Formulation of alginate/carrageenan microgels to encapsulate, protect and release immunoglobulins: Egg Yolk IgY

Luping Gu,David Julian McClements,Jiaying Li,Yujie Su,Yanjun Yang,Junhua Li

In this study, alginate/κ-carrageenan hydrogel beads were developed to encapsulate and release egg yolk immunoglobulin Y (IgY) under simulated gastrointestinal conditions. The effects of κ-carrageenan concentration on the encapsulation and loading efficiency of the hydrogel beads was assessed, as well as on their swelling and release behavior in a simulated human gut. An κ-carrageenan concentration of 0.30% gave the optimum encapsulation and loading efficiency. After incubation in simulated gastric fluids, there was no loss in IgY activity when it was encapsulated within alginate/carrageenan beads but around 35% loss when it was encapsulated in pure alginate beads. This effect was attributed to a stronger electrostatic attraction and a more closely packed biopolymer network formed within the composite beads, based on measurements of their electrical and microstructural properties. The composite beads were resistant to swelling in the stomach phase, but not in the intestine phase. The kinetics of swelling under simulated gastric conditions was found to follow Schott's second-order model. The gastrointestinal digestion of the IgY was slower in the composite beads than in the pure alginate beads. The release kinetics of IgY from the composite beads was fitted by an irregular release model. Our results suggest that alginate/κ-carrageenan composite beads are efficient carriers for the delivery systems of hydrophilic immunoglobins.

Ultrasensitive and selective detection of Staphylococcus aureus using a novel IgY-based colorimetric platform

Yun Zhang,Wenqing Tan,Yang Zhang,Huili Mao,Shuyou Shi,Liangwei Duan,Hui Wang,Junping Yu

To develop a specific method for the detection of S. aureus , chicken anti-protein A IgY was adopted for specifically capturing S. aureus , depending on the specific recognition of staphylococcal protein A (SPA) by chicken anti-protein A IgY, which can eliminate the interference from protein G-producing Streptococcus . HRP labeled IgG , Fc region of which has a high affinity towards SPA, was paired with IgY for the colorimeter analysis of the system. By optimizing the system, a super-low detection limit of 11  CFU of S. aureus in 100 μL PBS without enrichment, with a linear range from 5.0 × 10 2  CFU mL −1 to 5.0 × 10 4  CFU mL −1 was obtained. The entire assay was accomplished in less than 90 min and no cross-reactivity with the other tested bacterial species was observed. Moreover, the developed assay has been applied for the detection of S. aureus in three different types of real samples (sodium chloride injection, apple juice and human urine) with satisfactory results. To the best of our knowledge, it is the first time to report using chicken anti-protein A IgY and any IgG to detect S. aureus based on the dual-recognition mode of SPA. The novel method opened up a way for monitoring S. aureus in food samples with high sensitivity, specificity and simple operation.

Ultrasensitive colorimetric detection of Staphylococcus aureus using wheat germ agglutinin and IgY as a dual-recognition strategy

Zhang Yun,Tian Ge,Sun Xueni,Yang Xiaoli,Zhang Yi,Tan Wenqing,Duan Liangwei,Gao Shunxiang,Yu Junping

A novel colorimetric platform was designed for the determination of S. aureus by utilizing a dual-recognition strategy, where wheat germ agglutinin (WGA)-functionalized magnetic beads were served as separation elements to capture and enrich S. aureus efficiently from the matrix. Horseradish peroxidase (HRP) labeled chicken anti-protein A IgY (HRP-IgY) was used to label the captured S. aureus . A chicken IgY was introduced as a signal tracer to bind with staphylococcal protein A (SPA) on the surface of S. aureus , which can circumvent the interference from protein G-producing Streptococcus . Subsequently, the colorimetric signal was achieved by an HRP-catalyzed reaction, which was amplified by HRP-IgY bound by approximately 80,000 SPA molecules on one S. aureus . The entire detection process could be accomplished within 90 min. Under optimal conditions, the linear response of different S. aureus concentrations ranged from 7.8 × 10 2 to 2.0 × 10 5  CFU/mL and the limit of detection reached down to 3.9 × 10 2  CFU/mL. Some common non-target bacteria yielded negative results, indicating the excellent specificity of the method. The developed strategy was successfully applied to the determination of S. aureus in various types of samples with satisfactory recoveries. Therefore, the novel dual-recognition strategy possessed the advantages of high sensitivity, specificity, and low cost and exhibited considerable potential as a promising tool to defend public health. Sensitive and selective detection of Staphylococcus aureus via a dual-recognition colorimetric platform based on phenylboronic acid and IgY

