Ilex asprella , a valued plant traditionally used as food and medicine, is recognized for its effect on liver protection. Polysaccharides, as the key active components in Ilex asprella (IAP), its biological activities are still unexplored. Therefore, this study investigated the preventive effects of IAP on liver injury in vivo . H&E and TUNEL staining revealed IAP could ameliorate the histomorphology changes and liver apoptosis. For antioxidant ability, IAP preconditioning released D-GalN/LPS-induced oxidative stress via Nrf2/HO-1 signaling pathway in liver, as evidenced by increasing superoxide dismutase, total antioxidant capacity, glutathione peroxidase, and inhibiting the activation of Nrf2 and HO-1. Furthermore, IAP reduced the expression of IL-6, IL-1β and TNF-α, showing good anti-inflammatory effects. According to the results from western blot, the phosphorylation of TLR4, p65, IκB, p38, ERK, and JNK were effectively inhibited with IAP pre-treatment, indicating IAP can ameliorate the liver inflammation through TLR4/NF-κB and MAPK signaling pathway. 16S RNA sequencing revealed IAP re-regulated composition of gut microbiota, including inhibiting the harmful bacteria ( Parasutterella and Erysipelatoclostridium ) enrichment and restoring the probiotics ( Lactobacillus, Dubosiella and Bacteroide ). In conclusion, this study revealed IAP can improve liver injury by inhibiting liver oxidative stress, releasing inflammation and modulating the gut microbiota.

The polysaccharides obtained from the root of Ilex asprella, namely IAPS-1 and IAPS-2, with immunoregulatory activity were studied. Two polysaccharides were isolated and purified by Cellulose DEAE-52 and Sephadex columns. The structure of IAPS-1 was elucidated as 1,6-linked α- d -glucopyranosyl main chain with branch chain substituted at C-2 and/or C-4 position. For IAPS-2, the backbone is composed of 1, 4-linked α- d -glucose, galactose and rhamnose, and branched chains consists of arabinose, rhamnose and galacturonic acid, as confirmed by partial acid hydrolysis and NMR study. Further, the immunoregulatory activity of IAPS-1 and IAPS-2 was tested with the murine macrophages. Particularly, IAPS-2 polysaccharide can more effectively enhance the secretion of major inflammatory cytokines in macrophages, such as TNF-α, IL-1β, IL-12, compared with IAPS-1.

Taking the extraction amount of rutin as the index, the extraction process of rutin from Ilex asprella (Hook. et Arn.) Champ. ex Benth. using the ultrasonic-assisted deep eutectic solvent method was optimized through response surface method. The effects of various factors, including the types of deep eutectic solvent, molar ratio of the solvent, water content, liquid-solid ratio, the temperature, power and time of ultrasound on the extraction amount of rutin were investigated. In accordance with the result of univariate experiments, the extraction process was optimized through employing the Box-Behnken response surface design method. A three-factor, three-level experimental model was established with the liquid-solid ratio, water content and ultrasonic time as variables. The findings indicated that the optimal technological conditions were as stated below: the molar ratio of lactic acid to choline chloride at 1:1, the ultrasonic extraction temperature at 40 °C, the ultrasonic extraction time at 31 min, the water content at 28%, and the liquid-solid ratio at 20:1 mL/g. Under optimal conditions, the validation experimental result revealed that the extraction amount of rutin was 86.553 ± 1.35 µg/g, with an absolute error of less than 0.6% in comparison with the predicted value of the model. The use of deep eutectic solvents for the extraction of rutin and the application of response surface method for optimizing the extraction process of rutin from Ilex asprella (Hook. et Arn.) Champ. ex Benth. were demonstrated. The present study provided a reference for the comprehensive utilization of Ilex asprella (Hook. et Arn.) Champ. ex Benth. .

Plant polyphenols have gained increasing attention due to their abundant and powerful biological activities. However, polyphenols from roots of Ilex asprella (RIAPs) remain largely unexplored, with limited research on their composition and bioactivity, making it necessary to focus on RIAPs. In the present study, single - factor analysis and response surface optimization were performed for the extraction of RIAPs via an ultrasonic bath extraction method. The findings demonstrated that at 50 % ethanol concentration, 30 min of ultrasonic time, and 300 W of ultrasonic power, the maximum extraction content of RIAPs was 30.14 ± 0.41 mg/g. Compound and structural analysis via high-resolution liquid chromatography-mass spectrometry (LC-MS/MS) and a Fourier transform infrared (FT-IR) spectrometer confirmed that polyphenols were the majority of compounds comprising RIAPs, in addition to small amounts of alkaloids and trace amounts of other compounds such as amino acids. Scanning electron microscope (SEM) images revealed that RIAPs exhibited a densely reticulated and fragmented microstructure. In addition, bioactivity experiments demonstrated significant antioxidant, antibacterial, anti-inflammatory, and anti-hyperlipidemic activities of RIAPs, supporting their potential pharmaceutical applications. This study provides an experimental foundation for further pharmacological research and potential drug development of RIAPs.

Offline LC × LC-ECD coupled with HRMS screens antioxidants in Ilex asprella leaves. • 48 antioxidants were characterized by HRMS, mainly phenolic acids and flavonoid glycosides. • ECD intensity shows strong positive correlation with DPPH and ABTS activities. • LC-ECD fingerprinting reveals distinct geographical differences in sample profiles. Ilex asprella is a traditional medicinal and edible plant with multiple pharmacological activities such as anti-inflammatory properties. Previous studies on its chemical constituents and pharmacology have mostly focused on roots and stems, whereas research on the leaves remains relatively limited. In this study, an offline comprehensive two-dimensional liquid chromatography‑electrochemical detection (LC × LC-ECD) method combined with LC-HRMS was established for the rapid screening and structural identification of antioxidant components in Ilex asprella leaves. LC × LC-ECD achieved efficient separation of components with wide polarity ranges and significant concentration differences. Using LC-HRMS, a total of 48 compounds were identified or annotated, primarily including 19 hydroxycinnamoyl quinic acids, 14 flavonol glycosides, 6 dihydroflavone glycosides, 5 simple phenolic acids and their glycosides, 2 coumarins, and 2 other compounds. In vitro assays indicated that all electrochemically active fractions obtained from the first-dimension separation exhibited varying degrees of antioxidant activity, and their ABTS and DPPH radical scavenging activities were positively correlated with the electrochemical detection response intensity ( R 2 > 0.95). Subsequently, liquid chromatography-electrochemical detection was utilized to compare the antioxidant components among 29 batches of Ilex asprella leaves from different origins. Multivariate statistical analyses, including hierarchical cluster analysis, principal component analysis, and orthogonal partial least squares-discriminant analysis, consistently demonstrated that the 29 batches of samples could be clearly grouped into three categories based on their antioxidant component profiles, and the classification results were highly correlated with geographical origins, confirming significant variations in the antioxidant component profiles of Ilex asprella leaves from different origins.
