Upconversion-nanoparticle-functionalized Janus micromotors for efficient detection of uric acid

Ye Yuan,Changyong Gao,Zhexu Wang,Jianming Fan,Haofei Zhou,Daolin Wang,Chang Zhou,Baohua Zhu,Qiang He

We report enzyme-powered upconversion-nanoparticle-functionalized Janus micromotors, which are prepared by immobilizing uricase asymmetrically onto the surface of silicon particles, to actively and rapidly detect uric acid. The asymmetric distribution of uricase on silicon particles allows the Janus micromotors to display efficient motion in urine under the propulsion of biocatalytic decomposition of uric acid and simultaneously detect uric acid based on the luminescence quenching effect of the UCNPs modified on the other side of SiO2. The efficient motion of the motors greatly enhances the interaction between UCNPs and the quenching substrate and improves the uric acid detection efficiency. Overall, such a platform using uric acid simultaneously as the detected substrate and motion fuel offers considerable promise for developing multifunctional micro/nanomotors for a variety of bioassay and biomedical applications.

Multi-response biocompatible Janus micromotor for ultrasonic imaging contrast enhancement

Yiwen Feng,Xiaocong Chang,Hao Liu,Ying Hu,Tianlong Li,Longqiu Li

The acid-base environment of the biological digestive system is formed with the help of hydrogen ions and bicarbonate. At the same time, the chloride capacity in the blood is not ignorable. These ionic environments provide a new horizon for the medical application of bubble-propelled micromotors. Herein, a Janus Mg-based micromotor is reported as a novel ultrasonic contrast agent and its potential advantages in medical imaging are explored. With the capability of being powered by the magnetic field, these micromotors could be navigated to the corresponding ionic environment, and then they would rapidly generate continuous bubbles. These bubbles achieve an efficient and guided propulsion of micromotor, also provide support for contrast-enhanced ultrasound imaging. Besides owning the dual functions of imaging and targeted transport, the micromotors present aggregation response under the action of ultrasound, which is helpful for local reinforcement of the imaging effect. Such multi-response biocompatible Janus micromotor provides a new perspective for medical imaging and diagnosis.

Enhanced and Robust Directional Propulsion of Light-Activated Janus Micromotors by Magnetic Spinning and the Magnus Effect

Jianjie Li,Xiaoli He,Huaide Jiang,Yi Xing,Bi Fu,Chengzhi Hu

Advances in the versatile design and synthesis of nanomaterials have imparted diverse functionalities to Janus micromotors as autonomous vehicles. However, a significant challenge remains in maneuvering Janus micromotors by following desired trajectories for on-demand motility and intelligent control due to the inherent rotational Brownian motion. Here, we present the enhanced and robust directional propulsion of light-activated Fe3O4@TiO2/Pt Janus micromotors by magnetic spinning and the Magnus effect. Once exposed to a low-intensity rotating magnetic field, the micromotors become physically actuated, and their rotational Brownian diffusion is quenched by the magnetic rotation. Photocatalytic propulsion can be triggered by unidirectional irradiation based on a self-electrophoretic mechanism. Thus, a transverse Magnus force can be generated due to the rotational motion and ballistic motion (photocatalytic propulsion) of the micromotors. Both the self-electrophoretic propulsion and the Magnus force are periodically changed due to the magnetic rotation, which results in an overall directed motion moving toward a trajectory with a deflection angle from the direction of incident light with enhanced speed, maneuverability, and steering robustness. Our study illustrates the admirable directional motion capabilities of light-driven Janus micromotors based on magnetic spinning and the Magnus effect, which unfolds a new paradigm for addressing the limitations of directionality control in the current asymmetric micromotors.

Ion-imprinted MnO2/CoFe2O4 Janus magnetic micromotors synthesized by a lotus pollen template for highly selective recognition and capture of Pb(II) ions

Yang Han,Yangsai Lyu,Ningning Xing,Xiaolei Zhang,Kaiyuan Hu,Hui Luo,Dickon H. L. Ng,Jia Li

Micromotors that combine the merits of micro-devices with specific functions and autonomous motion have numerous applications in environmental remediation. However, their practical application is blocked by low productivity, a complex preparation process and high cost. Herein, we developed a facile biomass route to synthesize ion-imprinted MnO2/CoFe2O4 Janus micromotors with appealing recognition and capture capacities for Pb(II) ions. Owing to the combination of autonomous motion, the ion imprinting technique, and magnetic recovery, the ion-imprinted MnO2/CoFe2O4 Janus micromotors exhibited highly sensitive and selective adsorption and quick removal of lead ions from water. Such a Janus micromotor reveals great potential in environmental remediation.

