Anthocyanins are plant pigments widely present in fruits and vegetables that exert health benefits as part of human diet. They exist in not glycosylated forms (anthocyanidins) and glycosylated forms. Anthocyanins have been studied for their ability to inhibit polysaccharide metabolism enzymes (such as α-glucosidase) and, therefore, attenuate hyperglycemia and modulate type II diabetes. The aim of the present work was to compare the inhibitory activity of anthocyanins with that of acarbose, a common α-glucosidase inhibitor, by evaluating IC50 and Km, to compare the inhibitory capabilities among different compounds in an in vitro study. Our results showed that most of the anthocyanins studied presented lower IC50 values than acarbose; only Pn-3-glc presented IC50 value close to acarbose. Moreover, there was no difference in Km between complete competitive and complete non-competitive types of inhibition (T-test, p > 0.05). In conclusion, the anthocyanins and their degradation products work together on attenuating abilities of this sugar metabolism key enzyme, throughout competitive and non-competitive modes.

Traditional Chinese medicine Glycyrrhiza uralensis ( G. uralensis ) exerts anti-inflammatory and anti-allergic effects. However, there is currently no valid evidence regarding its anti-histamine effect. This study aimed to elucidate the antihistamine effect of Glycyrrhiza uralensis Fisch essential oils (GUEOs) and glabridin (Gla) extracted from G. uralensis in SPF KM mice and investigate the underlying mechanisms. The chemical components of GUEOs were analyzed using gas chromatography–mass spectrometry (GC–MS), and quality control was conducted. An acute histamine allergy model was established in SPF KM mice following one week of continuous histamine administration. Scratching behavior and appearance of back scratches were observed. Hematoxylin and eosin staining was used to detect the thickness of the mouse dermis. Toluidine blue staining was performed on dermal tissues to count the number of mast cells. Immunohistochemistry was used to determine the integrated optical density (IOD) values of nerve growth factor (NGF) and interleukin-1β (IL-1β). Quantitative Real-time polymerase chain reaction (qRT-PCR) was used to determine the relative intensity of IL-4 and IL-6. Enzyme-linked immunosorbent assay (ELISA) was employed to detect the content of interferon-γ (IFN-γ),IL-1 and filaggrin (FLG). Compared with the model group, both the GUEOs and Gla groups exhibited reduced scratching behavior in mice, accompanied by epidermal thinning, decreased mast cell numbers, downregulated expression of IL-1β, NGF, IL-4, IL-6, IFN-γ, and IL-1, and upregulated FLG expression. We showed that GUEOs and Gla exert a favorable inhibitory effect on histamine-induced allergic inflammation in mice, suggesting that G. uralensis can serve as a promising natural anti-histamine agent.

A new kind of hetero-bifunctional reactive dye containing 2-ethoxy-4-chloro- s -triazine, with better activity matching with β-hydroxyethyl sulphone sulphate at a low fixation temperature, was successfully synthesised and characterised. An ethoxy group was designed to be introduced into triazine to increase the substantivity and the reactivity of the dyes. Thereby, the fixation of mono- s -chlorotriazine/hydroxyethyl sulphone sulphate (KM-type) bifunctional reactive dyes on cotton was improved, and dyeing under mild conditions with high fixation was realised. Nineteen dyes with orange, red and blue colours were synthesised and characterised by UV-Vis and infrared spectroscopy and mass spectrometry. Synthesised dyes were dyed at 60, 75 and 90°C, respectively. Exhaustion and reactivity for all of the ethoxy-containing KM-type dyes was over 90% at the optimal temperature, and fixation was over 87%, which was higher than the ethoxy-free comparative dyes. The optimal fixation temperature of ethoxy-containing dyes was initially reduced to 75°C, and then to 60°C. Compared with comparative dyes, the wash fastness and light fastness were basically unchanged, and the wet rub fastness of the H-acid series was reduced by one grade.