Zhang Yun,Huo Chunhui,Yang Xiaohong,Cui Yanjie,Hou Yiqing,Guo Qinghe,Wang Qiongzi,Wang Mingyong,Zhang Yi

Staphylococcus aureus ( S. aureus ) is one of the major food-borne pathogenic bacteria that causes epidemic and food poisoning. In this study, a colorimetric sensing platform was developed for the sensitive and selective detection of S. aureus based on the synergy between 3-aminophenylboronic acid-functionalized magnetic beads (APBA-MBs) and horseradish peroxidase labeled anti-protein A IgY (HRP-IgY). Target S. aureus bacteria can be captured on the APBA-MBs and then labelled by HRP-IgY through formation of a sandwich structure. Detection of the bacteria then involves oxidation of the colorimetric HRP substrate, thereby converting S. aureus concentrations into a detectable signal. The optimized method can be performed within 120 min, with a detection limit of 9.4 × 10 2  CFU/mL . The proposed strategy was successfully applied to detection of S. aureus in spring water and apple juice samples. The dual-recognition platform showed selectivity towards S. aureus , and other common foodborne pathogens yielded negative results. The incorporation of IgY into the proposed strategy can address the lack of selectivity observed in previously reported APBA-based methods. Our strategy has potential as a versatile platform for the sensitive detection of various bacteria using corresponding IgY.

IgY antibodies against Ebola virus possess post-exposure protection in a murine pseudovirus challenge model and excellent thermostability

Yuan Zhang,Yanqiu Wei,Yunlong Li,Xuan Wang,Yang Liu,Deyu Tian,Xiaojuan Jia,Rui Gong,Wenjun Liu,Limin Yang

Ebola virus (EBOV) is one of the most virulent pathogens that causes hemorrhagic fever and displays high mortality rates and low prognosis rates in both humans and nonhuman primates. The post-exposure antibody therapies to prevent EBOV infection are considered effective as of yet. However, owing to the poor thermal stability of mammalian antibodies, their application in the tropics has remained limited. Therefore, a thermostable therapeutic antibody against EBOV was developed modelled on the poultry(chicken) immunoglobulin Y (IgY). The IgY antibodies retaining their neutralising activity at 25°C for one year, displayed excellent thermal stability, opposed to conventional polyclonal antibodies (pAbs) or monoclonal antibodies (mAbs). Laying hens were immunised with a variety of EBOV vaccine candidates and it was confirmed that VSVΔG/EBOVGP encoding the EBOV glycoprotein could induce high titer neutralising antibodies against EBOV. The therapeutic efficacy of immune IgY antibodies in vivo was evaluated in the newborn Balb/c mice who have been challenged with the VSVΔG/EBOVGP model. Mice that have been challenged with a lethal dose of the pseudovirus were treated 2 or 24 h post-infection with different doses of anti-EBOV IgY. The group receiving a high dose of 10 6 NAU/kg (neutralising antibody units/kilogram) showed complete protection with no symptoms of a disease, while the low-dose group was only partially protected. Conversely, all mice receiving naive IgY died within 10 days. In conclusion, the anti-EBOV IgY exhibits excellent thermostability and protective efficacy. Anti-EBOV IgY shows a lot of promise in entering the realm of efficient Ebola virus treatment regimens. Despite the amount of efficient Ebola virus therapeutic antibodies reported in recent years, their application in tropical endemic areas has remained limited due to the low thermal stability of mammalian antibodies. A highly thermostable therapeutic polyclonal antibody against EBOV was developed based on chicken immunoglobulin Y (IgY). The EBOV specific IgY antibodies displayed excellent thermal stability, retaining their neutralising activity at 25°C for one year. The newborn mice receiving the passive transfer of IgY achieved complete protection against a lethal dose of virus challenge proving that the anti-EBOV IgY provides a promising recourse to solve some of the current clinical application hindrances of Ebola antibody-based treatments in Africa due to thermal stability.

