Several key virulence factors of Pseudomonas aeruginosa are regulated by quorum-sensing systems, small noncoding RNAs (sRNAs), and environmental stress, leading to a high mortality rate. Our previous studies indicated that the alkaline phosphatase LapA regulated P. aeruginosa PAO1 biofilm formation in a chronic wound model established with ex vivo porcine skin explants. Notably, one particular sRNA located upstream of the lapA gene was highly expressed in the model. Therefore, the sRNA was further characterized via northern blotting and rapid amplification of cDNA ends. The results revealed that the sRNA we named LapS is 197 nucleotides in length and is derived from the 5´ UTR and sequence internal to the lapA gene. Next, LapS mutation, overexpression, and complementation strains were constructed from the PAO1 strain, and phenotypic experiments associated with lapA were performed and compared with those of the ΔlapA and wild-type strains. The results indicated that LapS is involved in regulating swarming motility, rhamnolipid and alkaline phosphatase production, las/rhl quorum-sensing systems, and biofilm formation by controlling the level of lapA mRNA under phosphate-depleted conditions. Therefore, this LapS-lapA signaling cascade is beneficial for balancing the virulence regulation of P. aeruginosa. Additionally, an in vitro study indicated that LapS directly and post-transcriptionally regulated at least one unlinked gene, putA, which encodes bifunctional proline dehydrogenase, a virulence factor of P. aeruginosa. Collectively, our findings reveal a previously unstudied regulatory sRNA and advance the understanding of the roles of sRNAs in the pathogenicity of P. aeruginosa.

In this study, a label-free glucose (Glu) biosensor was designed by modifying reduced graphene oxide-carboxymethyl chitosan-platinum nanoparticles (RGO-CMCS-Pt NPs) on a silicon-based light-addressable potentiometric sensor (LAPS) based on the field effect tube principle. Under the bias voltage , a modulated light source is directed onto the semiconductor layer of the sensor, generating a weak photocurrent . Simultaneously, Glu is catalyzed by glucose oxidase (GOD) to produce gluconic acid and H 2 O 2 . The H 2 O 2 further decomposes to facilitate electron transfer, disrupting the potential balance on the biosensor surface and causing changes in the photocurrent-bias voltage (I-V) curve. The voltage offset demonstrated a linear relationship with the Glu concentration ranging from 0.01 mg/mL to 4.0 mg/mL. The linear equation was △V= 19.17331 C+ 75.46574 (R 2 =0.98831) with a high sensitivity of 19.17 mV*mL/mg and a low detection limit (LOD) of 0.01 mg/mL (S/N = 3). Furthermore, the Glu biosensor exhibited good specificity, reproducibility, and stability, the biosensor also realized the detection of glucose in human serum samples with a relative standard deviation ranging from 0.63% to 3.47%, and the results were consistent with the hospital's biochemical indicators, which demonstrated the potential application of the prepared biosensor in the clinical detection of Glu.

Glucose (Glu) is an important source of energy and metabolic intermediate for animal and plant cells. In this study, a high-performance biosensor for specific detection of Glu was constructed by combining a light-addressable potentiometric biosensor (LAPS) and a specific sensitive unit. The specific sensitive unit was adopted a silicon-based capacitive structure, and the glucose oxidase/reduced graphene oxide -chitosan-ferrooxidase/gold nanoparticles (GOX/RGO-CS-Fc/AuNPs) was modified onto the silicon-based working electrode to construct an nanosensing membrane. The interaction between the specific sensitive unit and Glu resulted in the formation of a bilayer potential, and the field effect generated by the bilayer potential caused a change in the capacitance of the silicon structure, which resulted in a change in the photocurrent-voltage characteristics of the LAPS. Through the potential shift in the photocurrent-voltage characteristics, the LAPS was capable of detection the concentration of Glu. Under optimal conditions, the potential shift was linearly correlated with the Glu concentration from 0.01 mg/mL to 4.00 mg/mL with R 2 of 0.9821, and the limited detection was 0.001 mg/mL. In addition, the LAPS showed good recovery (97.92 %–105.72 %), admirable specificity and stability in detection Glu in human serum samples.

