Fabrication and evaluation of Lambda-Cyhalothrin nanosuspoemulsion with pH- and temperature-responsive based on polyethylene wax as carrier

Yanmin Huang,Xiangying Li,Dongmei Mo,Haiyan Yuan,Qiang Hu,Yang Jiang,Chunfang Gan,Yong Chen,Weiguo Li,Rui Lu,Jianguo Cui

Using polyethylene wax (PW) as the coating matrix, the lambda-cyhalothrin-PW nanosuspoemulsion (LC-PW) with a particle size of 80-150nm was prepared through high-speed stirring, hot melt emulsification and ultrasonic dispersion. The formulation and composition of the LC-PW were optimized, the morphology of the LC-PW was analyzed by dynamic light scattering (DLS) and TEM, and the structure of the LC-PW was characterized by UV and IR. The anti-photolysis test showed that LC-PW had a good anti-photolysis performance. Furthermore, LC-PW could sustainably release Lambda-cyhalothrin, which was pH- and temperature dependent. The insecticidal activity analysis in the greenhouse indicated that the toxic strength between LC-PW and LC-SC (lambda-cyhalothrin-suspension concentrate) to Mythimna separata was similar within the same concentration ranges tested, but the insecticidal duration of LC-PW was significantly longer than LC-SC. Thus, the new type of LC-PW with the properties of anti-photolysis and controlled release is suitable for application in the field as a better insecticide.

Overexpression of CpGPCRs contributing to lambda-cyhalothrin resistance in Cydia pomonella

Yu-Xi Liu,Chao Hu,Zi-Nan Xia,Yan Wang,Yu-Ting Li,Ping Gao,Yun-Tong Lv,Jia Li,Xue-Qing Yang

G protein-coupled receptors (GPCRs) constitute a diverse and crucial family of membrane receptors, regulating a wide array of physiological processes. Although the involvement of GPCR signaling pathways in modulating key genes associated with insecticide resistance has been documented in various insect species, the molecular mechanisms underlying GPCR-mediated resistance in Cydia pomonella remain largely unknown. To elucidate the molecular basis of lambda-cyhalothrin (LCT) resistance in C. pomonella , we performed comparative transcriptome sequencing on whole-body, head, and midgut tissues of larvae from a susceptible population (SS) and a laboratory-selected LCT-resistant population (LCR) derived from SS. Five GPCR candidates ( CpGPCRs ) exhibiting overexpression in LCR were identified, including two members of Family A ( CpGPCR4 and CpGPCR55 ), two of Family B ( CpGPCR68 and CpGPCR76 ), and one of Family F ( CpGPCR94 ). Spatiotemporal expression profiling revealed that these genes were predominantly expressed during the first-instar larval and pupal-adult stages, with notably high expression levels in the larval heads. Notably, all genes exhibited significantly elevated expression in the midgut of LCR larvae compared to SS larvae. RNA interference (RNAi)-mediated knockdown of CpGPCR4 , CpGPCR55 , and CpGPCR68 increased larval susceptibility to LCT, while knockdown of CpGPCR4 and CpGPCR55 significantly reduced larval survival rates. Furthermore, larvae treated with dsGPCR55 exhibited molting defects and a decline in 20-hydroxyecdysone (20E) titers. These findings demonstrate that CpGPCRs play essential roles in the survival and development of C. pomonella and are critically involved in mediating resistance to LCT. This study enhances our understanding of the molecular mechanisms driving insecticide resistance and identifies potential targets for the development of novel pest management strategies against C. pomonella .

