Developing a High-Umami, Low-Salt Soy Sauce through Accelerated Moromi Fermentation with Corynebacterium and Lactiplantibacillus Strains

Li-Hao Wang,Wen-Hui Qu,Ya-Nan Xu,Song-Gang Xia,Qian-Qian Xue,Xiao-Ming Jiang,Hong-Ying Liu,Chang-Hu Xue,Yun-Qi Wen

The traditional fermentation process of soy sauce employs a hyperhaline model and has a long fermentation period. A hyperhaline model can improve fermentation speed, but easily leads to the contamination of miscellaneous bacteria and fermentation failure. In this study, after the conventional koji and moromi fermentation, the fermentation broth was pasteurized and diluted, and then inoculated with three selected microorganisms includingCorynebacterium glutamicum,Corynebacterium ammoniagenes, andLactiplantibacillus plantarumfor secondary fermentation. During this ten-day fermentation, the pH, free amino acids, organic acids, nucleotide acids, fatty acids, and volatile compounds were analyzed. The fermentation group inoculated withC. glutamicumaccumulated the high content of amino acid nitrogen of 0.92 g/100 mL and glutamic acid of 509.4 mg/100 mL. TheC. ammoniagenesgroup andL. plantarumgroup were rich in nucleotide and organic acid, respectively. The fermentation group inoculated with three microorganisms exhibited the best sensory attributes, showing the potential to develop a suitable fermentation method. The brewing speed of the proposed process in this study was faster than that of the traditional method, and the umami substances could be significantly accumulated in this low-salt fermented model (7%w/vNaCl). This study provides a reference for the low-salt and rapid fermentation of seasoning.

Phytochemical and Flavor Characteristics of Mulberry Juice Fermented with Lactiplantibacillus plantarum BXM2

Xuefang Guan,Dazhou Zhao,Tian Yu,Shaoquan Liu,Shuying Chen,Junyang Huang,Gongti Lai,Bin Lin,Juqing Huang,Chengchun Lai,Qi Wang

Fermentation of mulberry juice not only improves its shelf life, but also effectively enhances their flavor and nutritional quality. This study elucidated the phytochemical and flavor characteristics of mulberry juice fermented withLactiplantibacillus plantarumBXM2, originally isolated from naturally fermented fruit beverage, through widely targeted metabolomics. The fermentation produced the unique flavor of fermented juice and decreased the pH from 4.15 to 3.19. The metabolomic analysis detected 907 non-volatile metabolites, from which 359 significantly different non-volatile metabolites (up 238, down 121) were screened out. Among 731 identified volatile metabolites, 26 flavor substances were the major contributors to the flavor differences between fermented and unfermented mulberry juices. It is hypothesized that lipid metabolism and amino acid catabolism are crucial pathways for the flavor enhancement of mulberry juice fermented withL. plantarumBXM2. Meanwhile, significant increases of the contents of a variety of bioactive substances, such as indole-3-lactic acid, octadeca-9,12,15-trienoic acid, di-/tri-peptides, etc., conferred additional health potential to BXM2-fermented mulberry juice.

Genomic and biogenic amine-reducing characterization of Lactiplantibacillus planatraum JB1 isolated from fermented dry sausage

Shan Qin,Xian Ming Zeng,Mei Jiang,Xin Rui,Wei Li,Ming Sheng Dong,Xiao Hong Chen,Qiu Qin Zhang

Biogenic amines are naturally present as hazardous, low-molecular organic basic compounds in various foods . Lactic acid bacteria show excellent potential to prevent biogenic amines accumulation. In this study, the amine-reducing mechanism of Lactiplantibacillus planatraum JB1, a strain isolated from fermented sausages , was explored according to its genome, and its ability to degrade biogenic amines was also evaluated in culture medium and dry sausage. The results showed that L. plantarum JB1 possessed a myriad of desirable properties, including tolerance to harsh environments and safety for assessment in vitro . Its genome contained genes encoding transport proteins and multicopper oxidase, which may be involved in biogenic amine degradation. In PBS and MRS medium, L. plantarum JB1 could degrade eight biogenic amines with high degradation rates for tyramine and putrescine. By inoculating sausages with L. plantarum JB1, the total content of biogenic amine was decreased by 32.84% after preparation and 32.07% after storage. Due to its good adaptability, security and amine-reducing capacity, L. plantarum JB1 could be considered a starter culture to ensure the safety of meat products.

