Polysaccharides from Lyophyllum decastes reduce obesity by altering gut microbiota and increasing energy expenditure

Tao Wang,Junjie Han,Huanqin Dai,Jingzu Sun,Jinwei Ren,Wenzhao Wang,Shanshan Qiao,Chang Liu,Li Sun,Shuangjiang Liu,Dianpeng Li,Shenglong Wei,Hongwei Liu

Polysaccharides are known to confer protection against obesity via modulation of gut microbiota . To expand our knowledge of mushroom-derived prebiotics , we investigated the structural characteristics and anti-obesity effects of Lyophyllum decastes polysaccharides. Two heteroglycans were purified and characterized. The isolated polysaccharides effectively reduced obesity and the related disorders in the diet-induced obese (DIO) mice. An altered gut microbiota with enrichments of Bacteroides intestinalis and Lactobacillus johnsonii and an increase of secondary bile acids were detected in the polysaccharide-treated mice. Supplementation of B. intestinalis and L. johnsonii prevented the obesity and hyperlipidemia in DIO mice, demonstrating their causal linkage to the efficacy of polysaccharides. An enhancement of energy expenditure in the brown adipose tissues due to up-regulation of the secondary bile acids-activated TGR5 pathway was deduced to be one of the mechanisms underlying the effect of polysaccharides. These results confirmed Lyophyllum decastes -derived polysaccharides as new prebiotics for preventing and treating obesity.

Synergistic protective effects of β-glucan and Lactobacillus johnsonii NSP009 in ulcerative colitis

Shanshan Zhang,Yonggan Sun,Linlin Gao,Ziyi Zhang,Yongkang Yu,Dechang Xu,Shaoping Nie

The combination of probiotics and prebiotics has emerged as a promising strategy for managing ulcerative colitis (UC). Among these, β-glucan (BG) and Lactobacillus johnsonii have individually shown potential in alleviating colitis, with BG notably enriching L. johnsonii populations, suggesting a synergistic therapeutic potential when combined. This study investigates the protective effects of a novel synbiotic formulation, BG and L. johnsonii NSP009 (LJ), termed BGLJ, in a dextran sulfate sodium (DSS)-induced colitis mouse model. Our results reveal that BGLJ significantly mitigated DSS-induced weight loss, reduced the disease activity index, and alleviated intestinal inflammation and barrier dysfunction. Notably, BGLJ markedly increased L. johnsonii abundance in mouse feces, with LJ producing substantial levels of indole-3-lactic acid (ILA), an aryl hydrocarbon receptor (AhR) agonist. BGLJ treatment robustly elevated ILA levels in the gut, activating AhR and upregulating IL-22 production. Collectively, our findings highlight the therapeutic potential of the BGLJ-based synbiotic in maintaining intestinal homeostasis and underscore its promise as a novel intervention for UC management.

Postbiotics From Lactobacillus Johnsonii Activates Gut Innate Immunity to Mitigate Alcohol-Associated Liver Disease

Ruopeng Yin,Tao Wang,Jingzu Sun,Huanqin Dai,Yuting Zhang,Ningning Liu,Hongwei Liu

Prolonged alcohol consumption disrupts the gut microbiota and the immune system, contributing to the pathogenesis of alcohol-associated liver disease (ALD). Probiotic-postbiotic intervention strategies can effectively relieve ALD by maintaining gut homeostasis. Herein, the efficacy of heat-killed Lactobacillus johnsonii (HKLJ) in mitigating alcoholic liver damage is demonstrated in mouse models of ALD. The gut-liver axis is identified as a pivotal pathway for the protective effects of L. johnsonii against ALD. Specifically, HKLJ is found to upregulate the expression of intestinal lysozymes, thereby enhancing the production of immunoregulatory substances from gut bacteria, which subsequently activated the Nucleotide-binding oligomerization domain 2 (NOD2)-interleukin (IL-23)-IL-22 innate immune axis. The elevated IL-22 upregulated the antimicrobial peptide synthesis to maintain intestinal homeostasis and moreover activated the Signal transducer and activator of Transcription3 (STAT3) pathway in the liver to facilitate the repair of hepatic injuries. The heat-killed L. johnsonii provoked immunity helps correct the gut microbiota dysbiosis, specifically by reversing the reduction of butyrate-producing bacteria (such as Faecalibaculum rodentium ) and the expansion of opportunistic pathogens (such as Helicobacter sp. and Pichia kudriavzevii ) induced by ethanol. The findings provide novel insights into the gut microbiota-liver axis that may be leveraged to enhance the treatment of ALD.

