Recently, ultrasensitive detection and multi-guest sensing have received extensive attention due to their high sensitivity and efficiency. Herein, we report a novel approach to achieve ultrasensitive detection of multi-analyte. This approach is concluded as “rationally introduce Aggregation-Induced Emission (AIE) into chemosensor”. According to this approach, by rationally introducing self-assembly moiety, the obtained chemosensor DNS could serve as a novel AIEgen and show strong AIE in DMSO/H 2 O (water fraction 80%) binary solution. Interestingly, a simple fluorescent sensor array based on the DNS has been developed. This sensor array could selectively sense Fe 3+ , Al 3+ , H 2 PO 4 − and L-Arg in water solution. More importantly, this sensor array shows ultrasensitive detection for Fe 3+ , Al 3+ and L-Arg. The LODs of the sensor array for Fe 3+ , Al 3+ and L-Arg are in the range of 3.54 × 10 −9 M to 9.42 × 10 −9 M. Moreover, H 2 PO 4 − could realize the reversible detection of Fe 3+ in the DMSO/H 2 O (water fraction 80%) solution. Meanwhile, DNS -based test papers and thin films were prepared, which could serve as test kits for convenient detection Fe 3+ , Al 3+ , and L-Arg in water. In addition, they could also act as efficient erasable fluorescent display materials.

The application of chiral ionic liquids (ILs) in capillary electrophoresis (CE) for enantioseparation has received considerable attention in the past few years; however, the use of chiral ILs as ligands in ligand-exchange capillary electrophoresis (LE-CE) has been reported only in a few papers. In this work, several tetraalkylammonium amino acid ILs (TAA-AAILs) including tetramethylammonium- l -arginine (TMA- l -Arg), tetramethylammonium- l -proline (TMA- l -Pro) and tetramethylammonium- l -glutamic acid (TMA- l -Glu) were first applied in LE-CE to act as chiral ligands for enantioseparation. The results show that TMA- l -Arg-is an excellent chiral ligand, and able to yield equal or even better resolutions (Rs) in much shorter migration times for most tested analytes compared with previously reported AAILs ligands or free l -Arg-ligand. Parameters of the new TAA-AAILs-based LE-CE system such as the alkane chain length of TAA-AAILs, ILs configuration, ILs concentration, the type and concentration of central metal ion, buffer pH and applied voltage were optimized. Satisfactory enantioseparations were achieved for most selected free- or dansyl-amino acids. The method was then successfully applied to the enantiomeric impurity test of a commercial amino acid sample. This work indicates that the application of TAA-AAILs as chiral ligands in LE-CE is of great potential for enantioseparation.

Critical limb ischemia (CLI) remains refractory to current revascularization strategies due to its complex pathology. Here, we report an injectable alginate hydrogel that orchestrates a dual-pathway regenerative cascade by simultaneously reprogramming the immune microenvironment and activating direct tissue repair. The system is rationally designed by integrating two functional modules: (i) immunomodulatory carbon dots (PNS-AA@CQDs-COS) that selectively target M1 macrophages and drive them to the M2 phenotype, and (ii) intrinsically bioactive cross-linkers (Zn2+/l-Arg) that serve as sustained-release pro-regenerative signals. Critically, this dual-component architecture enables a synergistic interplay─while the carbon dots establish an anti-inflammatory microenvironment (path 1), the liberated Zn2+ and l-Arg directly activate endothelial cells and muscle satellite cells, thereby initiating coordinated vascular and myogenic regeneration (path 2). In vitro, the hydrogel significantly amplified reparative crosstalk within macrophage-endothelial/myogenic cells’ cocultures. In a murine CLI model, a single injection of the hydrogel resulted in durable recovery of hindlimb perfusion (80.4% by day 14), improved motor function, and enhanced tissue repair. Mechanistic studies revealed that these benefits stem from microenvironment remodeling (M2 polarization and IL-10 upregulation) and modulation of key signaling pathways─suppressing NF-κB while potentiating PI3K/Akt/eNOS. This work presents a material-based strategy that couples immunomodulation with direct cellular activation, offering a synergistic therapeutic platform for CLI and potentially other ischemic diseases.

