A simple but highly sensitive electropolymerization of L-citrulline and β-cyclodextrin based voltammetric sensor for metribuzin

Huimin Li,Yapan Shi,Yudong Gao

A new voltammetric sensor, simultaneous electropolymerization of L-Citrulline (L-Cit) and β-cyclodextrin (β-CD) film modified glassy carbon electrode (GCE) was built for the first time. It was used to construct this sensor for quantification of these important pesticides and sensitive determination of metribuzin. The proposed metribuzin sensor displays excellent performance with wide linear range (3.0 × 10−8 ~ 1.0 × 10−6 mol L−1), ultralow detection limit of 10 nM (S/N = 3), and favourable specificity. Also, the sensor was applied for determination of metribuzin in the real samples, the satisfying results are achieved. The results imply that the proposed sensor can be potentially used for the environmental pollution science and environmental analytical chemistry.

Metabolomic and Transcriptomic Changes Underlying the Effects of L-Citrulline Supplementation on Ram Semen Quality

Guodong Zhao,Xi Zhao,Jiachen Bai,Airixiati Dilixiati,Yukun Song,Aerman Haire,Shangshang Zhao,Aikebaier Aihemaiti,Xiangwei Fu,Abulizi Wusiman

Simple SummaryOur previous study found that the addition ofL-citrulline (L-Cit) to feed improved ram semen density as well as vitality and sperm mitochondrial membrane potential. In this experiment, after feeding 12 g/d ofL-Cit to rams for 70 consecutive days, the contents of amino acids, sugars, and pyruvate in seminal plasma, in addition to the changes in metabolites in plasma, were analyzed by metabolomics, and the differentially expressed genes in the hypothalamus and testes were further studied through a transcriptome analysis. The results showed thatL-Cit could increase the contents of pyruvate and most amino acids in seminal plasma, and that the differentially expressed genes in the hypothalamus and testes were significantly enriched in protein-metabolism-related pathways. This shows thatL-Cit can improve the contents of amino acids in rams’ semen by promoting the protein metabolism of the hypothalamus–testes axis, thus improving semen quality.AbstractThis study examined the effects ofL-Cit supplementation on ram semen quality through metabolomics and transcriptomics. A total of 16 rams were randomly categorized into two groups. The control group was fed a basic diet, whereas the experimental group received feed supplemented with 12 g/d ofL-Cit. Semen and blood were collected from the rams on days 0 and 72 to measure sugar, pyruvate, amino acid, and nontargeted metabolite contents. Additionally, hypothalamic and testicular tissues were collected for a transcriptomic analysis. We found 27 differential metabolites between the control and experimental groups, of which 21 were downregulated (p< 0.05) and 6 were upregulated (p< 0.05). Compared with the control group, xylose and pyruvate contents in seminal plasma increased by 43.86% and 162.71%, respectively (p< 0.01). Additionally, the levels of 11 amino acids showed a significant increase in seminal plasma (p< 0.01). Furthermore, 961 and 715 differentially expressed genes were detected in the hypothalamic and testicular tissues, respectively. The pathways of significant enrichment in the hypothalamus and testes were protein digestion, absorption, glycolysis/gluconeogenesis, and amino as well as nucleotide sugar metabolisms. In the present study,L-Cit improved protein synthesis and blood metabolism, consequently increasing the contents of most amino acids in ram seminal plasma. Specifically, the hypothalamus controlled the expression of glycolysis/gluconeogenesis-related genes in the testes through its metabolites released into the serum, thereby providing energy for sperm production, which led to a decrease in the sugar content of seminal plasma.Keywords:metabolome;transcriptome;L-Cit;semen quality regulation;rams

A novel voltammetric sensor based on poly(l-Citrulline)/SWCNTs composite film modified electrode for sensitive determination of picroside II

