Sweet potato [Ipomoea batatas (L.) Lam] is grown as important cash and food crop worldwide and has been shown to exhibit allelopathic effects on other plants. However, its metabolome has not been studied extensively, particularly with respect to the production of phytotoxic bioactive secondary products. In this study, the chemical composition of petroleum ether extract of sweet potato was characterized, and the morphological and physiological effects of some individual components against four invasive alien weeds Bidens pilosa L., Galinsoga parviflora Cav., Lolium multiflorum Lam., and Phalaris minor Retz. were determined. Twenty-one components were identified by GS-MS, constituting 96.08% of petroleum ether extract in sweet potato. The major components were palmitic acid (PA) (17.48%), ethyl linoleate (EL) (13.19%), linoleic acid (LA) (12.55%), ethyl palmitate (EP) (11.77%), ethyl linolenate (ELL) (8.29%) oleic acid (5.82%), ethyl stearate (4.19%), and 3-methylphenol acetate (3.19%). The five most abundant compounds exhibited strong inhibition activity against the four invasive weeds tested. The highest inhibition rates were seen for LA, followed by PA and EP, respectively. Catalase (CAT), malondialdehyde (MDA), and peroxidase (POD) content of L. multiflorum were increased by the three allelochemicals, i.e., LA, PA and EP, but superoxide dismutase (SOD), chlorophyll-a and chlorophyll-b levels declined. Overall, the combined impact of all five compounds could be quite effective in suppressing the invasive weeds of concern.

The sweet potato weevil ( Cylas formicarius (Fab.) (Coleoptera: Brentidae)) is a pest that feeds on sweet potato ( Ipomoea batatas (L.) Lam. (Solanales: Convolvulaceae)), causing substantial economic losses annually. However, no safe and effective methods have been found to protect sweet potato from this pest. Herbivore-induced plant volatiles (HIPVs) promote various defensive bioactivities, but their formation and the defense mechanisms in sweet potato have not been investigated. To identify the defensive HIPVs in sweet potato, the release dynamics of volatiles was monitored. The biosynthetic pathways and regulatory factors of the candidate HIPVs were revealed via stable isotope tracing and analyses at the transcriptional and metabolic levels. Finally, the anti-insect activities and the defense mechanisms of the gaseous candidates were evaluated. The production of ( Z )-3-hexenyl acetate ( z 3HAC) and allo -ocimene was induced by sweet potato weevil feeding, with a distinct circadian rhythm. Ipomoea batatas ocimene synthase (IbOS) is first reported here as a key gene in allo -ocimene synthesis. Insect-induced wounding promoted the production of the substrate, ( Z )-3-hexenol, and upregulated the expression of IbOS , which resulted in higher contents of z 3HAC and allo -ocimene, respectively. Gaseous z 3HAC and allo -ocimene primed nearby plants to defend themselves against sweet potato weevils. These results provide important data regarding the formation, regulation, and signal transduction mechanisms of defensive volatiles in sweet potato, with potential implications for improving sweet potato weevil management strategies.

Sweet potato ( Ipomoea batatas L.) leaves are rich in polyphenols and consumed as leaf vegetable in world. The content and composition of polyphenols are diversity at different harvesting period. This study investigated the changes of total polyphenols content (TPC), antioxidant capacity, and polyphenol fractions content of sixteen varieties of SPLs during growth stage. The results showed significant diversity in the concentrations of active compounds in different genotypes and growth stages. TPC ranged from 27.45 to 243.82 mg CAE/g DW among the varieties over various growth stages, with G3 and E1 varieties exhibiting relatively high TPC and antioxidant capacity. Twelve polyphenol fractions were identified in SPLs. Correlation analysis demonstrated a significant positive relationship between antioxidant capacity and the content of monosubstituted caffeoylquinic acids (3-caffeoylquinic acid, 5-caffeoylquinic acid, 4-caffeoylquinic acid) and disubstituted caffeoylquinic acids (3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, 4,5-di-O-caffeoylquinic acid). Principal component analysis highlighted the superior performance of the E1 variety in terms of polyphenol content and antioxidant capacity.

