Deproteinization is a crucial step for the purification of polysaccharides from natural biomass. However, traditional deproteinization technologies often suffer from complicated operating processes and the usage of toxic chemical reagents. Herein, an advanced and universal deproteinization method based on dual-functional adsorbent consisted of preferential protein adsorption coating of tannic acid-Fe III (TA-Fe III ) and magnetic Fe 3 O 4 (Fe 3 O 4 @TA-Fe III , abbreviated as FTF NPs) was developed. The proposed FTF NPs showed remarkable efficiency to remove protein from Lycium barbarum L. polysaccharides (LBPs) with deproteinization ratio of 96%, higher than the typical Sevag method (85%). Detailed adsorption kinetics studies demonstrated deproteinization process reached equilibrium after 10 min, faster than other reported deproteinization adsorbents. Furthermore, FTF NPs are structurally and functionally nondestructive as regards LBPs without using organic reagents. Also, it exhibited widespread-use deproteinization performance to several common plant species. Therefore, the proposed nano-separation based on TA-Fe III complex is an advanced and universal tool to high-efficiently deproteinize plant polysaccharides.

Yes-associated protein 1 ( YAP1 ), a core effector of the Hippo signaling pathway, has garnered increasing attention for its role in ferroptosis signaling in breast cancer. This study investigates the antitumor properties of Lycium barbarum polysaccharides (LBPs) and develops LBPs-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles (LBPs NPs), further exploring the mechanisms of free LBPs and LBPs NPs in normal and malignant breast cells. Results demonstrated that YAP1 overexpression drives malignant transformation in normal mammary cells, including aberrant proliferation, apoptosis resistance, cytoskeletal remodeling, and enhanced invasiveness, alongside mitochondrial hyperfunction and altered energy metabolism. However, LBPs effectively suppressed these malignant phenotypes and counteracted the associated mitochondrial and metabolic enhancements by inhibiting YAP1 upregulation. In breast cancer cells, YAP1 overexpression similarly promoted proliferation, invasion, cytoskeletal reorganization, and apoptosis resistance, accompanied by enhanced mitochondrial dysfunction and energy, and acquired ferroptosis resistance. Both LBPs and LBPs NPs significantly inhibited cell proliferation, invasion, and cytoskeletal reorganization, disrupted energy metabolism, and ultimately induced ferroptosis via downregulation of YAP1 expression. Notably, both free LBPs and LBPs NPs effectively suppressed YAP1 expression, with LBPs NPs exhibiting significantly superior efficacy compared to free LBPs. This enhancement is attributed to improved drug delivery efficiency facilitated by the PLGA-based nanocarrier system. Mechanistically, LBPs NPs target the Hippo-YAP1 signaling axis, leading to altered phosphorylation status of YAP1 (p-YAP1), and synergistically induce changes in energy metabolism and ferroptosis to inhibit breast cancer metastasis. These findings provide a proof-of-concept at the in vitro level and a potential nanoparticle-based formulation platform for the treatment of breast cancer.

This study was to assess the digestion and colonic fermentation of two bioactive polysaccharides, EPS-LM and LBPS, and the subsequent influences on human gut microbiota through simulated gastrointestinal systems. EPS-LM, an exopolysaccharide isolated from mycelial culture of a medicinal fungus Cordyceps sinensis Cs-HK1, was characterized as a heteropolysaccharide consisting of Man(108):Gal(52.7):Glc(29.2) (molar ratio) with an average molecular weight (MW) 5.513 × 10 6 . LBPS was isolated from a well-known medicinal plant ( Lycium barbarum L.) which was also characterized as a heteropolysaccharide (1.236 × 10 5 MW). Both polysaccharides were highly resistant to saliva, gastric and small-intestine digestion with negligible MW reduction and release of reducing sugars but were quickly degraded to lower MW during in vitro human fecal fermentation. They were consumed as a carbon source by the gut bacteria to produce short-chain fatty acids (SCFAs). In comparison, the carbohydrate content of EPS-LM was more completely consumed than LBPS and there were also notable differences in consumption of specific monosaccharides and production of specific SCFAs, propionic and butyric acid, and relative abundance of gut bacterial populations between EPS-LM and LBPS group. The results suggest that metabolic outcomes and modulating effects of EPS-LM and LBPS on the gut microbiota are highly dependent on their molecular composition.
