Comparison of Lactobacillus rhamnosus GG and luxS mutant on gut barrier function of mice challenged with Escherichia coli

Zhaoxi Deng,Yanfei Ma,Kangwei Hou,Jinyan Dai,Yifan Zhong,Yingying Mao,Jinzhi Zhang,Haifeng Wang

Quorum sensing (QS) is an essential pathway for bacterial communication through autoinducers (AIs). The luxS gene involved in QS has been proven to regulate different behavior of Lactobacillus rhamnosus GG (LGG), whereas it is unclear whether the AI-2/LuxS system have beneficial effects the host by regulating LGG. Our aim was to investigate whether luxS mutation affects the ability of LGG to maintain intestinal microbiota homeostasis, intestinal barrier integrity and inflammatory immune responses in mice infected with Escherichia coli ( E. coli ). This study showed that both wild-type (WT) and luxS mutant (ΔluxS) of LGG can alleviate changes in the microbiota caused by E. coli by down-regulating the relative abundance of Parasturtella . Supplementation with WT and ΔluxS increased the CD4 + /CD8 + ratio in spleen, improved goblet cell numbers and enhanced Muc 2 secretion. Moreover, WT and ΔluxS decreases proinflammatory mediator levels by inhibiting MAPK and NF-κB signal transduction.

Physicochemical properties and volatile components of pea flour fermented by Lactobacillus rhamnosus L08

Mengqi Pei,Zifu Zhao,Song Chen,E.I. Reshetnik,S.L. Gribanova,Chun Li,Guofang Zhang,Libo Liu,Lin Zhao

Pea ( Pisum sativum L.) is an important source of plant proteins . However, the use of pea flour as a food ingredient is greatly limited due to its strong beany flavor. In this work, we investigated the effects of Lactobacillus rhamnosus L08 ( L. rhamnosus L08) fermentation on the physicochemical characteristics, amino acids composition and volatile flavor compounds of pea flour. We found that L. rhamnosus L08 had the probiotic potential of utilizing pea flour as fermentation substrate. L. rhamnosus L08 fermentation significantly increased the content of most amino acids , and total amino acid content increased from 222.18 mg/g at 0 h to 262.35 mg/g at 6 h. A total of 44 volatile compounds were identified in pea flour. L. rhamnosus L08 fermentation increased the variety of acids and esters, while reduced the unpleasant flavor such as nonanal , decanal , octanal , 1-hexanol and 2-ethyl-1-hexanol. The emulsifying stability and foam stability of pea flour were slightly enhanced during fermentation. Our results illustrate that lactic acid bacteria fermentation is a promising approach to eliminate undesirable aroma of pea flour. Moreover, this study provides theoretical support for the application of peas as high-quality protein source.

A Novel Postbiotic From Lactobacillus rhamnosus GG With a Beneficial Effect on Intestinal Barrier Function

Gao Jie,Li Yubin,Wan Yu,Hu Tongtong,Liu Liting,Yang Shaojie,Gong Zelong,Zeng Qing,Wei Yi,Yang Weijun,Zeng Zhijie,He Xiaolong,Huang Sheng-He,Cao Hong