Comparative analysis of exopolysaccharide-producing Lactiplantibacillus plantarum with ropy and non-ropy phenotypes on the gel properties and protein conformation of fermented milk

Xueliang Zhang,Gisele LaPointe,Yang Liu,Xiaomeng Wang,Luyao Xiao,Xiaogan Zhao,Wei Li

In this study, we evaluated the impact of Lactiplantibacillus plantarum ( L. plantarum ) with ropy and non-ropy phenotypes on gel structure and protein conformation of fermented milk. Ropy L. plantarum (T1 & CL80) secreted EPS with high molecular weight (1.41 × 10 6 , 1.19 × 10 6 Da) and intrinsic viscosity (486.46, 316.32 mL/g), effectively enhances fermented milk viscosity and water holding capacity (WHC) (65.4%, 84.6%) by forming a dense gel structure. Non-ropy L. plantarum (CSK & S-1A) fermented milk gel’s high surface hydrophobicity and free sulfhydryl content caused high hardness and low WHC. Raman spectroscopy combined with circular dichroism analysis showed that high levels of α-helix (29.32–30.31%) and random roil (23.06–25.36%) protein structures are the intrinsic factors that contribute to the difference among fermented milk gels of ropy and non-ropy strains. This study provides a basis for understanding the structural variability of fermented milk gels using ropy or non-ropy lactic acid bacteria.

Wenjiao Xue,Chen Liu,Yao Liu,Hao Ding,Chao An,Shizhe Zhang,Saijian Ma,Qiwen Zhang

To assess the probiotic potential of strain 68-1 with rutin conversion capabilities, isolated from Chinese traditional Jiangshui, a complete genomic analysis and in vitro tests were conducted. The Oxford Nanopore Technologies (ONT, Oxford, UK)–Illumina (San Diego, CA, USA) hybrid sequencing platform was used for whole genome sequencing and the results showed that strain 68-1 had a chromosome sequence of 3,482,151 bp, with 46.53% GC content and five plasmids with a sequence length ranging from 2009 bp to 48,711 bp. Strain 68-1 was identified asLactiplantibacillus pentosusbased on the whole genome sequence. A total of 133 genes encoding for carbohydrate-active enzymes (CAZymes) were identified and genes that may be involved in rutin conversion were found in theL. pentosus68-1 genome.L. pentosus68-1 showed excellent tolerance to gastrointestinal juice and adhesion properties, and corresponding genes were identified. In addition,L. pentosus68-1 exhibited strong antibacterial and antifungal activity, where organic acids may play a crucial role in its antagonistic ability. Moreover, the gene cluster for plantaricin_EF production was detected. No high virulence factor was found in theL. pentosus68-1 genome and no hemolytic effect was observed. In addition,L. pentosus68-1 showed resistance to ampicillin, gentamycin, and kanamycin, and the genomic analysis indicated that horizontal ARG transfer should not be possible. The results show thatL. pentosus68-1 could be developed as a novel probiotic candidate to improve rutin bioavailability in the food and feed industry.

Co-encapsulation of probiotic Lactiplantibacillus plantarum and polyphenol within novel polyvinyl alcohol/fucoidan electrospun nanofibers with improved viability and antioxidation

Jiage Ma,Zhongmei Tan,Mengguo Wu,Zihao Tian,Cong Xu,Jing Zhang,Yue Ma,Zhibiao Feng,Wei Yu,Bailiang Li,Yuchang Yao,Zhanmei Jiang,Juncai Hou

