Fast Electrochemical Determination of Imidacloprid at an Activated Glassy Carbon Electrode

Weimeng Si,Zhen Han,Wu Lei,Qiuju Wu,Yuehua Zhang,Mingzhu Xia,Qingli Hao

A fast and sensitive method for the determination of Imidacloprid (IMI) based on a simply pretreated glassy carbon electrode (GCE) has been developed. The easy activation at 2.0 V for only 30s of GCE leads to obvious catalytic property to IMI. The electrocatalytic properties and the electron-transfer kinetics of the activated GCE were investigated by cyclic voltammetry, differential pulse voltammetry and electrochemical impedance spectroscopy. The reduction mechanism of IMI was put forward and the limit of detection (LoD) was estimated to be about 0.61 μM.

Extracellular enzyme mediated biotransformation of imidacloprid by white-rot fungus Phanerochaete chrysosporium: Mechanisms, pathways, and toxicity

Shiye Zhu,Anwei Chen,Youzheng Chai,Ruoyu Cao,Jianhua Zeng,Ma Bai,Liang Peng,Jihai Shao,Xiang Wang

As one of the widely used neonicotinoid insecticide, imidacloprid (IMI) poses serious threat on many kinds of non-target species. The acceleration of its removal from the contaminated environment is therefore an urgent task. Herein, the transformation of IMI by white-rot fungus Phanerochaete chrysosporium was examined, and the reaction mechanism underlying enzymes-mediated IMI biotransformation was elucidated. In addition, transformed products, pathways, and toxicity to Escherichia coli and Microcystis aeruginosa or the growth of rice seeds were investigated. P. chrysosporium could efficiently transform 93.5% of IMI for 6 days primarily through biotransformation, in which extracellular manganese peroxidase and lignin peroxidase substantially responded to transform IMI. IMI mainly underwent nitrate reduction , hydroxylation, and de-chlorination. A less neurologically toxic major metabolite of IMI, 6-chloronicotinic acid, was detected. Molecular dynamics simulation demonstrated that IMI stably bonded to active pocket of extracellular enzymes. This study reveals the key metabolic mechanism and fate of IMI in environment, and highlights the potential of fungi for IMI transformation.

Probing the interaction of pepsin with imidacloprid via DFT calculation, spectroscopic approaches and molecular docking

Xiaopei Wang,Yuanyuan Yue,Yanyan Zhang,Zhiyue Wang,Jianming Liu,Qinghu Tang

The use of the pesticide imidacloprid can be hazardous to human health. In this work, the impacts of imidacloprid on the secondary structure of pepsin were investigated through spectroscopic techniques and simulation studies. One binding site for pepsin with strong affinity was evident from changes in the fluorescence emission spectra of pepsin in the presence of imidacloprid. Calculated thermodynamic parameters based on the van't Hoff relationship indicate the predominance of hydrophobic and hydrogen bonding interactions contributing to the imidacloprid-pepsin complex. Structural changes in pepsin were also evident from changes in the UV, FTIR , CD , three-dimensional fluorescence (3D), and synchronous fluorescence spectroscopy (SFS) of pepsin upon titration with imidacloprid. Furthermore, density functional theory (DFT) calculations and molecular docking studies revealed predominant binding of imidacloprid in the hydrophobic pocket of pepsin.

A combination of Tropilaelaps mercedesae and imidacloprid negatively affects survival, pollen consumption and midgut bacterial composition of honey bee

Shilong Ma,Yang Yang,Zhongmin Fu,Qingyun Diao,Mengyue Wang,Qihua Luo,Xing Wang,Pingli Dai

Tropilaelaps mercedesae is not only a major threat to honey bees in Asia but also a potential risk to global apiculture due to trade. Imidacloprid is a systemic insecticide that negatively affects individual bees. Moreover, the health of honey bees may be threatened by imidacloprid exposure and T. mercedesae infestation. We studied the effects of T. mercedesae and imidacloprid on the survival, food consumption and midgut bacterial diversity of Apis mellifera in the laboratory . Illumina 16S rRNA gene sequencing was used to determine the bacterial composition in the honey bee midgut. T. mercedesae decreased survival in parasitized honey bees compared with nonparasitized honey bees, but there was no significant difference in food consumption. The imidacloprid 50 μg/L diet significantly decreased syrup consumption of A. mellifera compared with the control diet. The combination of T. mercedesae infestation and imidacloprid 50 μg/L exposure reduced survival and increased pollen consumption of A. mellifera . T. mercedesae infestation or a combination of T. mercedesae infestation and exposure to 25 μg/L imidacloprid affected the midgut bacterial composition of honey bees. T. mercedesae infestation and imidacloprid exposure may reduce the survival and affect honey bee health.

