Lactobacillus reuteri ZJ617 attenuates metabolic syndrome via microbiota-derived spermidine

Ma Yanfei,Zhong Yifan,Tang Wenjie,Valencak Teresa G.,Liu Jingliang,Deng Zhaoxi,Mao Jiangdi,Liu Daren,Wang Shanshan,Wang Yuhao,Wang Haifeng

Metabolic syndrome (MetS) is a difficult-to-manage disease that poses a significant risk to human health. Here, we show that the supplementation of Lactobacillus reuteri ZJ617 ameliorates symptoms of MetS in mice induced by the high-fat diet. L. reuteri ZJ617 modulates host metabolism by interacting with the microbiome, resulting in the production of spermidine synthesized by the microbiota. L. reuteri ZJ617 serves as a source of substrates for the microbiota to synthesize spermidine, hence preventing the decline of bacteria responsible for spermidine production. Spermidine treatment mimics the metabolic effects of L. reuteri ZJ617, whereas pharmacological inhibition of spermidine biosynthesis in mice abolishes these benefits. Our findings reveal the mechanism by which L. reuteri ZJ617 alleviates MetS symptoms and provide support for its potential use as a probiotic for promoting metabolic health.

Characterisation of aldo-keto reductases from Lactobacillus reuteri DSM20016

Wenhao Qu,Yajun Bai,Tai-Ping Fan,Xiaohui Zheng,Yujie Cai

Lactobacillus reuteri DSM20016 has several aldo-keto reductases (AKRs), and they were heterologously expressed in E. coli BL21 (DE3) in this study. Subsequently, these AKRs were purified, and their enzymatic properties were investigated. It was found that these enzymes used NADPH as coenzyme and did not depend on metal ions. The optimum pH of these AKRs was a slightly acidic (AKR-1 and AKR-6: 5.5; AKR-3 and AKR-8: 6; AKR-2 and AKR-4: 6.5). These enzymes had a broad substrate spectrum and could convert a variety of aldehydes to alcohols; however, they did not catalyze the reverse reactions. All results showed that Lactobacillus reuteri had the ability to produce various volatile compounds, which allowed Lactobacillus reuteri to impart complex aroma to something.

Lactobacillus reuteri DSM 17938 alleviates d-galactosamine-induced liver failure in rats

Huiyong Jiang,Ren Yan,Kaicen Wang,Qiangqiang Wang,Xiaoxiao Chen,Lifeng Chen,Lanjuan Li,Longxian Lv

Liver failure is a serious hepatic dysfunction with high mortality. This work aimed to investigate the effect of a famous probiotic and drug, Lactobacillus reuteri DSM 17938, on liver failure in rats. Sprague-Dawley rats were gavaged with 3 × 10 9 CFU of DSM 17938 for 7 days. d -galactosamine was intraperitoneally injected to induce acute liver failure on the eighth day. Samples were collected to determine the liver function, serum cytokines levels, terminal ileum and liver histology, gut microbiota, metabolome and transcriptome. Our results showed that pretreatment with DSM 17938 not only reduced the elevation in serum alanine aminotransferase, aspartate aminotransferase, gamma-glutamyl transferase, IL-1α, IL-2, IL-18, M-CSF, and MIP-3α levels but also alleviated histological abnormalities of both the terminal ileum and liver induced by d -galactosamine. Additionally, DSM 17938 reduced d -galactosamine-induced enrichment of some taxa of gut Actinobacteria or Firmicutes, including abundant pathogens such as Actinomycetales , Coriobacteriaceae , Staphylococcaceae and Enterococcaceae . Furthermore, DSM 17938 reduced the d -galactosamine-induced increase in not only fecal metabolites such as trisaminol and lithocholic acid but also the transcription of liver inflammatory genes, such as Ccl2 , Ccl7 , Ccl11 , Ccl12 , Il6 , Il11 , Il20rb , Mmp3 and Mmp10 . Downregulation of retinol metabolism and PPAR signaling pathway as well as upregulation of viral protein interaction with cytokine and cytokine receptor and central carbon metabolism in cancer signaling pathway were involved in the mechanism of L. reuteri DSM 17938 alleviating liver failure. Our findings suggested that DSM 17938 is a potential probiotic for the prevention or treatment of liver failure.

