Investigations have shown that storage bugs seriously harm grains during storage. In the interim, essential oils (EOs) have been proven to be a good botanical pesticide. The anti-Lasioderma serricorneproperties ofElsholtzia ciliataessential oil, which was obtained by steam distillation, were evaluated usingDL-limonene, carvone, and their two optical isomer components using contact, repelling, and fumigation techniques. Simultaneously, the fumigation, contact, and repellent activities of carvone and its two optical isomers mixed withDL-limonene againstL. serruricornewere evaluated. The results showed thatE. ciliata, its main components (R-carvone,DL-limonene), andS-carvone exhibited both fumigations (LC50= 14.47, 4.42, 20.9 and 3.78 mg/L) and contact (LD50= 7.31, 4.03, 28.62 and 5.63 µg/adult) activity againstL.serricorne.A binary mixture (1:1) ofR-carvone andDL-limonene displayed an obvious synergistic effect. A binary mixture (1:1) of carvone and its two optical isomers exhibited an obvious synergistic effect, too. Furthermore, the repellent activity of the EO, carvone, and its two optical isomers,DL-limonene, and a combination of them varied. To stop insect damage during storage,E. ciliataand its components can be utilized as bio-insecticides.

In this study, four kinds of chemical substances (2,3,5,6-tetramethylpyrazine, β-ionone, citronellal, and paeonol), three kinds of plant essential oils (tea tree essential oil, lavender essential oil, and myrrh essential oil), and their combinations were selected to explore their synergistic effects on tobacco beetle [Lasioderma serricorne (Fabricius) (Coleoptera: Anobiidae)] adults by the behavioral test and laboratory simulation test. Behavioral test results showed that some of the combinations revealed a synergistic effect on tobacco beetle adults, especially the sexual attractant +2,3,5,6-tetramethylpyrazine + β-ionone + citronellal + paeonol (SABCD, one portion of sexual attractant, and 1 mg/L synergistic substances) combination and the food attractant +2,3,5,6-tetramethylpyrazine + paeonol (FAD, 1 ml of food attractant and 1 mg/L synergistic substances) combination showed the best behavioral effect on tobacco beetle adults with average dwell times of 120.97 and 126.74 s, respectively, compared to those of other combinations. Meanwhile, SABCD had the highest selection rate [89.47%, about 1.5 times that of the sexual attractant (S)] on tobacco beetle adults compared with those of other combinations. In addition, laboratory simulation test results showed that the SABCD combination had the highest average selection rate (37.31%, about 2 times that of S) on tobacco beetle adults at 1 mg/L. However, our results showed that there was no significant difference in the indoor simulation results of food attractant synergistic substances. Our results will provide guidance for the development of new pesticides for tobacco beetle adults.

Laccase-2 (Lac2) is a key phenoloxidase that catalyzes cuticular sclerotization and pigmentation, which are processes essential for insect molting and metamorphosis. Although the alternatively spliced isoform Lac2A is known to function in cuticular tanning, the physiological role of Lac2B has remained largely unresolved. Here, we identified and functionally characterized two Lac2 isoforms, LsLac2A and LsLac2B , from the cigarette beetle Lasioderma serricorne , a globally important stored-product pest. Both isoforms are highly expressed during the pupal and eclosion stages, with transcripts localized mainly in the cuticle and wing tissues, and are transcriptionally induced by 20-hydroxyecdysone (20E), linking their activity to endocrine regulation of development. Functional analyses revealed that LsLac2A and LsLac2B have distinct and complementary roles in molting and metamorphosis. During larval molting, LsLac2A promotes the synthesis of new cuticle by regulating genes involved in cuticular protein and chitin biosynthesis, whereas LsLac2B facilitates degradation of the old cuticle through modulation of chitinolytic and cuticular protein-degrading genes. In the pupal stage, both isoforms influence elytral pigmentation, sclerotization, and wing morphogenesis via the coordinated regulation of wing development-related and yellow family genes. These results reveal isoform-specific functions of LsLac2 in cuticle renewal and morphogenesis, uncovering a novel mechanism of hormonal and genetic integration during insect development. This work provides new insight into the molecular specialization of Lac2 and identifies promising RNAi-based targets for the sustainable control of L. serricorne .