Preparation and Characterization of a Novel Waterborne Lambda-Cyhalothrin/Alkyd Nanoemulsion

He Qin,Xiaoteng Zhou,Danfei Gu,Lingxiao Li,Chengyou Kan

Inefficient usage and overdosage of conventional pesticide formulations has resulted in large economic losses and environmental pollution due to their poor water solubility and weak adhesion to foliage. In order to develop a green and efficient pesticide formulation, a kind of alkyd resin (AR) based on vegetable oil was first synthesized and used to fabricate the lambda-cyhalothrin/AR (LC/AR) nanoemulsion via in situ phase inverse emulsification, and its properties were then investigated. Results showed that the particle size of the LC/AR nanoemulsion was 50–150 nm with maximum LC loading capacity of as much as 40.9 wt %, high encapsulation efficiency >90%, and great stability in multiple environments. The LC/AR nanoemulsion exhibited better controlled release characteristics compared with LC commercial formulations, and a stronger adhesion on the foliage of the resulted nanoemulsion was also observed, which was attributed to low surface tension and strong interactions with foliar surfaces.

Water-Based Nanoparticulate Formulation of Lambda-Cyhalothrin and Its Enhancement of Bioavailability

Xiaokang Tian,Rongrong Fan,Wenyi Wei,Dan Wang,Yuan Le,Jian-Feng Chen

Low efficacy and frequent use of conventional pesticide formulations have led to various food safety and ecological concerns. Green and effective nanoformulation offers promising solutions to water-insoluble pesticides. Herein, we prepared a water-based nanoparticulate formulation of lambda-cyhalothrin (LC WBNF) using high-gravity rotating packed bed (RPB) combined with lyophilization. The prepared formulation could be dispersed in water with an average particle size of 54.5 ± 0.7 nm and maintained long-term stability. LC WBNF demonstrated significant improvement in foliar wettability, adhesion, and resistance to rain-induced washout. LC WBNF enhanced the uptake and translocation of LC in broad bean plants due to the presence of nanoparticles. Furthermore, the insecticidal activity of LC WBNF was 2.8 and 2.3 times higher than that of commercial LC WP and LC WDG, respectively. This study presents a novel strategy for the development of nanoinsecticides for practical application in agricultural production.

LC50 of lambda-cyhalothrin stimulates reproduction on the moth Mythimna separata (Walker)

Xin-Ru Li,Yue Li,Wei Wang,Ning He,Xiao-Ling Tan,Xue-Qing Yang

Lambda-cyhalothrin has long been recommended as an effective insecticide to control the oriental armyworm , Mythimna separata (Walker), a notorious migratory pest of agricultural plants. Previous researches have suggested that survival, development, and reproduction of insects are influenced by sublethal concentrations of insecticides. However, the effects of sublethal concentrations of lambda-cyhalothrin on M. separata are less known. In this study, we determined the toxicity and effects of LC 20 and LC 50 concentration of lambda-cyhalothrin on development and reproduction of M. separata . Results indicate that LC 20 of lambda-cyhalothrin tends to decrease the life traits of M. separate , with a shortening larvae period of offspring and oviposition period, whereas LC 50 of lambda-cyhalothrin stimulates daily maximal fecundity and forwards the oviposition peak, suggesting a stimulation of reproduction by LC 50 of lambda-cyhalothrin. The M. separata population was increased by an LC 50 concentration of lambda-cyhalothrin, resulting in a net reproductive rate ( R 0 ) and intrinsic rate of increase ( r m ) significantly higher than that of the control. Transcripts of vitellogenin ( MsVg ) and vitellogenin receptor ( MsVgR ) genes were suppressed at day 1 after emergence of moth which developed from the larvae exposed to LC 20 and LC 50 of lambda-cyhalothrin, but were significant induced when the moth begin to lay eggs (day 4), with a more remarkable induction by LC 50 of lambda-cyhalothrin than those of LC 20 of lambda-cyhalothrin. Our results indicate that the observed stimulation of reproduction is therefore the results of up-regulation of MsVg and MsVgR by LC 50 of lambda-cyhalothrin.

CpGSTd3 is a lambda-Cyhalothrin Metabolizing Glutathione S-Transferase from Cydia pomonella (L.)