It has long been known that probiotics can be used to maintain intestinal homeostasis and treat a number of gastrointestinal disorders, but the underlying mechanism has remained obscure. Recently, increasing evidence supports the notion that certain probiotic-derived components, such as bacteriocins, lipoteichoic acids, surface layer protein and secreted protein, have a similar protective role on intestinal barrier function as that of live probiotics. These bioactive components have been named ‘postbiotics’ in the most recent publications. We previously found that the Lactobacillus rhamnosus GG (LGG) culture supernatant is able to accelerate the maturation of neonatal intestinal defense and prevent neonatal rats from oral Escherichia coli K1 infection. However, the identity of the bioactive constituents has not yet been determined. In this study, using liquid chromatography-tandem mass spectrometry analysis, we identified a novel secreted protein (named HM0539 here) involved in the beneficial effect of LGG culture supernatant. HM0539 was recombinated, purified, and applied for exploring its potential bioactivity in vitro and in vivo. Our results showed that HM0539 exhibits a potent protective effect on the intestinal barrier, as reflected by enhancing intestinal mucin expression and preventing against lipopolysaccharide (LPS)- or tumor necrosis factor α (TNF-α)-induced intestinal barrier injury, including downregulation of intestinal mucin (MUC2), zonula occludens-1 (ZO-1) and disruption of the intestinal integrity. Using a neonatal rat model of E. coli K1 infection via the oral route, we verified that HM0539 is sufficient to promote development of neonatal intestinal defense and prevent against E. coli K1 pathogenesis. Moreover, we further extended the role of HM0539 and found it has potential to prevent dextran sulfate sodium (DSS)-induced colitis as well as LPS/D-galactosamine-induced bacterial translocation and liver injury. In conclusion, we identified a novel LGG postbiotic HM0539 which exerts a protective effect on intestinal barrier function. Our findings indicated that HM0539 has potential to become a useful agent for prevention and treatment of intestinal barrier dysfunction- related diseases.

Lactobacillus rhamnosus GG alleviates radiation-induced intestinal injury by modulating intestinal immunity and remodeling gut microbiota

Li-Li Zhang,Jia-Ying Xu,Yifei Xing,Pengcheng Wu,Yi-Wen Jin,Wei Wei,Lin Zhao,Jing Yang,Guo-Chong Chen,Li-Qiang Qin

Radiation injury to the intestine is one of the most common complications in patients undergoing abdominal or pelvic cavity radiotherapy. In this study, we investigated the potential protective effect of Lactobacillus rhamnosus GG (LGG) on radiation-induced intestinal injury and its underlying mechanisms. Mice were assigned to a control group, a 10 Gy total abdominal irradiation (TAI) group, or a group pretreated with 10 8 CFU LGG for three days before TAI. Small intestine and gut microbiota were analyzed 3.5 days post-exposure. LGG intervention improved intestinal structure, reduced jejunal DNA damage, and inhibited the inflammatory cGAS/STING pathway. Furthermore, LGG reduced M1 proinflammatory macrophage and CD8+ T cell infiltration, restoring the balance between Th17 and Treg cells in the inflamed jejunum. LGG also partially restored the gut microbiota. These findings suggest the possible therapeutic radioprotective effect of probiotics LGG in alleviating radiation-induced intestinal injury by maintaining immune homeostasis and reshaping gut microbiota.

Study of the alleviation effects of a combination of Lactobacillus rhamnosus and inulin on mice with colitis

Zhijing Liu,Fei Liu,Wan Wang,Changbao Sun,Da Gao,Jiage Ma,Muhammad Altaf Hussain,Cong Xu,Zhanmei Jiang,Juncai Hou

Ulcerative colitis (UC) is a common inflammatory bowel disease (IBD) that has serious harmful effects on human health. Lactobacillus rhamnosus, a probiotic, has a strong colonization and adhesion effect and improves the intestinal health of the host. Inulin has good anti-inflammatory effects and can promote the proliferation of beneficial intestinal bacteria. The purpose of this study was to investigate the alleviating effects of L. rhamnosus 1.0320 in combination with inulin on UC, examining the resulting changes in intestinal flora. A UC model was established by having mice freely drink a 3% (w/v) dextran sodium sulphate (DSS) solution for seven days. After successful modeling, the mice were given antibiotics, L. rhamnosus 1.0320 by itself, inulin by itself, and L. rhamnosus 1.0320 combined with inulin as an intragastric intervention for 28 days. The abundance and structural changes of bacteria in the intestinal content of mice were analyzed by 16S rDNA high-throughput sequencing. The study found that male BALB/c mice can successfully establish a typical model of small intestinal inflammation by freely drinking a 3% DSS solution for one week. L. rhamnosus 1.0320 combined with inulin can alleviate DSS-induced colitis, reduce the Disease Activity Index (DAI) score of the pathological damage of colon tissue, decrease myeloperoxidase (MPO) activity, increase hemoglobin content, and regulate the expression levels of inflammatory cytokines IL-1β, IL-6, TNF-α and IL-10. The intestinal flora of mice is reduced after enteritis, and its structure gets disordered. The combination of L. rhamnosus 1.0320 and inulin can increase the abundance and diversity of intestinal flora, and increase the content of beneficial bacteria. Prebiotics promote the colonization ability of probiotics. L. rhamnosus 1.0320 combined with inulin can change the intestinal flora to relieve ulcerative colitis, providing a new theoretical basis for the study of UC mechanism.