Overcoming resistance in insect pest with a nanoparticle-mediated dsRNA and insecticide co-delivery system

Haixiang Lv,Xuchao Li,Jiaqing Li,Chang Yu,Qinghong Zeng,Guogui Ning,Hu Wan,Jianhong Li,Kangsheng Ma,Shun He

The cotton aphid Aphis gossypii Glover (Homoptera: Aphididae) is a notorious agricultural pest that has evolved high levels of resistance to various insecticides. Innovative RNA insecticides, as potential green plant protection products, are anticipated to alleviate the significant issue of pest resistance. Herein, a rough-surface hollow mesoporous silica (RHMS)-based gene/insecticide delivery system was constructed for the co-delivery of CYP6CY13 double-stranded RNA (dsRNA) and the chemical insecticide imidacloprid for the effective control of A. gossypii . The hollow mesoporous structure of RHMS facilitates insecticide loading and further combines with dsRNA through electrostatic interactions to form nanoscale complexes, while protecting the dsRNA from complete degradation by nucleases. The screened ds CYP6CY13 fragment was successfully expressed in the L4440-HT1115 (DE3) RNaseIII - system and inhibiting CYP6CY13 expression by RNA interference (RNAi) significantly increased the susceptibility of A. gossypii adults to imidacloprid. The toxicity of the RHMS/IMI/ds CYP6CY13 complex was significantly higher (1.95-fold) than that of imidacloprid. The results of the pot test showed that the effectiveness of the RHMS/IMI/ds CYP6CY13 complex against A. gossypii improved by 19.95 % in 5 days. Therefore, this nanoparticle-mediated gene/insecticide co-delivery system is expected to be an effective tool for pest control.

The binding interactions of imidacloprid with earthworm fibrinolytic enzyme

Yan-Qing Wang,Hong-Mei Zhang,Tao Chen

In this paper, several studies were conducted to elucidate the binding mechanism of earthworm fibrinolytic enzyme (EFE) with imidocloprid (IMI) by using theoretical calculation, fluorescence, UV–vis, circular dichroism spectroscopy and an enzymatic inhibition assay. The spectral data showed that the binding interactions existed between IMI and EFE. The binding constants, binding site, thermodynamic parameters and binding forces were analyzed in detail. The results indicate a single class of binding sites for IMI in EFE and that this binding interaction is a spontaneous process with the estimated enthalpy and entropy changes being 2.195   kJ   mol −1 and 94.480   J   mol −1   K −1 , respectively. A single class of binding site existed for IMI in EFE. The tertiary or secondary structure of EFE was partly destroyed by IMI. The visualized binding details were also exhibited by the theoretical calculation and the results indicated that the interaction between IMI and Phe (Tyr, or Trp) or EFE occurred. Combining the experimental data with the theoretical calculation data, we showed that the binding forces between IMI and EFE were mainly hydrophobic force accompanied by hydrogen binding, and π–π stacking. In addition, IMI did not obviously influence the activity of EFE. In a word, the above analysis offered insights into the binding mechanism of IMI with EFE and could provide some important information for the molecular toxicity of IMI for earthworms.

Flexible terahertz Metamaterial sensor for sensitive detection of imidacloprid

Tingting Lang,JinHui Zhang,Yanqing Qiu,Zhi Hong,Jianjun Liu

In this paper, a flexible terahertz (THz) Metamaterial (MM) with high sensitivity is proposed for the detection of the pesticide imidacloprid. The MM sensor is composed of a polyimide (PI) substrate and a periodic aluminum (Al) split ring resonator array. The sensing performance of the MM is verified by both simulation and experiment. The simulation results show that the refractive index (RI) sensitivity of the proposed MM sensor is 220.7 GHz/RIU, and the figure of merit (FOM) of this MM is 1.52. Then, the sensing performance of the MM sensor is verified by using imidacloprid solutions with different concentrations. The experimental results show that the detection sensitivity of the MM sensor is 12.3 GHz/(mg/ml), and the limit of detection (LOD) is 0.0112 mg/ml. The THz MM sensor proposed in this paper is flexible, simple to make, cost-effective, highly sensitive and has a very broad application prospect in the concentration detection of pesticides such as imidacloprid.

