A signal-enhanced LFIA based on tyramine (TYR)-induced AuNPs aggregation has been developed for the sensitive detection of danofloxacin (DAN). In the model, the hydroxyl radical produced by HRP catalyzing H 2 O 2 can trigger the TYR-AuNPs to aggregate on the T or C line for enhancing the detection signal. The linear range of TYR-AuNPs LFIA was 0.25–5 ng mL −1 with the limit of detection (LOD) of 0.032 ng mL −1 , and the LOD was 8-fold lower than that of the traditional AuNPs LFIA (0.26 ng mL −1 ). The TYR-AuNPs LFIA could be used with the naked eyes to qualitatively detect DAN with a cut-off limit of 2.5 ng mL −1 , which was 4-fold lower than that of the traditional AuNPs LFIA (10 ng mL −1 ). The recoveries of TYR-AuNPs LFIA were 86.04–105.14% and 92.41–110.19%, with the coefficient of variation of 1.71–2.05% and 4.42–5.89% in chicken and pork, respectively.

Typical symptoms of acute myocardial infarction (AMI) and acute aortic dissection (AAD) are similar chest pain, leading to higher clinical misdiagnosis rate, evenly the mortality rate of patients. In this work, ZrO 2 @PEI-QDs nanospheres with higher fluorescence were prepared by sequential electrostatic adsorption of branched PEI chains and CdSe/ZnS-COOH QDs on hollow ZrO 2 nanospheres. Three multi-channel LFIA strips (on sST2 and cTnI, cTnI and h-FABP, HMGB1 and sST2, respectively) based on ZrO 2 @PEI-QDs for rapid and correct distinguishing of AMI and ADD were constructed. Their visual limit of detection (vLOD for sST2 and cTnI, cTnI and h-FABP, HMGB1 and sST2) within 10–15 min reached 0.075 and 0.1 ng/mL, 0.1 and 1 ng/mL, 1 and 0.1 ng/mL, respectively, which were all lower than the normal level of healthy people. Compared with previous studies, ZrO 2 surface adsorbs more QDs, the fluorescence intensity was increased by about 2.84 times, and vLODs of three strips based on the quantitative fluorescence analysis were also increased by up to one orders of magnitude. The test results of five clinical serum samples indicated that three multi-channel LFIA strips could quantitatively detect four biomarkers of chest pain, and successfully distinguish AMI patients from AAD patients. These results indicated that this set of multi-channel LFIA strips could become an effective tool for the early and accurate detection or distinguish of ADD and AMI.

Tropomyosin (TM), a major shrimp allergen, poses a significant challenge to food safety. This study aimed to develop a biosynthetic recombinant tropomyosin (rTM) as a standardized alternative to the natural tropomyosin (nTM) and to establish a sensitive detection method. Structural analyses using ultraviolet, fluorescence, and circular dichroism spectroscopies confirmed high similarity between rTM and nTM. Comparable allergenicity was further validated in a murine model. A novel lateral flow immunoassay (LFIA) employing Prussian blue nanoparticles (PBNPs) as probes was subsequently developed. This assay exhibited a wide linear range of 0.1–50 μg/mL and a low detection limit of 0.051 μg/mL, and excellent specificity, accuracy, and precision in spiked food matrices. This work establishes a foundation for developing nTM alternatives and provides a reliable, portable platform for sensitively detecting macromolecular food allergens.

Since December 2019, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has caused millions of deaths and seriously threatened the safety of human life; indeed, this situation is worsening and many people are infected with the new coronavirus every day. Therefore, it is very important to understand patients’ degree of infection and infection history through antibody testing. Such information is useful also for the government and hospitals to formulate reasonable prevention policies and treatment plans. In this paper, we develop a lateral flow immunoassay (LFIA) method based on superparamagnetic nanoparticles (SMNPs) and a giant magnetoresistance (GMR) sensing system for the simultaneously quantitative detection of anti -SARS-CoV-2 immunoglobulin M (IgM) and G (IgG). A simple and time-effective co-precipitation method was utilized to prepare the SMNPs, which have good dispersibility and magnetic property, with an average diameter of 68 nm. The Internet of Medical Things-supported GMR could transmit medical data to a smartphone through the Bluetooth protocol, making patient information available for medical staff. The proposed GMR system, based on SMNP-supported LFIA, has an outstanding advantage in cost-effectiveness and time-efficiency, and is easy to operate. We believe that the suggested GMR based LFIA system will be very useful for medical staff to analyze and to preserve as a record of infection in COVID-19 patients.