Formate oxidase (FOX) has applications in enzymatic assays, pollutant biodegradation, and bioorganic chemistry. However, its sources are few and none of them have significant activity above pH 7. We performed bioinformatic mining of FOX and identified 579 potential sequences. Two FOXs from Aspergillus nomiae (AnFOX) and Trichophyton rubrum (TrFOX) were expressed in E. coli. AnFOX had an alkalinity-tolerant pH activity range. It not only had a Km value for formate one order of magnitude lower, but also had far higher catalytic activity under non-acidic conditions. Spectroscopic and mass spectrometry analyses found that AnFOX contains an 8-formyl FAD cofactor. AnFOX enabled the H2O2-dependent sulfoxidation of thioanisole over pH 6-8. Structural analysis and kinetic studies revealed that the acidic residue E142, positioned on the surface adjacent to the isoalloxazine ring of flavin, could contribute to the unique pH preference. This novel FOX could greatly expand the applicability under non-acidic conditions.

Ulcerative colitis is a chronic gastrointestinal disease characterized by intestinal inflammation and serious mucosal damage. As a naturally hydroxycinnamic acid, sinapic acid (SA) has antioxidant, anticancer, and neuroprotective activities. We investigated the anticolitic effect and potential mechanisms of SA in DSS-induced colitis in Kunming (KM) mice. SA treatment significantly reduced body weight loss, colon shortening, and intestinal wall thickening in colitis mice. SA treatment also significantly reduced the histological infiltration of inflammatory cells and decreased myeloperoxidase (MPO) activity in the colons of colitis mice. The administration of SA attenuated oxidative damage by enhancing the activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase and reduced the serum and colonic mRNA levels of proinflammatory cytokines in colitis mice. We used qRT-PCR and Western blotting assays and demonstrated that SA reduced the activation of the NLRP3 inflammasome and attenuated intestinal permeability by enhancing the expression of ZO-1, occludin, and claudin-1 in colitis mice. Here, we conclude that SA exhibits great anticolitic activity against DSS-induced colitis by enhancing the activity of antioxidant enzymes, reducing intestinal inflammation, and maintaining the intestinal barrier. Finally, we suggest that SA may be a safe adjuvant for the prevention of clinical colitis.

l -arabinose isomerases (L-AIS) are enzymes that mediate the conversion of l -arabinose into l -ribulose and D-galactose into D-tagatose. The potential of L-AI from Lactobacillus parabuchneri CICC 6004 (LPAI) in D-galactose biotransformation using E. coli whole-cells was studied. The parameters investigated in this study were 1) the optimum pH, temperature, and cofactor specificity for maximum enzyme activity ; 2) the D-tagatose biosynthesis from D-galactose using whole-cells containing LPAI. The results showed that with D-galactose, the purified LPAI showed the highest activity, 15.52 U/mg at pH 6.7, 45 °C, and necessitates 3 mM Ca 2+ as its cofactor. The K m , V max and K cat /K m values of LPAI for D-galactose were 5.4 mM, 35.5 U/mg and 2.8 mM −1 min −1 , respectively, while the respective values for l -arabinose were found to be 4.3 mM, 63.1 U/mg and 5.9 mM −1 min −1 .The applicability of LPAI for D-tagatose synthesis was shown by using whole-cell biocatalyst approach. Under the optimum conditions, during one-stage biotransformation of D-galactose by E. coli whole cells, 39.0 g/L of D-tagatose was produced from 140 g/L of D-galactose after 50 h, which corresponded to the yield and productivity of 0.28 g/g and 0.78 g/L/h, respectively. Further addition of new cells into the reaction mixture in the second stage produced 54.0 g/L of D-tagatose after 100 h. The D-tagatose total yield and productivity were 0.39 g/g and 0.54 g/L/h, respectively. The results attained in this study showed LPAI as a potential candidate for the industrial production of D-tagatose.

