Rapid self-healing nanocomposite gel crosslinked by LDH for lost circulation control

Cheng Wang,Jinsheng Sun,Yifu Long,Li Peng,Yingying Li,Ren Wang,Yuanzhi Qu,Xumin Yang

Lost circulation presents a complex challenge in drilling operations. Ascribed to the favorable remaining in fracture, strong resistance to efflux, and entire gel block rather than individual particles, the self-healing gels have been broadly applied as lost circulation material (LCM). In this study, we utilized layered bimetallic hydroxide (LDH) as a dynamic crosslinking agent to synthesize a novel nanocomposite (NC) gel through in-situ polymerization following the stripping of LDH in acrylic acid (AA) solution. LDH/PAA NC gels exhibit excellent self-healing properties, rheological strength, mechanical properties and adhesion properties, with these attributes being influenced by the LDH content. At 110 °C, the optimal LDH/PAA NC gel rapidly self-healed in bentonite mud within 1 h, revealing storage modulus (G′) of 4041 Pa, tensile strength of 8.04 kPa, and adhesion strength of 2.1 kPa. Visual plugging tests revealed that the LDH/PAA NC gel could withstand pressures up to 4 MPa, surpassing the capabilities of MA gel, which lacked self-healing properties. These findings highlight the potential of LDH/PAA NC gels as efficient LCM, offering a promising solution for controlling lost circulation during drilling operations.

Nano-magnetoelectric biomimetic system promotes neural differentiation and reverses dopaminergic neuron damage for Parkinson's disease therapy

Jie Zhang,Jiahui Shi,Qiubo Yang,Haogang Guo,Lingfeng Guo,Guoliang Zhou,Yinyi Liu,Xiaolei Wang,Xian Hao,Yifei Yang

Parkinson's disease (PD), a neurodegenerative disorder characterized by the depletion of dopaminergic neurons, has conventionally been managed through pharmacological intervention and deep brain stimulation (DBS) surgery. However, drug resistance and the risks of surgical implantation have hindered the effective of current treatment. Herein, we endeavor to engineer a wireless, non-invasive nanoplatform for electrostimulation-enhanced drug therapy, termed BCFO@LCM. This innovative platform integrates magnetoelectric nanomaterials (BaTiO 3 -CoFe 2 O 4 ) encapsulated within BV2 cell membranes (CM), enabling traversal of the blood-brain barrier (BBB) and dual-targeting of the PD microenvironment through magnetic and membrane fusion mechanisms. When exposed to an external magnetic field, this system facilitates the controlled release of the therapeutic agent L-DOPA for pharmacotherapy, concurrently generating electrical signals via the magnetoelectric nanomaterials to effectuate neuromodulation. Our findings reveal that BCFO@LCM markedly ameliorates dyskinesia in PD model mice, diminishes the attrition of dopaminergic neurons in the substantia nigra and striatum, augments tyrosine hydroxylase levels, and ultimately reverses the damage to dopaminergic neurons. Furthermore, the non-invasive electrical stimulation enhances neuronal activity within deep brain tissues and fosters the regeneration and differentiation of neurons.

Multifunctional Artificial Peroxisome Basing on Lactate Oxidase as a Self-Cascade Enhancing Active Oxygen Amplifier for Tumor Therapy

Huihui Wang,Xueping Huang,Ran Gao,Ke Li,Dandan Li,Zhuobin Xu,Zemin Ling,Chun Pan,Lizeng Gao,Hao Chen

The intricacy, diversity, and heterogeneity of cancers make research focus on developing multimodal synergistic therapy strategies. Herein, an oxygen (O2) self-feeding peroxisomal lactate oxidase (LOX)-based LOX–Ce6–Mn (LCM) was synthesized using a biomineralization approach, which was used for cascade chemodynamic therapy (CDT)/photodynamic therapy (PDT) combination therapies through dual depletion of lactate (Lac) and reactive oxygen species (ROS) generation. After endocytosis into tumor cells, the endogenous hydrogen peroxide (H2O2) can be converted to O2 by the catalase-like (CAT) activity of LCM, which can facilitate the catalytic reaction of LOX to consume more Lac and alleviate tumor hypoxia to enhance the generation of singlet oxygen (1O2) upon light irradiation. In addition, the H2O2 produced by LOX catalysis and oxidase-like (OXD) activity of LCM can be catalyzed into highly toxic hydroxyl radicals (•OH) via the Fenton-like reaction, enhancing oxidative damage to tumor cells. Both in vitro and in vivo experiments confirmed that LCM significantly promoted ROS accumulation and effectively inhibited tumor growth by inducing tumor cell autophagy under the combined effect of Lac depletion and CDT with PDT. Therefore, integrally designed LCM for reprogramming metabolism and the tumor microenvironment offers a promising multimodal strategy for tumor treatments.
