The soybean residue (okara) with high moisture content has generally been discarded, which has led to economic loss and socio-environmental problems. The effects of solid state fermentation using Kluyveromyces marxianus on the nutritional components, beany odor and processing properties of soybean residue were studied. The results showed that K. marxianus fermented soybean residue (FSR) had an increase in crude fat (24.5%), soluble dietary fibers (158%) and polysaccharides (262%), while phytic acid (61.7%) and the activity of a trypsin inhibitor (92.7%) decreased. A significant reduction in the beany odor was also observed. FSR showed higher water absorption and better emulsifying properties. Overall, the study indicated that fermentation with K. marxianus resulted in improved nutritional values and processing characteristics of the soybean residue.

Artemisinin, a vital antimalarial agent, is traditionally extracted from Artemisia annua L. by plant-based extraction. The microbial biosynthesis of the artemisinin precursor, amorpha-4,11-diene (AD), followed by its chemical conversion into artemisinin provides a sustainable solution to meet the demand for artemisinin while overcoming the limitations of plant-based extraction. Kluyveromyces marxianus is a promising host for microbial cell factories owing to its robust thermotolerance, rapid growth kinetics, and ability to utilize a wide range of carbon sources. Herein, genetic modification tools for K. marxianus , including transformation methods and expression vectors, were identified and optimized, while the sesquiterpene biosynthetic capacity of K. marxianus was preliminarily evaluated. To enhance the precursor metabolite supply, eight key genes from the endogenous mevalonate pathway were overexpressed within the K. marxianus genome. A dual co-expression strategy, involving both extrachromosomal and chromosomally integrated expression of amorpha-4,11-diene synthase ( ADS ) was employed, followed by high-temperature fermentation to optimize production. Finally, an engineered K. marxianus strain was developed for the first time, capable of stable, efficient, and cost-effective production of AD, with a titer of 66.78 mg/L, a 113-fold increase compared with the wild-type strain. This engineered strain serves as a robust chassis and a valuable reference for the production and study of other terpenoids, laying the groundwork for further exploration of K. marxianus characteristics and the biosynthesis of AD.

Background:Whey fermentation could produce bioactive substances with immunomodulatory effects, metabolic syndrome modulation, and antioxidant properties, thereby imparting functional characteristics to products and facilitating the development of novel foods with health-promoting potential. Methods:A non-targeted metabolomics approach using liquid chromatography–mass spectrometry (LC-MS) was employed to investigate changes in the metabolite profiles of whey fermented byKluyveromyces marxianusstrain KM812 over varying fermentation durations. Results:The findings demonstrated a progressive enrichment of metabolites over time. A total of 151 differential metabolites were identified and categorized primarily into amino acids, peptides, and analogues, fatty acids and conjugates, and carbohydrates and conjugates, as well as benzoic acids and derivatives. The highest relative content of whey metabolites was observed at 48 h of fermentation, with a cumulative increase of 1.45-fold, 1.49-fold, 3.39-fold, and 1.24-fold for peptides and amino acids, peptides, and analogues, fatty acids and conjugates, and carbohydrates and conjugates, respectively. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed associations with 23 specific metabolites and delineated 9 metabolic pathways, predominantly involved in amino acid and lipid metabolism. Conclusions:Based on the above, KM812 could effectively degrade macromolecular substances in whey into small molecules such as L-isoleucine, ornithine, betaine, α-linolenic acid, and palmitoleic acid, thereby influencing the nutritional and functional properties of whey. In-depth analysis of the metabolic products in KM812-fermented whey could provide a theoretical basis for the development of functional foods derived from small molecules in the future.

The demand for controllable fragrance materials is substantial owing to their potential to impart enduring scents in a variety of applications. However, the practical application of such materials has been limited by challenges in tunable morphogenesis, structural variability, and adaptability to diverse conditions. In our study, we introduce a hybrid living material that integrates a genetically engineered strain of Kluyveromyces marxianus CBS6556 with an adaptive hydrogel. The engineered K. marxianus achieved temperature stability in 2-phenylethanol (2-PE) and 2-phenylethyl acetate (2-PEAc) production by expressing relevant genes in the 2-PE metabolic pathway using the high-temperature preferential promoter SSE1. The enhanced water retention capacity supports the metabolic activities of the encapsulated yeast cells, ensuring their survival and functionality over an extended period. Fragrance-releasing living material (FLM) is designed to controllably emit fragrance 2-PE by adjusting the microbial concentration within the hydrogel matrix. The FLM exhibits versatile adhesion capabilities, effectively binding to a spectrum of surfaces such as wood, textiles, and glass as well as to natural substrates like leaves. This adaptability enhances the material’s applicability across various settings. Furthermore, FLM can be crafted into various forms, including microbeads, fibers, and films. This research opens up new horizons for controlled fragrance release of living materials.