Wenjing Wang,Lu Wang,Lina Zou,Gaiping Li,Baoxian Ye

A novel voltammetric sensor was constructed by simple dripping single-walled carbon nanotubes (SWCNTs) on to the glass carbon electrode (GCE) firstly and electro-polymerizing l -Citrulline film subsequently. The resulting poly( l -Citrulline)/SWCNTs/GCE showed a significant voltammetric response to picroside II due to the synergistic effect of SWCNTs and poly( l -Citrulline) film. The first electroanalytical method of picroside II was proposed with detection linear range from 8.0×10 −8 to 5.0×10 −6   mol   L −1 and a detection limit of 3×10 −8   mol   L −1 .The high sensitivity, selectivity and long-term stability made the sensor suitable for the determination of picroside II. Moreover, based on the systematically investigation and some kinetics parameters calculated in the experimentation, the reaction mechanism of picroside II at the poly( l -Citrulline)/SWCNTs modified GCE was obtained reliably. Lastly, the proposed sensor was used for the determination of picroside II in real sample with satisfactory results. This work promoted the potential applications of amino acid materials and SWCNTs in electro-chemical sensors.

Voltammetric Detection of 3-Nitropropionic Acid in Sugarcane Juices Using a Poly(L-citrulline)/Lanthanide-Containing Cyano-Bridged Heterometallic Coordination Polymer Modified Electrode

Suxia Wang,Min Zhou,Yongxi Zhu,Bo Peng,Jia Yang,Yongjun Ma

In this study, a novel poly(L-citrulline)/lanthanide-containing cyano-bridged heterometallic coordination polymer modified glassy carbon electrode, poly(L-Cit)/(Ho(III)-Fe(III)-WO42− CyHMCP/GCE, was constructed as a sensing platform for the electrochemical detection of 3-nitropropionic acid (3-NPA) using the two-step electrosynthesis method. The surface morphology of materials coated on the chemically modified electrode was characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Based on the excellent synergistic electrocatalytic performance for electrochemical reduction of 3-NPA, the voltammetric detection of 3-NPA in real sugarcane juice samples was achieved with a facile sample pretreatment process and less matrix interference. Furthermore, by means of water-phase infrared spectra technique, the electrocatalytic reduction pathway of 3-NPA was preliminarily studied. Under the optimized detection conditions, a good linear relationship was obtained between the reduction peak current of 3-NPA and testing concentration in the range of 40.0–4.00 × 103 μM, with a regression equation of Ip (μA) = −3.126 + 0.08804C3-NPA (μM), R2 = 0.9997. The lower detection limit (LOD) of 3-NPA was estimated to be 16.0 μM (S/N = 3). Therefore, the method had been employed to analyze raw sugarcane samples, with the average recovery of 96.8%–104%, and the actual concentration level of free 3-NPA in the raw sugarcane juices was revealed.

Facile synthesis of a high efficiency and durability L-citrulline flame retardant for cotton

Yu Chen,Shidong Liu,Caiyan Wan,Guangxian Zhang

A novel flame retardant (FR), the ammonium salt of citrulline-penta (methylphosphonic acid) (ACPMPA) based on L-citrulline was synthesized, and its structure was characterized by 13 C, 1 H, and 31 P nuclear magnetic resonance (NMR) spectroscopy. The ACPMPA flame retardant molecule contains five ammonium salts of phosphorus acid and one ammonium salt of carboxylic acid, which allowed the covalent attachment of ACPMPA onto cellulose via -P=O(-O-C) and –COOC bonds. The results showed that the treated cotton fabrics had very high flame retardance and excellent durability. The limiting oxygen index (LOI) of cotton fabric treated with 35%-ACPMPA reached 49.2% and only decreased to 34.2% after 50 laundry cycles. Vertical flame tests also demonstrated that the treated cotton fabric acquired good flame retardance. The thermogravimetry (TG) and TG-IR results showed that the treated cotton left more residues and released almost no flammable volatiles at high temperatures. The cone calorimetry results showed that the treated cotton released less heat than pure cotton. Scanning electron microscopy (SEM) and X-ray diffraction (XRD) results demonstrated that the structure of the treated cotton fabric was almost unchanged, and no free formaldehyde was detected, indicating that the treated cotton was safe. The treated cotton fabric also retained good tensile strength and whiteness.

