An eco-friendly and effective method for isolating essential oil from fresh Kaempferia galanga L.: Process optimization based on entropy weight method and response surface methodology

Qingfen Zhang,Dan Ge,Shengcheng Mi,Qi Lu

The bioactive properties of essential oil are strongly associated with their chemical composition. In this work, a novel method of salt-enhanced homogenization pretreatment coupled with solvent-free microwave extraction (HP-SSFME) was developed for extracting essential oil (KGEO) from fresh Kaempferia galanga rhizomes, which significantly improved the extraction efficiency. Gas chromatography was employed to measure the contents of the main active components, ethyl p -methoxycinnamate (EPMC) and ethyl cinnamate (EC) in the essential oil. The extraction process was optimized by the Box-Behnken Design using the comprehensive scores of the yields of EPMC, EC, and KGEO obtained by the entropy weight method. The optimal parameters were determined as follows: amount of sodium chloride was 1.60 %, homogenization speed of 15000 r/min, homogenization time of 4 min, microwave irradiation power of 300 W, microwave irradiation time of 23 min. Under these conditions, the KGEO yield was 5.10 %, EPMC yield was 3.81 %, EC yield was 0.89 %. Compared with the conventional methods, the HP-SSFME method not only significantly increased the extraction yield of KGEO but also remarkably enhanced the yields of EPMC and EC. Additionally, the HP-SSFME method has low CO 2 emission and energy consumption, highlighting its environmental friendliness. Moreover, in terms of antibacterial activity, the HP-SSFME outperformed other methods, demonstrating a more powerful and extensive inhibitory effect against a range of bacteria. This study provides a valuable reference for the efficient extraction and quality control of essential oils from Kaempferia galanga rhizomes.

Repellent and Insecticidal Effects of the Essential Oil of Kaempferia galanga Rhizomes to Liposcelis bostrychophila (Psocoptera: Liposcelidae)

Liu Xin Chao,Liang Yan,Shi Wang Peng,Liu Qi Zhi,Zhou Ligang,Liu Zhi Long

The aim of this research was to determine chemical composition and repellent and insecticidal activities of the essential oil of Kaempferia galanga L. rhizomes against the booklouse, Liposcelis bostrychophila Badonnel, and to isolate insecticidal or repellent constituents from the oil. The essential oil was obtained by hydrodistillation and analyzed by gas chromatography–mass spectrometry. Twenty-eight components of the oil were identified. The major compounds in the oil were ethyl-ρ-methoxycinnamate (38.6%), ethyl cinnamate (23.2%), 1,8-cineole (11.5%), trans-cinnamaldehyde (5.3%), and borneol (5.2%). Based on bioactivity-guided fractionation, four active constituents were isolated from the oil and identified as 1,8-cineole, ethyl cinnamate, ethyl ρ-methoxycinnamate, and trans-cinnamaldehyde. The essential oil exhibited contact toxicity against the booklouse with an LC50 value of 68.6 μg/cm2. Ethyl cinnamate (LC50 = 21.4 μg/cm2) exhibited stronger contact toxicity than ethyl ρ-methoxycinnamate and trans-cinnamaldehyde (LC50 = 44.6 and 43.4 μg/cm2, respectively) while 1,8-cineole showed weak acute toxicity. The essential oil also possessed fumigant toxicity against the booklouse with a LC50 value of 1.5 mg/liter air. 1,8-Cineole and trans-cinnamaldehyde (LC50 = 1.1 and 1.3 mg/liter, respectively) possessed stronger fumigant toxicity against the booklouse than ethyl cinnamate and ethyl ρ-methoxycinnamate (LC50 = 10.2 and 10.2 mg/liter air, respectively). trans-Cinnamaldehyde was strongly repellent to booklice, whereas ethyl cinnamate and ethyl ρ-methoxycinnamate were weakly repellent and 1,8-cineole did not repel booklice. The results indicate that the essential oil and its constituent compounds have potential for development into natural insecticides or fumigants and repellents for control of insects in stored grains.

Extracellular vesicles derived from Kaempferia Galanga L. show promise for targeted oral therapy in the treatment of ulcerative colitis

Lizhen Lin,Xinghong Zhou,Baizhao Peng,Jing Ma,Zhuping Zheng,Cuiping Jiang,Zhuoen He,Shuyuan Li,Wen Fang,Chuanghai Wu,Zaihao Jiang,Yao Chen,Lina Zhong,Hiu Yee Kwan,Chunyan Shen,Shenhai Gong,Xiaoshan Zhao

Ulcerative colitis (UC) is characterised by chronic intestinal inflammation and its global prevalence is increasing. Although a variety of drugs have been approved for the clinical treatment of UC, their application is often limited by unsatisfactory long-term effects, side effects, and high treatment costs. Therefore, there is an urgent need for the development of effective drugs with fewer side effects. In this study, we found that the extracellular vesicles extracted and purified from rhizomes of Kaempferia galanga L. (KGEVs) exhibit promising therapeutic effects in treating UC disease. With an average diameter of 133.8 nm, KGEVs are rich in functional components, including lipids, proteins, and pharmacologically and immunologically active molecules. In vivo experiments revealed that KGEVs accumulated in the colorectal region 6 h after oral administration, demonstrating targeted enrichment at the site of enteritis. Moreover, we found that KGEVs effectively alleviate DSS-induced colitis in mice, as indicated by reductions in body weight loss, DAI score, spleen index and colon length shortening. Mechanistically, KGEVs may alleviate colitis by repairing the intestinal barrier, inhibiting oxidative stress and colonic inflammation regulating gut microbiota and inhibiting the polarisation of macrophages into pro-inflammatory M1 macrophages during the inflammatory response, indicating significant anti-inflammatory effects. These results suggest the potential of KGEVs as a promising, cost-effective, and efficient oral therapeutic agents for UC treatment.

Optimized essential oil extraction method based on salt solution-enhanced homogenization pretreatment and microwave extraction for Kaempferia galanga L

Qingfen Zhang,Chenglin Li,Shengcheng Mi,Xiaojie Xu,Zhiqiang He,Lin Wang,Zhonghua Tang,Qi Lu

The primary purpose of this study is to develop a highly efficient extraction method to meet the growing demand for Kaempferia galanga essential oil (KGEO) in various industries. In this study, an innovative approach for extracting essential oil from fresh K. galanga rhizomes using salt solution-enhanced homogenization pretreatment and microwave extraction (HP-SOME) was proposed. A kinetic study was conducted on the extraction process of essential oil. The key factors were screened using the Plackett–Burman Design and optimized using the Box-Behnken Design. Under the optimization parameters: sodium chloride concentration of 1.5 %, microwave power of 475 W, and microwave time of 23 min, the actual yield was 5.26 ± 0.10 %. The HP-SOME method exhibited a significant increase of 58.91 % in essential oil yield compared to that of the hydrodistillation method (3.31 ± 0.10 %). In addition, the composition of the KGEO was determined using Fourier transform infrared spectroscopy, Raman spectroscopy, and gas chromatography-mass spectrometry. The KGEO obtained by HP-SOME had a 6.26 % increase in the main component ethyl p-methoxycinnamate and a 4.13 % increase in ethyl cinnamate compared to that obtained by the HD method. Investigations into antimicrobial activity revealed that KGEO extracted by the HP-SOME method exhibited superior and more comprehensive antimicrobial characteristics. In conclusion, the HP-SOME method demonstrated remarkable capabilities in obtaining KGEO with notable efficiency, yield, and quality. This method holds great promise in the field of essential oil extraction.