Poly-β-hydroxybutyrate alleviated diarrhea and colitis via Lactobacillus johnsonii biofilm-mediated maturation of sulfomucin

Ma Ning,Guo Pingting,Chen Jingyu,Qi Zengkai,Liu Chunchen,Shen Jiakun,Sun Yiwei,Chen Xiyue,Chen Guo-Qiang,Ma Xi

Maintainance of sulfomucin is a key end point in the treatment of diarrhea and inflammatory bowel disease (IBD). However, the mechanisms underlying the microbial sense to sulfomucin are poorly understood, and to date, there are no therapies targeting the secretion and maturation of sulfomucin in IBD. Herein, we biosynthesized poly-β-hydroxybutyrate (PHB) and found that PHB could alleviate inflammation caused by diarrhea and colitis by enhancing the differentiation of sulfomucin. Microbiota transplantation and clearance together demonstrate that PHB promoting sulfomucin is mediated by Lactobacillus johnsonii ( L. johnsonii ). Further studies revealed that PHB provides a favorable niche for L. johnsonii biofilm formation to resist disturbance and support its growth. L. johnsonii -biofilm alleviates colitis by regulating fucose residues to promote goblet cell differentiation and subsequent sulfomucin maturation. Importantly, PHB alleviates colitis by enhancing sulfomucin secretion and maturation in a L. johnsonii -dependent manner. PHB represents a class of guardians, acting as a safe probiotic-biofilm delivery system that significantly promotes probiotic proliferation. Altogether, this study adds weight to the possible role of probiotics and functional materials in the treatment of intestinal inflammation. The application of PHB and biofilm self-coating L. johnsonii carries high translational potential and may be of clinical relevance.

Biocompatible Chitosan-Based Nanoparticles for the Delivery of Lactobacillus johnsonii Lipoteichoic Acid: Enhancing Stability and Biofunctional Properties

Yini Liu,Min Guo,Jingyu Lu,Jianxin Zhao,Gang Wang,Ruojie Zhang,Hao Zhang,Zipei Zhang

Lipoteichoic acid (LTA), a component of the Gram-positive bacterial cell wall, exhibits potent immunomodulatory properties. However, it is highly unstable and susceptible to degradation under gastrointestinal conditions, which limits its in vivo functional application. In this study, LTA from theLactobacillus johnsonii was encapsulated into chitosan-tripolyphosphate (CS-TPP) nanoparticles for the first time, establishing a robust delivery system. The resulting LTA@CS-TPP nanoparticles displayed uniform nano size, favorable stability, high encapsulation efficiency, and sustained release behavior. Simulated digestion assays demonstrated that nanoencapsulation effectively protected LTA from gastric degradation and enabled controlled release in the intestinal phase. Cellular experiments showed that LTA@CS-TPP enhanced antioxidative stress defenses, reduced intracellular ROS levels, and alleviated LPS-induced inflammatory injury. Immunofluorescence analysis further revealed that LTA@CS-TPP significantly suppressed NF-κB activation and nuclear translocation, suggesting interference with the TLR4/MyD88/NF-κB signaling cascade. Compared with free LTA, nanoencapsulation not only preserved but also amplified its bioactivity, whereas blank CS-TPP nanoparticles exerted minimal effects. Collectively, these findings provide the first evidence of chitosan-based nanoencapsulation of LTA and highlight its potential as a bioactive delivery platform for inflammation modulation, laying the groundwork for future oral administration studies with promising applications in functional foods and therapeutic strategies.

