Phototherapy is gradually becoming a primary technique for treating tumors due to increased efficacy and less side effects. It largely depends on the photosensitizers. Nevertheless, most of photosensitizers show poor stability, how to encapsulate or load them, especially multiple ones, for effective delivery is of paramount importance. To address the challenge, a hyperbranched polymer was prepared by a reversible addition-fragmentation chain transfer polymerization, which is composed of 2,2,3,3-Tetrafluoropropyl methacrylate (TFPMA), bis(2-methacryloyl)oxyethyl disulfide (DSDMA) and poly(oligo ethylene glycol) methyl ether methacrylate (PEGMA). Benefiting from hydrophobic performance of fluorinated segments, this hyperbranched polymers can load hydrophobic IR780 (9%) and Ce6 (12%) due to self-assembly and hydrophobic-hydrophobic interaction. The final formed composite micelles showed good water dispersity, well biocompatibility, and strong stability. Remarkably, they had good photodynamic and photothemal abilities, enabling them to synergistically induce tumor cell apoptosis by oxidative damage and thermal ablation. This work offers a promising composites and strategy for loading photosensitizers stably and effectively towards cancer therapy.

IR780 iodide is a commercially available targeted near-infrared contrast agent for in vivo imaging and cancer photodynamic or photothermal therapy, whereas the accumulation, dynamics, and retention of IR780 in biological tissue, especially in tumor is still under-explored. Diffuse fluorescence tomography (DFT) can be used for localization and quantification of the three-dimensional distribution of NIR fluorophores. Herein, a homemade DFT imaging system combined with tumor-targeted IR780 was utilized for cancer imaging and pharmacokinetic evaluation. The aim of this study is to comprehensively assess the biochemical and pharmacokinetic characteristics of IR780 with the aid of DFT imaging. The optimal IR780 concentration (20 μg/mL) was achieved first. Subsequently, the good biocompatibility and cellar uptake of IR780 was demonstrated through the mouse acute toxic test and cell assay. In vivo, DFT imaging effectively identified various subcutaneous tumors and revealed the long-term retention of IR780 in tumors and rapid metabolism in the liver. Ex vivo imaging indicated IR780 was mainly concentrated in tumor and lung with significantly different from the distribution in other organs. DFT imaging allowed sensitive tumor detection and pharmacokinetic rates analysis. Simultaneously, the kinetics of IR780 in tumors and liver provided more valuable information for application and development of IR780.

The combination of chemotherapy and photothermal therapy can provide better tumor treatment effect than monotherapy , while the combination of imaging technology can achieve diagnostic effect during the treatment. In this paper, biocompatible mesoporous polydopamine (MPDA) were combined with the near-infrared fluorescent dye IR780 under alkaline condition to form an integrated drug delivery platform for tumor diagnosis and photothermal co-therapy. The particles size and mesoporous pore size of the nanospheres were adjusted by changing the preparation conditions of MPDA, so the platform had strong permeability and large drug loading capacity in tumor cells as drug carriers. The loading and release results of doxorubicin hydrochloride (DOX) proved that the carrier had a high drug loading and sustained release rate, as well as a faster dependent release under acidic and lighting conditions. Photothermal experiments showed that the carrier had high photothermal conversion efficiency, which was very critical in its application in photothermal therapy and fluorescence imaging of tumors. The vitro cytotoxicity test evaluated the survival rates of LX-2 cells and HepG2 cells after drug loading. The results showed that the MPDA@IR780-DOX + NIR carrier had a significant inhibitory effect on HepG2 cells (12.57%), but had little effect on LX-2 cells, which was beneficial for its application in integration of biological diagnosis and treatment.