Biodegradable magnesium fuel-based Janus micromotors with surfactant induced motion direction reversal

Zewei Zhao,Tieyan Si,Anna I. Kozelskaya,Igor O. Akimchenko,Sergei I. Tverdokhlebov,Sven Rutkowski,Johannes Frueh

The speed and motion directionality of bubble-propelled micromotors is dependent on bubble lifetime, bubble formation frequency and bubble stabilization. Absence and presence of bubble stabilizing agents should significantly influence speed and propulsion pattern of a micromotor, especially for fast-diffusing molecules like hydrogen. This study demonstrates a fully biodegradable Janus structured micromotor, propelled by hydrogen bubbles generated by the chemical reaction between hydrochloric acid and magnesium. Six different concentrations of hydrochloric acid and five different concentrations of the surfactant Triton X-100 were tested, which also cover the critical micelle concentration at a pH corresponding to an empty stomach. The Janus micromotor reverses its propulsion direction depending on the availability and concentration of a surfactant. Upon surfactant-free condition, the Janus micromotor is propelled by bubble cavitation , causing the micromotor to be pulled at high speed for short time intervals into the direction of the imploding bubble and thus backwards. In case of available surfactant above the critical micelle concentration, the Janus micromotor is pushed forward by the generated bubbles, which emerge at high frequency and form a bubble trail. The finding of the propulsion direction reversal effect demonstrates the importance to investigate the motion properties of artificial micromotors in a variety of different environments prior to application, especially with surfactants, since biological media often contain large amounts of surface-active components.

γ-Fe2O3@Ag-mSiO2NH2 magnetic Janus micromotor for active water remediation

Jie Yang,Yong Liu,Jia Li,Min Zuo,Wenzheng Li,Ningning Xing,Chengyan Wang,Tingting Li

Micromotors have hold great promise for environmental remediation. Herein, we reported a novel γ-Fe 2 O 3 @Ag-mSiO 2 NH 2 magnetic Janus micromotor for dynamic removal of heavy metal ions and organic pollutants from water. The as-prepared Janus micromotors displayed unique core-shell structure composed of amino-functionalized meso‑SiO 2 shell and γ-Fe 2 O 3 magnetic core, and could be propelled by O 2 bubbles generated by Ag-catalyzed decomposition of H 2 O 2 . This Janus micromotor could move with a high velocity of 203.06 ± 10.6 μm s −1 (about 253.8 body lengths s –1 ) in 10 wt.% H 2 O 2 solution. γ-Fe 2 O 3 @Ag-mSiO 2 NH 2 magnetic Janus micromotor showed much higher removal capacities for Cu 2+ (15.59 mg g −1 ) and doxycycline (31.45 mg g −1 ) than its static control samples due to the synergy of self-propulsive movement and amino-functionalized porous core-shell structure. Furthermore, the magnetic γ-Fe 2 O 3 cores enabled the remote magnetic control on the movement direction and magnetic recycling. This work provides a new strategy for environmental pollutants treatment.

Janus micromotors for motion-capture-ratiometric fluorescence detection of circulating tumor cells

Long Zhao,Yuan Liu,Songzhi Xie,Pan Ran,Jiaojun Wei,Qingjie Liu,Xiaohong Li

The rapid and sensitive detection of circulating tumor cells (CTCs) remains technical challenges due to extremely low abundance. In the current study, Janus micromotors (JMs) are developed to achieve motion-enhanced CTC capture and capture-induced ratiometric fluorescence signaling. JMs are constructed via catalase grafting on one side of Janus rods (JRs) to catalyze the decomposition of H 2 O 2 as power source. JMs with different aspect ratios show random, spiral, rotational or linear motion trajectories, and JMs with the aspect ratio of 2 (JM-2) display rotational motion with significantly larger mean-square-displacement (MSD) values than other JMs. TLS11a aptamers are conjugated on the other side of JRs for specific capture of tumor cells, and tetraphenylethene (TPE) derivatives and fluorescein isothiocyanate (FITC) are labeled on aptamers via base-pair interactions. The competitive binding of tumor cells with aptamers causes the release of TPE and FITC from JMs, which relieves the aggregation-induced emission (AIE) effect of TPE and aggregation-caused quenching (ACQ) profile of FITC on JMs. Thus, JMs displays apparent ratiometric fluorescence changes from blue ( I 450 ) to green ( I 526 ) after capture of tumor cells. The fluorescence intensity ratios of I 526 / I 450 could be fitted against cell levels, and the limit of detection is around 25 cells/mL within 1 min. In addition, the HepG2 detection by JM-2 exhibits no interference in the presence of other tumor cells (4T1 and H22), and the recovery rate of HepG2 cells in blood samples was over 95%. Thus, JMs demonstrate the motion-capture-ratiometric fluorescence sensing capabilities and provide a feasible strategy for real-time, rapid and sensitive detection of cells in laboratory, hospital and remote settings.

