Currently black pigments face challenges of non-ideal luminance factor (L-value), and the presence of potentially hazardous cobalt (Co). A novel stable Na 2 NiMn 2 O 6 (NNMO) black pigment is reported in this work. The pigment exhibits excellent properties, including an ideal low L-value, Co-free composition, and stable bright black hue. A range of analytical techniques are employed to evaluate the properties of the novel pigment, including XRD, SEM, EDS-Mapping, XPS, UV–V is , and CIELAB colorimetry. The chromaticity parameters of the black pigment are L∗ = 24.39, a∗ = 0.21, and b∗ = 0.39. The bright black color of the pigment and the glazing sample was attributed to the combined contributions of Ni 2+ and Mn 3+ /Mn 4+ in the Na 2 NiMn 2 O 6 pigment and NiMnO 3 , Na 4 Mn 9 O 18 , Mn 2 O 3 ingredients in the glazing sample. This research offering valuable guidance for the development of novel, Co-free, and low L-value black pigment.

Efficient methods and universal DNA elements are eagerly required for the expression of proteins and the production of target chemicals in synthetic biology and metabolic engineering. This paper develops a customized-design approach by utilizing the host-independent T7 expression system (HITES), which facilitates the rational design and rapid construction of T7 expression systems. Firstly, the E i L (Upper-limit value of initial enzyme activity) value is discovered to play a pivotal factor in the successful construction of the T7 expression system, different host strains exhibit varying E i L values, and this study presents a method to measure the E i L values. Secondly, E. coli DH5α is chosen as the host strain, and it demonstrates that various strategies to modulate T7 RNA polymerase activity can efficiently construct the HITES T7 expression system in E. coli DH5α under the guidance of E i L. Lastly, the customized-design of HITES enables the efficient expression of sfGFP and D-MIase proteins across 13 host strains, guided by E i L values. This customized-design method of HITES offers a streamlined pathway for T7 system construction across a broad range of hosts and serves as an enabling tool for synthetic biology, metabolic engineering, and enzyme engineering.

Summary The effects of ultrasound (US) pretreatment combined with drying process on storage quality maintenance and bioactive compound accumulation of broccoli stems were investigated. Results indicated that US pretreatment (λ = 8.5 mm) delayed the decline of L* value and ascorbic acid content but inhibited the increase of a* and b* values, browning index, and microbial counts of fresh-cut broccoli stems. Furthermore, the US application enhanced total phenolics content by activating phenylalanine ammonia-lyase, cinnamate-4-hydroxylase, and 4-coumarate-CoA ligase in phenylpropane metabolism. Meanwhile, it also promoted γ-aminobutyric acid (GABA) accumulation by upregulating activities of diamine oxidase, polyamine oxidase, and aminoaldehyde dehydrogenase in GABA shunt and polyamine degradation pathway, respectively, which in turn, accelerated the biosynthesis of GABA from polyamines and glutamate. In addition, compared with other three drying methods, the combined vacuum freeze drying (VFD) could further retain the sensory attributes and bioactive compounds to a maximum extent in dehydrated broccoli stems. Hence, current findings illustrated that US pretreatment combined with VFD process is beneficial for commercial quality maintenance and nutritional value enhancement, thus providing new perspectives on the comprehensive utilisation of broccoli stems.