L-Citrulline Supplementation Restrains Ferritinophagy-Mediated Ferroptosis to Alleviate Iron Overload-Induced Thymus Oxidative Damage and Immune Dysfunction

Tongtong Ba,Dai Zhao,Yiqin Chen,Cuiping Zeng,Cheng Zhang,Sai Niu,Hanchuan Dai

L-citrulline (L-cit) is a key intermediate in the urea cycle and is known to possess antioxidant and anti-inflammation characteristics. However, the role of L-cit in ameliorating oxidative damage and immune dysfunction against iron overload in the thymus remains unclear. This study explored the underlying mechanism of the antioxidant and anti-inflammation qualities of L-cit on iron overload induced in the thymus. We reported that L-cit administration could robustly alleviate thymus histological damage and reduce iron deposition, as evidenced by the elevation of the CD8+T lymphocyte number and antioxidative capacity. Moreover, the NF-κB pathway, NCOA4-mediated ferritinophagy, and ferroptosis were attenuated. We further demonstrated that L-cit supplementation significantly elevated the mTEC1 cells’ viability and reversed LDH activity, iron levels, and lipid peroxidation caused by FAC. Importantly, NCOA4 knockdown could reduce the intracellular cytoplasmic ROS, which probably relied on the Nfr2 activation. The results subsequently indicated that NCOA4-mediated ferritinophagy was required for ferroptosis by showing that NCOA4 knockdown reduced ferroptosis and lipid ROS, accompanied with mitochondrial membrane potential elevation. Intriguingly, L-cit treatment significantly inhibited the NF-κB pathway, which might depend on restraining ferritinophagy-mediated ferroptosis. Overall, this study indicated that L-cit might target ferritinophagy-mediated ferroptosis to exert antioxidant and anti-inflammation capacities, which could be a therapeutic strategy against iron overload-induced thymus oxidative damage and immune dysfunction.Keywords:L-citrulline;thymus;iron overload;ferritinophagy;ferroptosis

L-Citrulline Ameliorates Iron Metabolism and Mitochondrial Quality Control via Activating AMPK Pathway in Intestine and Improves Microbiota in Mice with Iron Overload

Dai Zhao,Yuan Gao,Yiqin Chen,Yingsi Zhang,Yian Deng,Sai Niu,Hanchuan Dai

Scope Oxidative stress caused by iron overload tends to result in intestinal mucosal barrier dysfunction and intestinal microbiota imbalance. As a neutral and nonprotein amino acid, L-Citrulline (L-cit) has been implicated in antioxidant and mitochondrial amelioration properties. This study investigates whether L-cit can alleviate iron overload-induced intestinal injury and explores the underlying mechanisms. Methods and results C57BL/6J mice are intraperitoneally injected with iron dextran, then gavaged with different dose of L-cit for 2 weeks. L-cit treatment significantly alleviates intestine pathological injury, oxidative stress, ATP level, and mitochondrial respiratory chain complex activities, accompanied by ameliorating mitochondrial quality control. L-cit-mediated protection is associated with the upregulation of Glutathione Peroxidase 4 (GPX4) expression, inhibition Nuclear Receptor Coactivator 4 (NCOA4)-mediated ferritinophagy and ferroptosis, and improvement of gut microbiota. To investigate the underlying molecular mechanisms, Intestinal Porcine Epithelial Cell line-J2 (IPEC-J2) cells are treated with L-cit or AMP-activated Protein Kinase (AMPK) inhibitor. AMPK signaling has been activated by L-cit. Notably, Compound C abolishes L-cit's protection on intestinal barrier, mitochondrial function, and antioxidative capacity in IPEC-J2 cells. Conclusion L-cit may restrain ferritinophagy and ferroptosis to regulate iron metabolism, and induce AMPK pathway activation, which contributes to exert antioxidation, ameliorate iron metabolism and mitochondrial quality control, and improve intestinal microbiota. L-cit is a promising therapeutic strategy for iron overload-induced intestinal injury.