A synergistic regulation strategy combining phonon-electron modulation and interfacial wear-resistance is developed to tackle the long-term stability bottlenecks. • The introduction of thermally conductive filler realizes efficient phonon heat dissipation, leading to a 38.1 °C reduction in interface temperature and thus effectively suppressing electron thermionic emission for stable energy output. • The optimized TENG achieves superior output stability, with merely 0.46% decay in electrical output even after undergoing 16 km sliding under a 20 N load. • It represents a 324-fold enhancement in output stability compared to the device without MoS 2 incorporation. The sliding-mode triboelectric nanogenerators (S-TENGs) are highly promising for mechanical-to-electrical energy conversion, owing to their simple structural design and high charge transfer efficiency. However, their practical applications are severely restricted by inadequate long-term stability, which primarily stems from material wear and frictional heat-induced electron thermionic emission. Herein, we tackle both challenges via the synergistic regulation of phonon-mediated heat dissipation and interfacial wear resistance. Specifically, thermally conductive fillers are incorporated into a tetrafluoroethylene-hexafluoropropylene-vinylidene fluoride terpolymer matrix to significantly enhance phonon heat transfer, leading to a remarkable 38.1 °C reduction in the interface temperature, effectively restraining the electron thermionic emission effect. Notably, the optimized S-TENG demonstrates superior output stability, exhibiting only 0.46% output decay after 16 km sliding under a 20 N load, which corresponds to a 324-fold enhancement in operational stability. Meanwhile, a liquid lubricant is introduced into a porous thermoplastic polyurethane film to effectively reduce frictional resistance, thereby mitigating material wear. The resulting S-TENG exhibits exceptional mechanical robustness, as evidenced by minimal surface wear even following 100000 sliding cycles. This work presents an innovative strategy to enhance the durability and extend the lifespan of S-TENG devices, which has also been successfully implemented in a self-powered system. The authors showcase an innovative dual-regulation strategy integrating phonon-mediated-driven efficient heat dissipation and interfacial wear resistance to resolve S-TENG stability issues induced by heat and material wear. The optimized S-TENG exhibited merely 0.46% output decay after continuous sliding 16 km, and the material surface remained undamaged, enabling high-performance energy harvesting. The newly semi-synthetic β -lactam antibiotics play critical roles in prevention and treatment of β -lactam antibiotic-resistant bacteria. Acetyl xylan esterases belong to carbohydrate esterase 7 family can specifically hydrolyze 7-aminocephalosporanic acid (7-ACA) to produce deacetyl-7-aminocephalosporanic acid (D-7ACA) which is one of the most important starting materials to synthesize newly semi-synthetic β -lactam antibiotics. Here, an effective acetyl xylan esterase, ThAXE, from Thermobifida halotolerans YIM 90462 T , was firstly identified and characterized. ThAXE exhibited its highest catalytic activity against p -nitrophenyl acetate ( p -NPC 2 ) at pH 8.5 and 50 °C, with K m and K cat / K m were 1.0 ± 0.1 mM and 2262.9 mM −1 s −1 , respectively. Additionally, ThAXE can hydrolyze the acetyl group of 7-ACA at its C3' position with the specific activity of 7.4 U mg −1 . Furthermore, molecular docking combined with site-directed mutagenesis validation demonstrated that the pocket of catalytic residues Ser186, Asp272, and His301 could accommodate p -NPC 2 and 7-ACA ligands. This is the first report of a novel acetyl xylan esterase from T. halotolerans that has considerable potential biotechnological values.

Purpose: To explore the potential relationship between salivary amino acid concentrations and 8 km running performance in male undergraduate students. Methods: Thirty male undergraduate students were recruited. Participants completed an 8 km run while wearing smart bracelets. Saliva samples were collected before, immediately after, and 24 h after the run. Ultra-High Performance Liquid Chromatography (UHPLC) was used to quantify salivary amino acids. Results: The fast group (average speed > 12.80 km/h) had a significantly shorter running time (35.66 ± 1.30 min,p< 0.001) and higher speed (13.59 ± 0.46 km/h,p< 0.001) than the slow group. Before the run, salivary serine concentration (20.19 µg/mL,p= 0.013) was higher in the fast group. After 24 h, salivary glutamine concentration (6.65 µg/mL,p= 0.047) was lower in the fast group. Salivary threonine concentration was positively correlated with running speed. For every 1 µg/mL increase in salivary threonine concentration, average running speed increased by 0.011 km/h, and this correlation persisted after adjusting for age and heart rate. Conclusions: This study found a positive correlation between salivary threonine and 8 km running speed, along with differences in serine and glutamine concentrations among runners with different speeds. These findings provide preliminary evidence for the relationship between salivary amino acid concentrations and running performance, though further research with larger samples and diverse exercise types is needed.

