L-theanine, a non-protein amino acid naturally occurring in tea leaves, is recognized for its antioxidant, anti-inflammatory, and neuroprotective properties. Despite its known benefits, the mechanisms by which L-theanine influences lifespan extension remain poorly understood. This study investigated the effects of L-theanine on the lifespan ofCaenorhabditis elegansand explored the underlying mechanisms. Our findings indicate that L-theanine significantly diminishes the accumulation of advanced glycation end products (AGEs), which are biomarkers closely linked to aging and age-related diseases. Through an AGE-level analysis, we observed that L-theanine, when administered during early adulthood, notably extended the lifespan ofCaenorhabditis elegansunder both normal and high-glucose-induced stress conditions. L-theanine enhanced the lifespan under typical conditions and provided protective effects against high-glucose-induced stress. A further analysis demonstrated that L-theanine extends the lifespan ofCaenorhabditis elegansby modulating the DAF-2/DAF-16 insulin-like signaling pathway and reducing the accumulation of advanced glycation end products (AGEs). In summary, this study identified L-theanine as a potential anti-aging intervention that extends the lifespan by reducing AGE accumulation and regulating insulin-like signaling pathways. These findings provide new insights for developing anti-aging strategies and lay the groundwork for further research on the potential benefits of L-theanine in mammals. Future studies could explore the molecular mechanisms, test L-theanine in mammalian models, and assess the long-term side effects.

With the increasing number of patients with type II diabetes associated with starchy foods, the inhibition of starch digestion has become one of the most important strategies for limiting postprandial glucose elevation. However, indirect inhibition of starch digestibility through the starch digestive enzymes pathway remains poorly studied. Based on this, we found that L-theanine (THE) had the potential to inhibit starch digestive enzyme activity and investigated the mechanism based on multi-spectral and computer simulations. The results showed that the half-inhibitory concentration (IC 50 ) values of THE on α-amylase and α-glucosidase were 1.22 mmol/L and 2.77 mmol/L, respectively, which exhibited a mixed type of inhibition. Fluorescence quenching results and analyses showed that the mechanism of inhibition of THE with digestive enzymes had both static quenching, mainly based on non-covalent interactions such as van der Waals forces and hydrogen bonding, with binding energies of −3.19 and −7.76 kcal/mol, respectively (Molecular docking results). The multi-spectral analysis showed that the binding of THE changed the secondary structure of starch digestive enzymes and the conformation of polypeptide chains, thus inhibiting the activity. The findings of this study are expected to guide the development of food products with hypoglycaemic function using THE.

Diacylglycerol (DAG) is commonly used as fat substitute in food manufacture due to its functional properties, but DAG has poor emulsification and oxidation stability, which limits its wide application in food industry. In this work, fluorescence quenching data and thermodynamic parameters were analyzed to investigate the interaction mechanism between l -theanine (L-Th) and β-lactoglobulin (β-LG). DAG emulsion was prepared by using β-lactoglobulin-theanine (β-LG-Th) as surface stabilizer, and its emulsification and oxidation stability were evaluated. The results showed that the hydrophobic interaction played an important role on the conjugate of β-LG and L-Th due to the negative values for ΔG, positive values for ΔH and ΔS at pH 4.0, pH 6.0 and pH 8.0. The DAG has been better embedded by using β-LG-Th as surface stabilizer, and the droplet size was about 0.2 µm to 1.5 µm when the pH was 6.0, the ratio of L-Th to β-LG was 1:1. β-LG-Th as surface stabilizer for DAG can increase the ζ-potential and emulsion index, make the emulsion droplet size distribution more uniform. The l -theanine was better to be used to improve the emulsification stability and antioxidant capacity of DAG by binding β-LG as surface stabilizer.