This study aimed to compare the extraction efficiency of dynamic high pressure microfluidization-assisted extraction (DHPMAE) and other commonly used extraction methods on the antioxidant activities (AAs) of sweet potato leaves ( Ipomoea batatas L., SPL), and to identify the antioxidant compounds by HPLC-QTOF-MS 2 . Two highly consumed commercial varieties, orange-fleshed and cream-fleshed sweet potato leaves (OFL and CFL), were taken as materials. OFL was evidenced to be a better antioxidants resource than CFL due to higher polyphenols and AAs. DHPMAE had the best extraction efficiency on total phenolics and total flavonoids , with the values in OFL extract at 16.35 mg GAE/g DM and 16.51 mg RE/g DM, respectively. DHPMAE extract also exhibited the strongest DPPH· and ABTS· + scavenging ability, chelating ability and reducing power. Multiple regression analysis revealed that polyphenols played an important contribution to the AAs of SPL extracts. After successive fractionation of DHPMAE extract by chloroform, ethyl acetate, and n -butanol, the ethyl acetate fraction showed the highest total phenolic (569.38 mg GAE/g Extract) and total flavonoid (43.15 mg RE/g Extract) content, as well as the highest AAs. HPLC-QTOF-MS 2 analysis of the ethyl acetate fraction resulted in identifying 37 compounds including 20 phenolic acids , 12 flavonoids, 3 organic acids, 1 nucleoside and 1 ester, and 20 of them were firstly detected in SPL. Caffeoylquinic acid derivatives and flavonoids were the most abundant antioxidant compounds. The presented results suggested that DHPM might be a promising assisted technology in extracting polyphenols from plants, and SPL could be utilized for the development of functional food.

We studied the relative efficacy of different forms of foliar iron (Fe) fertilisation on leaf re-greening in Fe-deficient, purple-fleshed sweet potato (Ipomoea batatas (L.) Lam.) varieties xuzi8 and xuzi6. Activities of ferric chelate reductase (FCR) and concentrations of Fe were measured in the leaves and roots at intervals over 5 days to quantify recovery from leaf chlorosis. Freshly expanded and chlorotic leaves were immersed in one of three different fertiliser compounds containing 9 mm Fe: FeSO4, Fe2(SO4)3, Fe(III)-EDTA. An Fe-sufficient treatment and an Fe-deficient control were included. The experiment had a completely randomised block design with five replications per treatment and was conducted in a sunlit glasshouse. For variety xuzi8, leaf FCR activity in the Fe2(SO4)3 treatment was highest at 1 h after application, and higher than all other treatments, whereas FeSO4 and Fe(III)-EDTA treatments showed their highest FCR at day 5 after application, both significantly higher than the Fe2(SO4)3 and control treatments. Furthermore, leaf Fe concentration reached a maximum in the FeSO4 treatment at day 1, and in the Fe2(SO4)3 treatment at day 3. By contrast, root Fe concentration was relatively constant and lower in the foliar Fe treatments than the Fe-sufficient and -deficient treatments. For variety xuzi6, leaf SPAD was higher with the Fe2(SO4)3 than the FeSO4 treatment at day 5 after application. In general, FCR activity and Fe concentrations in roots and leaves of xuzi6 were higher than those of xuzi8. Variations in leaf Fe concentrations were similar for both the FeSO4 and Fe2(SO4)3 treatments of the two varieties. Maximum leaf Fe levels in xuzi6 were ~4-fold those in xuzi8. The results of the study suggest that foliar-applied Fe2(SO4)3 was the most effective compound at correcting Fe-deficiency symptoms. The higher leaf and root FCR activity and Fe concentration in xuzi6 might explain its higher tolerance to Fe deficiency and better re-greening than xuzi8.