In this paper, a novel silicon-based light-addressable potentiometric sensor (LAPS) has been designed for the detection of 1,5-anhydroglucitol (1,5-AG) in human serum. Reduced graphene oxide-chitosan-ferrocene (RGO-CS-Fc)/AuNPs nanohybrids and pyranose oxidase (PROD) enzyme is used to fabricate biological sensitive membrane unit by layer-by-layer assembly technology. When a bias voltage is provided to the LAPS system, the catalytic oxidation reaction between 1,5-AG and PROD to produce H 2 O 2 . The by-product H 2 O 2 can oxidize Fc(Fe 2+ ) ions in RGO-CS-Fc nanohybrids into Fc(Fe 3+ ) ions, which cause the potential of the sensitive membrane surface to change and the potential shift of I-V curve will generate a corresponding offset response. Under the optimal conditions, the potential shift of the LAPS is linearly related to the concentration of 1,5-AG at 10 µg·mL −1 –350 µg·mL −1 with the correlation coefficient of 0.97414. The sensitivity is 0.44273 mV/µg·mL −1 and the lowest detection limit is 10 µg·mL −1 . In addition, the biosensor showed good specificity, acceptable stability and satisfactory recovery rates (91.28%-107.66%), which would be a potential testing methods in actual clinical samples.

1,5-anhydroglucitol (1,5-AG) is an important indicator in the early detection of diabetes. In this study, a high-performance light addressable potentiometric sensor (LAPS) for the detection of 1,5-AG based on reduced graphene oxide-polyacrylamide-ferrocene/gold nanoparticles (rGO-PAM-Fc/AuNPs) as sensing membrane and pyranose oxidase (PROD) as the specific recognition substance was constructed. Among them, rGO-PAM-Fc/AuNPs modification LAPS chip can not only effectively increase the specific surface area and adsorb more PROD , but also enhance the capacitive effect of LAPS and improve the detection sensitivity. 1,5-AG is hydrolyzed by PROD and produces hydrogen peroxide (H 2 O 2 ) that can oxidize divalent ferrocene ions in the rGO-PAM-Fc composite to trivalent ferrocene ions, therefore leads to a change in the membrane potential on the surface of the LAPS chip and causes a shift in the I-V curve, which reflect the change of 1,5-AG concentration. The designed LAPS had a good linearity with 1,5-AG concentration in the range of 100.0–1000.0 μg/mL with a correlation coefficient of 0.99068. The sensitivity was calculated to be 0.1908 mV/μg/mL and a limit of detection (LOD) of 21.74 μg/mL was obtained using three signal-to-noise ratio (S/N = 3). Also, the 1,5-AG biosensor has good specificity, reproducibility and stability, and can be used in human serum samples with the recoveries of 93.62 %–114.52 %. This work provides a highly sensitive, portable and low-cost assay for the detection of 1,5-AG. Atherosclerotic Cardiovascular Disease (ASCVD) is one of the major fatal diseases for humans and the fatality rate is increasing rapidly. Low density lipoprotein (LDL) is a serum key marker in the diagnosis of ASCVD because LDL level is associated with the prevalence of ASCVD. Herein, a light addressable potentiometric sensor (LAPS) was constructed for rapid detection of LDL using reduced graphene oxide-polyaniline-hemin nanocomposites (RGO-PANI-HNCs) and low density lipoprotein aptamer (LDLapt) as sensitive membrane of LAPS. Under the effect of bias voltage, the specific binding between LDL and LDLapt, and forming the LDL-LDLapt complex will cause the voltage change on the sensitive membrane surface of the LAPS system, which will cause the change of original current–voltage (I-V) balance in the circuit, resulting in the shift of I-V curve. The correlation between the potential shift of LAPS and LDL concentration was linear from 1.0 to 100.0 μg/mL under optimal conditions. The sensitivity is 4.7671 mV/μg/mL, and the detection limit is 0.79 μg/mL. Also, the LDL biosensor showed satisfactory specificity, selectivity, reproducibility, and showed a good precision in real human serum samples, which provides a simple and effective way for the early diagnosis of ASCVD.