Phanerochaete chrysosporium-driven quinone redox cycling promotes degradation of imidacloprid

Qingru Xie,Chao Xue,Anwei Chen,Cui Shang,Si Luo

This study systematically investigated the advanced oxidation processes driven by the white-rot fungus (WRF) Phanerochaete chrysosporium in the removal of the neonicotinoid pesticide imidacloprid (IMI). The quinone redox cycling driven by P. chrysosporium promotes the degradation of IMI in presence of Mn 2+ and Fe (III)-oxalate. Results showed that the degradation efficiency varied with the type of quinine mediator, and gallic acid (GA) was a more effective mediator than 1,4-benzoquinone (BQ). The removal rate of IMI was increased by 1.8-fold in the presence of 100 μM of Mn 2+ compared with that of the system without Mn 2+ . The addition of oxalic acid and EDTA promoted the degradation of IMI, and the effect of oxalic acid was more pronounced at the later stage (day 5 and day 6); the optimal Fe 3+ /oxalate ratio was 1:3. Time-course experiments revealed the maximum removal rates of IMI were 97.37%, 82.0% and 73.52% at the initial IMI concentration of 10, 20, and 30 mg/L, respectively, in the presence of 100 μM Mn 2+ , 100 μM Fe 3+ -300 μM of oxalate and 500 μM gallic acid. Under quinone redox cycling conditions, the good correlation between ·OH production rates and the enzymes activity was concluded, the quinines redox cycles promote enzyme activity, as well as the rate of ·OH production. Results from this study indicated the GA mediator-based system using P. chrysosporium offers a new method to advance the bio-oxidation process owing to its simple culture system, low-cost precursors, unique fungi, and degradation stability.

Biodegradation of imidacloprid and diuron by Simplicillium sp. QHSH-33

Ke Xu,Xin-Ran Ke,Wan-Ting Zhang,Xin-Yuan Wu,Zhi-Jun Song,Mei-Juan Jiao,Xiao-Juan Gao,Li Zhou,Hong-Yan Ji,Fang Wang,Xiu-Li Wu

Imidacloprid (IMI) and diuron (DIU) are widely used pesticides in agricultural production. However, their excessive use and high residues have caused harm to the ecological environment and human health. Microbial remediation as an efficient and low-toxic method has become a research hotspot for controlling environmental pollutants. A fungus QHSH-33, identified as Simplicillium sp., has the ability to degrade neonicotinoids IMI and phenylurea DIU. When QHSH-33 and pesticide were co-cultured in liquid medium for 7 days, the degradation rates of IMI and DIU by QHSH-33 in simulated field soil microenvironment were 50.19 % and 70.57 %, respectively. Through HPLC-MS analysis, it was found that the degradation of IMI mainly involved nitro reduction, hydroxylation and other reactions. Three degradation pathways and eight degradation products were identified, among which two metabolites were obtained by microbial transformation of IMI for the first time. The degradation of DIU mainly involved demethylation and dehalogenation reactions, and two degradation pathways and four degradation products were identified, one of which was a new degradation product of DIU. Toxicity assessment demonstrated that most of the degradation products might be considerably less harmful than IMI and DIU. Whole genome sequencing of QHSH-33 revealed a genome size of 33.2 Mbp with 11,707 genes. The genome of QHSH-33 was annotated by KEGG to reveal 128 genes related to exogenous degradation and metabolism. After local blast with reported IMI and DIU degrading enzymes, seven IMI-degrading related genes and seven DIU-degrading related genes were identified in the QHSH-33 genome. The results of this study will help to expand our knowledge on the microbial decomposition metabolism of IMI and DIU, and provide new insights into the degradation mechanism of IMI and DIU in soil and pure culture system, laying a foundation for QHSH-33 strain applied to the removal, biotransformation or detoxification of IMI and DIU.