Wei Wang,Chao Hu,Xin-Ru Li,Xiao-Qi Wang,Xue-Qing Yang

Little is known about the role of specific delta GST genes in the detoxification of lambda-cyhalothrin in the global quarantine fruit pest codling moth, Cydia pomonella (L.). Real-time quantitative PCR shows that CpGSTd3 was ubiquitously expressed at all developmental stages and is most abundant in the larval stage and lowest in the egg stage; the mRNA level of CpGSTd3 is higher in the midgut and Malpighian tubules of fourth-instar larvae and abdomens of adults than in other tissues. Exposure of fourth-instar larvae to an LD10 dosage of lambda-cyhalothrin significantly induced the transcript of CpGSTd3 at 3 h, but the mRNA level was down-regulated after 12 h of treatment. Recombinant CpGSTd3 expressed in Escherichia coli was able to catalyze the conjugation of 1-chloro-2,4-dinitrobenzene (CDNB) and with an IC50 value of 0.65 mM for lambda-cyhalothrin. Metabolism assays indicate that recombinant CpGSTd3 could metabolize lambda-cyhalothrin. These results suggest that CpGSTd3 is probably a lambda-cyhalothrin metabolizing GST in C. pomonella.

A common mechanism of detoxification for lambda-cyhalothrin and abamectin in Cydia pomonella

Ju Di,Hu Chao,Li Peirong,Gao Ping,Li Yuting,Yang Xueqing

Background The primary method utilized by orchard owners to combat Cydia pomonella is the application of various chemical insecticides. However, this has resulted in the development of resistance. The resistance mechanisms to insecticides from different chemical classes are diverse but interconnected. Therefore, it is crucial to comprehend the commonalities in these mechanisms to effectively develop strategies for managing resistance.Materials and methodsTo determine whether target-site insensitivity to LCT and AM plays a role in resistance, the sequences of voltage-gated sodium channel (VGSC) and glutamate-gated chloride channel (GluCl) containing the mutation domains were detected. To validate whether similar mechanisms were involved in the detoxification process of lambda-cyhalothrin (LCT) and abamectin (AM) at sublethal doses (specifically LD10 and LD30), cytochrome P450 monooxygenases (P450), glutathione S-transferases (GST), and carboxylesterases (CarE) activities were evaluated after insecticides exposure; synergistic experiments were conducted using piperonyl butoxide (PBO), diethyl maleate (DEM), and triphenyl phosphate (TPP) as inhibitors of P450, GST, and CarE respectively. RNA-sequencing (RNA-Seq) was performed to compare the expression levels of detoxification-related genes between susceptible (SS) and resistant strains. Results The best known target-site mutations caused by LCT and AM, including L1014F in VGSC and V263I, A309V, I321T, and G326E in GluCl were not occurred. We observed that PBO had a strong synergistic effect on LCT and AM, while DEM on LCT. The activities of detoxification enzymes increased after insecticide exposures, indicating that the detoxification of LCT was primarily carried out by P450 and CarE enzymes, while P450 and GST enzymes played a major role in the detoxification of AM. A total of 72 P450 genes and 75 CarE genes were identified in the C. pomonella transcriptome, with 43 of these genes (including 11 P450, 3 GST, 10 CarE, 11 ABC transporters, and 8 UDP-glycosyl transferases) being over-expressed in response to both insecticides Interestingly, ABC transporters were predominantly induced by AM treatment, while GST showed higher induction levels with LCT treatment. Furthermore, LCT-resistant strains of C. pomonella exhibited higher levels of induction of detoxification-related genes compared to susceptible strains. Conclusion The up-regulation of these detoxification genes is a common metabolic mechanism employed by C. pomonella to counteract the effects of insecticides, although the extent of gene expression change varies depending on the specific insecticide.

Characterization of a lambda-cyhalothrin metabolizing glutathione S-transferase CpGSTd1 from Cydia pomonella (L.)

