Residual pesticides in agricultural production are one of the major food safety concerns around the world. In this study, we investigated the amount of isofenphos-methyl in corns by using surface-enhanced Raman spectroscopy (SERS) coupled with chemometric methods. The highest isofenphos-methyl peak at 1043 cm −1 was used to detect and quantify the amount of isofenphos-methyl spiked in methanol-water (1:1) solvent and corns. The limits of detection (LODs) of isofenphos-methyl in methanol-water solvent and corns were 0.01 μg/mL and 0.01 μg/g, respectively. Quantitative detection of isofenphos-methyl residues (0–5 μg/mL) was conducted using partial least squares (PLS) model. Based upon the results of leave-one-out cross-validation, isofenphos-methyl could be predicted by PLS (the correlation coefficient was 0.9964) with a low root mean square error of calibration (RMSEC=0.146). The developed SERS method provides a much simpler and more sensitive way to monitor isofenphos-methyl for safe production of corns, which can be extended to the other agricultural products.

Isofenphos-methyl (IFP) is widely used as an organophosphorus for controlling underground insects and nematodes. However, excessive use of IFP may pose potential risks to the environment and humans, but little information is available on its sublethal toxicity to aquatic organisms. To address this knowledge gap, the current study exposed zebrafish embryos to 2, 4, and 8 mg/L IFP within 6–96 h past fertilization (hpf) and measured mortality, hatching, developmental abnormalities, oxidative stress, gene expressions, and locomotor activity. The results showed that IFP exposure reduced the rates of heart and survival rate, hatchability, and body length of embryos and induced uninflated swim bladder and developmental malformations. Reduction in locomotive behavior and inhibition of AChE activity indicated that IFP exposure may induce behavioral defects and neurotoxicity in zebrafish larvae. IFP exposure also led to pericardial edema, longer venous sinus-arterial bulb (SV-BA) distance, and apoptosis of the heart cells. Moreover, IFP exposure increased the accumulation of reactive oxygen species (ROS) and the content of malonaldehyde (MDA), also elevated the levels of antioxidant enzymes of superoxide dismutase (SOD) and catalase (CAT), but decreased glutathione (GSH) levels in zebrafish embryos. The relative expressions of heart development-related genes ( nkx2.5 , nppa , gata4 , and tbx2b ), apoptosis-related genes ( bcl2 , p53 , bax , and puma ), and swim bladder development-related genes ( foxA3 , anxa5b , mnx1 , and has2 ) were significantly altered by IFP exposure. Collectively, our results indicated that IFP induced developmental toxicity and neurotoxicity to zebrafish embryos and the mechanisms may be relevant to the activation of oxidative stress and reduction of acetylcholinesterase (AChE) content.

Isofenphos-methyl (IFP), an organophosphorus pesticide widely used in agriculture, has raised public health concerns because of its potential risks to human health. Although recent studies have linked IFP exposure to developmental toxicity, the underlying toxicological mechanisms remain insufficiently understood. To address this gap, this study employed a zebrafish model to investigate the developmental toxicity of IFP through an integrated approach combining behavioral, histopathological, biochemical, and transcriptomic analyses. Exposure to IFP from 2 to 96 hours post-fertilization (hpf) adversely affected early zebrafish embryonic development, with a calculated 96-h LC₅₀ of 6.407 mg/L. Embryos exposed to 0.1, 2, 4, 8 mg/L IFP diminished responsiveness to light/dark stimulation, and disrupted circadian rhythms. Moreover, IFP exposure impaired immune function, significantly reducing in macrophage and neutrophil numbers. Transcriptomic analysis revealed disruptions in neuroactive ligand–receptor interaction, leading to dysfunctional synaptic signalling and neuronal communication; and it also disturbed protein digestion and absorption, thereby limiting the availability of essential amino acid precursors (e.g., tryptophan and tyrosine) required for the synthesis of key neurotransmitters such as serotonin and dopamine (DA). These transcriptomic alterations were supported by altered neurotransmitter levels, brain mitochondrial damage, and histopathological abnormalities in both brain and ocular structures. RNA-sequencing results were further validated using quantitative real-time reverse transcription polymerase chain reaction (RT-qPCR), confirming the robustness of the data. Collectively, these findings demonstrate that IFP exerts pronounced neurotoxic and immunotoxic effects on developing zebrafish, underscoring potential risks during early developmental stages and the urgent need for stricter regulation of IFP contamination in aquatic environments.