Microalgae fuel cells enhanced biodegradation of imidacloprid by Chlorella sp.

Zhikang Deng,Jinyao Zhu,Lie Yang,Zulin Zhang,Bolin Li,Ling Xia,Li Wu

The widespread use of neonicotinoid pesticides has caused environmental threat that has attracted scientific concern. This study developed a microalgae fuel cells (MFCs) with Chlorella sp. as the cathode synergistically enhanced the removal of imidacloprid (IMI). The MFCs were operated at open-circuit and closed-circuit. Under closed-circuit condition (with an external load), 57.20–61.66% of IMI (10–50 mg/L) was removed within 12 days. However, only 39.66–42.66% was removed in the open-circuit (without an external load). Additionally, a voltage of 200 mV was also detected in the closed-circuit MFCs, indicating the green bioenergy was produced during the degradation of IMI. Cyclic voltammetry revealed that the cathodic electroactivity was higher with the addition of IMI. Biomass accumulation (indicated by chlorophyll and dry cell weight) were significantly enhanced by IMI. On the 12th day, there was no difference in MDA content between the different IMI concentration (10–50 mg/L), while a significantly upregulated superoxide dismutase (SOD) of each group was also found at the same time. Six intermediates were detected and five major degradation pathways were proposed. Based on the identified products and microalgae growth, the degradation products of IMI were less toxic /non-toxic than IMI. Considering the efficient removal of IMI and energy production, the MFCs could be a promising technique for controlling IMI contamination by effective detoxification.

Machine learning driven decoding of impurity fingerprint in imidacloprid material

Mengling Tu,Wen Ma,Jia Chen,Yunxiao Zhu,Yang Liu,Xiaoli Ni,Xianjiang Li

In recent years, imidacloprid, a first generation neonicotinoid pesticide, has attracted considerable attention due to its high detection rate in the environment and food, as well as its detrimental impact on human beings. The identification of structure-related impurities constitutes an essential aspect of pesticide registration and product standard development, and is of paramount importance for exposure risk and safety evaluation. In this study, the analysis of impurities in high-purity imidacloprid material was conducted using a combination of liquid chromatography-high resolution mass spectrometry (Orbitrap) and machine learning platform (SIRIUS + CSI:FingerID). The structure of IMI, as well as the feature fragments, were served as the foundation for decoding the chemical structures of the impurities, and this procedure was guided by the molecular formulas and fragment tree by SIRIUS + CSI:FingerID. Ultimately, 28 impurities were annotated and assigned confidence levels, of which 10 had CAS registry numbers, 5 had PubChem CIDs, and 4 were further verified by three dimension indexes (retention time, ultraviolet spectra and mass spectra) using standards. These impurities have been categorized into three levels (4 in level 1, 12 in level 2a and 12 in level 2b) according to their credibility. This approach was capable of providing accurate molecular formulas and possible structures of impurities, thereby greatly saving the time taken to resolve mass spectra. The elucidated impurity fingerprint can assist in the assessment of the potential health and ecological risks associated with imidacloprid and related impurities.

Imidacloprid detection using terahertz metamaterial based on electromagnetically induced transparency

Jiaojiao Yu,Gan Liu,Xuan Zhang,Jianyuan Qin

Imidacloprid is a common insecticide which can remain in water, soil, and plant parts with long-term consumption, threaten environment and human health. Therefore, it is necessary to propose an effective method for imidacloprid detection. Here, a terahertz metamaterial based on electromagnetically induced transparency (EIT) is theoretically and experimentally demonstrated. This metamaterial displays an EIT resonance at 0.89 THz and the physical mechanism of the EIT effect is investigated by analyzing the electric fields and surface current distribution. The EIT resonance can be modulated by changing the geometric structure parameters of the metamaterial. This metamaterial can be used to detect the thickness and the refractive index of the dielectric layer based on the EIT resonance, and can achieve a refractive index sensitivity of 210 GHz/RIU. The detection of imidacloprid shows that the frequency shift and the transmittance change at peak are depend on the change of imidacloprid concentration. The minimum detectable imidacloprid concentration reaches a level of 0.01 ppm in this case. These results theoretically and experimentally prove the feasibility of terahertz metamaterial based on EIT resonance applied in the detection of imidacloprid concentration.