Lateral flow immunoassay (LFIA) has been widely used for the early diagnosis of diseases. However, conventional colorimetric LFIA possesses limited sensitivity, and the single-mode readout signal is easily affected by the external environment, leading to insufficient accuracy. Herein, multifunctional Fe3O4@MoS2@Pt nanotags with a unique “pompon mum”-like structure were triumphantly prepared, exhibiting excellent peroxidase (POD)-like activity, photothermal properties, and magnetic separation capability. Furthermore, the Fe3O4@MoS2@Pt nanotags were used to establish dual-mode LFIA (dLFIA) for the first time, enabling the catalytic colorimetric and photothermal dual-mode detection of severe acute respiratory syndrome coronavirus 2 nucleocapsid protein (SARS-CoV-2 NP) and influenza A (H1N1). The calculated limits of detection (cLODs) of SARS-CoV-2 NP and H1N1 were 80 and 20 ng/mL in catalytic colorimetric mode and 10 and 8 ng/mL in photothermal mode, respectively, demonstrating about 100 times more sensitive than the commercial colloidal Au-LFIA strips (1 ng/mL for SARS-CoV-2 NP; 1 μg/mL for H1N1). The recovery rates of dLFIA in simulated nose swab samples were 95.2–103.8% with a coefficient of variance of 2.3–10.1%. These results indicated that the proposed dLFIA platform showed great potential for the rapid diagnosis of respiratory viruses.

The coronavirus disease 2019 (COVID-19) pandemic highlighted the need for rapid and accurate viral detection at the point-of-care testing (POCT). Compared with nucleic acid detection, lateral flow immunoassay (LFIA) is a rapid and flexible method for POCT detection. However, the sensitivity of LFIA limits its use for early identification of patients with COVID-19. Here, an innovative surface-enhanced Raman scattering (SERS)-LFIA platform based on two-dimensional black phosphorus decorated with Ag nanoparticles as important antigen-capturing and Raman-signal-amplification unit was developed for detection of SARS-CoV-2 variants within 5–20 min. The novel SERS-LFIA platform realized a limit of detection of 0.5 pg/mL and 100 copies/mL for N protein and SARS-CoV-2, demonstrating 1000 times more sensitivity than the commercial LFIA strips. It could reliably detect seven different SARS-CoV-2 variants with cycle threshold (Ct) < 38, with sensitivity and specificity of 97 and 100%, respectively, exhibiting the same sensitivity with q-PCR. Furthermore, the detection results for 48 SARS-CoV-2-positive nasopharyngeal swabs (Ct = 19.8–38.95) and 96 negative nasopharyngeal swabs proved the reliability of the strips in clinical application. The method also had good specificity in double-blind experiments involving several other coronaviruses, respiratory viruses, and respiratory medications. The results showed that the innovative SERS-LFIA platform is expected to be the next-generation antigen detection technology. The inexpensive amplification-free assay combines the advantages of rapid low-cost POCT and highly sensitive nucleic acid detection, and it is suitable for rapid detection of SARS-CoV-2 variants and other pathogens. Thus, it could replace existing antigens and nucleic acids to some extent.