Improving the flame retardancy and accelerating the degradation of poly (lactic acid) in soil by introducing fully bio-based additives

Shuang Qiu,Yuchun Li,Peng Qi,Dan Meng,Jun Sun,Hongfei Li,Zheng Cui,Xiaoyu Gu,Sheng Zhang

In this study, a novel bio-based flame retardant LC-PA is prepared by the Mannich reaction between phytic acid (PA) and L-citrulline (LC). LC-PA is combined with tannic acid (TA) and introduced into PLA to improve fire performance and accelerate biodegradability . Compared with control PLA, the PLA composite containing 10% LC-PA/TA increases the LOI value to 26.9%, reaches a V -0 rating in the UL-94 test, and reduces the peak heat release rate and total heat release by 24.5% and 21.1%, respectively. More importantly, the introduction of LC-PA/TA accelerates the degradation rate of PLA in soil, which is of significance for biodegradable materials. The addition of LC-PA/TA can attract water and provide a suitable energy source for microbial proliferation, accelerating the hydrolysis and microbial degradation of PLA. This work provides a practical approach for high flame retardancy and rapid biodegradability in the soil to the bio-based polymer.

Benzylic rearrangement for urinary analysis of guanidino and ureido compounds in cardiac surgery–associated acute kidney injury using high-performance liquid chromatography–tandem mass spectrometry

Yunpeng Bai,Yuming Zou,Yingjia Zeng,Linhui Hu,Sumei Huang,Kunyong Wu,Qingxia Yi,Jingchun Chen,Guowu Liang,Yingbang Li,Wendong Huang,Chunbo Chen

Rationale Because acute kidney injury (AKI) is closely related to poor prognosis in critically ill patients, developing biomarkers for its prediction and early diagnosis is particularly important. Endogenous guanidino compounds (GCs) and ureido compounds (UCs) can participate in various biochemical processes because of their important physiological activities. The aim of this study was to investigate the alteration profiles of urinary GCs/UCs as potential biomarkers in patients with cardiac surgery–associated acute kidney injury (CSA-AKI) at different stages. Methods GCs/UCs were reacted with benzil via benzylic rearrangement, and their derivatives were used to investigate fragmentation mechanisms using tandem mass spectrometry (MS/MS) in positive ion mode. Furthermore, a high-performance liquid chromatography (HPLC)–MS/MS method was developed to measure the concentrations of GCs/UCs in urine samples taken from patients with CSA-AKI at different time points. Results MS/MS analysis in positive ion mode showed that benzylic GCs/UCs exhibited similar fragmentation processes, which could produce the characteristic ion (C 13 H 12 N + ) at m/z 182.0. Furthermore, an obviously different fragmentation pattern of benzylic UCs in the positive ion mode might be due to the neutral loss of the H 2 CO 2 group under low collision energy. Of the eight selected GCs/UCs, methylguanidine exhibited significantly increased concentrations in urine when CSA-AKI occurred, whereas guanidinoethyl sulfonate (GDS), homoarginine (HArg) and homocitrulline (HCit) exhibited decreased concentrations. After recovery from AKI, the urinary concentrations of the aforementioned GCs/UCs returned to normal. Some of the aforementioned metabolites with significant changes (GDS, HArg and HCit) had large areas under the curve in the receiver operating characteristic curve for distinguishing AKI stages on the third day after surgery. Conclusions In patients with CSA-AKI, urinary GCs/UCs were significantly disrupted due to injured kidney, and some GC/UC metabolites exhibited a good ability to become potential biomarkers for AKI stages. The present study provides essential resources and new therapeutic targets for further research on CSA-AKI.