Highly sensitive and selective colorimetric determination of Staphylococcus aureus via chicken anti-protein A IgY antibody

Yun Zhang,Wenqing Tan,Lin Zhang,Shuyou Shi,Yuna Niu,Xue Yang,Jinjuan Qiao,Hui Wang

Staphylococcus aureus (S. aureus) is a food-borne pathogenic organism that causes an array of infectious diseases ranging from superficial skin infections to potentially fatal diseases. Thus quantitative detection of S. aureus with high selectivity and sensitivity is of great significance. In this study, Staphylococcus protein A (SPA), which is specifically secreted on the surface of S. aureus, was selected as an epitope. On the basis of the specific recognition of SPA by chicken anti-protein A immunoglobulin Y (IgY), a facile colorimetric immunoassay for the detection of S. aureus was established. To the best of our knowledge, this is the first time chicken anti-protein A IgY has been utilized as an antibody pair for the quantitative detection of S. aureus. Chicken anti-protein A IgY was conjugated with magnetic particles for bacterial capture. Then horseradish peroxidase (HRP) labeled chicken anti-protein A IgY was employed as the revealing partner to perform a sandwich format assay, followed by an HRP catalyzed colorimetric reaction. Optimized detection conditions produced a calibration curve for S. aureus ranging from 7.8 × 102 CFU mL−1 to 2.0 × 105 CFU mL−1 with a detection limit of 2.3 × 102 CFU mL−1 (3σ) in phosphate-buffered saline (PBS). The detection limits (3σ) in spiked samples were very close to those in PBS. The entire assay was accomplished in less than 60 min and no cross-reactivity with the other tested bacterial species was observed. These results demonstrate that the proposed method is effective for rapid and selective screening of S. aureus in food samples.

Preparation of IgY Oriented Conjugated Fe3O4 MNPs as Immunomagnetic Nanoprobe for Increasing Enrichment Efficiency of Staphylococcus aureus Based on Adjusting the pH of the Solution System

Shi Xuening,Sun Hongbin,Li Hang,Wei Shengnan,Jin Jin,Zhao Chao,Wang Juan,Li Hui

Immunomagnetic separation based on Fe3O4 magnetic nanoparticles (MNPs) has been widely performed in sample pretreatment. The oriented conjugation strategy can achieve a better capture effect than the N-(3-dimethylamlnopropyl)-N'-ethylcarbodiimide hydrochloride (EDC) /N-hydroxysuccinimide (NHS) method. However, immunoglobulin yolk (IgY) cannot be oriented through an SPA strategy like immunoglobulin G (IgG). In this article, an oriented conjugation nanoprobe was prepared for the enrichment of bacteria based on pH adjusting. The main factors affecting the enrichment efficiency were studied, such as the pH of the buffer system, the concentration of IgY, the concentration of nanoprobe, and the enrichment time. Under the optimal conditions, the enrichment efficiency toward target bacteria could reach 92.8%. Combined with PCR, the limit of detection (LOD) was found to be 103 CFU/ml, which was lower than the PCR only. In conclusion, we provided a new protocol for the oriented conjugation of IgY and high sensitivity detection with simple pretreatment.

