylated dextran amine-indocyanine green (BDA-ICG) was prepared and characterized with better long-term stability and higher nerve-to-background ratio than ICG in vivo, and successfully imaged the injured peripheral nerve from the healthy one within 24 h. Our results show that BDA-ICG are promising neural tracers and clinically available dyes with NIR-II emission tail characteristics as ICG.

ICG/MnO2-HFn-mPEG-DSPE-Lip enhances the anticancer activity of ICG phototherapy

Ma Dan,Lu Huixiang,Zhang Hai,Liao Anru,Peng Qianrong,Yang Min

Hypoxia poses a significant challenge to the efficacy of photodynamic therapy (PDT) for cancer treatment. This study aims to design and synthesize PEGylated liposomes encapsulating MnO₂, indocyanine green (ICG), and H-chain ferritin (HFn) to potentially address hypoxia and enhance the therapeutic outcomes of PDT. PEGylated liposomes (ICG/MnO₂-HFn-mPEG-DSPE-Lip) were constructed with a rod-like structure, incorporating MnO₂ as a hypoxia-modulating agent and ICG as a photosensitizer. The drug loading capacity, stability and safety of liposomes were characterized. Singlet oxygen quantum yield (Φ Δ ) was measured under simulated tumor microenvironment conditions. In vitro phototoxicity was evaluated using A549 human lung adenocarcinoma cells. Liposomes have a high drug loading capacity, good biocompatibility and good long-term stability. Under tumor-simulated conditions, Φ Δ was significantly improved, increasing from 0.210 for free ICG to 0.507. The liposomes demonstrated remarkable phototoxic effects on A549 cells (90.5% cell death under combined PDT/PTT vs. 15% for free ICG). This nanoplatform proposes a novel strategy to overcome hypoxia-induced PDT resistance, and the enhanced efficacy may be attributed to the increased oxygen supply mediated by MnO 2 carried by HFn. These findings provide important insights for the development of next-generation therapeutic systems targeting tumor hypoxia.

Nanosheet-shaped WS2/ICG nanocomposite for photodynamic/photothermal synergistic bacterial clearance and cutaneous regeneration on infectious wounds

Zhiling Chen,Rui Zhang,Tao Wang,Yanan Peng,Qionglin Zhou,Peipei Cao,Xinxin Xiao,Fengling Li,Ziming Wei,Yuanyuan Wang,Dan Xu,Bin Qiao,Shaowen Cheng,Qiang Wu,Lina Niu

Bacterial infections present a significant threat to human health, a challenge that is intensified by the slow pace of novel antibiotic development and the swift emergence of bacterial resistance. The development of novel antibacterial agents is crucial. Indocyanine green (ICG), a widely used imaging dye, efficiently generates reactive oxygen species (ROS) and heat for treating bacterial infections but suffers from aggregation and instability, limiting its efficacy. In this study, tungsten disulfide (WS₂) nanosheet with a high surface area was used to load ICG, creating a multifunctional nanocomposite, WS 2 /ICG, aimed at treating bacteria-infected wounds. The two-dimensional surface structure of WS₂ provides dispersible binding sites for ICG, and the synthesized nanocomposite exhibits excellent stability. Under near-infrared (NIR) laser excitation, the generated heat further synergistically enhances the yield of singlet oxygen. Additionally, the WS₂/ICG nanoplatform synergistically combines photothermal effect with photodynamic effect, achieving a “1 + 1 > 2” enhancement. Upon NIR laser excitation, the nanocomposite disrupts bacterial cell membranes through localized heating and ROS accumulation, leading to energy metabolism system disruption and subsequent bacterial lysis and death. The findings demonstrate WS₂/ICG's outstanding antibacterial properties and biocompatibility, effectively treating skin infections and promoting tissue regeneration, providing a simple and promising solution for bacteria-infected wounds.

Design, preparation and pharmacodynamics of ICG-Fe(Ⅲ) based HCPT nanocrystals against cancer

Qiongzhe Ren,Xuefeng Tang,Yi Lu,Qing Li,Zhiqian Liao,Shinan Jiang,Haoli Zhang,Zhigang Xu,Lei Luo