The sweetpotato (Ipomoea batatas[L.] Lam) is a globally significant crop, valued for its nutritional and economic importance. The tuberous roots of the sweetpotato are rich in carotenoids, which contribute to their vibrant colors and health benefits. This study focuses on three elite fresh-consumption sweetpotato cultivars: “Kokei No. 14,” “Xinxiang,” and “Zheshu81” with distinct flesh colors. To elucidate the metabolic pathways and genetic mechanisms underlying carotenoid biosynthesis in the sweetpotato, 20 types of carotenoids were quantified using targeted metabolomic analyses, and the key genes involved in carotenoid synthesis were identified with transcriptomic analyses. The results revealed significant differences in carotenoid content and composition among the cultivars, with “Zheshu81” exhibiting the highest carotenoid levels. Weighted gene co-expression network analysis further highlighted key regulatory genes and transcription factors influencing carotenoid accumulation. This study identifies key transcriptional regulators associated with carotenoid accumulation, sheds light on sweetpotato carotenoid biosynthesis mechanisms, and lays a foundation for breeding to improve its nutritional quality and flesh color.

Natural deep eutectic solvents (NADESs) coupled with microwave-assisted extraction (MAE) were applied to extract total flavonoid compounds from spent sweet potato (Ipomoea batatasL.) leaves. In this study, ten different NADESs were successfully synthesized for the MAE. Based on single-factor experiments, the response surface methodology (RSM) was applied, and the microwave power, extraction temperature, extraction time, and solid–liquid ratio were further evaluated in order to optimize the yields of total flavonoid compounds. Besides, the extracts were recovered by macroporous resin for the biological activity detection of flavonoid compounds. As a result, NADES-2, synthesized by choline chloride and malic acid (molar ratio 1:2), exhibited the highest extraction yield. After that, the NADES-2-based MAE process was optimized and the optimal conditions were as follows: microwave power of 470 W, extraction temperature of 54 °C, extraction time of 21 min, and solid–liquid ratio of 70 mg/mL. The extraction yield (40.21 ± 0.23 mg rutin equivalents/g sweet potato leaves) of the model validation experiment was demonstrated to be in accordance with the predicted value (40.49 mg rutin equivalents/g sweet potato leaves). In addition, flavonoid compounds were efficiently recovered from NADES-extracts with a high recovery yield (>85%) using AB-8 macroporous resin. The bioactivity experiments in vitro confirmed that total flavonoid compounds had good DPPH and O2−· radical-scavenging activity, as well as inhibitory effects onE. coli,S. aureus,E. carotovora, andB. subtilis. In conclusion, this study provides a green and efficient method to extract flavonoid compounds from spent sweet potato leaves, providing technical support for the development and utilization of sweet potato leaves’ waste. Keywords:natural deep eutectic solvents;response surface;spent sweet potato leaves;antioxidant activity

Black spot disease, which is caused by the pathogenic fungalCeratocystis fimbriata, seriously affects the production of sweet potato and its quality during postharvest storage. In this study, the preliminary identification of the rhizosphere actinomycete strain SPS-33, and its antifungal activity of volatilesin vitroandin vivowas investigated. Based on morphological identification and phylogenetic analysis of the 16S rRNA gene sequence, strain SPS-33 was identified asStreptomyces lavendulae.Volatile organic compounds (VOCs) emitted by SPS-33 inhibited mycelial growth and sporulation ofC. fimbriatain vitroand also induced a series of observable hyphae morphological changes. In anin vivopathogenicity assay, exposure to SPS-33 significantly decreased the lesion diameter and water loss rate in sweet potato tuberous roots (TRs) inoculated withC. fimbriata. It increased the antioxidant enzymes’ activities of peroxidase, catalase, and superoxide dismutase as well as decreased malondialdehyde and increased total soluble sugar. In the VOC profile of SPS-33 detected by a headspace solid-phase micro extraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS), heptadecane, tetradecane, and 3-methyl-1-butanol were the most abundant compounds. 2-Methyl-1-butanol, 3-methyl-1-butanol, pyridine, and phenylethyl alcohol showed strong antifungal effects againstC. fimbriata. These findings suggest that VOCs fromS. lavendulaeSPS-33 have the potential for pathogenC. fimbriatacontrol in sweet potato postharvest storage by fumigant action. Keywords:Ipomoea batatas(L.)