Kluyveromyces marxianus is a fast-growing, food-grade yeast with broad substrate utilization, however, its limited expression tools hinder its synthetic biology applications. Here, we developed single- and dual-gene expression systems based on screened elements, including a promoter pENO and a bidirectional promoter pHTX. These systems enabled stable expression across multiple K. marxianus strains, with expression levels affected by genetic element combinations, carbon source, and culture time. Using the single-gene system with a short intergenic sequence (IGG) for bicistronic expression, the production of leghemoglobin from Vigna angularis (VaHB) reached 30.4 mg/L using galactose as carbon source. In contrast, the dual-gene system achieved 48.4 mg/L of VaHB with the co-expression of HEM1 , enhancing heme biosynthesis using glucose as a carbon source. Additionally, Simian Virus 40 (SV40) nuclear localization signals (NLS) directed fluorescent proteins to the nucleus, enabling subcellular targeting. This toolkit supports efficient and context-responsive gene expression in K. marxianus , facilitating its development as a versatile microbial chassis for industrial protein production and synthetic biology applications. Superior blue mold inhibition by K. marxianus NCWP versus S. cerevisiae ’s. • Unique rough, folded surface microstructure of the β-glucan NCWP. • Enhanced pear antioxidant capacity and early defense response activation. • Transcriptome revealed K. marxianus NCWP activation of hormone and immune pathways. • Coordinated upregulation of defense genes underpinning boosted pear resistance. Yeast cell wall polysaccharides represent a green and effective strategy for inducing fruit disease resistance, although their underlying mechanisms remain to be fully elucidated. This study investigated the efficacy and defense mechanisms of insoluble cell wall polysaccharides (NCWP) derived from Kluyveromyces marxianus in enhancing pear resistance against Penicillium expansum . Structural analysis revealed that K. marxianus NCWP is a β-glucan characterized by rough, folded, and protuberant surface morphology. This distinctive microstructure was associated with a stronger inhibitory effect on blue mold in pear fruit compared to the smoother NCWP from Saccharomyces cerevisiae . Treatment with K. marxianus NCWP triggered early defense responses, including a reactive oxygen species (ROS) burst, increased activities of antioxidant and defense-related enzymes, and upregulation of the corresponding genes. It also promoted endogenous γ-aminobutyric acid (GABA) accumulation and activated the GABA shunt pathway. Transcriptomic analysis further demonstrated that NCWP significantly induced jasmonic acid, abscisic acid, and ethylene signaling pathways, and upregulated pattern-triggered immunity (PTI)-related genes such as MKK5, PTI5 , and WRKY1 . Collectively, these findings indicate that K. marxianus NCWP enhances fruit resistance by concurrently activating immune signaling, hormone pathways, and metabolic defense processes, providing new insights into the mode of action of yeast-derived resistance inducers.

Hyperuricemia (HUA) is a systemic metabolic disorder closely linked to disrupted host-microbiota interplay. Current first-line drugs (e.g., allopurinol) primarily target urate synthesis but lack tissue-repair capabilities and cause long-term adverse effects. Although microbial metabolites circumvent the inflammatory risks of live probiotics, the urate-lowering mechanisms of Kluyveromyces marxianus fermentation metabolites remain largely uncharacterized. Here, we integrated in vitro bioassays, in vivo animal experiments, and multi-omics profiling to elucidate the anti-hyperuricemic efficacy of an ethyl acetate extract from K. marxianus XZ1. In vitro , the XZ1 extract exhibited significant antioxidant and xanthine oxidase (XOD) inhibitory activities. The cell-free extract exerted a dual-action mechanism: directly suppressing urate synthesis by inhibiting XOD and adenosine deaminase (ADA) activities, and enhancing systemic urate clearance through bidirectional regulation of renal and intestinal urate transporters (upregulating excretory ABCG2/OAT1 and downregulating reabsorptive URAT1/GLUT9). Integrated 16S rRNA sequencing and untargeted metabolomics revealed these systemic benefits are driven by gut microbiota remodeling, forming a “microbiota–short-chain fatty acid (SCFA)–gut–kidney urate transporter” regulatory axis. The intervention enriched SCFA-producing taxa, normalized fecal SCFA profiles, strengthened intestinal barrier integrity and reprogrammed host purine and redox metabolic networks. Collectively, K. marxianus XZ1 fermentation products lower uric acid and protect against HUA-induced multi-organ injury, inflammation, and oxidative stress. These findings highlight its potential as a safe, industrially stable, and multi-target functional food ingredient for early dietary intervention in asymptomatic hyperuricemia.