ATP, an important cofactor, is involved in many biocatalytic reactions that require energy. Polyphosphate kinases (PPK) can provide energy for ATP-consuming reactions due to their cheap and readily available substrate polyphosphate. We determined the catalytic properties of PPK from different sources and found that PPK from Cytophaga hutchinsonii ( Ch PPK) had the best catalytic activity for the substrates ADP and polyP 6 . An extracellular–intracellular dual system was constructed to high-throughput screen for better catalytic activity of Ch PPK mutants. Finally, the specific activity of Ch PPK D82N-K103E mutant was increased by 4.3 times. Therefore, we focused on the production of L-theanine catalyzed by GMAS as a model of ATP regeneration. Supplying 150 mM ATP, GMAS enzyme could produce 16.8 ± 1.3 g/L L-theanine from 100 mM glutamate. When 5 mM ATP and 5 U/mL Ch PPK D82N-K103E were added, the yield of L-theanine was 16.6 ± 0.79 g/L with the conversion rate of 95.6 ± 4.5% at 4 h. Subsequently, this system was scaled up to 200 mM and 400 mM glutamate, resulting in the yields of L-theanine for 32.3 ± 1.6 g/L and 62.7 ± 1.1 g/L, with the conversion rate of 92.8 ± 4.6% and 90.1 ± 1.6%, respectively. In addition, we also constructed an efficient ATP regeneration system from glutamate to glutamine, and 13.8 ± 0.2 g/L glutamine was obtained with the conversion rate of 94.4 ± 1.4% in 4 h after adding 6 U/ mL GS enzyme and 5 U/ mL Ch PPK D82N-K103E , which further laid the foundation from glutamine to L-theanine catalyzed by GGT enzyme. This proved that giving the reaction an efficient ATP supply driven by the mutant enzyme enhanced the conversion rate of substrate to product and maximized the substrate value. This is a positively combination of high yield, high conversion rate and high economic value of enzyme catalysis. The mutant enzyme will further power the ATP-consuming biocatalytic reaction platform sustainably.

The study aimed to prepare the chelate of L-theanine (LT) with zinc (Zn) and evaluate its stability, antioxidant and antidiabetic activity. The chelation mechanism between Zn 2+ and LT was investigated by ultraviolet-visible, Fourier transform infrared, scanning electron microscopy/energy dispersive spectrometer and 1 H nuclear magnetic resonance spectroscopy. L-theanine-Zn chelate (LT-Zn) was obtained using LT to Zn citrate in a mass ratio of 3:1 at pH 6 and 70 °C for 180 min with the chelation efficiency of 61.86 ± 1.26%. The results revealed the amino nitrogen atoms and carboxyl oxygen atoms in LT chelated with Zn ions, and the mass of the chelate was confirmed by liquid chromatography with tandem mass spectrometry. Scanning electron microscopy with the element analyzed by energy dispersive spectroscopy showed that the morphology of LT-Zn consisted of irregular granular particles with rough surface. The LT-Zn exhibited higher solubility and improved stability under 100 °C, various pH condition and different concentrations of glucose and NaCl ( p < 0.05) than that of sulfate Zn and Zn gluconate. Compared to sulfate Zn and Zn gluconate, the LT-Zn showed better stability during simulated gastrointestinal digestion. The chelate also exhibited antioxidant activity, as well as an inhibitory effect on α-glucosidase and α-amylase. Overall, the study provided a novel approach to prepare Zn nutraceuticals with antioxidant and antidiabetic properties.

The ultrasound microbubbles modified with the nanomaterial GO not only allow real-time monitoring of liver blood flow for disease diagnosis but also serve as drug delivery carriers, offering the dual benefits of diagnostic imaging and targeted therapy. This presents a highly promising strategy for the integrated diagnosis and treatment of hepatocellular carcinoma (HCC) patients. In this study, L-theanine (TH), a natural polyphenol with antioxidant, and antitumor, was used as a “green” reducing agent for graphene oxide (GO) to prepare nanoscale reduced graphene oxide (rGO-TH), which was then blended with SonoVue microbubbles to prepare composite microbubbles (SV@rGO-TH MBs) for the integrated diagnosis and treatment of HCC. The SV@rGO-TH MBs remain stable at higher Mechanical Index (MI), thereby enhancing contrast-enhanced ultrasound(CEUS) imaging effects and improving the accuracy of disease diagnosis. SV@rGO-TH MBs show significant antitumor activity, inhibiting HepG2 cancer cell growth in vivo and in vitro. Furthermore, combined with ultrasound irradiation, nanoscale rGO-TH more easily penetrates the vascular walls and accumulates in tumor tissues, enhancing the antitumor effect. Our research demonstrates that SV@rGO-TH MBs are a safe and effective ultrasound contrast agent (UCA) for the integrated diagnosis and treatment of HCC, providing insights for the early diagnosis and precise treatment of tumors.