Chronic and infected wounds, particularly those caused by bacterial infections, present significant challenges in medical treatment. This study aimed to develop a novel nanoparticle formulation to enhance wound healing by combining antimicrobial and photothermal therapy using albumin as a carrier for Tanshinone IIA and the near-infrared photothermal agent IR780. The nanoparticles were synthesized to exploit the antimicrobial effects of Tanshinone IIA and the photothermal properties of IR780 when exposed to near-infrared laser irradiation. Characterization of the nanoparticles was performed using Transmission Electron Microscopy (TEM) and spectroscopic analysis to confirm their successful synthesis. In vitro antibacterial activity was evaluated using cultures of methicillin-resistant Staphylococcus aureus (MRSA), and in vivo efficacy was tested in a mouse model of MRSA-infected wounds. Wound healing progression was assessed over 16 days, with statistical analysis performed using two-way ANOVA followed by Tukey’s post-hoc test. The nanoparticles demonstrated significant photothermal properties, enhancing bacterial eradication and promoting the controlled release of Tanshinone IIA. In vitro studies showed superior antibacterial activity, especially under photothermal activation, leading to a substantial reduction in bacterial viability in MRSA cultures. In vivo, nanoparticle treatment combined with near-infrared laser irradiation significantly improved wound closure rates compared to controls and treatments without photothermal activation. By the 16th day post-treatment, significant improvements in wound healing were observed, highlighting the potential of the combined photothermal and pharmacological approach. These findings suggest that albumin-loaded nanoparticles containing Tanshinone IIA and IR780, activated by near-infrared light, could offer an effective therapeutic strategy for managing chronic and infected wounds, promoting both infection control and tissue repair.

The IR780 iodide (IR780) is recognized as an effective theranostic agent for simultaneous near-infrared fluorescence imaging and photothermal therapy (PTT). However, the rigid chloro-cyclohexenyl ring makes IR780 insoluble in almost all pharmaceutically acceptable solvents, which inevitably limits its clinical application. We report folic acid (FA)-functionalized graphene quantum dots (GQDs-FA) containing a large and intact sp2 domain with carboxyl groups around the edge. Such GQDs-FA possess exceptionally high loading capacity for IR780 via strong π–π stacking interactions, and the water solubility of IR780 is improved by over 2400-fold after loading onto GQDs-FA (IR780/GQDs-FA). IR780/GQDs-FA with an improved photostability, an enhanced tumor-targeting ability, and a high photothermal conversion efficiency of 87.9% were capable of producing sufficient hyperthermia to effectively kill cancer cells and completely eradicate tumors upon 808 nm laser irradiation. The present IR780/GQDs-FA may open up great opportunities for the effective PTT to treat cancer.

Combined photo-induced cancer therapy (photothermal therapy (PTT) and photodynamic therapy (PDT)) is a promising anti-cancer strategy for its non-invasiveness property. However, severe hypoxia in tumor microenvironment (TME) would weaken the PDT effect and limit the clinical efficiency of PTT/PDT treatment. To solve this problem, denatured BSA-hemin-IR780 (dBHI) was designed and synthesized for anti-cancer treatment in this work. Denatured bovine serum albumin (dBSA) was used as the binding matrix to bind hemin and IR780 via covalent reaction and hydrophobic self-assembly, respectively. The obtained dBHI nanoparticles possess both PTT and PDT effect due to the presence of IR780 in the composites. Moreover, because of the existence of hemin, dBHI also exhibited photo-enhanced Fenton-like catalytic performance, which could decompose H 2 O 2 to form oxygen for improving the PDT effect and generate hydroxyl radical (·OH) for chemodynamic therapy (CDT). Therefore, by using dBHI, synergistic effect of PTT/PDT/CDT combined treatment has been achieved for cancer therapy.