The cigarette beetle, Lasioderma serricorne (F.), presents a significant management challenge as a stored product pest. The beetle's increasing resistance to conventional fumigants necessitates the search for alternative solutions to mitigate losses in agro-based industries. Here, we constructed a small molecule compounds library containing twenty-eight methyl benzoate-related compounds (MBCs) and evaluated their repellent and toxic effects against cigarette beetles by Y-tube olfactometer behavioral assays, free-choice box assays and fumigation assays. We found that six MBCs (methyl 3-methoxybenzoate, methyl benzoate, methyl 3-formylbenzoate, methyl 2-aminopyridine-3-carboxylate, methyl 2-aminobenzoate and methyl 4-amino-2-methoxybenzoate) act as repellents for cigarette beetles. Notably, methyl benzoate (MeBA) was effective across a broader range of concentrations (200–4000 ppm) compared to other MBCs. Four compounds (methyl 3-formylbenzoate, methyl 2-aminopyridine-3-carboxylate, methyl 2-aminobenzoate and methyl 4-amino-2-methoxybenzoate) required higher minimum concentrations (500–4000 ppm) than MeBA for repellence, while methyl 3-methoxybenzoate was effective at a concentration (10 ppm) lower than that for MeBA. Mixtures of MeBA with methyl 3-methoxybenzoate and methyl 3-formylbenzoate enhanced the repellent effect against cigarette beetles when compared to MeBA alone. Additionally, exposure to varying concentrations of MeBA, methyl 3-methoxybenzoate, methyl 3-formylbenzoate, and methyl 2-aminobenzoate resulted in 100% mortality of cigarette beetles. Our findings indicate that specific MBCs offer the potential for their use as repellents and fumigants for this problematic stored product pest.

In insects, glutathione S -transferases (GSTs) play a pivotal role in the detoxification of a wide range of pesticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest insect of stored products. Recently, pyrethroid insecticides have been used to control this pest. However, little is known concerning the responses and functions of GSTs in L. serricorne under pyrethroid exposure. In this study, transcriptome sequencing was performed on L. serricorne , and a total of 14 GSTs were identified by retrieving the unigene dataset. Of these, 13 predicted GSTs fell into six cytosolic classes, namely, delta, epsilon, omega, sigma, theta, and zeta, and one was assigned to an “unclassified” group. The GST genes were differentially expressed in various larval tissues and at different developmental stages. Exposure to the pyrethroid insecticide lambda-cyhalothrin (LCT) caused oxidative stress in L. serricorne larvae and led to significantly elevated expression levels of six genes, among which LsGSTe1 was the most upregulated. Recombinant LsGSTE1 protein displayed LCT-metabolizing activity. Furthermore, LsGSTE1 protects cells against oxidative stress. Moreover, knockdown of LsGSTe1 by RNA interference dramatically increased the susceptibility of L. serricorne larvae to LCT treatment. The results from this study provide sequence resources and expression data for GST genes in L. serricorne . Our findings indicate that LsGSTE1 plays a dual role in LCT detoxification by metabolizing the pesticide and by preventing LCT-induced oxidative stress. Thus, the LsGSTe1 gene could be used as a potential target for sustainable management of the cigarette beetle.