Anji Baicha ( Camellia sinensis ), one of the top 10 teas in China, possesses old leaves rich in bioactive ingredients yet is frequently discarded due to inferior taste. This study evaluated the effects of fermentation with Eurotium cristatum , a probiotic known to enhance bioactivity, on the primary nutrients and on the hypoglycemic and sedative properties of old leaves of Anji Baicha. Results indicated a considerable reduction in free amino acids and tea polyphenols post-fermentation. Conversely, the concentrations of flavonoids and theabrownins increased significantly, showing a positive correlation with the in vitro hypoglycemic effect. The contents of L-theanine and γ-aminobutyric acid (γ-GABA), known for their sedative effects, increased after fermentation, while caffeine and glutamate, renowned for their neural stimulation effect, decreased. Molecular docking revealed that L-theanine and γ-GABA may act as a sedative and sleep aid by competing for receptors with caffeine and glutamate, specifically NMDAR2A. This study not only introduces a novel approach for utilizing old leaves of Anji Baicha but also establishes a foundation for further investigating the hypoglycemic, sedative, and sleep-aiding effects of tea fermented with Eurotium cristatum . Practical Application This research offers a sustainable solution for repurposing the underutilized old leaves of Anji Baicha, enhancing their market value and contributing to the circular economy within the tea industry. The fermented tea could be integrated into health-conscious food and beverage products, targeting consumers seeking natural hypoglycemic and sleep-aiding alternatives. Additionally, the findings open avenues for the development of functional teas that cater to specific health needs, such as diabetes management and sleep improvement.

The digestibility of starch-based foods is receiving increased attention. To date, the full understanding of how including L-theanine (THE) can modify the structural and digestive properties of starch has not been fully achieved. Here, we investigated the multi-scale structure and digestibility of maize starch (MS) regulated by THE in ultrasound field and the molecular interactions . Ultrasound disrupted the structure of starch granules and opened the molecular chains of starch, promoting increased THE binding and producing more low-order or disordered crystal structures. In this case, the aggregation of starch molecules, especially amylose , was reduced, leading to increased mobility of the systems. As a result, the apparent viscosity, G', and G" were significantly decreased, which retarded the starch regeneration. Density functional theory calculations indicated that there were mainly non-covalent interactions between THE and MS, such as hydrogen bonding and van der Waals forces. These interactions were the main factors contributing to the decrease in the short-range ordering, the helical structure, and the enthalpy change (ΔH) of MS. Interestingly, the rapidly digestible starch (RDS) content of THE modified MS (MS-THE-30) decreased by 17.89 %, while the resistant starch increased to 26.65 %. These results provide new strategies for the safe production of resistant starch.

Tea (Camellia sinensis) is the most widely consumed beverage aside from water. The flavor of tea is conferred by certain metabolites, especially l-theanine, in C. sinensis. To determine why more l-theanine accumulates in C. sinensis than in other plants, we compare l-theanine contents between C. sinensis and other plant species (Camellia nitidissima, Camellia japonica, Zea mays, Arabidopsis thaliana, and Solanum lycopersicum) and use a stable isotope labeling approach to elucidate its biosynthetic route. We quantify relevant intermediates and metabolites by mass spectrometry. l-Glutamic acid, a precursor of l-theanine, is present in most plants, while ethylamine, another precursor of l-theanine, specifically accumulates in Camellia species, especially C. sinensis. Most plants contain the enzyme/gene catalyzing the conversion of ethylamine and l-glutamic acid to l-theanine. After supplementation with [2H5]ethylamine, all the plants produce [2H5]l-theanine, which suggests that ethylamine availability is the reason for the difference in l-theanine accumulation between C. sinensis and other plants.

