An Online-SPE/SEC/LCMS Method for the Detection of N-Nitrosamine Disinfection Byproducts in Wastewater Plant Tailwater

Huili Tang,Ziru Li,Hanqing Chen,Yang Xu,Xu Jiang,Erdeng Du,Zhen Lyu,Lu Zheng,Mingguo Peng

N-nitrosamines have recently attracted attention as a class of disinfection byproducts and are also a hot spot in environmental studies. Current N-nitrosamine analytical methods typically involve manual solid phase extraction (SPE) of samples followed by quantitative analysis using liquid chromatography-mass spectrometry (LCMS), which is time-consuming and may also fail to eliminate complex matrix effects. Size exclusion chromatography (SEC) is a technique that can separate compounds according to their molecular size. For the first time, this study developed an Online-SPE/SEC/LCMS quantitative analysis method to detect and analyze nine common N-nitrosamine disinfection byproducts in wastewater plant tailwater, including N-dimethylnitrosamine (NDMA) and N-nitrosodiethylamine (NDEA), etc. The samples of 1.0 mL can be directly injected after the simple 0.22 μm membrane filtration. This method reports the combination of SPE, SEC, and RP C18 columns to achieve several functions in a processing time of 20 min, including online enrichment, desalination, and matrix separation for the first time. The method provides good linearity (R2> 0.999), recoveries ranging from 91.67% to 105.88%, relative standard deviation (RSD) lower than 4.17%, and the limits of detection (LOD) are 0.12–6.60 ng/L. This method alleviates tedious human labor and can effectively overcome the matrix effect (ME< 20%). This method allows for the accurate quantitative analysis of N-nitrosamines with high compatibility in wastewater plant tailwater, rivers, and lakes with a high background matrix. Interested researchers can also use this method as a reference in the online analysis of other specific pollutants after necessary optimization. It can also be utilized for non-targeted screening and targeted analysis of contaminants in water with a wide range of applications, giving valuable information for environmental monitoring.Keywords:N-nitrosamines;analysis method;online solid-phase extraction (Online-SPE);size exclusion chromatography (SEC);LCMS;column switching;wastewater plant tailwater

Partitioning, renal clearance, and internal exposure assessment of liquid crystal monomers (LCMs): Based on paired serum and urine of general population in Northwest China

Kaiming Yang,Yiwei Gong,Guanlan Wu,Jiawen Zhang,Weici Quan,Yuxi Wu,Yadi Ai,Di Huang,Kai Yang,Hongguang Cheng,Qingrui Zhang,How Yong Ng

Liquid crystal monomers (LCMs), which are the key material of liquid crystal displays, have been recently considered as potential persistent, bioaccumulative, and toxic (PBT) chemicals. However, there is limited information on the accumulation and metabolism of LCMs in humans. In this study, a total of 162 paired serum and urine samples of a general population (aged 19 - 70 years) residing in Northwest China were collected and analyzed for the concentrations of 15 representative LCMs. The mean ∑15LCMs concentrations were 129.13 and 38.52   ng/mL in the serum and urine, respectively. BEEB was found to be the most P&B LCM and dominated both serum and urine. DFPrB accumulated remarkably in the serum, while PBIPHCN and BEEB were more easily eliminated by the kidneys. The strong correlations of some LCMs in the paired serum and urine suggest that urinary LCM concentrations can reflect their metabolic levels in the body. The estimated daily intake of ∑15LCMs for the study participants based on the serum data was 3.41   ng/kg body weight/day, and the hazard index of ∑15LCMs was 5.23×10 -5 which is acceptable (< 1). This study provides the evidence for the presence of LCMs in the serum and urine of general population and highlights the renal clearance of LCMs in the human body.

Liquid Crystal Monomers (LCMs) in Sediments: Method Validation and Detection in Sediment Samples from Three Typical Areas

Huijun Su,Shaobo Shi,Ming Zhu,Jianhua Li,Guanyong Su

Liquid crystal monomers (LCMs) have been proposed to be persistent, bioaccumulative, and toxic (PBT) substances; however, there is a dearth of information regarding their occurrence in sediment samples. Here, an analytical method was developed for the quantitative determination of LCMs in sediment samples, and n = 76 sediment samples were collected and analyzed to determine accurate concentrations of LCMs. Our results indicated that the developed pretreatment procedure was applicable for the determination of LCM concentrations in sediments. We observed that LCMs were detected in 75 out of the 76 sediment samples, and 23, 18, and 14 out of the 39 target LCMs were quantified in at least one of the analyzed sediments from rivers around LCM or liquid crystal device (LCD) manufacturers, Taihu Lake, and rivers around e-waste recycling sites, respectively. The LCMs in the samples from rivers around LCM/LCD manufacturers exhibited the greatest mean concentrations of 26.1 ng/g dry weight (dw), followed by those from e-waste recycling site areas (1.15 ng/g dw) and Taihu Lake (0.076 ng/g dw). Collectively, this study provided the first analytical method that was able to quantify the concentrations of LCMs in sediment samples and provided the first evidence for the occurrence of LCMs in sediment samples.