The use of nanocrystal technology to manufacture drug delivery systems intended to enhance therapeutic efficacy has attracted the attention of the pharmaceutical industry. However, the clinical application of nanocrystal drugs for injection is restricted by Ostwald ripening and the large-scale use of stabilizers such as polysorbate and lecithin, which have potential toxicity risks including hemolysis and allergies. Here, we designed an amorphous nanocrystal drug complex (IHNC), which is stabilizer-free and composed of indocyanine green (ICG) framework loading with a chemotherapeutic agent of 10-hydroxycamptothecin (HCPT). Considering the possibility of industrial manufacturing, IHNC was simply prepared with the assistance of ferric ion (III) via supramolecular assembly strategy. The theoretical result of Materials Studio simulation indicated that the prepared ICG-Fe(III) framework showed a stable spherical structure with the appropriate cavity for encapsulating the two drugs of HCPT and ICG with equal mass ratio. The IHNC was stable at physiological pH, with excellent PTT/PDT efficacy, and in vivo probing characteristics. The nanoscale size and reductive stimuli-responsiveness can be conducive to drug accumulation into the tumor site and rapid unloading of cargo. Moreover, such combination therapy showed synergistic photo/chemotherapy effect against 4T1 breast cancer and its tumor inhibition rate even up to 79.4%. These findings demonstrated that the nanocrystal drug delivery strategy could avoid the use of stabilizers and provide a new strategy for drug delivery for combination therapy.

Oxygen Nanobubbles Enhance ICG/Fe(III)-Mediated Dual-Modal Therapy To Induce Ferroptosis in Tumor Treatment

Li Yang,Wei-Hua Zhang,Yan Li,Yan-Li An,Ye-Ming Wu,Ning Gu,Gao-Jun Teng

Noninvasive therapies such as photodynamic therapy (PDT) and chemodynamic therapy (CDT), which rely on reactive oxygen species (ROS), are gaining attention for their low toxicity. However, single-modal treatments have individual limitations that restrict the therapeutic efficacy. Fe(III) can coordinate with the hydrophilic regions of indocyanine green (ICG) molecules to form the ICG/Fe(III) complex, making it a promising dual-modal agent for combined PDT and CDT. However, coordination with Fe(III) leads to the aggregation quenching of ICG, hindering its application in dual-modal therapy. We innovatively utilize oxygen nanobubbles, prepared solely from water and oxygen, to significantly reverse the aggregation-induced quenching of the ICG/Fe(III) complex, thereby enhancing its stability in aqueous environments. In this system, Fe(III) assembles at the nanobubble interface, coordinating with ICG’s hydrophilic regions to form the ICG/Fe(III)-NBs. The oxygen nanobubbles boost PDT efficiency by improving the ICG/Fe(III) complex stability and oxygen content, while Fe(III) achieves CDT by generating hydroxyl radicals (•OH) through the Fenton reaction. This dual-modality treatment significantly disrupts the tumor’s redox balance, induces ferroptosis, and demonstrates strong antitumor efficacy, reducing tumor volume to 34% of its initial size in mice. The strategy offers a promising and clinically viable approach to cancer treatment.

Polymer encapsulated clinical ICG nanoparticles for enhanced photothermal therapy and NIR fluorescence imaging in cervical cancer

Shuang Chen,Lijun Zhu,Zhong Du,Rong Ma,Ting Yan,Gulinigaer Alimu,Xueliang Zhang,Nuernisha Alifu,Cailing Ma

Photothermal therapy (PTT) is a popular tumor therapy method, which is based on efficient photothermal nanoagents (PTNs). Clinical Indocyanine Green (ICG), as a Food and Drug Administration (FDA) approved agent, is an often-used PTN, meanwhile it is also a good near-infrared (NIR) fluorescence contrast agent. However, the further applications of ICG in biomedical fields are limited due to its poor stability. In this study, ICG was encapsulated by the amphiphilic polymer poly(styrene-co-maleic anhydride) (PSMA) to form ICG@PSMA nanoparticles. Furthermore, optical and thermal characteristics of ICG@PSMA nanoparticles were studied in detail. Strong NIR fluorescence and excellent photothermal properties of ICG@PSMA nanoparticles under 808 nm laser irradiation were measured. Besides, favorable biocompatibility of ICG@PSMA nanoparticles was demonstrated on a human cervical cancer cell line (HeLa) via cell viability studies. Hence, ICG@PSMA nanoparticles were further applied to enhanced PTT of living HeLa cells under 808 nm excitation, and a high PTT efficiency of ∼70% was obtained. The novel ICG nanoparticles as a promising PTT nanoplatform could offer an opportunity for further tumour treatments.