Liu Jiyuan,Yang Xueqing,Zhang Yalin

In insects, glutathione S -transferases (GSTs) are enzymes involved in detoxification of insecticides. However, few data are available for the codling moth, Cydia pomonella (L.). In this study, we cloned a delta class GST gene CpGSTd1 from C. pomonella . Real-time quantitative PCR shows that CpGSTd1 was up-regulated with aging, and the mRNA level of CpGSTd1 was higher in the fat body and silk glands than in other tissues. The expression level of CpGSTd1 exposure to insecticide suggests that CpGSTd1 is up-regulated after chlorpyrifos-methyl and lambda-cyhalothrin treatments. Both lambda-cyhalothrin and chlorpyrifos-methyl altered GST activity in vivo. The purified CpGSTd1 protein exhibits a high catalytic efficiency with CDNB and was inhibited by lambda-cyhalothrin and chlorpyrifos-methyl in vitro. Metabolism assays indicate that lambda-cyhalothrin was significantly metabolized while chlorpyrifos-methyl was not metabolized by CpGSTd1. Binding free energy analysis suggests that CpGSTd1 binding is tighter with lambda-cyhalothrin than with chlorpyrifos-methyl. Our study suggests that CpGSTd1 plays a key role in the metabolism of insecticides in C. pomonella .

Visual lambda-cyhalothrin detection using yellow carbon dots molecularly imprinted polyvinylidene fluoride fluorescent membrane

Yahui He,Yajie Liu,Xinyu Liu,Yao Zhu,Yaling Yang,Jing Wang,Baoguo Sun

Due to its common use as an insecticide during agricultural production, residual lambda-cyhalothrin (LC) in food and the environment presents a significant human health risk. Developing an accurate LC detection method that does not require pretreatment remains a challenge. This study developed a novel yellow carbon dots molecularly imprinted (YCDs@MIPs) polyvinylidene fluoride (PVDF) composite (YCDs@MIPs@PVDF) fluorescent membrane for rapid visual LC detection. This method required no pretreatments during analysis since the LC could be specifically adsorbed when the samples passed through the YCDs@MIPs@PVDF membrane, causing fluorescence quenching, while matrix-interfering components could be removed for the porous structure of the membrane. A smartphone was used for rapid visual LC detection based on membrane fluorescence color changes in a dark-box ultraviolet analyzer. In food with a 15 % PVDF content and 6 mL added YCDs@MIPs, LC was quantitatively detected in a broad concentration range (5–500 μg/L) with a limit of detection (LOD) of 0.039 μg/L to 0.16 μg/L, which was lower than the strictest EU2019/92 limit of 10 μg/L. The analysis time was reduced from over 8 h when using the GB 23200.121–2021 and GB/T20769–2008 national standard methods to 30 min, while the analytical procedure decreased from six steps to two. The visual detection strategy combining the YCDs@MIPs@PVDF composite fluorescent membrane with a smartphone provided a new rapid detection technique for composite membrane purification and fluorescence signal output function.

Quick and convenient construction of lambda-cyhalothrin antigen for the generation of specific antibody

Nan Cui,Limin Cao,Jianxin Sui,Hong Lin,Xiangning Han,Xiangfeng Chen,Hanyi Xie,Xun Sun

Lambda-cyhalothrin is a pyrethroid widely used in crop, fruit and vegetable production, but has potential health threats to human. Immunoassay is a cheap, rapid and facile method to detect lambda-cyhalothrin, yet wide application of this method still requires improvement in the construction of antigen. In this study, we developed a one-step lambda-cyhalothrin hapten synthesis that transformed the cyanide group in lambda-cyhalothrin to amide. Complete antigen was assembled by coupling the amide with succinic-anhydride-activated carrier proteins, and corresponding polyclonal antibodies were generated using Balb/c mice. Using antibody generated by the method in this paper, the competitive ELISA demonstrated the lowest detection limit of 3.772 μg/L for lambda-cyhalothrin, and no significant cross-reactivity for other pyrethroid pesticides was observed. All the results suggested we have established a more efficient technique of generating lambda-cyhalothrin antibody. Furthermore, since the activated proteins used in this study are highly controllable, we believe these proteins could potentially be the prototype of a series of standardized carrier proteins for the synthesis of complete antigens.