Fingerprinting and characterization of the polysaccharides from Polygonatum odoratum and the in vitro fermented effects on Lactobacillus johnsonii

Jia-Rui Liu,Bo-Xue Chen,Jia-Qi Huang,Xue Li,Tian-Yi Cui,Bin Lv,Zhi-Fei Fu,Xin Zhao,Wen-Zhi Yang,Xiu-Mei Gao

Polygonatum odoratum (Yu-Zhu) can be utilized to treat the digestive and respiratory illness . Previous studies have revealed that the underlying therapeutic mechanism of P. odoratum polysaccharides (POPs) is associated with remodeling the gut microbiota . However, POPs in terms of the chemical composition and fermentation activities have been understudied. Here we developed the three-level fingerprinting approaches to characterize the structures of POPs and probed into the beneficial effects on promoting the growth and fermentation of Lactobacillus johnsonii . POPs were prepared by water decoction followed by alcohol sedimentation, while trifluoroacetic acid under different conditions to prepare the hydrolyzed oligosaccharides and monosaccharides . POPs exhibited three main molecular distribution of 601–620 kDa, 4.12–6.09 kDa, and 3.57–6.02 kDa. Hydrolyzed oligosaccharides with degree of polymerization (DP) 2–13 got primarily characterized by analyzing the rich fragmentation information obtained by hydrophilic interaction chromatography/ion mobility-quadrupole time-of-flight mass spectrometry (HILIC/IM-QTOF-MS). Amongst them, the DP5 oligosaccharide was characterized as 1,6,6-kestopentaose. The molecular ratio of Fru: Ara : Glc: Gal : Xyl was 87.72: 0.30: 11.56: 0.19: 0.23. In vitro fermentation demonstrated that 4.5 mg/mL of POPs could significantly promote the growth of L. johnsonii . Co-cultivated with 4.5 mg/mL of POPs, L. johnsonii exhibited stronger antimicrobial activity against Klebsiella pneumoniae . The concentrations of short-chain fatty acids in the POPs-lactobacilli fermented products, including acetic acid, isobutyric acid , and isovaleric acid , were increased. Conclusively, POPs represent the promising prebiotic candidate to facilitate lactobacilli , which is associated with exerting the health benefits.

Lactobacillus johnsonii L531 Alleviates the Damage Caused by Salmonella Typhimurium via Inhibiting TLR4, NF-κB, and NLRP3 Inflammasome Signaling Pathways

Shiyan Chen,Yanan Li,Bingxin Chu,Lanxin Yuan,Ning Liu,Yaohong Zhu,Jiufeng Wang

SalmonellaTyphimurium (S.Typhimurium) is an aggressive zoonotic pathogen that causes enteritis and diarrhea. Antibiotic therapy is still the primary method at present. However, the increasing emergence of multi-drug resistant bacteria weakens the therapeutic efficacy of antibiotics. Probiotics have been widely studied as an alternative antibiotic therapy. In this study, we established an IPEC-J2 cell model ofS.Typhimurium infection, aiming to determine the protective effect ofLactobacillus johnsoniiL531 (L. johnsoniiL531) onS.Typhimurium infection. As our data showed,S.Typhimurium infection resulted in a robust inflammatory response demonstrated by promoted protein levels of the inflammatory-related pathway (TLR4, MyD88, p-IκBα, and p-p65), increased cytokine levels of IL-6, IL-1β, IL-18, and TNF-α, and activated the NLRP3 inflammasome via promoting its assembly. However,L. johnsoniiL531 pre-incubation inhibited the activation of the above inflammatory signaling pathways and reduced the expression levels of pro-inflammatory cytokines. In addition,L. johnsoniiL531 alleviated the damage ofS.Typhimurium to tight junctions ZO-1, Occludin, and Claudin-1. In summary, our findings suggested thatL. johnsoniiL531 alleviatedS.Typhimurium-induced tight junction injury by inhibiting the TLR4/NF-κB/NLRP3 inflammasome signaling pathway. Keywords:SalmonellaTyphimurium;Lactobacillus johnsonii;TLR4;NF-κB;NLRP3 inflammasome;tight junctions