Polyvinyl alcohol (PVA)/fucoidan (FUC) blend nanofibers were systematically fabricated to co-encapsulate probiotic Lactiplantibacillus plantarum 69–2 (LP69–2) and four kinds of polyphenols by electrospinning for the first time. Scanning electron microscopy showed that some areas of PVA/FUC nanofibers encapsulated with LP69–2 were locally broadened. Attenuated total reflectance-Fourier transform infrared spectroscopy and X-ray diffraction suggested that LP69–2 and polyphenol were successfully encapsulated in PVA/FUC electrospun nanofibers. Thermogravimetric analysis revealed that the addition of LP69–2 and polyphenol enhanced the thermal stability of nanofibers. Moreover, the incorporation of FUC and polyphenol significantly increased the ABTS + and DPPH radical scavenging ability of PVA nanofibers ( P  < 0.05). Notably, PVA/FUC/LP69–2/DMY nanofibers displayed the highest DPPH radical scavenging ability. After 21 d, these nanofibers loaded with polyphenols could maintain viability of LP69–2 over 7 lg CFU/g at 4 °C and the viability of LP69–2 in PVA/FUC/DMY nanofibers was the highest. Overall, the co -encapsulation of probiotic and polyphenol within PVA/FUC electrospun nanofibers increased the viability of probiotics and enhanced antioxidant activity of nanofibers. This study provided unique insights for protecting probiotics and developing novel functional foods with higher probiotics.

Effects of the Exopolysaccharide from Lactiplantibacillus plantarum HMX2 on the Growth Performance, Immune Response, and Intestinal Microbiota of Juvenile Turbot, Scophthalmus maximus

Gege Hu,Yihui Wang,Rui Xue,Tongji Liu,Zengjia Zhou,Zhennai Yang

In this study, the exopolysaccharide (EPS) fromLactiplantibacillus plantarum(HMX2) was isolated from Chinese Northeast Sauerkraut. Its effects on juvenile turbot were investigated by adding different concentrations of HMX2-EPS (C: 0 mg/kg, H1: 100 mg/kg, H2: 500 mg/kg) to the feed. Compared with the control group, HMX2-EPS significantly improved the growth performance of juvenile turbot. The activities of antioxidant enzymes, digestive enzymes, and immune-related enzymes were significantly increased. HMX2-EPS could also increase the secretion of inflammatory factors and enhance the immune response of turbot by regulating the IFN signal transduction pathway and exhibit stronger survival rates after theA. hydrophilachallenge. Moreover, HMX2-EPS could improve the diversity of intestinal microbiota in juvenile fish, increase the abundance of potential probiotics, and reduce the abundance of pathogenic bacteria. The function of gut microbes in metabolism and the immune system could also be improved. All results showed better effects with high concentrations of HMX2-EPS. These results indicated that HMX2-EPS supplementation in the diet could promote growth, improve antioxidant activity, digestive capacity, and immunity capacity, and actively regulate the intestinal microbiota of juvenile turbot. In conclusion, this study might provide basic technical and scientific support for the application ofL. plantarumin aquatic feed. Keywords:Lactiplantibacillus plantarum;exopolysaccharide (EPS);immunity;intestinal health;Scophthalmus maximus

A novel strain Lactiplantibacillus plantarum LPP95 isolated from Chinese pickles: Antifungal effect, mechanism, and potential application in yogurt

Qiang Wang,Feng Zhang,Yuhong Zhang,Yu Zhang,Hongwei Wang,Jiajia Song,Huayi Suo

This study investigated the antifungal effect, mechanism, and potential application in yogurt of Lactiplantibacillus plantarum ( L. plantarum ) LPP95 isolated from Chinese pickles. The L. plantarum LPP95 showed a high inhibitory effect on the growth of five molds , with the strongest inhibitory effect observed on Penicillium sp. The cell-free supernatants (CFS) of L. plantarum LPP95 resulted in the shrinking of Penicillium sp. spores , and destroyed the integrity of the cell wall and membrane of spores, causing the leakage of nucleic acid from cells. The antifungal activity of CFS was not decreased after heat or proteolytic enzyme treatment but was affected by pH change. The inhibitory activity of CFS was associated with organic acids, and 66 organic acids were identified by LC-MS analysis. In addition, the validation experiments showed that trans -cinnamic acid, citric acid , trans -3-indoleacrylic acid, and DL-4-hydroxyphenyllactic acid all exhibited antifungal activity. Moreover, L. plantarum LPP95 effectively inhibited the growth of Penicillium sp. in yogurt. Overall, L. plantarum LPP95 and its CFS have the potential as biological preservatives.