The development of alternative strategies for the efficient treatment of subcutaneous abscesses that do not require the massive use of antibiotics and surgical intervention is urgently needed. Herein, a novel synergistic antibacterial strategy based on photodynamic (PDT) and NO gas therapy is reported, in which, a PDT-driven NO controllable generation system (Ce6@Arg-ADP) is developed with l -Arg-rich amphiphilic dendritic peptide (Arg-ADP) as a carrier. This carrier not only displays superior bacterial association and biofilm penetration performance, but also acts as a versatile NO donor. Following efficient penetration into the interior of the biofilms, Ce6@Arg-ADP can rapidly produce massive NO via utilizing the H 2 O 2 generated during PDT to oxidize Arg-ADP to NO and l -citrulline, without affecting singlet oxygen ( 1 O 2 ) production. The combination of 1 O 2 and the reactive by-products of NO offers notable synergistic antibacterial and biofilm eradication effects. Importantly, following efficient elimination of all bacteria from the abscess site, Arg-ADP can further generate trace quantities of NO to facilitate the angiogenesis and epithelialization of the wound tissues, thereby notably promotes wound healing. Together, this study clearly suggests that Arg-ADP is a versatile NO donor, and the combination of PDT and NO represents a promising strategy for the efficient treatment of subcutaneous abscesses.

Multidrug-resistant (MDR) bacterial infections remain a critical public health concern worldwide, with conventional antibacterial strategies presenting limitations such as inconsistent target localization, suboptimal therapeutic efficiency, and progressive antimicrobial resistance development. Nitric oxide (NO), a bioactive molecule, exhibits potent antibacterial effects, resulting in the emergence of NO-based gas therapy as a promising and innovative therapeutic strategy. However, these therapies present the principal challenge of effective NO delivery, owing to its short half-life and limited diffusion capacity. Hence, this study aimed to develop AI‑molybdenum disulfide (L-Arg/IR780@MoS 2 ) nanoflowers (NFs) employing L-arginine (L-Arg) as an endogenous NO precursor and MoS 2 as a dual carrier for L-Arg and IR780 iodide (IR780). Following near-infrared irradiation during photodynamic therapy, generated reactive oxygen species initiated an oxidation cascade for L-Arg-to-NO conversion. The produced NO, in combination with photothermal therapy, modulated the bacterial microenvironment. This significantly altered the expression of key genes in MDR bacteria associated with nucleotide metabolism and biofilm formation, thereby effectively disrupting biofilms and suppressing bacterial growth. Furthermore, the AI-MoS 2 NFs exhibited notable ancillary therapeutic benefits, including anti-inflammatory activity, enhanced re-epithelialization, increased collagen deposition, and angiogenesis modulation, which collectively accelerated wound healing. Altogether, the findings of this study present a synergistic therapeutic strategy designed to address the dual challenges posed by bacterial biofilms and antimicrobial resistance.

Muscle metabolism produces creatinine, a waste product whose levels in the blood and urine are crucial markers of kidney health. Herein, a smartphone-based electrochemical detection strategy was developed to quantify creatinine in human blood serum. Since creatinine was electrochemically inactive, a standard copper solution was added as an electro-activator to produce an electrochemically active creatinine–copper complex. At a pH of 7.4, the creatinine–copper composite was oxidized in a phosphate buffer solution (PBS). Electrochemical oxidation of the free Cu+ ion in PBS is tested by the surface modification of Ti2C2Tx@poly(L-Arg) nanocomposite. The analytical performance of the developed electrochemical sensor was evaluated by differential pulse voltammetry. The developed electrochemical sensor was evaluated using a combination of techniques: electrochemical methods like cyclic voltammetry and electrochemical impedance spectroscopy, morphological analysis with scanning electron microscopy, and structural analysis with attenuated total reflectance Fourier transform infrared spectroscopy and X-ray diffraction. Notably, the developed sensor demonstrated an impressively low detection limit of 0.05 μM and a linear range of 1–200 μM. Moreover, the sensor remarkably exhibited a stable creatinine detection response with an acceptable reproducibility for two two-week periods and demonstrated a robust immunity against interfering molecules. This is the first report on the synthesis of Ti2C2Tx@poly(L-Arg) nanocomposites and their application in the electrochemical detection of creatinine. This smartphone-based creatinine sensor offers a promising, rapid, and reliable technique for creatinine detection, with potential applications in clinical diagnostics and biomedical research, due to its high sensitivity, selectivity, and portability.