Comprehensive Profiling of Amino Acids and Derivatives in Biological Samples: A Robust UHPLC-MS/MS Method for Investigating Acute Lung Injury

Yamin Liu,Xunjiang Wang,Jiaojiao Wei,Kangning Fu,Yilin Chen,Linnan Li,Zhengtao Wang,Li Yang

The severe respiratory dysfunctions associated with acute lung injury (ALI) and its sequelae have a high morbidity and mortality rate, are multifactorial, and lack a viable treatment. Considering the critical function that amino acids and derivatives play in the genesis of illnesses and the regulation of metabolic processes, monitoring the levels of metabolites associated with amino acids in biological matrices is necessary and interesting to study their pathological mechanisms. Exploring the dynamics of amino acid and derivatives levels and searching for biomarkers provides improved clinical ideas for the diagnosis and treatment of ALI. Therefore, we developed an ultra-high-performance liquid chromatography-electrospray tandem mass spectrometry (UHPLC-MS/MS) method that can simultaneously determine the amino acid and derivatives metabolic levels to study amino acid profiles in different biological samples to facilitate clinical research of ALI. In this study, 48 amino acids and derivatives, including neurotransmitters, polyamines, purines, and other types, were quantified simultaneously in a fast, high-throughput, sensitive, and reliable manner within a 15-minute run time without derivatization. No relevant studies have been reported to quantify these 48 amino acid metabolites in three biological samples simultaneously. Satisfactory linearity (R > 0.995), inter-day and intra-day accuracy (85.17–112.67% and 85.29–111.60%, respectively), inter-day and intra-day precision (RSD < 13.80% and RSD < 12.01%, respectively), matrix effects (81.00%–118.00%), recovery (85.09%–114.65%) and stability (RSD < 14.72%) were all demonstrated by the optimized method's successful validation for all analytes. In addition, the suggested method was effectively implemented in plasma, urine, and lung tissue from normal mice and mice with ALI, with the aim of finding potential biomarkers associated with ALI. Potential biomarkers were screened through multivariate statistical analysis and volcanic map analysis, and the changes of markers in ALI were again identified through heat map analysis and correlation analysis with biochemical indicators, which provided ideas and references for subsequent mechanism studies. Here, the technique created in this work offers a quick and dependable way to perform an integrated analysis of amino acids in a variety of biological materials, which can provide research ideas for understanding the physiopathological state of various diseases.

Dynamic profiling of intra- and extra-cellular L/D-amino acids metabolism in colorectal cell and intestinal epithelial cell

Wenchan Deng,Rongrong Huang,Yuanjiang Pan,Cuirong Sun

The metabolism process of amino acids is closely related to the growth of normal and cancer cells. It is still not clear how L/D-configuration amino acids participate in the metabolism of colorectal cell. Herein, intra- and extra-cellular metabolic distribution of L/D-amino acids in colorectal cell (HCT116) and human normal intestinal epithelial cell (NCM460) were profiled utilizing HPLC-MS/MS coupled with a chiral probe. The results displayed the differential metabolic portrayal for the two cell lines. Compared with NCM460 cell, 13 kinds of significant differential amino acids were founded in a lower concentration within HCT116 cell, and L-Gln was even not detected for intra-cell; as for extra-cell culture medium, the HCT116 cell consumed more L-Gln, D-Phe and D-Leu, while L-Met was low ingested in HCT116 cell. L-Ala and Gly were excretion in both two cell lines, excepted L-Cit which was uptake in HCT116 and excretion in NCM460 cell. Furthermore, the dynamic changes of chiral amino acids displayed that phenylalanine, tyrosine and tryptophan biosynthesis and arginine biosynthesis is the major pathway for intra-cellular metabolites and extra-cellular metabolites, respectively. Moreover, with additional D-amino acids in culture medium, the results exhibited that high concentration of D-amino acids have no significant effect on the proliferation of NCM460 cell, but could influence the profiling of amino acids metabolites, and further affect the proliferation of HCT116 cell. This present work enhances the understanding of these differential amino acids metabolic network and depicts a dynamic process of metabolic dysregulation of HCT116 and NCM460 cell.