Inertial cavitation is crucial for the therapeutic effects of sonodynamic. Therefore, approaches that can induce highly efficient inertial cavitation should be of benefit for sonodynamic effect. Our previous study demonstrated that highly efficient inertial cavitation activity can be achieved through the combinatorial use of a short-pulsed focused ultrasound (SPFU) sequence and perfluorohexane (PFH) nanodroplets. Herein, we applied the SPFU sequence and PFH nanodroplets in sonodynamic. A hydrophobic sonosensitizer, IR780 iodine, was loaded inside denatured bovine serum albumin-shelled PFH (PFH@BSA-IR780) nanodroplets. The sonodynamic efficacy was validated by treating HeLa cervical cancer cells. Under SPFU exposure, PFH@BSA-IR780 nanodroplets were highly effective in promoting reactive oxygen species generation and inducing cancer cell death. A significant decrease in cell viability was achieved within just 10 s. Besides the cytotoxicity of ROS, the mechanical bioeffects of inertial cavitation also led to severe cell death resulting from higher acoustic power or the longer treatment time. The application of the SPFU sequence coupled with PFH@BSA-IR780 nanodroplets is a promising strategy for efficient sonodynamic.

Chloride double perovskites have sparked significant interest and opened new avenues for luminescent materials hosts due to their lower phonon energy, flexible compositions, and excellent luminescent property. However, the instability to water has limited its fundamental applications, especially in the biological fields, although its stability has been improved in comparison with lead−based perovskites. Here, nanophosphor Cs 2 NaBiCl 6 : Nd 3+ , Yb 3+ (CNBC: Nd 3+ , Yb 3+ ) is prepared by a hot-inject method, and then its stability to water is enhanced by coating a layer of SiO 2 shell. For improving its photothermal conversion ability, organic photothermal agents IR780 is adsorbed on the surface of CNBC: Nd 3+ , Yb 3+ @SiO 2 to form monodispersed temperature self−monitoring photothermal agents CNBC: Nd 3+ , Yb 3+ @SiO 2 @IR780, where the temperature sensing property is based on the temperature−dependent lifetimes of Yb 3+ and the photothermal conversion property comes from the synergistic effect of IR780 and the nonradiative transition of Nd 3+ during luminescence. The maximum relative sensitivity and photothermal conversion efficiency of CNBC: Nd 3+ , Yb 3+ @SiO 2 @IR780 reach 1.44% K −1 at 280 K and 36.8%. Finally, the temperature detection and photothermal antimicrobial ability of CNBC: Nd 3+ , Yb 3+ @SiO 2 @IR780 are evaluated through chicken tissue temperature measurement and photothermal ablation effect against Escherichia coli and Staphylococcus aureus (93.3% and 95.1%).

Micellar nanoparticles have unique advantages as carriers for therapeutic or imaging agents, owing to their smaller size and better penetration of tumors. However, some agents, due to their physical or chemical properties, are difficult to load into micelles. IR780 is one of these agents, and is also a promising near-infrared dye for fluorescence imaging (FI)/photoacoustic imaging (PAI) and cancer photothermal therapy (PTT). Its hydrophobic and high crystallization structure results in limited bioavailability in vivo. It is difficult to load into micelles constructed from an amphiphilic block polymer with relatively low molecular weight. In this study, we use computer simulation and introduce another small biomolecule, α-lipoic acid, into the micelles constructed from a mPEG-PCL copolymer, to lower the energy of molecular interaction between MPEG-PCL and IR780, and expect to enhance the loading capacity of the micelles to IR780. The introduction of α-lipoic acid decreases the energy of molecular interaction between MEPG-PCL and IR780 from −46.18 kJ mol−1 to −196.52 kJ mol−1 and increases the loading capacity and stability of the mPEG-PCL micelles to IR780, which also maintains the loading capacity to DTX. We further construct DTX/IR780 co-loaded mPEG-PCL micelles for FI/PAI dual modal imaging guided PTT/chemotherapy of cancer. By FI and PAI evaluation in vitro and in vivo, we demonstrate that the DTX/IR780 co-loaded micelles can be used as FI and PAI probes. By further evaluating the therapeutic outcome of PTT/chemotherapy co-therapy of breast cancer, we demonstrate that the DTX/IR780 co-loaded mPEG-PCL micelles can serve as promising candidates for FI and PAI guided PTT/chemotherapy of breast cancer.