Lactiplantibacillus plantarum MPB-65 combined with epicatechin alleviates hyperuricemia by modulating the gut–kidney axis

Zihao Tian,Jing Zhang,Zhongmei Tan,Xiaolei Duan,He Zhang,Bailiang Li,Enyu Wang,Vicky Yang Wang,Dong Xu,Jiage Ma,Wei Yu

Hyperuricemia (HUA) is a metabolic disorder induced by excessive generation or underexcretion of uric acid (UA), leading to abnormally elevated levels of serum UA beyond the normal range. This study explored the potential effects of Lactiplantibacillus plantarum MPB-65 combined with epicatechin (EC) as a synbiotic supplement in alleviating HUA. The results showed that the combination of L. plantarum MPB-65 and EC effectively reduced the levels of serum UA, creatinine (CRE), and blood urea nitrogen (BUN). It also alleviated kidney and colon inflammatory responses, reduced kidney oxidative stress, and relieved tissue injury in the kidneys and colon. Furthermore, L. plantarum MPB-65 combined with EC could inhibit liver xanthine oxidase (XOD) activity, downregulate the expression of URAT1 and GLUT9 in the kidneys and upregulate the expression of ABCG2 in the colon. It was also shown to significantly suppress the expression of key proteins in the JAK2/STAT3 signaling pathway in the kidneys. Meanwhile, the HUA-induced gut microbiota dysbiosis was ameliorated by intervention with L. plantarum MPB-65 combined with EC, particularly in the species composition and structure of the gut microbiota. L. plantarum MPB-65 combined with EC increased the short-chain fatty acid levels. Overall, L. plantarum MPB-65 combined with EC exerted alleviating effects on potassium oxonate- and adenine-induced HUA through modulating the gut–kidney axis.

Synergistic action of non-digestible xylooligosaccharide and Lactiplantibacillus plantarum ZDY2013 against high fat diet and streptozocin-induced type 2 diabetes mellitus in rats

Ying Zhan,Na Zhang,Kaiming Wang,Jinmei Li,Mingliang Jin,Nagendra P. Shah,Hua Wei,Zhihong Zhang

Patients with type 2 diabetes mellitus (T2DM) often exhibit reduced Lactobacillus abundance, dysregulated immune responses, disrupted intestinal barrier integrity, and increased insulin resistance. Consumption of non-digestible oligosaccharides has been shown to support the persistence of Lactobacillus in the gut and improve gut homeostasis. Lactiplantibacillus plantarum ZDY2013, a probiotic capable of metabolizing various oligosaccharides, serves as a potent regulator of intestinal mucosal immunity. In this study, we investigated the potential ameliorative effects of xylooligosaccharides combined with L. plantarum ZDY2013 (synbiotic) on T2DM-induced intestinal injury and explored the underlying mechanisms. Our results showed that synbiotic improved glucose metabolism, reduced lipid accumulation, and alleviated insulin resistance in T2DM rats. Moreover, synbiotic outperformed L. plantarum ZDY2013 alone in restoring intestinal barrier integrity by suppressing oxidative stress and intestinal inflammation, while significantly enhancing the colonization of L. plantarum ZDY2013 and altering the abundance of key bacterial genera. Interestingly, synbiotic treatment also increased the production of short-chain fatty acids (SCFAs), which were strongly associated with specific bacterial taxa. Furthermore, gut microbiota-derived SCFAs were confirmed to ameliorate insulin resistance by promoting glucose uptake and glycogen synthesis in IR-HepG2 cells. Collectively, these findings suggest the potential use of synbiotics as a clinical intervention to ameliorate T2DM. This study provides a rationale for exploring dietary approaches as a mitigating strategy for managing long-standing diabetes.