Light responsive Fe-Tcpp@ICG for hydrogen peroxide detection and inhibition of tumor cell growth

Yu Wang,Xitong Zhou,Wanxin Dong,Qinsong Zhong,Xinxin Mo,He Li

In this work, we synthesized Fe-Tcpp@ICG(ICG, Indocyanine green) with light stimuli-response through 5,10,15,20-tetrakis (4-carboxyphenyl) porphyrin (Fe-Tcpp) loaded ICG by electrostatic adsorption. The morphology and properties of Fe-Tcpp and Fe-Tcpp@ICG were characterised by ultraviolet–visible absorption spectrometer, X-ray diffraction, thermogravimetric analyzer and transmission electron microscope , respectively. A non-enzymatic photoelectrochemical sensor based on Fe-Tcpp@ICG was constructed to quantitatively detect hydrogen peroxide in tumor microenvironment . Under the optimal conditions, the linear range of detecting hydrogen peroxide was 0.01–50 mmol/L with detection limit of 0.2 μmol/L (S/N = 3). This sensor proposed a simple, fast, sensitive and label-free method for the detection of hydrogen peroxide. Moreover, the results also showed that the Fe-Tcpp@ICG can catalyze the decomposition of hydrogen peroxide to generate singlet oxygen, which can kill tumor cells. These indicated that this material was expected to be used for detecting hydrogen peroxide and inhibition of tumor cell growth.

Mahounga Didel M.,Shan Lingling,Jie Cao,Du Changli,Wan Shunan,Gu Yueqing

Purpose A novel near infrared fluorescent probe, l -methyl-methionine (Met)–ICG-Der-02, was synthesized and characterized for in vivo imaging of tumors and early diagnosis of cancers. Method Met was conjugated with ICG-Der-02 dye through the amide bond function by ethyl-3-(3-dimethyllaminopropyl) carbodiimide hydrochloride/ N -hydroxysuccinimide catalysis chemistry. Met–ICG-Der-02 probe uptake was evaluated on PC3, MDA-MB-231, and human embryonic lung fibroblast cell lines. The dynamics of Met–ICG-Der-02 was investigated in athymic nude mice prior to evaluation of the probe targeting capability in prostate and breast cancer models. Results Met–ICG-Der-02 was successfully synthesized. Cell experiments demonstrated excellent cellular uptake of Met–ICG-Der-02 on cancer cell lines without cytotoxicity. Optical imaging showed a distinguishable fluorescence signal in the tumor area at 2 h while maximal tumor-to-normal tissue contrast ratio was at 12 h Met–ICG-Der-02 post-injection. Additionally, dynamic study of the probe indicated intestinal and liver–kidney clearance pathways. Conclusion Met–ICG-Der-02 probe is a promising optical imaging agent for tumor diagnosis, especially in their early stage.

ICG@ZIF-8/PDA/Ag composites as chemo-photothermal antibacterial agents for efficient sterilization and enhanced wound disinfection

Shuhan Hui,Qiqi Liu,Yandong Han,Lijuan Zhang,Jun Yang,Shan Jiang,Haisheng Qian,Wensheng Yang

Bacterial infection has increasingly affected people's lives, therefore it is significant to explore novel antibacterial agents and strategies for efficient disinfection. Herein, we designed ZIF-8 based composites ICG@ZIF-8/PDA/Ag, which encapsulate photothermal agent indocyanine green (ICG) and grow polydopamine (PDA) on their surface for in situ reduction to generate Ag nanoparticles. With 20 min of 808 nm laser irradiation at 1.5 W cm−2, 100 μg mL−1 ICG@ZIF-8/PDA/Ag exhibited 100% bactericidal effects toward E. coli and S. aureus bacteria resulting from both hyperthermia of ICG and PDA and chemical toxicity of the released Ag and Zn ions. When the bacterial incubation period was extended to 12 h, the minimum bactericidal concentration (MBC) of ICG@ZIF-8/PDA/Ag was reduced to 6.25 μg mL−1, and this extremely low MBC was due to the long-term chemo-photothermal combinational effect induced by NIR irradiation. Additionally, the composites successfully promote the healing of S. aureus infected wounds on mice. This work constructed photo-responsive antibacterial composites that realize chemo-photothermal synergistic therapy.

ZnS@ZIF-8 core-shell nanoparticles incorporated with ICG and TPZ to enable H2S-amplified synergistic therapy

Chao Fang,Dong Cen,Yifan Wang,Yongjun Wu,Xiujun Cai,Xiang Li,Gaorong Han

Abnormal tumor microenvironment, such as hypoxia, interstitial hypertension and low pH, leads to unexpected resistance for current tumor treatment. The development of versatile drug delivery systems which present responsive characteristics to tumor microenvironment (TME) has been extensively carried out, but remains challenging. In this study, zeolitic imidazolate framework-8 (ZIF-8) coated ZnS nanoparticles have been designed and prepared for co-delivery of ICG/TPZ molecules, denoted as ZSZIT, for H 2 S-amplified synergistic therapy. Methods: The ZSZ nanoparticles were characterized using SEM, TEM and XRD. The in vitro viabilities of cancer cells cultured with ZSZIT under normoxia/hypoxia conditions were evaluated by cell counting kit-8 (CCK-8) assay. In addition, in vivo anti-tumor effect was also performed using male Balb/c nude mice as animal model. Results: ZSZIT shows cascade PDT and hypoxia-activated chemotherapeutic effect under an 808nm NIR irradiation. Meanwhile, ZSZIT degrades under tumor acidic environment, and H 2 S produced by ZnS cores could inhibit the expression of catalase, which subsequently favors the hypoxia and antitumor effect of TPZ drug. Both in vitro and in vivo studies demonstrate the H 2 S-sensitized synergistic antitumor effect based on cascade PDT/chemotherapy. Conclusion: This cascade H 2 S-sensitized synergistic nanoplatform has enabled more effective and lasting anticancer treatment.