Yeast-derived cell wall polysaccharides possess numerous biological activities, but their application in postharvest preservation is rarely reported. The aim of this research was to investigate the effects of Kluyveromyces marxianus soluble cell wall polysaccharide (SCWP) on preventing the infection of Penicillium expansum in pear fruit. The results showed that K. marxianus SCWP treatment could significantly improve the resistance of pear fruit to P. expansum , with respect to Saccharomyces cerevisiae -derived SCWP. Composition of both SCWPs was mannan with the main chains consisting of a → 6)-α-D-Manp-(1 → unit and the branch structure formed by → 2)-α-D-Manp-(1 except that K. marxianus SCWP took on a shorter side chain and a rougher surface than S. cerevisiae SCWP. In addition, mechanisms of K. marxianus SCWP on stimulating resistance response were associated with the apparent oxidative burst, increased gene expression and enzyme activity of antioxidant and defense systems in pear fruit. Our findings suggest that K. marxianus SCWP can be used as an innovative and promising candidate for preventing postharvest fungal decay and extending fruit shelf life.

To improve the flavor of tamarind and enhance its market prospects, Kluyveromyces marxianus was used to ferment tamarind pulp. The physicochemical properties, bioactive components, and flavor compounds of the tamarind pulp were determined. The mechanism underlying flavor formation in the fermented tamarind pulp was further explored from the perspectives of metabolomics and transcriptomics. During fermentation, the malic acid content increased significantly from 85.92 mg/100 mL to 546.76 mg/100 mL. Electronic nose analyses indicated a time-dependent enhancement of aroma complexity in fermented tamarind pulp. 3-Methyl-1-butanol, phenethyl alcohol, isoamyl acetate and phenethyl acetate were the dominated volatile compounds. The emergence of these flavor compounds markedly altered and enriched the aroma profile of tamarind pulp. However, the levels of flavor-contributing amino acids in the tamarind pulp decreased, though they were likely utilized by the yeast to generate new flavor compounds and support its growth. Transcriptome profiling revealed significant enrichment of pyruvate metabolism, amino acid degradation, and redox-related processes, providing molecular evidence for metabolic reprogramming in the yeast strain.

Postharvest fungal infestation poses a significant threat to fresh fruit, leading to substantial economic losses. This study investigated the efficacy and mechanism of yeast cell walls in inducing fruit resistance, with a focus on extracts from Kluyveromyces marxianus. Compared to the commonly referenced Saccharomyces cerevisiae cell wall, application of the K. marxianus cell wall provided superior protection in pear fruit against Penicillium expansum infection, markedly reducing disease incidence. This enhanced biocontrol activity is attributed to its distinct polysaccharide composition, characterized by 60.37% mannan, 38.34% glucan, and 1.29% chitin, and its unique structural properties. The treatment triggered a rapid wound oxidative burst, upregulated key antioxidant and defense-related enzymes, and promoted the accumulation of γ-aminobutyric acid in pear tissue. Transcriptomic analysis revealed that β-glucan and mannan components synergistically activated plant hormone signaling and plant–pathogen interaction pathways. Notably, mannan specifically modulated carbohydrate metabolism and MAPK-associated defense responses. These findings establish the K. marxianus cell wall as a potent and novel biocontrol agent, offering a sustainable strategy for managing postharvest decay through the targeted induction of fruit innate immunity.

Kluyveromyces marxianus is a traditional fermentation starter, but the effects of K. marxianus fermentation on tomato remain largely unknown. Here, K. marxianus was employed for tomato puree fermentation under sterile conditions. Sensory evaluation revealed that K. marxianus decreased the unpleasant flavor components in fermented tomato puree (FTP). GC–MS analysis demonstrated that K. marxianus fermentation improved the flavor and aroma, primarily due to increases in alcohols (48.84 %) and esters (43.31 %) and enrichment of aroma-associated metabolites in volatile organic compounds. Widely targeted metabolomics showed that the fermentation significantly increased 37 phenolic compounds, potentially enhancing the antioxidant effects of FTP. The contents of lycopene isomers including cis -lycopene significantly increased in FTP (179.52 μg/g). Notably, FTP showed enrichment of beneficial genera, including Akkermansia and Pediococcus , in fecal microbiota. Collectively, these results indicate that K. marxianus fermentation is a viable approach to improving the sensory profile and potential health benefits of fermented tomato.