With the increasing misuse of antibiotic drugs, the convenient and ultrasensitive detection of chloramphenicol (CAP) is essential for ensuring food safety and human health. In this study, a comprehensive platform was developed integrating detection and discrimination capabilities by combining surface-enhanced Raman scattering (SERS) with lateral flow immunoassay (LFIA) technology, along with a support vector machine (SVM) model for data analysis, to detect CAP residues in aquatic products. The CAP-specific antibody-modified gold nanostars (Au NSs) were synthesized as SERS–LFIA nanoprobes, achieving an ultralow limit of detection (LOD) of 10 pg/mL and a theoretical LOD of 1.6 pg/mL for simulated aquatic samples. This sensitivity surpasses that of the currently available commercial strips and demonstrates high specificity. Additionally, the automated discrimination model exhibited high sensitivity (95.83%) and specificity (96.43%) as well as an accuracy of 96% on the test set, marking an 8% improvement over traditional Raman intensity discrimination methods. This developed platform is a simple, fast, and automated technique that eliminates the need for sample pretreatment, offering a promising approach for the trace detection of CAP in aquatic products.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder requiring early diagnosis for effective intervention. The combination of blood Aβ 42 /Aβ 40 ratio and p-tau-181 is recognized as a promising core biomarker panel for AD diagnosis. However, conventional detection methods suffer from limitations including time-consuming procedures, lack of multiplexing capability, and insufficient sensitivity. Herein, we developed a magnetic surface-enhanced Raman scattering (SERS)-encoded tags-based lateral flow immunoassay (LFIA) for simultaneous detection of Aβ 42 , Aβ 40 , and p-tau-181 on a single test line. Magnetic Fe 3 O 4 @Au nanoparticles were synthesized and modified with three distinct Raman reporters (DTNB for Aβ 42 , 4-MBA for Aβ 40 , and 2-MPY for p-tau-181) and corresponding detection antibodies. Under optimal conditions, the assay achieved rapid detection (∼28 min) and high sensitivity, with limits of detection of 93.33 fg/mL for Aβ 42 , 218.78 fg/mL for Aβ 40 , and 56.23 fg/mL for p-tau-181. The method exhibited excellent specificity against interfering substances (BSA, human IgG) and closely related species (Aβ 38 , p-tau-217), as well as good reproducibility (RSD <5%) and stability (≥4 weeks). In clinical plasma samples (n = 40), the combined score (p-tau-181/(Aβ 42 /Aβ 40 )) progressively increased with disease severity and correlated well with MMSE scores and MRI MTA grades, capturing both cognitive decline and hippocampal atrophy. This magnetic SERS-encoded LFIA platform offers a rapid, sensitive, user-friendly, and multiplex-capable approach for early AD diagnosis and progression monitoring, showing promise for future point-of-care applications.

Highly infectious and pathogenic viruses seriously threaten global public health, underscoring the need for rapid and accurate diagnostic methods to effectively manage and control outbreaks. In this study, we developed a comprehensive Surface-Enhanced Raman Scattering–Lateral Flow Immunoassay (SERS-LFIA) detection system that integrates SERS scanning imaging with artificial intelligence (AI)-based result discrimination. This system was based on an ultra-sensitive SERS-LFIA strip with SiO2-Au NSs as the immunoprobe (with a theoretical limit of detection (LOD) of 1.8 pg/mL). On this basis, a negative–positive discrimination method combining SERS scanning imaging with a deep learning model (ResNet-18) was developed to analyze probe distribution patterns near the T line. The proposed machine learning method significantly reduced the interference of abnormal signals and achieved reliable detection at concentrations as low as 2.5 pg/mL, which was close to the theoretical Raman LOD. The accuracy of the proposed ResNet-18 image recognition model was 100% for the training set and 94.52% for the testing set, respectively. In summary, the proposed SERS-LFIA detection system that integrates detection, scanning, imaging, and AI automated result determination can achieve the simplification of detection process, elimination of the need for specialized personnel, reduction in test time, and improvement of diagnostic reliability, which exhibits great clinical potential and offers a robust technical foundation for detecting other highly pathogenic viruses, providing a versatile and highly sensitive detection method adaptable for future pandemic prevention.