Lactiplantibacillus plantarum FRT4 protects against fatty liver hemorrhage syndrome: regulating gut microbiota and FoxO/TLR-4/NF-κB signaling pathway in laying hens

Li Daojie,Meng Kun,Liu Guohua,Wen Zhiguo,Han Yunsheng,Liu Weiwei,Xu Xin,Song Liye,Cai Hongying,Yang Peilong

Background Fatty liver hemorrhage syndrome (FLHS) has become one of the major factors leading to the death of laying hen in caged egg production. FLHS is commonly associated with lipid peroxidation, hepatocyte injury, decreased antioxidant capacity, and inflammation. However, there are limited evidences regarding the preventive effect of Lactiplantibacillus plantarum on FLHS in laying hens and its mechanisms. Our previous results showed that Lp. plantarum FRT4 alleviated FLHS by regulating lipid metabolism, but did not focus on its antioxidant and anti-inflammatory functions and mechanisms. Therefore, this study aimed to investigate the preventive mechanisms of Lp. plantarum FRT4 in alleviating FLHS, with a focus on its role in antioxidant activity and inflammation regulation. Results Supplementation with Lp. plantarum FRT4 enhanced the levels of T-AOC, T-SOD, and GSH-Px, while reducing the levels of TNF-α, IL-1β, IL-8, and NLRP3 in the liver and ovary of laying hens. Additionally, Lp. plantarum FRT4 upregulated the mRNA expressions of SOD1 , SOD2 , CAT , and GPX1 , downregulated the mRNA expressions of pro-inflammatory factors IL-1β , IL-6 , and NLRP3 , and upregulated the mRNA expressions of anti-inflammatory factors IL-4 and IL-10 . Lp. plantarum FRT4 improved the structure and metabolic functions of gut microbiota, and regulated the relative abundances of dominant phyla (Bacteroidetes, Firmicute, and Proteobacteria) and genera ( Prevotella and Alistipes ). Additionally, it influenced key KEGG pathways, including tryptophan metabolism, amino sugar and nucleotide sugar metabolism, insulin signaling pathway, FoxO signaling pathway. Spearman analysis revealed that the abundance of microbiota at different taxonomic levels was closely related to antioxidant enzymes and inflammatory factors. Furthermore, Lp. plantarum FRT4 modulated the mRNA expressions of related factors in the FoxO / TLR - 4 / NF-κB signaling pathway by regulating gut microbiota. Moreover, the levels of E 2 , FSH, and VTG were significantly increased in the ovary after Lp. plantarum FRT4 intervention. Conclusions Lp. plantarum FRT4 effectively ameliorates FLHS in laying hens. This efficacy is attributed to its antioxidant and anti-inflammatory properties, which are mediated by modulating the structure and function of gut microbiota, and further intervening in the FoxO / TLR - 4 / NF-κB signaling pathway. These actions enhance hepatic and ovarian function and increase estrogen levels. Video

Lactiplantibacillus plantarum P101 Alleviated Alcohol-Induced Hepatic Lipid Accumulation in Mice via AMPK Signaling Pathway: Gut Microbiota and Metabolomics Analysis

Feng Xiaoyan,Wang Mengqi,Wen Siyue,Hu Liehai,Lan Yuzhi,Xu Hengyi

Mitigating steatosis is essential for delaying the progression of alcoholic liver disease. The effect and mechanism of Lactiplantibacillus plantarum P101 (LP.P101) on alleviating alcohol-induced hepatic lipid accumulation were investigated in our study. The mouse model was constructed by a short-term (10-day)-plus-binge ethanol feeding and gavaged with 10 8 CFU/mL of LP.P101 daily. Lipid droplet in the liver was significantly reduced by LP.101 intervention on AMPK activation. However, when AMPK was inhibited by dorsomorphin, the levels of related indicators (ALT, TG, etc.) and the expression levels of AMPK and relevant genes in the liver converged to that of the alcohol-fed group. Compared with the alcohol-fed group, LP.P101 reduced the relative abundance of Firmicutes and increased that of Bacteroidetes. Parabacteroides merdae was negatively correlated with lipid accumulation, and unclassified Negativibacillus was negatively associated with AMPK activation. Importantly, LP.P101 modified the compositions of the serum metabolites. The potential biomarker stercobilinogen was positively correlated with AMPK activation and negatively associated with lipid accumulation. This work confirmed that LP.P101 attenuated alcohol-induced hepatic lipid accumulation in mice through AMPK activation, and the alterations in gut microbiota and metabolites may play a significant role on AMPK activation.