Lactobacillus rhamnosus RL-H3-005 and Pediococcus acidilactici RP-H3-006 ameliorate Atopic Dermatitis in offspring mice by modulating the gut microbiota

Mingxin Li,Shugang Li,Chunqing Ai,Wenxiu Zhu,Donglin Sui,Yixuan Li,Binbin Hou,Xiaomeng Ren

The increasing incidence of atopic dermatitis (AD) presents a public health issue, and thus there is an urgent need for safe and effective preventive strategies. In this study, a strategy to reduce the risk of AD in offspring by administering probiotics to the mother was explored. Female mice were given Lactobacillus rhamnosus RL-H3-005 (RL) and Pediococcus acidilactici RP-H3-006 (RP) during pregnancy and lactation period, and their inhibition effects on AD symptoms in offspring mice were evaluated. The results showed that supplementation with RL and RP during pregnancy and lactation period altered not only the gut microbiota community but also microbial community in milk in pregnant mice. Further study showed that pups from RL- and RP-treated pregnant mice had mild symptoms of AD, characterized by reduced epidermal thickness in the ear tissues and numbers of mast cells, which was associated with the reduction in secretory immunoglobulin A (sIgA), immunoglobulin E (IgE), IL-4, IL-5, IL-13, thymic stromal lymphopoietin (TSLP), and eosinophil cationic protein (Ecp). The underlying mechanism of RL and RP on the inhibition of AD symptom in offspring was closely associated with the alterations of the gut microbiota community in offspring, including an increase in Ligilactobacillus and Bacteroides . This study implies that RL and RP hold the potential to reduce the risk of AD in offspring via gut-mammary axis during the pregnancy and lactation period.

Purification and characterization of bacteriocin produced by Lactobacillus rhamnosus zrx01

Ruixiang Zhao,Yanqing Lu,Junjian Ran,Gang Li,Shuang Lei,Yang Zhu,Baocheng Xu

The bacteriocin-zrx01 produced by Lactobacillus rhamnosus zrx01 was purified, its basic biochemical characteristics and bacterial antibacterial sensitivity were studied. The bacteriocin-zrx01 was separated and purified using ethyl acetate, AKTA purifier™ 10 protein purifier with HiTrap™ Capto™ Q anion exchange chromatography and Superdex™ 75 10/300 GL gel chromatography. The purity of bacteriocin-zrx01 was analyzed using high performance liquid chromatography . The sensitivity to temperature, pH and enzymes of the purified bacteriocin-zrx01 was measured as the diameter of the antimicrobial ring. The results showed that the purified bacteriocin-zrx01 bacterial antibacterial activity was good, and the specific vitality was raised from the original fermentation broth at 625 to 5330 AU/mg. The bacteriocin-zrx01 had a higher stability at pH 3–9, and the antibacterial activity was the strongest at pH 4. When pH was 9, it still retained 86% of the antibacterial activity of the control group. Bacteriocin-zrx01 was cold-stable at −70 and −20 °C for 30 min. It still retained 57% antimicrobial activity compared with the control after being autoclaved at 121 °C for 30 min. Some enzymes, such as protease K, neutral protease, trypsin, thrombin, alkaline protease and papain , affected its antibacterial activity, but it was sensitive to pepsin , and the diameter of the antimicrobial ring decreased by 50% after 2 h of pepsin treatment. These results showed that the bacteriocin-zrx01 produced by Lactobacillus rhamnosus zrx01 might have a potential as a natural food preservative , although in vivo studies are needed.