Commercial gold nanoparticle-based lateral flow immunoassays (LFIAs) are widely used for point-of-care (POCT) diagnostics; however, inefficient signal transduction and the absence of intelligent interpretation result in low sensitivity and poor quantification. Here, we present an on-device platform (Multimatrix QuantAI-LFIA) that integrates deep learning for the intelligent quantitative detection of staphylococcal enterotoxin B (SEB) across multiple biological matrices. By employing Fe-polydopamine (Fe-PDA) as a charge-transfer-active label for signal transduction, this system displayed superior colorimetric performance owing to efficient ligand–metal charge transfer (LMCT) transitions of Fe(III), achieving an initial visual limit of detection (LOD) of 5 ng/mL within 15 min. Further, the deep learning model enabled robust quantitative analysis under varying illumination conditions, extending the dynamic range to 0.1–800 ng/mL and reducing the LOD to 0.1 ng/mL, which was 50-fold lower than the visual readout. Through transfer learning, the model demonstrated reliable adaptation to serum and urine samples, yielding strong correlations between predicted and actual SEB concentrations (R2 > 0.90). In blind tests, the Multimatrix QuantAI-LFIA achieved an area under the curve (AUC) of 0.924 via a self-developed smartphone application, substantially outperforming naked-eye interpretation (0.700). This on-device platform provides rapid, accurate, and matrix-adaptive quantification, offering a practical pathway toward next-generation POCTs.

Herein, a novel core-shell material Fe 3 O 4 @MIL-100(Fe) was synthesized for developing a novel LFIA by a two-step solvothermal method with good magnetic properties and high specific surface area. It was used as both magnetic separation and label agents in LFIA for the detection of Listeria monocytogenes . The enrichment of target bacteria can be achieved quickly and easily by direct adsorption of Fe 3 O 4 @MIL-100(Fe) on the bacteria and by the action of an applied magnetic field. It was beneficial in separation and concentration of target bacteria from food samples. The Fe 3 O 4 @MIL-100(Fe)-bacterial was then detected by the anti- Listeria monocytogenes antibody on the T line under the capillary action of the LFIA strip, resulting in the formation of a visible orange band. Under the optimal conditions, there was a good linear relationship between the intensity of the T line and the quantity of Listeria monocytogenes in the range of 10 5 -10 8 CFU mL − 1 , with a visual detection limit of 3.3 × 10 6 CFU mL − 1 . The strip showed strong specificity for L. monocytogenes , and the detection could be finished in 45 min, without the needing of pre-treatment. Thus, the proposed Fe 3 O 4 @MIL-100(Fe)-based strip was simple prepared, label-free, low cost, rapid and convenience.

Eu-complex-swollen polystyrene (PS) fluorescent microspheres have attracted considerable attention in the field of lateral flow immunoassay (LFIA) bio-detection. However, the excessive swelling and induced concentration quenching effect of Eu complexes often deteriorate their fluorescence intensity and fluorescence efficiency. To address this issue, herein, we synthesized Eu(TTA)3(TPPO)2 and introduced divinylbenzene (DVB) to fabricate three-dimensionally cross-linked PS-DVB-Eu microspheres in order to obtain a moderate swelling capacity of the Eu complexes without obvious fluorescence concentration quench. This cross-linked structure effectively promotes the homogeneous distribution of swollen Eu(TTA)3(TPPO)2 in PS-DVB-Eu microspheres. This thus resulted in improved fluorescence properties with a high internal quantum efficiency (IQE) of 52.36% and a lifetime of 539 μs. The fluorescence of the cross-linked PS-DVB-Eu microspheres was 3.85 times stronger than that of the non-crosslinked PS-Eu microspheres. The cross-linked structures also helped the PS-DVB-Eu microspheres maintain a relatively high zeta potential and a large surface carboxyl concentration. These merits support PS-DVB-Eu microspheres in obtaining an enhanced lateral flow immunoassay (LFIA) performance, including an LOD of 0.08 ng mL−1 and a CV value of less than 7.34% on detecting C-reactive protein (CRP). This demonstrates that cross-linked PS-DVB-Eu microspheres are promising materials for high performance LFIA point-of-care testing techniques.