BACKGROUND: Chitin is the second largest carbon source on the earth, and chitosan oligosaccharides produced by its degradation have good application prospects in medicine, cosmetics, and agricultural production. OBJECTIVE: The discovery of a chitinase with high efficiency,
high stability and clear degradation mechanism is of great help to promote the research of chitin derivatives and the development of the industrial chain. MATERIALS AND METHODS: In this experiment, a lowtemperature chitinase-producing strain Photobacterium sp. LG-29 was isolated
from deep-sea mud in the Bohai Sea, and studied by means of molecular biology, biochemistry and bioinformatics. RESULTS: Purification of chitinase yielded an enzyme solution with a concentration of 0.918 mg/mL and a specific activity of 21.036 U/mg. The optimum action temperature is
35°C, and it is still active at 4°C, showing low-temperature enzymatic activity, and also has certain thermal stability. The optimum pH is 8.0, and it maintains more than 70% of the enzyme activity at pH 11, which is very stable in an alkaline environment. Mn2+, Ca2+,
and Mg2+ are the main activators of enzymes, while Fe2+, Zn2+, etc. have extremely significant inhibitory effects on enzymes. The Km and Kcat of chitinase were determined to be 269.05 μ mol/L and 0.49 min-1, respectively. Chitinase PbCHI5 has
both endonuclease and exonuclease activity. The theoretical pI of the enzyme is 4.16, which is a stable hydrophilic protein. CONCLUSION: This experiment laid a theoretical foundation for the development and utilization of new low-temperature chitinases.

Abstract N -Acety-L-cysteine (NAC), enriched with multiple electron-rich groups involving –SH, –C=O, and –COOH, exhibits strong affinities to transition metal ions. Herein, NAC was adopted as the stabilizing agent to synthesize a series of peroxidase-like Pt nanoclusters (NCs) with different physicochemical properties. Both the cluster size and the charge state of NAC–Pt NCs are highly dependent upon the molar ratio of [K 2 PtCl 4 ]/[NAC], contributing to different peroxidase mimicking activities. The most active Pt nanozyme (~ 1.7 nm), composed of 60% metallic Pt 0 and 40% positively charged Pt 2+ respectively, exhibits the K m of 0.132 mM for 3,3′,5,5′-tetramethylbenzidine and 35 mM for H 2 O 2 . Based on the heparin-activated enzymatic activity of NAC–Pt at pH 6.0, an ultrasensitive test was established for quantitative determination of heparin, giving the linear response of 0.5–20 μg/mL as well as the limit of detection of 2 × 10 −3 μg/mL. This proposed method is also applicable in biological fluid for practical applications. Such a colorimetric method possesses several advantages involving high sensitivity, short response, eco-friendly synthesis, low consumption of material and energy, free of expensive instrument, as well as simple operation. Chondroitin AC lyase can efficiently hydrolyze chondroitin sulfate (CS) to low molecule weight chondroitin sulfate, which has been widely used in clinical therapy, including anti-tumor, anti-oxidation, hypolipidemic, and anti-inflammatory. In this work, a novel chondroitin AC lyase from Pedobacter xixiisoli ( Px chonAC) was cloned and overexpressed in Escherichia coli BL21 (DE3). The characterization of Px chonAC showed that it has specific activities on chondroitin sulfate A, Chondroitin sulfate C and hyaluronic acid with 428.77, 270.57, and 136.06 U mg −1 , respectively. The K m and V max of Px chonAC were 0.61 mg mL -1 and 670.18 U mg -1 using chondroitin sulfate A as the substrate. The enzyme had a half-life of roughly 660 min at 37 °C in the presence of Ca 2+ and remained a residual activity of 54 % after incubated at 4 °C for 25 days. Molecular docking revealed that Asn123, His223, Tyr232, Arg286, Arg290, Asn372, and Glu374 were mainly involved in the substrate binding. The enzymatic hydrolysis product was analyzed by gel permeation chromatography, demonstrating Px chonAC could hydrolyze CS efficiently.