Microfluidic Generation of Near-Infrared Photothermal Vitexin/ICG Liposome with Amplified Photodynamic Therapy

Cao Xia,Liu Qi,Adu-Frimpong Michael,Shi Wenwan,liu Kai,Deng Tianwen,Yuan Hui,Weng Xuedi,Gao Yihong,Yu Qingtong,Deng Wenwen,Yu Jiangnan,Wang Qilong,Xiao Gao,Xu Ximing

Glioma, in which a malignant tumor cell occurs in neural mesenchymal cells, has a rapid progression and poor prognosis, which is still far from desirable in clinical treatments. We developed a lab-on-a-chip (LOC) device for the rapid and efficient preparation of vitexin/indocyanine green (ICG) liposomes. Vitexin could be released from liposome to kill cancer cell, which can potentially improve the glioma therapeutic effect and reduce the treatment time through synergistic photodynamic/photothermal therapies (PDT/PTT). The vitexin/ICG liposome was fabricated via LOC and its physicochemical property and release in vitro were evaluated. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method and live/dead staining were used to examine the enhanced antitumor effect of vitexin/ICG liposome in cooperation with PDT/PTT, while the related mechanism was explored by flow cytometry and western blot. The results were as follows: (1) The prepared vitexin/ICG liposome was smaller in size, homogenous in particle size distribution with significant low polydispersity index (PDI), and enhanced cumulative release in vitro . (2) We found that the formulated liposome presented strong cancer cell inhibition and suppression of its migration in a dose-dependent manner. (3) Further mechanistic studies showed that liposome combined with near-infrared irradiation could significantly upregulate levels of B cell lymphoma 2-associated X (Bax) protein and decrease B cell lymphoma 2 (Bcl-2) at protein levels. The vitexin/ICG liposomes prepared based on a simple LOC platform can effectively enhance the solubility of insoluble drugs, and the combined effect of PTT/PDT can effectively increase their antitumor effect, which provides a simple and valid method for the clinical translation of liposomes. β-catenin/TCF4 inhibitors ICG-001 and LF3 alleviate BDL-induced liver fibrosis by suppressing LECT2 signaling

Yue Gao,Shicheng Fan,Pengfei Zhao,Huilin Li,Chenghui Cai,Xuan Li,Yanying Zhou,Min Huang,Huichang Bi

Liver fibrosis can be characterized by the over-deposition of extracellular matrix (ECM). It has been reported that β-catenin/TCF4 interaction was enhanced in bile duct ligation (BDL) model, which implicated the critical role of β-catenin/TCF4 interaction during the progression of fibrosis . However, whether inhibiting β-catenin/TCF4 signaling attenuates liver fibrosis remains unknown. In the current study, we used ICG-001, an inhibitor that disrupts the interaction between CREB binding protein (CBP) and β-catenin, to inhibit β-catenin/TCF4 transcriptional activity. We also used LF3, a small molecule antagonist, to inhibit β-catenin/TCF4 interaction. The antifibrotic effect of ICG-001 and LF3 was assessed on BDL-induced liver fibrosis model. The results indicated both ICG-001 and LF3 significantly reduced the positive staining area of Sirius Red and α-SMA. The protein expression levels of α-SMA, Collagen Ⅰ and CD31 were also significantly downregulated in BDL + ICG-001 and BDL + LF3 groups. Besides, ICG-001 and LF3 promoted portal angiogenesis and inhibited sinusoids capillarization in fibrotic livers. For mechanistic study, we measured the level of leukocyte cell-derived chemotaxin 2 (LECT2), a direct target of β-catenin/TCF4, which was recently reported to participate in hepatic fibrosis by regulating angiogenesis. The results showed that both ICG-001 and LF3 reduced LECT2 expression in BDL mice. LF3 also downregulated pSer 675 β-catenin and nuclear β-catenin. In conclusion, this study demonstrated that inhibiting β-catenin/TCF4 signaling by ICG-001 or LF3 mitigated liver fibrosis by downregulating LECT2, promoting portal angiogenesis and inhibiting sinusoids capillarization, which provided new evidence that β-catenin/TCF4 signaling might be a target for the treatment of liver fibrosis.