Prophylaxis and therapy of pandemic H1N1 virus infection using egg yolk antibody

Yuan-e Yang,Junlin Wen,Suqing Zhao,Kun Zhang,Yingliang Zhou

Influenza A virus infects the human respiratory system and causes acute and fatal pulmonary diseases. The emergence of drug-resistant viral strains highlights the need for alternative therapeutic approaches. In this work, IgY antibody was raised in immunized laying hens, and its antiviral activity was evaluated in the context of passive immunization. With inactivated whole H1N1 virus, high-titer IgY antibody 9.18   mg/mL egg yolk was induced by the eighth week after immunization. Western blotting and the hemagglutination inhibition (HI) test demonstrated that the IgY antibody could specifically bind the neuraminidase and hemagglutinin of the H1N1 virus. In the plaque reduction assay, the IgY antibody reduced the H1N1 viral infection in MDCK (Madin-Darby canine kidney) cells. In a mouse model, the anti-H1N1 IgY antibody exhibited in vivo protection by reducing the infectious titer of the virus in the lung while maintaining the weight and normal structure of the lung tissue. Additionally, the anti-H1N1 IgY antibody exhibited protective activity comparable to the neuraminidase inhibitor oseltamivir. These results demonstrated that IgY can be easily produced and can offers an effective alternative approach for influenza control.

Research Note: A novel method for preparation of egg yolk immunoglobulin Y against Porphyromonas gingivalis

Han Su,Kemin Wei,Minlei Zhao,Xin Li,Yibo Zhang

ABSTRACT Porphyromonas gingivalis ( P. gingivalis, P. g ) is the main pathogen of periodontal disease, which is treated with egg yolk immunoglobulin Y ( IgY ) against P. gingivalis . In order to quickly obtain IgY, 30 hens were immunized with inactivated P. gingivalis . The purification of IgY was carried out by the oleic acid ( OA ) method and the classical method ( AS ), respectively. The IgY antibody characteristics and antibacterial effects in HPDLF cells were detected by SDS-PAGE, indirect ELISA, Western blot and viability/toxicity assays. SDS-PAGE and Western blot analysis showed that IgY molecules which were rapidly purified by OA method were complete and specific to P. gingivalis . In addition, the results of crystal violet staining and bacterial staining indicated that IgY could agglutinate with P. gingivalis , inhibiting bacterial invasion of host cells. This study is the first to rapidly and efficiently purify IgY by OA method, and the purified IgY is expected to be used in the detection and treatment of P. gingivalis .

Preparation and characterization of IgY antibody and its application in immunochromatographic assay for bongkrekic acid and its isomer

Bin Wang,Xue-Ming Cao,Ting-Ting Yun,Xiao-Min Liang,Yi-Na Lu,Yu-Xin Zhong,Zhen-Lin Xu

Egg yolk immunoglobulin (IgY) has emerged as a promising alternative to monoclonal antibodies (mAbs) due to its facile extraction, higher yield, and greater tolerance to organic solvents. This work developed a selective IgY antibody against bongkrekic acid (BA) and isobongkrekic acid (IsoBA), the lethal toxins produced by Burkholderia gladioli pv. Cocovenenans (BGC), which led to severe food poisoning incidents and resulted in casualties. In detail, the purified cBA was first conjugated with a carrier protein to generate an immunogen and immunized hens, followed by an optimized purification protocol, attaining a high-affinity anti-BA IgY with minimal cross-reactivity. Based on the obtained anti-BA IgY, a rapid and sensitive immunochromatographic assay (ICA) was developed and integrated with a smartphone. The linear range of ICA was 0.72–4.41 μg/kg with a limit of detection (LOD) of 0.42 μg/kg. Recovery rates ranged from 82.2 % to 106.4 % in spiked rice flour, agaric, and tremella, and there was no statistically significant deviation from results obtained by LC-MS/MS in blind samples. Compared to mAb-based ICAs, IgY-based ICAs offer some significant advantages, including a non-invasive production process that adheres to ethical animal welfare standards, high production yields, and exceptional tolerance to organic solvents. These attributes position IgY as a viable alternative for future commercial ICA development.