Characterisation of five alcohol dehydrogenases from Lactobacillus reuteri DSM20016

Zhenghong Hu,Pu Jia,Yajun Bai,Tai-ping Fan,Xiaohui Zheng,Yujie Cai

Alcohol dehydrogenases convert aldehydes causing volatile aromas in cheese into corresponding alcohols. In this study, we cloned and recombinantly produced five novel ADHs from Lactobacillus reuteri DSM20016 in Escherichia coli BL21 (DE3). The resulting proteins have molecular weights between 35 and 39 kDa. The optimum pH for enzyme activity of ADH-1, ADH-2 and ADH-3 was weakly acidic (pH 5, 6 and 5, respectively), compared with pH 7 for ADH-4 and ADH-5. The optimum temperature range for ADH-1, ADH-3, ADH-4 and ADH-5 was 40 °C ―55 °C, compared with 35 °C for ADH-2. These enzymes reduced a broad range of substrates; the highest catalytic efficiency and substrate affinity for ADH-1 was observed with heptaldehyde (K cat /K m  = 39.4 s −1 ·mM −1 , K m  = 0.459 mM), while 3-methylbutanal was the best substrate for ADH-5 (K cat /K m  = 24.0 s −1 ·mM −1 , K m  = 1.35 × 10 −2  mM). ADH-2, ADH-3 and ADH-4 exhibited the highest substrate affinity for valeraldehyde, propionaldehyde, and hexanal, respectively (K m  = 1.19 × 10 −2  mM, 2.81 × 10 −2  mM, 1.35 × 10 −2  mM). The enzymatic properties indicated that these enzymes effectively converted fragrant compounds during cheese ripening.

Lactobacillus reuteri E9 Regulates Sleep Disorders Through Its Metabolite GABA

Yongcheng Jiang,Lina Guo,Houde He,Haonan Chen,Tao Chen,Yan Liu,Wang Zhao

Background:Insomnia, the most prevalent sleep disorder, is clinically defined as difficulty initiating or maintaining sleep. Although many medications are effective for insomnia treatment, they carry risks of drug dependence and abuse. The microbiota-gut-brain axis (MGBA) facilitates bidirectional signaling between the gastrointestinal tract and the central nervous system via gut microbes. Probiotics that provide mental and behavioral benefits through MGBA (psychobiotics) offer broad therapeutic potential. Methods:A non-toxic, drug-resistant strain of Lactobacillus reuteri E9 was isolated and characterized. Its effects were evaluated in a pentylenetetrazol (PTZ)-induced zebrafish model of sleep disorder. Neurotransmitter levels (glycine, serine, taurine, γ-aminobutyric acid (GABA)) and gene expression of GABA/melatonin receptors were analyzed.Results:E9 significantly upregulated inhibitory neurotransmitters, including GABA, taurine, glycine, and serine (p < 0.05). In PTZ-induced zebrafish, E9 exerted sedative effects by reducing seizures and hyperactivity. Concurrently, E9 upregulated the expression of GABA receptor genes and melatonin receptor (Mtnr1aa) genes in zebrafish neural tissue. Conclusions:Lactobacillus reuteri E9 demonstrates potential as a psychobiotic for sleep disorder management by modulating key inhibitory neurotransmitters and sleep-related receptor expression via the MGBA pathway, offering a non-pharmacological alternative to conventional treatments.

Multi-stage assembled Lactobacillus reuteri-capsule patch for persistent anti-infection

Chen Zhou,Yuxiang Wang,Ming Yang,Hao Guo,Xiang Ren,Gongbing Liu,Hailong Wang,Wei Wei,Yujiang Fan,Hongning Wang,Yong Sun

Recurrent bacterial invasion delayed the recovery of infected site so that provide one sustained antibacterial therapy strategy is essential. Here, we used one simple multi-stage assembly strategy to developed a Lactobacillus reuteri-capsule patch that could achieve sustained antibacterial applications. The encapsulated strain in sodium alginate (SA) capsule effectively maintained their activity and exhibited the sustained release behavior through responding to pH-change of infection microenvironment. In addition, the nanofiber structure of bacteria cellulose (BC) shielded the contact between strains and lesion to avoid additional inflammatory reactions (isolation rate reached 99.9%). This patch shown the sustained antibacterial capacity against the formation of common pathogen`s biofilm by hinder the biosynthesis (including E. coli, S. aureus, MRSA and P. aeruginosa), which avoid recurrent bacterial invasion. Transcriptomics results also demonstrates that this patch shown various bioactivities to promote the proliferation, migration and vascularization, in contrast with traditional antibiotic therapy. In vivo infection model verification further demonstrates that this patch effectively clearance biofilm to prevent the recurrent invasion of P. aeruginosa, while achieving substantial tissue regeneration through collagen deposition and neovascularization. This strategy provides a simple and efficient approach for persistent anti-infection application.