Glutathione S-Transferase Genes are Involved in Lambda-Cyhalothrin Resistance in Cydia pomonella via Sequestration

Chao Hu,Ji-Yuan Liu,Wei Wang,David Mota-Sanchez,Shun He,Yu Shi,Xue-Qing Yang

Pest management is mostly accomplished by the use of insecticides. However, the overuse of insecticides has led to the development of resistance. Glutathione S-transferases (GSTs) are vital detoxification enzymes involved in insecticide resistance in insects. In this study, we report the involvement of GSTs in insecticide resistance to lambda-cyhalothrin in Cydia pomonella, a globally quarantined fruit pest. A total of 25 GST, including 22 cytosolic genes and 3 microsomal genes, are identified from the genome database of C. pomonella. These cytosolic genes are further classified into six classes, including four in delta, eight in epsilon, three in omega, three in sigma, one in theta, and one in zeta class, as well as two unclassified genes. The real-time quantitative polymerase chain reaction (RT-qPCR) shows that the majority of these genes are mainly expressed throughout the larval stage and in the midgut of the fourth-instar larvae. Exposure to an LD10 dose of lambda-cyhalothrin resulted in the upregulation of 17 GST genes. Moreover, mRNA levels of most GST genes, with the exception of CpGSTe6, CpGSTd2, CpGSTd4, and CpGSTz1, are considerably higher in a lambda-cyhalothrin-resistant population (ZW_R) than those of susceptible strains. Recombinant CpGSTd1, CpGSTd3, CpGSTe3, and CpGSTs2 can bind and metabolize lambda-cyhalothrin, with the highest metabolic rate observed for CpGSTd3 but no metabolite(s) was detected, supporting the role of GSTs in sequestration of lambda-cyhalothrin. Molecular dynamics simulation analysis indicates that key residues of hydrophobic pocket-derived lipophilic energy S(lipo) interactions with a hydrophobic pharmacophore of lambda-cyhalothrin are crucial for metabolism by CpGSTd3 and further lead to resistance. Our study is the first to experimentally confirm the involvement of GSTs in lambda-cyhalothrin resistance via sequestration and provides new insights into resistance management in C. pomonella.

Transgenerational Effects of Elevated Temperature on Life History and Lambda-Cyhalothrin Sensitivity in Cydia Pomonella

Zheng Kang-Wu,Wei Zi-Han,Xia Zi-Nan,Wang Bo-Kun,Gao Ping,Li Yu-Ting,Yang Xue-Qing

Rising ambient temperatures driven by global warming are subjecting insect populations to thermal conditions exceeding their historical environmental ranges. Understanding how temperature alters key life-history traits is essential for optimizing integrated pest management strategies, particularly for invasive species. This study investigated the effects of short-term thermal stress (31 °C for the parental generation, with offspring returned to 26℃) on life history traits and insecticide sensitivity in the codling moth Cydia pomonella (L.), a major agricultural invasive pest. Results demonstrated that elevated temperature significantly shortened larval developmental duration and reduced pupal weight and overall fitness, with these phenotypic effects persisting across generations. Additionally, increased temperature attenuated the sensitivity of C. pomonella to lambda -cyhalothrin, especially in parental generation. In contrast, reproductive output and sensitivity to chlorantraniliprole remained largely unaffected by thermal elevation. Activities of detoxification enzymes including cytochrome P450 monooxygenases (P450s), carboxylesterases (CarEs), and the antioxidant enzyme catalase (CAT), were significantly upregulated at elevated temperatures, with transgenerational inheritance observed in progeny, potentially contributing to the observed decrease in insecticide sensitivity. These findings advance our understanding of the transgenerational physiological and biochemical adaptions of C. pomonella to thermal stress, with implications for resistance management under climate change scenarios.