Antimicrobial Substances and Mechanisms of Lactobacillus rhamnosus against Gardnerella vaginalis

Huang Yu-Ping,Shi Jie-Yan,Luo Si-Chen,Xu Shao-Yan,Zhang Jia-Dong,Molnár István,Yang Qiong-Qiong,Zhang Bo-Bo

Bacterial vaginosis (BV) is a common vaginal disease associated with abnormal changes in the vaginal microbiome. Our previous study found that Lactobacillus rhamnosus has a good therapeutic effect on bacterial vaginosis by inhibiting the most prominent bacterium associated with BV, Gardnerella vaginalis . In this study, we show that acetic acid and lactic acid are the main substances in the cell-free supernatant (CFS) of L. rhamnosus that inhibit the growth of G. vaginalis. Further study on the mechanism showed that acetic acid and lactic acid alter the morphology of the G. vaginalis cells, eventually causing the cells to shrink or burst, resulting in exudation of their intracellular contents. In addition, these two organic acids also dissipate the membrane potential of bacterial cells, affecting their synthesis of ATP. A reduced activity of the Na + /K + -ATPase leads to abnormal ATP metabolism, and ultimately inhibits the growth and reproduction of G. vaginalis . Our study provides valuable information for the widespread application of L. rhamnosus in the treatment of bacterial vaginosis.

Enhanced Osteoporosis Treatment via Nano Drug Coating Encapsulating Lactobacillus rhamnosus GG

Hafza Behroz Mehvish,Shirong Zhang,Yi Liang,Da Sun,Xu Qiu,Kai Qu,Xian Qin,Jiawei Li,Fei Yan,Chunhui Lang,Lixin Xu,Guixue Wang,Jian Chen,Bo He,Kun Zhang,Wei Wu

Osteoporosis is the most common systemic skeletal disorder, particularly associated with aging and postmenopausal women. With the growing knowledge about the gut–bone axis, the therapeutic strategies for osteoporosis have been shifted toward regulating gut microbiota to promote positive bone metabolism. AlthoughLactobacillus rhamnosus GG (LGG) is widely reported to positively regulate bone metabolism by restoring the dysbiotic microbiome, oral administration is associated with sensitivity to gastric fluid and low bioavailability. Other studies also demonstrated that bisphosphonates ameliorate osteoporosis by directly regulating bone metabolism, especially inhibiting osteoclast activity. However, the side effects caused by bisphosphonate treatment still represent a significant problem. In this study, we assembled alendronate, a clinically used bisphosphonate, with DSPE-phospholipid nanoencapsulation and LGG (ADB), to protect LGG from the acidic environment of the stomach, while simultaneously reducing the gastrointestinal side effects associated with the oral administration of alendronate. We further investigated the potential of these DSPE-phospholipid nanoencapsulated bacteria ADB to repair osteoporotic bone deterioration, and their ability to regulate gut microbiota in vivo, which is strongly associated with the intrinsic environment. Compared with the same dosage of LGG and alendronate alone, ADB positively regulated bone metabolism, and osteoporosis was significantly revised in ovariectomized mice models.