Lactobacillus reuteri Biofilms Inhibit Pathogens and Regulate Microbiota in In Vitro Fecal Fermentation

Meng-Xin Hu,Fei He,Ya-Xin Guo,Li-Zhen Mo,Xuan Zhu

Bacteria colonizing the gastrointestinal tract generally grow well in biofilms. In recent years, probiotic biofilms have been considered the most promising fourth-generation probiotics. However, the research into the functions of probiotic biofilms is just starting. In this study, Lactobacillus reuteri DSM 17938 biofilms formed on electrospun cellulose acetate nanofibrous scaffolds were contrasted with planktonic cells. Pathogen inhibition analysis of Escherichia coli, Staphylococcus aureus, and Listeria monocytogenes suggested a significant distinction between the planktonic and biofilm groups. In human fecal fermentation, L. reuteri remodeled the microbiota by decreasing the relative abundances of Proteobacteria, Escherichia–Shigella, and Desulfovibrio and increasing the relative abundances of Phascolarctobacterium, Bacteroides, and Lactobacillus. Moreover, L. reuteri biofilms played more positive roles in microbiota modulation and short-chain fatty acid production than planktonic L. reuteri. These findings provide an understanding of the beneficial effects of probiotic biofilms, laying a foundation for the application of probiotic biofilms as a health promoter.

Taking SCFAs produced by Lactobacillus reuteri orally reshapes gut microbiota and elicits antitumor responses

Li Nannan,Niu Lili,Liu Yao,Wang Yang,Su Xiaomin,Xu Ce,Sun Zanya,Guo Huishu,Gong Jingru,Shen Shun

Background Colorectal cancer (CRC) incidence is increasing in recent years due to intestinal flora imbalance, making oral probiotics a hotspot for research. However, numerous studies related to intestinal flora regulation ignore its internal mechanisms without in-depth research. Results Here, we developed a probiotic microgel delivery system (L.r@(SA-CS)2) through the layer-by-layer encapsulation technology of alginate (SA) and chitosan (CS) to improve gut microbiota dysbiosis and enhance anti-tumor therapeutic effect. Short chain fatty acids (SCFAs) produced by L.r have direct anti-tumor effects. Additionally, it reduces harmful bacteria such as Proteobacteria and Fusobacteriota, and through bacteria mutualophy increases beneficial bacteria such as Bacteroidota and Firmicutes which produce butyric acid. By binding to the G protein-coupled receptor 109A (GPR109A) on the surface of colonic epithelial cells, butyric acid can induce apoptosis in abnormal cells. Due to the low expression of GPR109A in colon cancer cells, MK-6892 (MK) can be used to stimulate GPR109A. With increased production of butyrate, activated GPR109A is able to bind more butyrate, which further promotes apoptosis of cancer cells and triggers an antitumor response. Conclusion It appears that the oral administration of L.r@(SA-CS)2 microgels may provide a treatment option for CRC by modifying the gut microbiota.

Lactobacillus reuteri biofilms formed on porous zein/cellulose scaffolds: Synbiotics to regulate intestinal microbiota

Fei He,Xue-Ke Ma,Cheng-Kai Tu,Hui Teng,Xin Shao,Jie Chen,Meng-Xin Hu

Supplementing probiotics or indigestible carbohydrates is a usual strategy to prevent or revert unhealthy states of the gut by reshaping gut microbiota. One criterion that probiotics are efficacious is the capacity to survive in the gastrointestinal tract. Biofilm is the common growth mode of microorganisms with high tolerances toward harsh environments. Suitable scaffolds are crucial for successful biofilm culture and large-scale production of biofilm-phenotype probiotics. However, the role of scaffolds containing indigestible carbohydrates in biofilm formation has not been studied. In this study, porous zein/cellulose composite scaffolds provided nitrogen sources and carbon sources simultaneously at the solid/liquid interfaces, being beneficial to the biofilm formation of Lactobacillus reuteri . The biofilms showed 2.1–17.4 times higher tolerances in different gastrointestinal conditions. In human fecal fermentation, the biofilms combined with the zein/cellulose composite scaffolds act as the “synbiotics” positively modulating the gut microbiota and the short-chain fatty acids (SCFAs), where biofilms provide probiotics and scaffolds provide prebiotics. The “synbiotics” show a more positive regulation ability than planktonic L. reuteri , presenting potential applications in gut health interventions. These results provide an understanding of the synergistic effects of biofilm-phenotype probiotics and indigestible carbohydrates contained in the “synbiotics” in gut microbiota modulation.