An LCMS/MS method for the simultaneous determination of ten antimicrobials and its application in critically ill patients

Tingting Liu,Xiaomin Chen,Guanxuanzi Zhang,Jing Zhao,Qian Lu,Fang Wang,Hongxia Li,Bing Liu,Ping Zhu

Significant pharmacokinetic variation occurs in critically ill patients, leading to underexposure to antibiotics and poor prognosis. In this study, we developed a simple, sensitive, and fast liquid chromatography tandem mass spectrometry (LC MS/MS) platform for the simultaneous quantification of 8 antibacterial and 2 antifungal drugs, which is optimally suited for clinically efficient, real-time therapeutic drug monitoring (TDM). Multiple reaction monitoring (MRM) mass spectrometry was used in this method, and samples were prepared via protein precipitation with methanol. Chromatographic separation was accomplished on a BGIU Column-U02 (2.1x50 mm, 3 µm), with six stable isotopes and one analog as an internal standard. The overall turnaround time of the assay was 5 minutes. All the drugs tested (piperacillin, cefoperazone, meropenem, levofloxacin, moxifloxacin, daptomycin, linezolid, vancomycin, fluconazole and voriconazole) were linear in the test concentration range (r ≥ 0.9900), the accuracy was 95 %-111 %, the precision variation coefficient was greater than or equal to 10 %, the lower limit of quantitation was 0.31–7.51 mg/L, and the coefficient of variation of the matrix factor was less than 10 %. The recovery rates ranged from 85 % to 115 %, and the antibiotics were stable at 4°C and −20°C for 6 days, with an offset of greater than or equal to 15 %. This method was successfully applied to routine TDM in 252 elderly critically ill patients.

Skin toxicity of liquid crystal monomers (LCMs): Mitochondrial dysfunction and metabolic dysregulation revealed by integrated multi-omics analysis

Chenxin Dongye,Shanshan Wang,Xiangrong Chen,Changjie Li,Yanfang Zhao,T.-W. Dominic Chan,Rabah Boukherroub,Xiangfeng Chen

Liquid crystal monomers (LCMs), as core components of liquid crystal displays (LCDs), are emerging as environmental materials due to their widespread use and potential for human and ecological exposure. Even as inquiries pertaining to the environmental and health risks of LCMs are progressing, their direct toxic effects on human organs and ecosystems persist in being inadequately comprehended. The present research underscores the hazards entailed by prolonged LCMs exposure, with specific reference to skin cells and animal models, under daily exposure magnitudes. This study reveals that long-term LCMs exposure disrupts mitochondrial function in skin cells, triggers inflammatory pathways (TNF signaling), and downregulates critical proteins (PLOD2, DDIT4) and metabolites (ATP, glutathione), indicating oxidative stress and cellular dysfunction. In vivo experiments further demonstrate histopathological damage in mouse skin, including disordered skin appendages and adipose disorganization, highlighting LCMs' hazardous potential. Multi-omics analysis links LCMs exposure to diseases such as lung cancer and Alzheimer’s, while untargeted metabolomics identifies β -alanylleucine downregulation as a promising biomarker for LCM-induced toxicity. Given LCD e-waste growth, improper disposal releases LCMs, risking ecosystem bioaccumulation. These findings underscore the need for stricter regulation of LCMs throughout their lifecycle-from production to waste management-to mitigate ecological and health risks, with β -alanylleucine serving as a potential monitoring tool for environmental contamination.

Experimental determination of octanol-water partition coefficient (KOW) of 39 liquid crystal monomers (LCMs) by use of the shake-flask method

Ming Zhu,Huijun Su,Yurong Bao,Jianhua Li,Guanyong Su

Liquid crystal monomers (LCMs) were recently proposed as persistent, bioaccumulative, and toxic substances; however, there is a dearth of information regarding their experimental octanol-water partition coefficients ( K OW ). In the present study, we determined the experimental K OW values of these 39 LCMs by use of a classic shake-flask method. We observed that experimental K OW values of LCMs largely varied depending on their specific structures, and the Log transformed K OW generally fall in the range of 4.94–7.62. The experimental K OW values were further compared with those predicted by Estimation Programs Interface (EPI) Suite software. Interestingly, we observed that experimental and estimated Log K OW values were generally comparable for LCMs containing two benzene or cyclohexane rings; however, the estimated values gradually deviated from the experimental ones as the number of benzene or cyclohexane rings of LCM structures increased. Based on the experimental Log K OW values of 39 LCMs, we established a quantitative structure activity relationship (QSAR) model for predicting Log K OW values of other LCMs, for which authentic standards are not available. Adjusted square of determination coefficient (R 2 ) of the developed model is 0.810, indicating its goodness-of-fit for estimation of Log K OW values of other substances with similar backbone structures. Overall, our present study provides the first insight on experimental Log K OW values of LCMs, and suggests that LCMs are inclined to accumulative in the fatty tissue of organisms in the aqueous environments.