Lactobacillus rhamnosus induces CYP3A and changes the pharmacokinetics of verapamil in rats

Jie Liu,Yi Cheng,Yuanjin Zhang,Shengbo Huang,Zongjun Liu,Xin Wang

Verapamil, a calcium channel blocker, has been approved as the first-line drug for treatment of angina pectoris, hypertension and supraventricular tachycardia. Lactobacillus rhamnosus , one of the normal strains in human intestinal tract, is very popular in the probiotic market for conferring a health benefit on the host. This report investigated the potential of gut microbiota-drug interactions between lactobacillus rhamnosus and verapamil via using wild type (WT) and Cyp3a1/2 knockout (KO) rats. In WT rats, administration of Lactobacillus rhamnosus for 14 days decreased systemic exposure of verapamil and increased its metabolite norverapamil in vivo , and resulted in gut microbiota-drug interactions. In Cyp3a1/2 KO rats, however, this interaction disappeared. Further studies found that Lactobacillus rhamnosus induced CYP3A activity and expression, and changed the composition of gut microbiota, thus changing the pharmacokinetics of verapamil. These results demonstrated the interaction between lactobacillus rhamnosus and verapamil, and indicated that the effect of gut microbiota on metabolic enzymes cannot be ignored.

Preparation of shell-core fiber-encapsulated Lactobacillus rhamnosus 1.0320 using coaxial electrospinning

Cong Xu,Jiage Ma,Zhijing Liu,Wan Wang,Xin Liu,Shanshan Qian,Lijun Chen,Liya Gu,Chuanqiang Sun,Juncai Hou,Zhanmei Jiang

In this study, shell-core fibers were successfully prepared by using Eudragit S100 (ES100) and poly(vinyl alcohol) (PVA)/pectin (PEC) through coaxial electrospinning technology. The electrospun fiber was characterized by attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) and X-ray diffraction (XRD). Thermo-gravimetric analysis (TGA) showed that the coaxial electrospun fiber encapsulated with Lactobacillus rhamnosus 1.0320 ( L. rhamnosus) had higher thermal stability than the electrospun fiber prepared by uniaxial electrospinning. L. rhamnosus encapsulated by coaxial electrospun fiber maintained 90.07% and 91.96% survivability in simulated gastric and intestinal fluids. After continuous simulated gastrointestinal fluid treatment, the survival rate of L. rhamnosus encapsulated by coaxial electrospun fiber was 81.40%. The results indicate that shell-core fiber-encapsulated probiotics can improve the tolerance of probiotics to the harsh environment of gastrointestinal tract . The fiber prepared in this study can be applied to the preparation of functional fermented food such as probiotic yogurt fermentation in the future.

Dual-Targeted Nanoparticles Hitchhiking on Lactobacillus rhamnosus Bacterial Ghosts to Alleviate Nonalcoholic Steatohepatitis

Xiumin Zhang,Ronggang Liu,Yannan Chen,Huihui Wang,Wentao Su,Yukun Song,Mingqian Tan

Oral nutritional interventions for nonalcoholic steatohepatitis (NASH) have garnered significant interest due to their potential benefits. Astaxanthin (AXT) has the potential to enhance liver function and act as an effective antioxidant for NASH intervention, but its application is limited by its stability and bioavailability. This study aims to develop dual-targeted AXT nanoparticles (AXT@TWG) for precise liver-targeted delivery by ″hitchhiking″ on Lactobacillus rhamnosus bacterial ghosts (LBGs) to effectively intervene in NASH. In vitro experiments demonstrated that AXT@TWG nanoparticles significantly reduced LPS-induced reactive oxygen species production and apoptosis while effectively alleviating lipid accumulation. In vivo experiments demonstrated that LBGs significantly enhanced the intestinal accumulation efficiency of AXT@TWG. Pharmacokinetic evaluations revealed that the efficiency of AXT@TWG@LBGs entering the bloodstream was approximately 2.7 times higher than that of AXT@TWG nanoparticles and their accumulation in the liver was about 1.3 times greater. AXT@TWG@LBGs effectively alleviated NASH by reducing triglycerides, free fatty acids, and malondialdehyde levels by 23.07, 65.32, and 21.42%, respectively, compared to the model group, thereby mitigating lipid accumulation and enhancing antioxidant capacity. Additionally, AXT@TWG@LBGs effectively reduced insulin resistance, lowered inflammatory cytokine levels, and corrected disturbances in lipid metabolism. Therefore, this study provides a potentially effective strategy for the treatment of NASH.