One-step synthesis of PCL/Mg Janus micromotor for precious metal ion sensing, removal and recycling

Zhang Dongmei,Wang Dan,Li Jieai,Xu Xiaoyi,Zhang Hui,Duan Ruomeng,Song Bo,Zhang Dafeng,Dong Bin

The field of the micromotor has undergone continuous progresses, especially in the environmental remediation application. However, to the best of our knowledge, there are few micromotors which can sense, remove and recycle metal ions simultaneously. In this paper, we develop a Janus micromotor consisting of polycaprolactone (PCL) and magnesium (Mg) microparticle synthesized by one-step method. Due to the displacement reaction between Mg and the precious metal ions, the resulting micromotor can be self-propelled in hydrogen peroxide (H 2 O 2 ) solution. Since the motion velocity is highly dependent on both ion species and their concentrations, the micromotor can function as a motion-based metal ion sensor with good selectivity and sensitivity. In addition, since Mg alone is capable of propelling the whole structure in the presence of chloride ion (Cl − ), the water-driven PCL/Mg micromotor exhibits higher efficiency when utilized in noble metal removal and recycling as compared to that of the stationary one. We thus believe the one-step fabrication, precious metal ion sensing, removal and recycling capabilities make the current micromotor potentially attractive for polluted water monitoring and treatment.

Light/gas cascade-propelled Janus micromotors that actively overcome sequential and multi-staged biological barriers for precise drug delivery

Xiang Zhou,Xiuhong Huang,Bincheng Wang,Lihui Tan,Yi Zhang,Yanpeng Jiao

Passive diffusion-based drug delivery systems have indeed shown impressive performance to overcome a cascade of biological barriers, but the delivery efficiency is still modest. Although artificial micro/nanomotors are capable of autonomous movement, oriented navigation and seeking target actively represent an alternative and attractive strategy to revolutionize drug delivery patterns potentially, the separated and single propulsion mode of motors still hampers their further biomedical application that needs to overcome successive biological barriers. Herein, we demonstrated a cascaded NIR-light/gas-driven Janus calcium carbonate particle micromotors (JCPMs) to overcome multi-staged biological barriers and achieved active drug delivery. These JCPMs exhibited NIR light-driven thermophoretic motion with precisely steered start/stop switch, speed, and motion trajectory in biological media and displayed CO 2 -bubbles-powered autonomous propulsion in an acidic environment. With the collaborative use of cascaded light/gas double-mode driving, JCPMs displayed a multistage self-adapted strategy to overcome sequential biological barriers during tumor-targeted drug delivery in a cascaded manner, involving of light-driven active tumor cells seeking, facilitated tumor penetration and cellular membranes traversal by virtue of photo-thermomechanical force and self-thermophoretic propulsion, gas-driven lysosomes escape and intracellular drug release successively. Such a strategy guaranteed trajectory-controllable active drug delivery to exert the selective tumor therapeutic effect as well as avoided side effects. We envisioned that the proposed cascaded driving mode of micromotors for traversing biological barriers would boost the delivery efficiency for tumor treatment, which may pave a convenient and versatile way to develop next generation active vehicles in the biomedical field.

Ultrasound-powered hydrogen peroxide-responsive Janus micromotors for targeted thrombolysis and recurrence inhibition

Wenxiong Cao,Wei Wei,Bo Qiu,Yuan Liu,Shuang Xie,Qibo Fang,Xiaohong Li

Thrombosis seriously endangers human health with high incidence and mortality worldwide, and the clinical treatment is challenged by low bioavailability of thrombolytic agents, high bleeding risk and thrombosis relapse. Herein, ultrasound-powered Janus micromotors with stimuli-responsive multiple release capabilities are developed for efficient thrombolysis and inhibition of thrombosis recurrence. Resveratrol-loaded hyaluronic acid (HA Res ) nanoparticles (NPs) and urokinase plasminogen activator (uPA) were entrapped into H 2 O 2 -sensitive poly(1,4-cyclohexanedimethanol-co-oxalate) (POX) microparticles (MPs), followed by polydopamine capping and RGD grafting to obtain Janus rJPox@u-HA Res MPs. After RGD-mediated delivery to the thrombus site, the peroxalate ester bonds of POX are oxidized by the elevated H 2 O 2 to produce CO 2 , and the MP collapse accelerates the local release of uPA and HA Res NPs. The resulting CO 2 bubbles amplify the cavitation effect of ultrasound to promote thrombus penetration and site-specific uPA thrombolysis. The endothelial cell targeting and sustained resveratrol release from HA Res NPs dramatically improve endothelial cell viability, nitric oxide production and migration. Compared with free drug administration, the uPA loading into MPs significantly extends the half-life (15 folds) and bioavailability (7 folds), while the resveratrol inoculation into NPs increases the bioavailability 29 times. On an acute lower limb thrombosis model, the thrombus accumulation of MPs is promoted by RGD-mediated interactions with the activated platelets and ultrasound-driven penetration into thrombi, leading to almost full removal of blood clots. The H 2 O 2 -scavenging capability of POX, sustained resveratrol release and efficient thrombolysis alleviate oxidative stresses, eliminate coagulation biomarkers and repair the damaged endothelial layer to effectively inhibit thrombosis relapse. It is demonstrated that rJPox@u-HA Res /US treatment could not only achieve safe and site-specific thrombolytic therapy at the early stage but also restore vascular homeostasis to effectively prevent thrombosis recurrence.