Investigation of the mechanism by which Tegillarca granosa polysaccharide regulates non-alcoholic fatty liver disease in mice by modulating Lactobacillus Johnsonii

Hanwen Zhang,Xingwen Yang,Jinyu Chen,Qihong Jiang,Shiwei Ya,Lin Chen,Xingwei Xiang

Non-alcoholic fatty liver disease (NAFLD), a prevalent chronic liver disease, is marked by excessive lipid deposition in the liver without alcohol abuse. Scapharca subcrenatum, a major Chinese farmed bivalve, yields S. subcrenatum polysaccharide (TGP), an active substance with known biological activity. Previous studies revealed TGP's significant regulatory effect on a high-fat diet (HFD)-induced NAFLD in mice. However, the precise mechanisms, particularly involving gut microbiota, remain unclear. In the current study, an antibiotic-treated mouse model was established to determine the mechanistic role of the gut microbiota in the observed anti-obesity effects of TGP. In addition, 16S rRNA genomic and metagenome-derived taxonomic analyses were performed to assess the gut microbial populations. The results showed that TGP selectively enhanced the number of the eosinophilic bacterium Lactobacillus johnsonii , which was reduced in HFD mice. Of note, the oral administration of L. johnsonii formulations to HFD mice alleviated NAFLD, and this was related to regulating lipid metabolism and the accumulation of lipids in the liver. Therefore, the current study uncovered a potential pathway for developing NAFLD treatment strategies based on the interaction between TGP and the gut microbiota.

Buyang Huanwu Decoction promotes neurorepair after spinal cord injury through a Lactobacillus johnsonii–indole-3-lactic acid–AhR–PI3K/Akt axis

Dong Jinwang,Cao Yang,Chen Xiujin,Xie Tao,Zhang Xiaobo,Zhao Qingpeng,Shi Cunhu,Miao Qiangqiang,Xu Zhengwei,Yan Liang,Dong Liang

Background Spinal cord injury (SCI) induces gut microbiota dysbiosis, which significantly affects recovery. Buyang Huanwu Decoction (BHD), a traditional Chinese medicine formula, has shown therapeutic effects on SCI. Although BHD is known to modulate gut microbiota, whether its benefits are mediated through the gut–spinal cord axis remains unclear. Methods A rat SCI model was established. BHD was administered orally, and fecal microbiota transplantation (FMT) from BHD-treated rats (BHD-FMT) was performed to assess neuroprotective and gut-protective effects. Behavioral testing, histology, and immunofluorescence evaluated motor recovery, inflammation, and neuroregeneration. Gut microbiota profiling was performed using 16S rDNA sequencing and metagenomics, while targeted metabolomics quantified tryptophan metabolites. Transcriptomics validated key pathways, and a microbiota–metabolite–signaling network was constructed. Results BHD significantly improved motor function, reduced spinal inflammation, and promoted neuronal survival and axonal regeneration. It restored gut function, reduced colonic inflammation, and enhanced ZO-1 and Occludin expression, which were further confirmed by FMT. BHD-FMT reshaped the gut microbiota and enriched Lactobacillus johnsonii , which correlated positively with recovery. Metabolomics showed increased tryptophan metabolites, including indole-3-lactic acid (ILA) and indole-3-propionic acid (IPA), with ILA strongly associated with functional improvement. Transcriptomic analysis and Western blot validation demonstrated that BHD-FMT activated the AhR–PI3K/Akt pathway, which was suppressed by an AhR antagonist. Conclusion BHD promotes neuroregeneration after SCI by reshaping gut microbiota and enhancing tryptophan metabolism, potentially exerting its effects through the L. johnsonii –ILA–AhR–PI3K/Akt network. These findings reveal a gut–spinal cord axis–mediated mechanism of BHD and highlight microecological targets for SCI therapy. Effects of fermentation with feruloyl esterase-producing Lactobacillus johnsonii on the composition, structure, physicochemical properties, and in vitro hypoglycemic activity of soluble dietary fiber from rice bran