Novel Immunosensor via Dual-Species Antibody Synergy: Guinea Pig Immunoglobulin G (IgG)/Chicken Immunoglobulin Y (IgY)-Mediated Specific Determination of Staphylococcus aureus

Yongsheng Chang,Xiane Guo,Xiuzhen Ge,Xuexi Yu,Shujun Ma,Canyu Liu,Yunwei Lou,Tingmin Chang,Yun Zhang

Traditional immunoassays for Staphylococcus aureus (S. aureus) suffer from significant limitations, particularly owing to cross-reactivity with protein G-producing Streptococcus species. To address this issue, here is reported an innovative colorimetric strategy that depended on the unique binding characteristics of guinea pig immunoglobulin G (IgG) toward staphylococcal protein A (SPA), a highly expressed epitope on S. aureus. Unlike conventional monoclonal or polyclonal antibodies, guinea pig IgG exhibits high specificity for SPA, while demonstrating negligible affinity for protein G. The method also integrated horseradish peroxidase (HRP)-conjugated chicken anti-protein A immunoglobulin Y (IgY) (HRP–IgY) in a sandwich immunoassay format, enabling the catalytic oxidation of 3, 3′, 5, 5′-tetramethylbenzidine (TMB) and hydrogen peroxide. The signal intensity correlated linearly with S. aureus concentration over a dynamic range of 7.8 × 102 CFU/mL–1.0 × 105 CFU/mL, achieving a detection limit of 6.9 × 102 CFU/mL. Notably, the assay demonstrated exceptional specificity, with no cross-reactivity with other common foodborne pathogens. Compared with mainstream methods, this assay completed the entire analytical process within 90 min-shorter than polymerase chain reaction (PCR) and traditional enzyme-linked immunosorbent assay (ELISA). It eliminated the need for complex DNA extraction or strict anti-cross-contamination controls and maintained stable performance in real samples. This advantage outperforms PCR (often susceptible to matrix inhibitors) and ELISA (typically exhibits reduced sensitivity in complex matrices). By offering significant improvements over conventional techniques, this strategy holds great promise for enhancing the efficiency and reliability of S. aureus detection in practical applications.

A rapid triple-mode fluorescence switch assay for immunoglobulin detection by using quantum dots-gold nanoparticles nanocomposites

Qi Wang,Xuan Fu,Xi Huang,Fangyi Wu,Meihu Ma,Zhaoxia Cai

The immunoglobulins, most of them are glycoprotein, play an important role on humoral immune. We herein report a triple-modenano sensor with a fluorometric readout based on CdTe quantum dots (QDs) and gold nanoparticles (AuNPs) for discriminative detection of immunoglobulin. Immunoglobulins fall into several classes, each with its own functional characteristics. In this research, we use chicken egg yolk immunoglobulin (IgY) as an example to establish the specific detection methods. IgY is the only antibody existing in chicken egg yolk, which has many significant advantages. We used anti-IgY and staphylococcal protein A (SPA) to make the detection discriminatively to IgY. The immune interaction between QDs-SPA and anti-IgY-AuNPs leads to drastic quenching (turning off) of the donor by a fluorescence resonance energy transfer (FRET) process. After the addition of IgY, anti-IgY-AuNPs peel off from the QDs-SPA surface and bind to IgY due to competitive immunoreactions, resulting in the restoration of the fluorescence intensity of QDs (turning on). Consequently, this process can be utilized for the selective detection of IgY via optical responses. This proposed assay demonstrated a favorable anti interference ability and highly selectivity toward IgY with a detection limit of 1.16   ng/mL. More importantly, the nanosensor could not only function in aqueous solution for IgY detection with high sensitivity but also exhibit sensitive responses toward IgY in complicated biological environments, demonstrating its potential in bioanalysis, which might be significant in biochemical field in the future.