Prophylactic Supplementation with Lactobacillus Reuteri or Its Metabolite GABA Protects Against Acute Ischemic Cardiac Injury

Jiawan Wang,Hao Zhang,Hailong Yuan,Siqi Chen,Ying Yu,Xuan Zhang,Zeyu Gao,Heng Du,Weitao Li,Yaohui Wang,Pengyan Xia,Jun Wang,Moshi Song

The gut microbiome has emerged as a potential target for the treatment of cardiovascular disease. Ischemia/reperfusion (I/R) after myocardial infarction is a serious complication and whether certain gut bacteria can serve as a treatment option remains unclear. Lactobacillus reuteri ( L. reuteri ) is a well-studied probiotic that can colonize mammals including humans with known cholesterol-lowering properties and anti-inflammatory effects. Here, the prophylactic cardioprotective effects of L. reuteri or its metabolite γ-aminobutyric acid (GABA) against acute ischemic cardiac injury caused by I/R surgery are demonstrated. The prophylactic gavage of L. reuteri or GABA confers cardioprotection mainly by suppressing cardiac inflammation upon I/R. Mechanistically, GABA gavage results in a decreased number of proinflammatory macrophages in I/R hearts and GABA gavage no longer confers any cardioprotection in I/R hearts upon the clearance of macrophages. In vitro studies with LPS-stimulated bone marrow-derived macrophages (BMDM) further reveal that GABA inhibits the polarization of macrophages toward the proinflammatory M1 phenotype by inhibiting lysosomal leakage and NLRP3 inflammasome activation. Together, this study demonstrates that the prophylactic oral administration of L. reuteri or its metabolite GABA attenuates macrophage-mediated cardiac inflammation and therefore alleviates cardiac dysfunction after I/R, thus providing a new prophylactic strategy to mitigate acute ischemic cardiac injury.

Engineered Lactobacillus reuteri for scavenging reactive oxygen species and modulating oral microflora in periodontitis therapy

Wang Yuqiang,Tang Ying,Huang Qianxiao,An Jiaxin,Zhou Yueli,Yang Hongye,Song Fangfang,Zhang Xianzheng,Huang Cui

The onset and progression of periodontitis are closely associated with subgingival dysbiosis and excessive localized oxidative stress. While some oral probiotics exhibit certain inhibitory effects on periodontitis-related pathogens, they often struggle to effectively colonize and antagonize these pathogens due to the complex oxidative stress at the site of periodontitis. In this study, we engineer Lactobacillus reuteri with a reactive oxygen species (ROS)-responsive adhesive polymer (phenylboric acid-dopamine-hyaluronic acid) (LR@PDH). In the periodontitis microenvironment, this polymer can consume ROS and then expose the phenolic hydroxyl group of dopamine, promoting the selective adhesion and colonization of Lactobacillus reuteri at the site of inflammation to antagonize pathogens. The results show that, compared to conventional probiotic therapy, inflammation-responsive adhesive Lactobacillus reuteri effectively alleviates local oxidative stress, reduces the abundance of pathogenic bacteria in the subgingival microbiome, and inhibits the progression of periodontitis. Additionally, its good biocompatibility and safety highlight its potential as a therapeutic approach for clinical treatment of periodontitis.

Manganese-engineered Lactobacillus Reuteri with enhanced antitumor and immunomodulatory activities for colorectal cancer prevention and treatment

Peng Cao,Qilin Li,Yuyu Li,Honglian Guo,Chunyu Wei,Rengui Xu,Chenke Ouyang,Wei Chen,Lin Wang,Zheng Wang

Probiotic-based biotherapy for colorectal cancer (CRC) faces significant challenges due to poor tumor-targeting and limited bioactivities within the complex tumor microenvironment. This study shows that Lactobacillus reuteri (L. reuteri )’s inherent bioactivities, including proliferation, metabolism, and colorectal colonization, can be enhanced by manganese, prompting the fabrication of a manganese-engineered strain (MnLR). Oral administration of MnLR with its metabolic substrate-glycerol (MnLR/Gly) promotes the enrichment of L. reuteri and its antitumor metabolites within colon tumors. Across multiple preclinical colon tumor models, MnLR/Gly alone achieves a 95.6% inhibition of orthotopic tumor growth, liver metastases reduction by 62.1%, and confers durable protection, with 75% of prophylactically treated mice remaining long-term tumor-free and 62.5% resisting tumor rechallenge. Mechanistically, MnLR/Gly induces both intratumoral and peripheral dendritic cell maturation, M1 macrophage polarization, and effector T-cell responses. In orthotopic CRC rabbit models, GlyMnLR enteric capsules demonstrate significant antitumor efficacy comparable to standard chemotherapy while exhibiting favorable safety. These findings highlight MnLR as an effective standalone probiotic therapy for CRC prevention and treatment. Probiotic therapy for colorectal cancer is limited by the poor tumor targeting and low intratumoral activity. The authors show that manganese-engineered L. reuteri with glycerol (MnLR/Gly) accumulates in colorectal tumors, produces cancer-selective metabolites, activates antitumor immunity, and achieves potent, durable tumor suppression in preclinical models.
