ZnO and ZnO nanoparticles (ZnO NPs) are widely used in food packaging, food preservation, cosmetic preparation, and animal feed. ZnO is alleged showing multiple bioactivities including antimicrobial and anti-inflammation. It is hypothesized in this study that bulk ZnO and ZnO NPs could attenuate symptoms associated with high-fat-diet–induced obesity. Bulk ZnO and ZnO NPs with diameters of 30 and 90 nm were administered to high-fat-diet (HFD)–induced obese mice. Body weight, liver and fat tissue indices of ZnO-treated mice were decreased compared with those of obese mice (MOD). Blood glucose levels in oral glucose tolerant test and insulin tolerant test of ZnO-treated mice were lower than those of MOD. Serum lipid profile of ZnO-treated mice was ameliorated with lower total cholesterol, total triglyceride, and low-density lipoprotein cholesterol levels compared with that of MOD. In addition, the levels of serum IL-1β and LPS-binding protein were also decreased by ZnO treatment. Both bulk and nanosized ZnO could attenuate HFD-induced phenotypes related with obesity, but ZnO NP is more efficient to lower the fat index and bulk ZnO is better to restore the disturbed serum lipid profile.

Antimicrobial peptides , as an integral part of the innate immune system, kill bacteria through a special mechanism of action, making them less susceptible to drug resistance. However, Lipopolysaccharide (LPS) as the permeation barrier on the bacterial membrane, inhibits the antibacterial activity of antimicrobial peptides and triggers the inflammatory response. GWKRKRFG is an LPS binding sequence with a β-boomerang motif that can be linked to antimicrobial peptides to enhance their LPS affinity and reduce the possibility of LPS-induced inflammatory responses. In this study, a series of hybrid peptides were designed by conjugating the reported LPS binding sequence to the C-/N-terminal sequences of the natural porcine antimicrobial peptide PMAP-23 to increase the LPS affinity of peptides. Among all the designed hybrid peptides, 4R-PP-G8 showed the best antibacterial activity, nonhemolytic activity, and excellent cell selectivity. The presence of LPS not only induced the secondary structure transformation of 4R-PP-G8 from a random structure to an α-helical structure but also reduced the antibacterial activity of 4R-PP-G8 in a dose-dependent manner, indicating the excellent binding ability of 4R-PP-G8 to LPS. The LPS/LTA binding assay further verified the interaction between the peptide and LPS. The membrane permeability test verified that 4R-PP-G8 possessed a strong capability to penetrate the bacterial membrane after interacting with LPS. More direct membrane disruption was observed under FE-SEM and TEM. In conclusion, we provided a simple and efficient method to improve the LPS binding ability of antimicrobial peptides and enhance their antimicrobial activity, resulting in the peptide 4R-PP-G8 with clinical application potential.

Blood infection can release toxic bacterial lipopolysaccharides (LPSs) into bloodstream, trigger a series of inflammatory reactions, and eventually lead to multiple organ dysfunction, irreversible shock, and even death, which seriously threatens human life and health. Herein, a functional block copolymer with excellent hemocompatibility is proposed to enable broad-spectrum clearance of LPSs from whole blood blindly before pathogen identification, facilitating timely rescue from sepsis. A dipeptide ligand of histidine–histidine (HH) was designed as the LPS binding unit, and poly[(trimethylamine N-oxide)-co-(histidine–histidine)], a functional block copolymer combining the LPS ligand of HH and a zwitterionic antifouling unit of trimethylamine N-oxide (TMAO), was then designed by reversible addition–fragmentation chain transfer (RAFT) polymerization. The functional polymer achieved effective clearance of LPSs from solutions and whole blood in a broad-spectrum manner and had good antifouling and anti-interference properties and hemocompatibility. The proposed functional dihistidine polymer provides a novel strategy for achieving broad-spectrum clearance of LPSs, with potential applications in clinical blood purification.