Xiangfei Li,Menghan Wang,Jie Liu,Fei Gao,Xinyi Pang,Jing Sun,Chi Chen,Yingjian Lu

Soluble dietary fiber (SDF) is a key functional component in rice bran, but its low natural abundance limits its application potential. In this study, two Lactobacillus johnsonii strains with distinct feruloyl esterase (FAE) activities were selected to investigate their effects on the modification of rice bran SDF through fermentation. The results showed that fermentation with the high-FAE strain L. johnsonii 46 (FSDF-46) increased the SDF yield from 6.39% to 11.45%. Structural characterization indicated that fermentation was associated with the formation of a porous structure, reduced particle size, and decreased crystallinity. Furthermore, FSDF-46 exhibited higher water holding capacity (0.87 g/g) and oil holding capacity (3.789 g/g). The glucose adsorption capacity of FSDF-46 increased by 44.8% and 37.52%. In vitro assays showed that FSDF-46 exhibited enhanced inhibitory effects on α-amylase (52.33%) and α-glucosidase (91.63%). These findings suggest a potential role of FAE-related fermentation in dietary fiber modification and provide a basis for the development of value-added products from rice bran.

Traditional Chinese medicine formulation Chang-Kang-Fang restores gut-brain homeostasis in IBS-D via the Lactobacillus johnsonii-indole-3-lactic acid-aryl hydrocarbon receptor/interleukin-22 axis

Fengge Yang,Danmei Tian,Yuting Zhang,Zixuan Xia,Yunjuan Li,Xinsheng Yao,Yanxia Xiong,Guirong Zhou,Hongwei Liu,Jinshan Tang

CKF ameliorates intestinal dysfunction and neurobehavioral abnormalities in IBS-D mice. • CKF boosts L. johnsonii and tryptophan-derived indoles, notably ILA. • L. johnsonii acts as a psychobiotic for IBS-D and comorbidities. • ILA improves IBS-D intestinal and behavioral deficits mainly via AhR activation. • CKF targets the L. johnsonii -ILA-AhR/IL-22 gut-brain axis. Background Gut dysbiosis contributes to irritable bowel syndrome (IBS) by disrupting intestinal barrier functions and promoting inflammation. Chang‑kang‑fang (CKF), a Chinese herbal formula proven effective for diarrhea‑predominant IBS (IBS‑D) in a phase III trial, is thought to modulate the gut microbiota, yet the underlying mechanism remains unclear. Purpose To elucidate how CKF ameliorates IBS‑D by modulating the gut microbiota. Methods IBS‑D was induced by restraint stress, tail pinch, and senna gavage. Intestinal function, inflammation, and behavior were evaluated. Gut microbiota and metabolites were profiled via 16S rRNA sequencing and metabolomics. FMT confirmed the causal involvement of the microbiota. The key microbe and metabolite were validated in vivo . Mechanistic analyses evaluated intestinal barrier integrity, systemic inflammation, hippocampal blood‑brain barrier (BBB) function, neuroinflammation, and synaptic plasticity. Results CKF alleviated diarrhea, visceral hypersensitivity, inflammation, barrier dysfunction, and anxiety‑ and depression‑like behaviors. These benefits were associated with gut microbiota remodeling, featuring enrichment of Lactobacillus johnsonii and enhanced tryptophan metabolism. FMT confirmed that this microbial remodeling mediates the therapeutic effects of CKF. Supplementation with L. johnsonii or its metabolite indole‑3‑lactic acid (ILA) largely recapitulated these gut-brain benefits via AhR/IL‑22 activation. Mechanistically, CKF‑induced L. johnsonii enrichment activated the ILA-AhR-IL‑22 pathway, which enhanced intestinal and BBB integrity, thereby attenuating systemic and neuroinflammation, and restoring hippocampal synaptic plasticity. Conclusion This study positions CKF as a microbiota‑modulating therapy for IBS‑D that restores gut‑brain homeostasis via the L. johnsonii -ILA-AhR-IL‑22 axis, highlighting L. johnsonii and ILA as potential therapeutics for IBS and its comorbidities.