In order to reduce the negative effects caused by oxidative stress on broilers, it is particularly important to find ways to alleviate oxidative stress. As a natural plant extract, L-theanine has a variety of biological effects, such as improving antioxidant capacity, promoting growth, and enhancing immunity and antitumor. This trial evaluated the effects of dietary supplementation of L-theanine on growth performance, antioxidation, meat quality, and intestinal microflora in 817 White Feather Broilers. A total of 108 21-day-old 817 broilers with similar body weight (BW) were randomly divided into three groups with six replicates per group and six chickens within each replicate. The three groups were corn-soybean-based diet (NC group); basal diet plus drinking water with 30 mg hydrocortisone/kg (PC group); and basal diet supplemented with 400 mg L-theanine/kg plus drinking water with 30 mg hydrocortisone/kg (LT group). Compared with the NC group, from 21 to 24 days of age, the PC and LT groups had decreased BW, average daily gain (ADG), and average daily feed intake (ADFI), and increased feed to gain ratio (F/G; p < 0.05). At 24 days of age, the LT group had improved superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in serum as compared to the NC group (p < 0.05). The LT group broilers also had significantly higher concentrations of malondialdehyde (MDA) in serum and liver (p < 0.05). On the 42nd days, the PC group had lower PH45min (p < 0.05) than the NC and LT groups and higher cooking loss and shear force (p < 0.05). Moreover, the villi height of the PC group was significantly lower in jejunum than the NC group (p < 0.05). The LT group had a higher ZO-1 content in duodenum than the NC and PC groups (p < 0.05). The activity of GSH-Px in the liver of the LT group was increased than in the PC group (p < 0.05). The relative abundance of Firmicutes in the LT group was significantly higher than in the NC and PC groups (p < 0.05). These results suggested that the effects of acute oxidative stress on growth performance and meat quality of broilers are continuous, and dietary supplementation of L-theanine could improve the growth performance and meat quality, enhance the intestinal mucosal barrier and antioxidant capacity, and improve the composition of the intestinal flora of broilers caused by acute oxidative stress.

BACKGROUND The quality of tea is influenced by numerous factors, especially l- theanine, which is one of the important markers used to evaluate the sweetness and freshness of tea. Sensitive, rapid, and accurate detection of l -theanine is therefore useful to identify the grade and quality of tea. RESULTS A high-sensitivity, paper-based fluorescent sensor combined with chemometrics was established to detect l- theanine in tea water based on CdTe quantum dots / corn carbon dots and nano tetra pyridel-porphine zinc (ZnTPyP). To verify the reliability of this method, fluorescence spectra and fluorescence-visualized paper-based sensors were compared. The fluorescence spectrum method demonstrated a linear range of 1 to 10 000 nmol L −1 and a limit of detection (LOD) of 0.19 nmol L −1 . In the fluorescence-visualized paper-based sensors there was a linear range of 10–1000 nmol L −1 , and the LOD was 10 nmol L −1 . Partial least squares discriminant analysis (PLSDA) and partial least squares regression analysis (PLSR) were used successfully to determine l -theanine accurately in tea water with this approach. The accuracy of the PLSDA model was 100% both in the training set and the predicting set, and the correlation coefficient between the actual concentration and the predicted concentration was greater than 0.9997 in the PLSR model. CONCLUSION This fluorescence-visualized paper-based sensor, combined with chemometrics, could be applied efficiently to the practical analysis of tea water samples, which provides a new idea to ensure the flavor and quality of tea. © 2020 Society of Chemical Industry

An ultrasensitive surface-enhanced Raman spectroscopy (SERS) aptamer sensor (aptasensor) using a noble metal nanoparticle-magnetic nanospheres composite was developed for L-theanine detection. It makes use of Fe 3 O 4 @Au MNPs and Au@Ag NPs embedded with the Raman reporter 4-mercaptobenzoic acid (4MBA). Au@4MBA@Ag NPs modified by aptamer and Fe 3 O 4 @Au MNPs modified by cDNA created the aptasensor with the strongest Raman signal of 4MBA through the specific binding of the aptamer. With the preferred binding of L-theanine aptamer to L-theanine, Au@4MBA@Ag NPs were released from Fe 3 O 4 @Au MNPs, causing a linear decrease in SERS intensity to achieve the SERS detection of the L-theanine. The SERS peak of 4MBA at 1078 cm −1 was used for quantitative determination. SERS intensity showed a good log-linear relationship within the range 10 −10 to 10 −6 M of L-theanine. The aptasensor has a high selectivity for L-theanine compared with other twelve tested analytes. Hence, this aptasensor is a promising analytical tool for L-theanine detection. The developed method was applied to the analysis of real samples, demonstrating excellent performance. The comparison with the standard liquid chromatography mass spectrometry method showed an error within 20%. Graphical abstract