In recent years, breast cancer attracts more and more attention because of its high mobility and high mortality for women. Phototherapy, including photothermal therapy (PTT) and photodynamic therapy (PDT), provides a new pathway to breast cancer therapy. Herein, we developed multifunctional
IR780 loaded nanoparticles based on folic acid modified chitosan (FASOC-IR780 NPs) for targeted imaging and phototherapy of cancer compatible with a single-nearinfrared (NIR) laser irradiation. In vitro characterizations demonstrated the self-assembled FASOC-IR780 NPs own narrow size
distribution and encouraging photothermal performance with enhanced generation of ROS under laser irradiation. The cytotoxicity studies on MDA-MB-231 cells showed enhanced toxicity upon NIR light irradiation. NIR small animal imaging demonstrated the significant tumor-selectivity of the nanoparticles
to breast cancer cells that over expressed folate receptor. In vivo treatments with FASOC-IR780 NPs resulted in effective tumor suppression. These results indicate that the multifunctional FASOC-IR780 NPs show notable targeting and theranostic potential in breast cancer therapy.

Multifunctional phototheranostics combining diagnostic and therapeutic modalities may provide a revolutionary opportunity for cancer treatment. As a promising tumor phototheranostic molecule, IR780 iodide (IR780) shows excellent photodynamic and photothermal performance under near-infrared laser irradiation; however, its hydrophobicity and instability limit its further use in organisms. This work demonstrates the design and development of a multifunctional nanoplatform (PMIDA, referring to polydopamine (PDA)-manganese dioxide (MnO 2 )-IR780) for imaging-guided phototherapy. The good biocompatibility of PDA greatly improves the water solubility and photostability of IR780, and its excellent photothermal properties make PMIDA a dual photothermal therapy (PTT). MnO 2 -induced generation of oxygen in the tumor microenvironment improves the hypoxia effect and photodynamic therapy (PDT) of IR780. Moreover, Mn 2+ serves as a decent T 1 -weighted magnetic resonance imaging (MRI) probe to guide treatment. Notably, in relevant cellular assays, PMIDA shows high photodynamic and photothermal effects contributing to the final therapeutic effect. The MRI-guided PDT/PTT synergistic therapy effect in vivo is demonstrated by precise tumor diagnosis and complete tumor elimination outcomes. Based on these experiments, PMIDA nanoparticles display promising effects in facilitating intravenous injection of IR780 and achieving magnetic resonance imaging (MRI)-guided phototheranostic efficacy for tumor treatment.

Combined chemo-phototherapy has shown considerable advantages and potential in cancer treatment. For this purpose, self-assembled nanoparticles by gambogic acid (GA) and IR780 (referred to as GA-IR780 NPs) were prepared. Herein, GA, an active compound derived from Garcinia hanburyi Hook.f, was selected as a chemo-agent. IR780 was used as a photothermal agent as well as a photosensitizer, which could kill tumor cells via photothermal effect and photodynamic effect. The obtained GA-IR780 NPs were uniform spheres with particle size of ca. 50 nm. The drug loading efficiency of GA and IR780 was 38.42 % and 56.64 %, respectively. The GA-IR780 NPs exhibited excellent photothermal properties as well as photodynamic effect when irradiated by near infrared (NIR) light (808 nm, 2.0 W/cm 2 ). Moreover, the GA-IR780 NPs showed enhanced cytotoxicity with NIR light activation. Results of animal experiments showed that GA-IR780 NPs had the most significant tumor inhibition when irradiated by laser, and the results of H&E , Ki-67 and TUNEL staining confirmed that the GA-IR780 NPs+Laser group caused the most severe tumor tissue damage. The above results indicated that GA-mediated chemotherapy combining with IR780-based phototherapy could significantly improve the anti-tumor efficacy.