Depleting intracellular glutathione (GSH) has emerged as a potent strategy to combat cancer. However, the existing GSH-depleting agents are too toxic or ineffective and standalone GSH depletion fails to yield a satisfactory curative effect. Herein, we present an intelligent nanoparticle that possesses GSH depletion, glucose consumption accompanied with H2O2 production, and NO generation properties for multimodal cancer therapy. The nanoparticle is constructed by synthesis of tetrasulfide bond-doped mesoporous silica nanoparticles followed by conjugating glucose oxidase (GOx) on the surface and loading l-arginine (l-Arg) into the mesopores. In this nanoparticle, the doped tetrasulfide bonds can quickly deplete GSH, which increases the cellular reactive oxygen species concentration to induce ferroptosis and meanwhile triggers particle biodegradation to expose the loaded l-Arg. Moreover, the elevated H2O2 level activates l-Arg to release NO for NO therapy. GOx consumes glucose to initiate starvation therapy and simultaneously produces a large amount of H2O2. Importantly, the produced H2O2 can not only potentiate ferroptosis but also promote NO release to enhance NO therapy. Besides, NO could in turn improve the efficacy of starvation therapy by damaging the mitochondria to block energy supply. In vitro and in vivo studies demonstrate that the nanoparticles show a great synergistic effect of ferroptosis/starvation/NO therapy, which can significantly kill cancer cells and remarkably inhibit tumor growth without obvious side effects. Therefore, we think that the designed nanoparticles may provide a promising paradigm for synergistic cancer therapy and hold a prospect in clinical trials.

Dual pathological barriers, including capillarized liver sinusoidal endothelial cells (LSECs) and deposited extracellular matrix (ECM), result in insufficient drug delivery, significantly compromising the anti-fibrosis efficacy. Additionally, excessive reactive oxygen species (ROS) in the hepatic microenvironment are crucial factors contributing to the progression of liver fibrosis. Hence, hyaluronic acid (HA) modified liposomes co-delivering all-trans retinoic acid (RA) and L-arginine (L-arg) were constructed to reverse hepatic fibrosis. By exhibiting exceptional responsiveness to the fibrotic microenvironment, our cleverly constructed liposomes efficiently disrupted the hepatic sinus pathological barrier, leading to enhanced accumulation of liposomes in activated hepatic stellate cells (HSCs) and subsequent induction of HSCs quiescence. Specially, excessive ROS in liver fibrosis promotes the conversion of loaded L-arg to nitric oxide (NO). The ensuing NO serves to reestablish the fenestrae structure of capillarized LSECs, thereby augmenting the likelihood of liposomes reaching the hepatic sinus space. Furthermore, subsequent oxidation of NO by ROS into peroxynitrite activates pro-matrix metalloproteinases into matrix metalloproteinases, which further disrupts the deposited ECM barrier. Consequently, this NO-induced cascade process greatly amplifies the accumulation of liposomes within activated HSCs. More importantly, the released RA could induce quiescence of activated HSCs by significantly downregulating the expression of myosin light chain-2, thereby effectively mitigating excessive collagen synthesis and ultimately leading to the reversal of liver fibrosis. Overall, this integrated systemic strategy has taken a significant step forward in advancing the treatment of liver fibrosis.

Introduction Sonodynamic therapy (SDT) has potential clinical applications for cancer therapy, and is yet restricted by complex tumor microenvironmental (TME) factors. Thus, the research problem of TME modulation as well as efficient tumor treatment still needs to be clarified. Method In this study, a calcium carbonate (CaCO 3 ) nanoplatform was designed for ultrasound (US) and TME response-triggered, which encapsulated Ag 2 S and loaded with l -Arg, and further wrapped with RBC/Platelet membrane, named as QD@Ca/M L-Arg . Results Non-invasive US-triggered SDT by Ag 2 S and acidic environment-responsive drug release were achieved. In vitro experiments validated the efficacy of SDT, Ca-ion interference and nitric oxide (NO) gas therapy as combined therapy for cancer treatment. By means of RNA sequencing, the cancer therapeutic mechanism of SDT in redox-related pathways was elucidated. The immunosuppressive TME was simulated with a M2-macrophage/cancer cell co-culture system to confirm the immune activating effect of immunogenic cell death (ICD). Conclusion Accordingly, the potential of QD@Ca/M L- Arg- was demonstrated for in vitro TME modulation, cancer therapeutic efficacy and clinical translation.
