Electrospun PLGA/PCL Nanofiber Film Loaded with LPA Promotes Full-Layer Wound Healing by Regulating the Keratinocyte Pyroptosis

Xinqi Huang,Jianghuiwen Lu,Yumei An,Mingyuan Xu,Xueshi Chen,Chao Liu,Xuefeng Zhou,Haiyan Shan,Yunzhu Qian,Mingyang Zhang

Electrospun nanofibers have a number of qualities that make them a suitable choice for skin wound healing. Lysophosphatidic acid (LPA) stimulates the keratinocytes and fibroblasts to proliferate, differentiate, and migrate and enhances skin wound healing. Here, we developed the electrospun scaffolds contained in polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA). The scaffolds loaded with LPA nanoparticles retained a porous nanofiber structure and showed better physicochemical properties and biocompatibility. The scaffold continuously releases LPA to quickly initiate cell signaling and maintain long-term anti-inflammatory activity. In this study, we found that PP scaffold with LPA reduces the disordered collagen deposition and the thickness of the newborn epidermis, improves skin healing, and reduces scar formation. Explaining the mechanism of LPA mineralized tissue regeneration in skin wound healing, LPA inhibited the pyroptosis of keratinocyte, a cell death process that induces inflammation and scar formation by inhibiting the expression of TNF-α and β-catenin proteins. Thus, the electrospun PP scaffold with LPA can be potentially developed as a therapeutic avenue to target skin wound healing.

Lysophosphatidic Acid Analogue rather than Lysophosphatidic Acid Promoted the Bone Formation In Vivo

Zi-Li Yu,Bin-Fang Jiao,Zu-Bing Li

Lysophosphatidic acid (LPA), a bioactive lipid molecule, has recently emerged as physiological and pathophysiological regulator in skeletal biology. Here we evaluate the effects of LPA on bone formation in vivo in murine femoral critical defect model. Primary femoral osteoblasts were isolated and treated with osteogenic induction conditional media supplemented with 20 μM LPA or LPA analogue. Mineralized nodules were visualized by Alizarin Red S staining. Forty-five C57BL/6 mice underwent unilateral osteotomy. The femoral osteotomy gap was filled with porous scaffolds of degradable chitosan/beta-tricalcium phosphate containing PBS, LPA, or LPA analogue. 2, 5, and 10 weeks after surgery, mice were sacrificed and femurs were harvested and prepared for Micro-Computed Tomography (Micro-CT) and histological analysis. Alizarin Red S staining showed that LPA and LPA analogue significantly enhanced the mineral deposition in osteoblasts. Micro-CT 3D reconstruction images and HE staining revealed that significantly more newly formed bone in osteotomy was treated with LPA analogue when compared to control and LPA group, which was verified by histological analysis and biomechanical characterization testing. In summary, our study demonstrated that although LPA promotes mineralized matrix formation in vitro, the locally administrated LPA was not effective in promoting bone formation in vivo. And bone formation was enhanced by LPA analogue, administrated locally in vivo. LPA analogue was a potent stimulating factor for bone formation in vivo due to its excellent stability.

An abnormal LPA/LPAR1-NHE1 axis leads to the autophagy deficiency of trophoblast cells in recurrent spontaneous abortion

Min Chen,Jia-Lu Shi,Zi-Meng Zheng,Zhi Lin,Ming-Qing Li,Jun Shao

The autophagy of trophoblasts is necessary for developing and maintaining of a healthy pregnancy. Autophagy dysfunction in trophoblast cells is linked to recurrent spontaneous abortion (RSA). However, the mechanism underlying trophoblast autophagy is unknown. In this study, we investigated the expression of autophagy-related genes in both normal and RSA villi. We also examined the production of lysophosphatidic acid (LPA) and lysophosphatidic acid receptor1 (LPAR1) in trophoblast cells during early pregnancy. We found that the activation of the LPA-LPAR1 axis triggered the autophagy of trophoblast cells and increased the expression of sodium/hydrogen exchanger 1 (NHE1). Inhibition of NHE1 suppressed the autophagy in trophoblast cells and we confirmed that NHE1 regulates LPA production in trophoblast cells. Additionally, we found decreased expression of autophagy-related genes and LPAR1 in villi from RSA patients. These observations indicate that the LPA/LPAR1-NHE1 axis regulates the autophagy of trophoblast cells during pregnancy. Insufficient autophagy and poor expression of LPAR1 in trophoblast cells may result in dysfunction of the trophoblasts and increased risk of spontaneous abortion. Overall, our research elucidated that a positive LPA/LPAR1-NHE1 axis can promote the autophagy of trophoblast cells and the abnormal axis leads to the autophagy deficiency of trophoblast cells in recurrent spontaneous abortion.

Programmable Microfluidic-Assisted Highly Conductive Hydrogel Patches for Customizable Soft Electronics

Junchen Liao,Zhiqiang Ma,Shiyuan Liu,Wei Li,Xiaodan Yang,Mohamed Elhousseini Hilal,Xiang Zhou,Zhengbao Yang,Bee Luan Khoo

The utilization of hydrogels in soft electronics has led to significant progress in the field of wearable and implantable devices. However, challenges persist in hydrogel electronics, including the delicate equilibrium between stretchability and electrical conductivity, intricacies in miniaturization, and susceptibility to dehydration. Here, a lignin-polyacrylamide (Ag-LPA) hydrogel composite endowed with anti-freeze, self-adhesive, exceptional water retention properties, and high stretchability (1072%) is presented. Notably, this composite demonstrated impressive electrical conductivity at room temperature (47.924 S cm −1 ) and extremely cold temperatures (42.507 S cm −1 ). It is further proposed for microfluidic-assisted hydrogel patches (MAHPs) to facilitate customizable designs of the Ag-LPA hydrogel composite. This approach enhances water retention and offers versatility in packaging materials, making it a promising choice for enduring soft electronics applications. As a proof-of-concept, soft electronics across diverse applications and dimensions, encompassing healthcare monitoring, environmental temperature sensing, and 3D-spring pressure monitoring electronics are successfully developed. The scenery of an extremely cold environment is further extended. The conductivity of the embedded Ag-LPA hydrogel composite unveils the potential of MAHPs in polar rescue missions. It is envisioned that MAHPs will impact the development of sophisticated and tailored soft electronics, thereby forging new frontiers in engineering applications.