This study investigated the biological activity of essential oils (EOs) and their main components (caryophyllene and cuminol) against Tribolium castaneum larvae and Lasioderma serricorne adults. EOs were extracted from both flowers and leaves of Elsholtzia stauntoni. The results indicated that the EOs derived from both flowers and leaves, as well as their main components (caryophyllene and cuminol), exhibited contact activities with LD50 values of 35.11, 43.66, 15.44, and 26.05 μg/larva, respectively, against T. castaneum larvae. Moreover, they demonstrated obvious contact toxicities with LD50 values of 14.15, 11.18, 17.81, and 13.91 μg/adult, respectively, against L. serricorne adults. Additionally, EOs from both flowers and leaves exhibited fumigant activities against T. castaneum larvae (LC50 = 24.42 and 62.54 mg/L) and L. serricorne adults (LC50 = 15.26 and 24.29 mg/L). Furthermore, the EOs of flowers and leaves and their main components (caryophyllene and cuminol) remained significantly repellency to T. castaneum larvae and L. serricorne adults within the testing concentration range of 78.63-15.83 nL/cm2 after 12 h of exposure. These findings suggested that the EO from E. stauntoni and its two major components could potentially be developed as a natural insecticide for controlling T. castaneum larvae and L. serricorne adults.

In total, 13 components were isolated from the aerial parts of R. thymifolium by chromatographic separation techniques. Based on the results of MS, 1 H NMR, and 13 C NMR spectrometry, their structures were determined as ent-kaurane-16β-ol ( 1 ), pentacosane ( 2 ), undecanoic acid, ethyl ester ( 3 ), 1-Tridecanethiol ( 4 ), 1-Dodecanol ( 5 ), 4-Methyl-7-hydroxycoumarin ( 6 ), tertradecan-1-ol ( 7 ), apocynin ( 8 ), 3-Methoxy-4-methylbenzaldehyde ( 9 ), scopoletin ( 10 ), hyperin ( 11 ), quercetin ( 12 ), acetophenone ( 13 ). In addition, ent-kaurane-16β-ol ( 1 ) was isolated from R. thymifolium for the first time. Then, the insecticidal toxicities against T. castaneum , L. serricorne , and D. destructor . of compounds 1–13 were evaluated, and the results showed that apocynin ( 8 ) and acetophenone ( 13 ) exhibited obvious contact activity against the storage pests with LD 50 values of 6.46 and 8.72 μg/adult respectively. Acetophenone ( 13 ) showed excellent contact activity against D. destructor with LC 50 values of 0.08 mg/mL. The study clearly showed the active fractions from R. thymifolium have might potential to be developed into potential compounds on storage insect prevention.

In insects, cytochrome P450 monooxygenases (CYPs) play a pivotal role in the detoxification of various insecticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest of stored products, for which pyrethroid insecticides are increasingly used. Nevertheless, the responses and functions of CYPs in L. serricorne under pyrethroid exposure remain poorly understood. In this study, three full-length CYP cDNAs were identified from L. serricorne and designated as CYP6BK29 , CYP6SY1 , and CYP6TE1 . Phylogenetic analysis revealed that these CYPs belong to the CYP3 clan and the CYP6 family. Reverse transcription–quantitative PCR showed that CYP6BK29 and CYP6SY1 were predominantly expressed in the larval midgut, whereas CYP6TE1 transcripts were enriched in the fat body. The expression of CYP6BK29 peaked in the third- and fourth-instar larvae, while CYP6SY1 was specifically highest in the fourth-instar. In contrast, CYP6TE1 showed the highest mRNA levels in adults. Exposure to a sublethal dose (LD 30 ) of lambda-cyhalothrin (LCT) significantly upregulated the mRNA levels of CYP6BK29 and CYP6TE1 , but not that of CYP6SY1 . RNAi-mediated knockdown of CYP6BK29 significantly increased the susceptibility of larvae to LCT. In contrast, silencing CYP6SY1 or CYP6TE1 had no significant effect on larval survival under LCT exposure. These results provide evidence that CYP6BK29 contributes to LCT detoxification in L. serricorne , highlighting its potential as a molecular target for novel pest management strategies. Summary Characterization of three novel CYP genes ( CYP6BK29 , CYP6SY1 , and CYP6TE1 ) in Lasioderma serricorne . Successful transcriptional knockdown of all three genes via RNAi. CYP6BK29 knockdown enhances larval susceptibility to lambda-cyhalothrin, implicating its role in detoxification.
